Verfahren zur Herstellung von 17-20,20-21-Bisalkylidendioxy-Steroiden der Pregnanreihe Bekanntlich sind die grosse Mehrzahl der physio logisch aktiven Steroide Glieder der Pregnanreihe mit einer freien oder veresterten Dioxyaceton-Grup- pierung in 17-Stellung. Diese Seitenkette kann eine Oxydation an C-21, eine Reduktion an C-20, eine Umlagerung unter Bildung eines D-Homosteroids und unter stark sauren oder basischen Bedingungen eine Zersetzung erfahren.
Da Reaktionen und Reak tionsbedingungen, welche diese Veränderungen her vorrufen können, oft notwendig sind, damit der Kern des Steroidmoleküls modifiziert oder mit funktio nellen Gruppen substituiert werden kann, kommt der Möglichkeit einer Entaktivierung oder Blockierung der Dioxyaceton-Seitenkette für die praktische Synthese von physiologisch aktiven Steroiden grosse Bedeutung zu.
Gewisse Erfolge wurden erzielt durch Bildung der C-20-Äthylendioxy-Steroide, doch sind diese Blockierungsgruppen nicht vollständig befrie digend, da sie einerseits ziemlich säurelabil sind und anderseits nur einen Teil der Dioxyaceton-Seitenkette entaktivieren, nämlich die Keton-Funktion.
Die vorliegende Erfindung bezweckt nun die Schaffung einer Reihe von neuen Steroiden der Pregnanreihe, in welchen die Dioxyaceton-Gruppie- rung durch eine sehr stabile Gruppe entaktiviert ist, wobei alle drei C-Atome der Dioxyaceton-Gruppe geschützt sind.
Dieser Schutz soll mit Hilfe einer Gruppe erfolgen, welche sich in passender Weise entfernen lässt zur Rückbildung der Dioxyaceton- Gruppierung, nämlich durch eine Bismethylendioxy- Gruppe oder eine substituierte Bismethylendioxy- Gruppe geschützt ist. Gegenstand der Erfindung bildet ein Verfahren zur Herstellung dieser neuen Art von Steroiden aus einem Steroid, welches eine Dioxyaceton-Gruppierung aufweist.
Es wurde gefunden, dass beim Behandeln eines Steroids der Pregnanreihe mit einer Dioxyaceton- Gruppierung in 17-Stellung mit Formaldehyd oder einem niedrigen Alkylaldehyd in Gegenwart einer starken Säure ein durch Methylendioxy-Gruppen oder substituierte Methylendioxy-Gruppen disubstituiertes Steroid entsteht,
in welchem alle drei C-Atome des Dioxyaceton-Teils zu Gliedern von mindestens einem der hierbei gebildeten 5gliedrigen Ringe werden.
Als Aldehyde verwendet man erfindungsgemäss Formaldehyd oder geradkettige aliphatische Aldehyde von der Formel RCHO, in welcher R ein niedriges Alkylradikal bedeutet.
Das niedrige Alkylradikal kann bis zu 6 C-Atomen aufweisen; Beispiele für derartige niedrige Alkylaldehyde sind Acetaldehyd, Propionaldehyd, n-Butyraldehyd, n-Valeraldehyd, n-Capronaldehyd und n-Heptaldehyd. Formaldehyd wird als Reagens bevorzugt, da man das entstehende 17-20;20-21 -Bismethylendioxy-Steroid in guter Ausbeute in kristalliner Form erhält.
Das erfindungsgemässe Verfahren lässt sich wie folgt darstellen:

Wenn R ein Wasserstoff ist, so erhält man als Produkt ein 17-20,20-21-Bismethylendioxy-Steroid, und wenn R ein niedriges Alkylradikal bedeutet, er hält man ein Methylen-substituiertes 17-20,20-21- Methylendioxy-Steroid. Hierbei handelt es sich um neue Substanzen. Obschon der Dioxyaceton-Teil, um in der dargestellten Weise reagieren zu können, un- substituiert sein muss, kann man, wenn dies zweck mässig ist, in das Reaktionsgemisch auch ein Steroid einführen, welches in einer oder beiden Oxygruppen verestert ist.
Das erfindungsgemässe Verfahren zur Herstel lung der 17-20,20-21-Bisalkylidendioxy-Steroide der Pregnanreihe mit der Teilformel

worin n eine ganze Zahl mit einem Wert von 0-6 inklusive bedeutet, ist dadurch gekennzeichnet, dass man ein 17a,21-Dioxy-20-keto-Steroid der Pregnan- reihe in Gegenwart einer starken Säure mit einem Aldehyd der Formel C"Hz"-1CH0 behandelt.
Um eine optimale Ausbeute an Bisalkylendioxy- Verbindung zu erhalten, verwendet man das Aldehyd im überschuss. Vorzugsweise verwendet man Form aldehyd (oder eine Formaldehyd liefernde Substanz) zur Bildung des 17-20,20-21-Bismethylendioxy- Steroids. Acetaldehyd, Propionaldehyd, Butyraldehyd und Valeraldehyd sind Beispiele für weitere ver wendbare Aldehyde, welche mit einem 17a,21-Dioxy- 20-keto-Steroid reagieren, wobei man die entspre chenden 17-20,
20-21-Bisäthylidendioxy-, 17-20,20-21-Bispropionaldioxy-, 17-20,20-21-Bisbutyraldioxyd- bzw. 17-20,20-21-Bisvaleraldioxy-Steroide der Pregnenreihe erhält.
Zur Durchführung des erfindungsgemässen Ver fahrens kann man reines Formaldehyd, 400/aige wässerige Formaldehydlösung (Formalin) oder eine andere Substanz verwenden, welche im Reaktions gemisch als Formaldehydquelle wirkt. Beispiele für derartige Substanzen, welche im Reaktionsgemisch als Quelle für Formaldehyd dienen, sind Paraform- aldehyd, Formal und Trioxan.
Die Reaktion wird in Gegenwart einer starken Säure, z. B. Salzsäure, Perchlorsäure, Bromwasser stoffsäure, Schwefelsäure, Phosphorsäure und der gleichen durchgeführt. Als starke Säuren kommen z. B. solche mit einem -log k von weniger als 2,25 in Frage. Lewis-Säuren und Ameisensäure lassen sich ebenfalls verwenden.
Die Reaktion des 17a,21-Dioxy-20-keto-Steroids mit Formaldehyd oder andern Aldehyden erfolgt zweckmässig in einem unter den Reaktionsbedingun gen inerten organischen Lösungsmittel. Es kann sich dabei um ein mit Wasser mischbares Lösungs mittel, wie Dioxan oder Tetrahydrofuran, handeln, in welchem Falle die Reaktion in einem einphasigen Lösungssystem erfolgt. Vorzugsweise führt man je doch die Reaktion in einem zweiphasigen Reaktions gemisch durch, wobei das Steroid in einem mit Wasser nicht mischbaren organischen Lösungsmittel gelöst ist und die zweite Phase von der wässerigen Säure-Formaldehyd-Phase gebildet wird.
Das zwei- phasige Reaktionsgemisch liefert sehr befriedigende Ergebnisse und wird zur Erzielung optimaler Aus beuten unter den angegebenen Bedingungen bevor zugt. Es ist günstig, wenn sowohl das Ausgangsmate rial wie auch das Reaktionsprodukt im organischen Lösungsmittel löslich sind, doch erhält man das 17-20,20-21-Bismethylendioxy-Steroid auch in guter Ausbeute und Qualität, wenn das Endprodukt im organischen Lösungsmittel löslich und das als Aus gangsmaterial verwendete 17a,21-Dioxy-20-keto- Steroid unlöslich ist.
Mit Wasser nicht mischbare organische Lösungsmittel, welche sich für die Durch führung des Verfahrens eignen, sind Chloroform, Me- thylenchlorid, Äthylendichlorid, Tetrachlorkohlen- stoff, Äthylendibromid, Methylenbromid, Benzol, Äthyläther und dergleichen, wobei die chlorierten Lösungsmittel am zweckmässigsten sind.
Die erfindungsgemässe Umsetzung erfolgt ge wöhnlich bei Temperaturen von etwa -30 bis etwa i-50 C. Am einfachsten arbeitet man bei Tempera- turen von 0 bis etwa 35 C, bei welchen mit den meisten Steroiden befriedigende Ausbeuten erzielt werden.
Die zur Erzielung optimaler Ausbeuten an 17-20, 20-21-Bisalkyliden-dioxy-Steroiden erforderliche Re aktionszeit hängt in gewissem Ausmass von der an gewendeten Reaktionstemperatur ab. Es hat sich auch gezeigt, dass die für gute Ausbeuten erforder liche Reaktionsdauer eine Funktion der Art des als Ausgangsmaterial verwendeten 17a,21-Dioxy-20-keto- Steroids ist.
Beispielsweise benötigt man in einem Chloroform-Formalin-Salzsäure-Reaktionssystem für die optimale Umwandlung von Cortison in 17-20, 20-21-Bismethylendioxy - 4 - pregnen-3,11-dion etwa 40-50 Stunden, während für die Umwandlung von Prednisolon in 17-20,20-21-Bismethylendioxy-1,4- pregnadien-11/3-ol-3-on in hoher Ausbeute nur 15 bis 30 Minuten nötig sind.
Diese Unterschiede in der Reaktionsfähigkeit der Steroide stellen jedoch der raschen Bestimmung der richtigen Reaktionsdauer unter beliebigen Reaktions bedingungen keinerlei Schwierigkeiten entgegen. Der Verlauf der Umwandlung des 17a,21-Dioxy-20-keto- Teils eines Steroids der Pregnanreihe in das 17-20, 20-21-Bisalkylidendioxy-Derivat lässt sich leicht ver folgen durch Verwendung von Dianisol-bisdiphenyl- tetrazolium-chlorid-Reagens, das als B. T.-Reagens für die Durchführung des B. T.-Tests bekannt ist.
Dieses Reagens reagiert spezifisch mit einer Dioxy- acetongruppe unter Bildung einer tiefblauen Farbe. Mit den 17-20,20-21-Bisalkylidendioxy-Steroiden bildet es keine Farbe. Mit Hilfe dieses B. T.-Tests lässt sich somit der Fortschritt der Reaktion quanti tativ oder gewünschtenfalls auch nur halbquantitativ verfolgen, da die Intensität der blauen Farbe eine Funktion der in der untersuchten Lösung vorhan denen Menge an 17a,21-Dioxy-20-keto-Steroid ist.
Diese Farbintensität lässt sich leicht bestimmen auf Grund einer Vergleichsmessung mit einer bekannten Standardlösung, worauf sich das Ausmass, in wel chem sich die Reaktion vollzogen hat, berechnen lässt. Die Verwendung von Tetrazoliumblau (B. T.)- Reagens zur Prüfung auf Anwesenheit einer Dioxy- acetongruppierung ist in der Literatur näher be schrieben.
Nach dem oben diskutierten Verfahren und wenn nötig unter Verfolgung des Reaktionsverlaufs mit Hilfe des B. T.-Tests lässt sich jedes beliebige 17a,21- Dioxy-20-keto-Steroid der Pregnanreihe in das ent sprechende 17-20,20-2l -Bisalkylidendioxy-Steroid überführen.
Funktionelle Gruppen oder Substituenten in an dern Teilen des Pregnanmoleküls stören den Prozess nicht und werden in den meisten Fällen auch ihrer seits unter den Reaktionsbedingungen nicht zerstört oder verändert. Da die Reaktion in Gegenwart einer starken Säure verläuft, können während deren Durchführung Substituenten in andern Teilen des Moleküls, welche stark säureempfindlich sind, reagie- ren;
wo solche Nebenreaktionen eintreten können, jedoch unerwünscht sind, bildet man, wenn es an geht, vorzugsweise vorübergehend ein säurebestän diges Derivat solcher Substituenten. Im Falle von 11-Oxy-Steroiden kann eine Reaktion der 11-Hy- droxylgruppe vermieden werden, indem man den Prozess in einem Lösungsmittelsystem durchführt, welches von Alkoholen, insbesondere von niedrigen Alkanolen, frei ist.
So lassen sich in einem alkohol freien System beispielsweise Hydrocortison und Prednisolon ohne weiteres durch Behandeln mit Formaldehyd und starker Säure in 17 20,20-21- Bismethylendioxy-4-pregnen-lli3-ol-3-on bzw. 17-20, 20-21-Bismethylendioxy-1,4-pregnadien-1 lss-ol-3-on überführen.
Ist bei der Reaktion Alkohol zugegen, so erhält man natürlich ebenfalls das 17-20,20-21- Bismethylendioxy-Derivat, doch wird die 11-Oxy- gruppe gleichzeitig zur Alkoxymethylgruppe ver- äthert. Diese wird indessen bei der Entfernung der Bismethylendioxy-Funktion wieder gespalten.
Befinden sich an andern Stellen des Moleküls funktionelle Substituenten, welche gewöhnlich mit Aldehyden reagieren, z. B. Amine, so tritt bei Durch führung des erfindungsgemässen Verfahrens eine Re aktion ein. Dies führt jedoch zu keinen ernsthaften Schwierigkeiten, da sich die ursprüngliche funk tionelle Gruppe in den meisten Fällen leicht regenerie ren lässt. Das erfindungsgemässe Verfahren lässt sich somit an Steroiden der Pregnanreihe allgemein durch führen.
Das für die Reaktion verwendete Steroid weist gewöhnlich in 3-Stellung eine sauerstoffhaltige Funktion oder eine durch Hydrolyse in eine solche überführbare Funktion auf. Die Steroide können in andern Stellungen des Kerns aus kondensierten Ringen Substituenten, wie Oxy-, Keto-, Halogen oder niedrige Alkylgruppen aufweisen, beispielsweise in den Stellungen 2, 3, 4, 6, 7, 9, 11, 12, 14 oder 15.
Sie können vollständig gesättigt sein oder eine oder mehrere Doppelbindungen aufweisen, beispiels weise in 1-, 3-, 5-, 7-, 11- oder 14-Stellung. Als Bei spiele für nach dem erfindungsgemässen Verfahren aus den entsprechenden 17a,21-Dioxy-20-keto- Steroiden hergestellte neue Steroide seien erwähnt:

Gewisse der in 3- und 11 -Stellung oxydierten und im Ring A ungesättigten 17-20,20-21-Bismethylen- dioxy-Steroide der Pregnanreihe zeigen cortisonartige Wirkung und können ihrerseits zur Behandlung von Krankheiten verwendet werden, welche auf entzün dungshemmende Stoffe ansprechen.
Meistens liegt jedoch der grösste Wert der neuen Verbindungen darin, dass sie Zwischenprodukte darstellen, in wel chen die wichtige 17-Dioxyaceton-Seitenkette che misch inert und unzugänglich gemacht ist, so dass andere Teile des Steroidmoleküls gefahrlos Reak tionen und Umwandlungen unterworfen werd-.n kön nen, welche eine ungeschützte Dioxyacetongruppie- rung zerstören würden. Beispielsweise lassen sich Claisen-Kondensationen in a-Stellung zu einer Carbonylgruppe durchführen, ohne dass in der Sei tenkette eine unerwünschte Reaktion eintritt.
In dieser Weise kann 2-Methylcortison oder 2-Methyl- hydrocortison aus Cortison bzw. Hydrocortison hergestellt werden, indem man zunächst das 17-20, 20-21-Bisalkylidendioxy-Derivat herstellt, dieses nacheinander mit Methyloxalat und einem Methylie- rungsmittel behandelt und schliesslich das 2-Methyl- 17-20,20-21-Bismethylendioxy-Steroid mit Schwe felsäure behandelt,
um die Bismethylendioxy-Funk- tion wieder zu spalten und das 2-Methylcortison bzw. -hydrocortison zu erhalten. Dieser Prozess wäre mit einer ungeschützten Dioxyaceton-Seitenkette nicht durchführbar, da in der 21-Stellung eine tiefgreifende Nebenreaktion erfolgen würde.
Ferner ist es nach Inaktivierung der Seitenkette auf Grund des erfindungsgemässen Verfahrens nun mehr möglich, ein 17a,21-Dioxy-20-keto-Steroid der Pregnanreihe mit einer Carbonyl- oder Oxyfunktion am Kern mit einem Metallalkyl oder einem Metall- Grignard-Reagens zu behandeln, wobei der Kern alkyliert wird, ohne dass in der Seitenkette uner wünschte Reaktionen eintreten.
Die neuen Verbin dungen ermöglichen auch die Anwendung eines günstigen Verfahrens zum selektiven Reduzieren einer Carbonylgruppe im Ring A des Steroidkerns, ohne dass gleichzeitig der 20-Keto-Substituent reduziert wird.
Sind die gewünschten Reaktionen am Steroid mit der geschützten Dioxyaceton-Seitenkette durch geführt, so lässt sich die Seitenkette leicht regenerie ren durch Behandeln der 17-20,20-21-Bisalkyliden- dioxy-Verbindung mit Säure.
<I>Beispiel 1</I> 17-20,20-21-Bismethylendioxy-4-pregnen- 3,11-dion Man suspendiert 500 mg Cortison in 40 cm3 Chloroform und 10 cm3 Formalin (40o/oige wässe rige Formaldehydlösung) und setzt 10 cm3 konzen trierte Salzsäure zu. Dieses Zweiphasensystem wird bei Zimmertemperatur 52 Stunden lang kräftig ge rührt. Die wässerige Phase wird dann mit wässerigem Natriumhydroxyd alkalisch gemacht, abgetrennt und mit Chloroform extrahiert.
Nach Waschen mit Natriumbisulfitlösung dampft man das Chloroform ab, wobei man 685 mg eines kristallinen Rückstandes erhält. Nach dem Umkristallisieren aus Aceton und Methanol erhält man 17-20,20-21-Bismethylen- dioxy-4-pregnen-3,11-dion mit einem Schmelzpunkt von 253-259 C. Durch weitere Reinigung wird der Schmelzpunkt auf 259-262 C erhöht.
Wenn man das erhaltene Bismethylendioxy-De- rivat hydrolysiert und acetyliert, erhält man Cortison- acetat.
<I>Beispiel 2</I> 17-20,20-21-Bismethylendioxy-4-pregnen- 3,11-dion Man suspendiert 500 mg Cortison (freier Alkohol) in 25 cm3 Chloroform. Zu diesem Gemisch gibt man eine Lösung aus 10 cm3 konzentrierter Salzsäure und 10 cm3 Formalin. Das aus zwei Phasen bestehende Reaktionsgemisch wird 19 Stunden lang bei Zimmer temperatur gerührt, worauf man die Chloroform schicht abtrennt. Diese wird mit gesättigter Natrium- bicarbonatlösung gewaschen, getrocknet und unter Vakuum zu einem amorphen Rückstand konzentriert.
Beim Behandeln mit siedendem Methanol scheidet sich ein kristallines Produkt mit einem Schmelz punkt von 220-247 C ab, von welchem man 328 mg erhält. Durch weiteres Umkristallisieren aus Aceton- Methylenchlorid erhält man reines 17-20,20-21- Bismethylendioxy-4-pregnen-3,11-dion.
Das gleiche Produkt erhält man, wenn man eine Lösung von Cortison in Methylenchlorid unter den beschriebenen Bedingungen mit Formalin und Per chlorsäure erhitzt.
<I>Beispiel 3</I> 17-20,20-21-Bismethylendioxy-4-pregnen- 3,11-dion Zu einer Lösung von 500 mg Cortisonacetat und 20 cm3 Chloroform gibt man ein Gemisch aus 10 cm3 Formalin und 10 cm3 konzentrierter Salz säure. Dieses zweiphasige Reaktionsgemisch. wird 70 Stunden lang bei Zimmertemperatur gerührt. Darauf trennt man die chloroformische Schicht ab und extrahiert die anorganische Schicht mit Chloro form.
Den chloroformischen Auszug setzt man der ursprünglichen chloroformischen Schicht zu, wäscht die gesamte Lösung mit verdünntem Natriumbisulfat und trocknet dann über Magnesiumsulfat. Das Chloroform wird unter reduziertem Druck entfernt, wobei man einen amorphen Gummi im Gewicht von 475 mg erhält. Nach dem Reinigen durch Chromato- graphieren auf mit Säure gewaschener Tonerde und Eluieren mit Äther erhält man 180 mg l7-20, 20-21-Bismethylendioxy-4-pregnen-3,11-dion.
In gleicher Weise entsteht aus 4,9(11)-Pregnadien- 17(x,21-diol-3,20-dion 17-20,20-21-Bismethylen- dioxy-4,9(11)-pregnadien-3-on, und aus 4-Pregnen- 17a,21-diol-3,20-dion-21-acetat 17-20,20-21-Bis- methylendioxy-4-pregnen-3-on.
<I>Beispiel 4</I> 17-20,20-21-Bismethylendioxy-4-pregnen- l lss-ol-3-on Zu 500 mg in 25 cm3 Chloroform suspendiertem Hydrocortison gibt man ein Gemisch aus 10 cm3 Formalin und 10 cm3 konzentrierter Salzsäure. Das zweiphasige Reaktionsgemisch wird bei Zimmer temperatur 72 Stunden lang gerührt. Hiernach trennt man die Schichten und wäscht die anorganische Schicht mit Chloroform.
Die vereinigten chlöro- formischen Auszüge werden mit gesättigter Natrium- bicarbonatlösung gewaschen, getrocknet und unter reduziertem Druck konzentriert. Der entstehende Gummi wird auf mit Säure gewaschener Tonerde chromatographiert, wobei man ein nichtkristallines Produkt erhält, welches im B. T.-Test negativ ist und das für 17-20,20-21 - Bismethylendioxy - 4 - pregnen-llss-ol-3-on zu erwartende Infrarot-Spek- trum zeigt.
Wenn man dieses nichtkristalline Material in 2 cm3 Essigsäure löst, mit 60 mg Chromtrioxyd ver setzt, das Reaktionsgemisch 10 Minuten lang bei Zimmertemperatur stehenlässt, dem Gemisch 5 cm?' Wasser zusetzt, dann mehrmals mit Methylenchlorid extrahiert, die Methylenchloridauszüge mit verdünn tem Natriumbicarbonat wäscht, trocknet und kon zentriert, erhält man l7-20,20-21 - Bismethylen- dioxy-4-pregnen-3,11-dion.
<I>Beispiel 5</I> 17-20,20-21-Bismethylendioxy-1,4- pregnadien-3,11-dion Zu einer Suspension von 500 mg Prednison in 25 cm3 Chloroform gibt man ein Gemisch aus 10 cm3 Formalin und 10 cms konzentrierter Salzsäure. Man rührt das zweiphasige Gemisch 70 Stunden lang bei Zimmertemperatur und trennt dann die beiden Schichten.
Die wässerige Schicht wird einmal mit Chloroform extrahiert, und der chloroformische Aus zug wird mit der ursprünglichen organischen Schicht vereinigt. Die chloroformische Lösung wird mit ge sättigter Natriumbicarbonatlösung gewaschen, ge trocknet und unter reduziertem Druck konzentriert, wobei man 712 mg eines semikristallinen Feststoffes erhält. Dieses Rohprodukt wird mit siedendem Me thanol geläutert, wobei man 352 mg eines kristallinen Materials mit dem Schmelzpunkt 175-195 C er hält.
Nach dem Umkristallisieren dieses Materials aus Aceton und Methanol erhält man eine Probe reines 17-20,20-21-Bismethylendioxy-1,4-pregnadien-3,11- dion mit dem Schmelzpunkt 214-217 C.
Das gleiche Produkt erhält man bei Verwendung von Schwefelsäure anstelle von Salzsäure.
<I>Beispiel 6</I> 17-20,20-21-Bismethylendioxy-1,4- pregnadien-11 ss-ol-3-on-11-methoxy-methyläther Zu einer Suspension von 500 mg Prednisolon in 25 cm3 Chloroform gibt man ein Gemisch aus 10 cm3 Formalin und 10 cm3 konzentrierter Salz säure. Dieses zweiphasige System wird 45 Stunden lang bei Zimmertemperatur gerührt. Hiernach wer den die beiden Schichten getrennt. Man extrahiert die wässerige Schicht einmal mit Chloroform und vereinigt den chloroformischen Auszug mit der ur sprünglichen organischen Phase.
Die vereinigte chloroformische Lösung wird mit gesättigter Natrium- bicarbonatlösung gewaschen, getrocknet und unter reduziertem Druck konzentriert, wobei man 500 mg eines semikristallinen Feststoffes erhält. Durch Um kristallisieren dieses- Produktes aus Methanol erhält man 300 mg eines bei 191-202 C schmelzenden Produktes.
Durch weiteres Umkristallisieren aus Aceton und Methanol erhält man eine Probe aus reinem 17-20,20-21-Bismethylendioxy-1,4-pregna- dien-llss-ol-3-on-11-methoxymethyl-äther mit dem Schmelzpunkt 217-220 C.
<I>Beispiel 7</I> 17-20,20-21-Bismethylendioxy-4,6- pregnadien-3,11-dion Eine Lösung aus 1,106 g 4,6-Pregnadien-3,11,20- trion-17a,21-diacetat in 50 em3 wasserfreiem Me thanol wird unter Stickstoffatmosphäre eine halbe Stunde lang gerührt.
Darauf setzt man 2,5 cm?, einer 2n methanolischen Lösung von Natriummethoxyd zu und setzt das Rühren weitere 5 Minuten lang fort. Nach Zusatz von 0,3 cm3 Wasser rührt man weitere 5 Minuten lang und säuert die Lösung dann mit Essigsäure an.
Man entfernt das Lösungsmittel unter reduziertem Druck, löst den Rückstand in Chloro form und extrahiert die chloroformische Lösung nacheinander mit Wasser, l0fl/Qiger Natriumbicar- bonatlösung und nochmals mit Wasser. Man trocknet die chloroformische Lösung über Natriumsulfat und entfernt das Lösungsmittel unter reduziertem Druck, wobei man 1,09 g rohes 4,6-Pregnadien-17a,21-diol- 3,11,20-trion erhält.
Ein Gemisch aus diesem Diol, 50 cm3 Chloro form, 20 cm3 konzentrierter Salzsäure und 20 cm- Formalin wird bei Zimmertemperatur 64 Stunden lang kräftig gerührt. Dann fügt man 50 cm3 Chloroform und 50 cm3 Wasser zu, trennt die Schich ten und wäscht die organische Phase nacheinander mit Wasser, 10ahIger Natriumbicarbonatlösung und nochmals mit Wasser.
Nach dem Trocknen über Natriumsulfat entfernt man das Chloroform. unter reduziertem Druck, wobei man einen Rückstand von 17-20,20-21-Bismethylendioxy-4,6-pregnadien-3,11- dion erhält.
Dieses Produkt wird in Benzol gelöst, an mit Säure gewaschener Tonerde adsorbiert und mit Äther eluiert. Durch Abdampfen des Äthers erhält man die gewünschte Verbindung, welche nach Kristalli- sieren aus Methanol einen Schmelzpunkt von 239 bis 243 C (Zersetzung) und die spezifische Drehung [a] D + 119 (Chloroform) besitzt.
Unter den im zweiten und dritten Absatz dieses Beispiels beschriebenen Bedingungen und unter Ver wendung von Benzol als ursprüngliches organisches Lösungsmittel und von Bromwasserstoffsäure anstelle von Salzsäure erhält man aus 6-Methyl-9a-fluor-1,4- pregnadien-llss,17a,21-triol-3,20-dion 17 20,20-21 Bismethylendioxy-6-methyl-9a-fluor-1,4-pregnadien- 11 ss-ol-3-on.
<I>Beispiel 8</I> 17-20.20 21-Bismethylendioxy-1,4-pregnadien-- l lss-ol-3-on Zu einer Suspension von 500 mg 1,4-Pregnadien- llss,17a,21-triol-3,20-dion in 25 ein?- Chloroform gibt man ein Gemisch von 10 cm3 Formalin und 10 cm?- konzentrierter Salzsäure. Man rührt das Re aktionsgemisch eine Stunde lang kräftig bei Zimmer- temperatur,
trennt dann die chloroformische Schicht ab und wäscht sie mit gesättigter Natriumbicarbonat- lösung. Nach dem Trocknen konzentriert man unter vermindertem Druck, wobei man einen Gummi er hält, welcher beim Läutern mit siedendem Methanol kristallisiert. Dieses Produkt wird mehrmals aus Äthylacetat umkristallisiert, wobei man 17-20, 20-21-Bismethylendioxy-1,4-pregnadien-11 ss-ol-3-on erhält.
<I>Beispiel 9</I> 17-20,20-21-Bismethylendioxy-9a-fluor-4-pregnen- 1 lss-ol-3-on Man löst 5 g 9a-Fluor-hydrocortisonacetat in 250 cm3 Chloroform. Unter Rühren setzt man dieser Lösung 100 cm3 konzentrierte Salzsäure und 100 cm3 37a/aige wässerige Formaldehydlösung (Formalin) zu.
Nach 18stündigem Rühren bei Zimmertemperatur trennt man die chloroformische Schicht ab, wäscht sie mit gesättigter Natriumbicarbonatlösung, trocknet und dampft zur Trockne ein. Durch Läutern des Öls mit Methanol erhält man 1,1 g 17-20,20-21-Bis- methylendioxy - 9a - fluor -4 - pregnen - 11@-ol-3-on, Schmelzpunkt (245 ) 248-255 C.
<I>Beispiel 10</I> Man löst 20 g 9a-Fluorhydrocortison-alkohol in 1 Liter Chloroform und gibt zu dieser Lösung unter Rühren 400 em3 konzentrierte Salzsäure und 400 cm3 Formalin. Das Reaktionsgemisch wird bei Zimmertemperatur 1 Stunde lang gerührt und dann wie in Beispiel 9 beschrieben aufgearbeitet. Das Pro dukt schmilzt bei 250-260 bzw. 285-290 C.
<I>Beispiel 11</I> 17-20,20-21-Bismethylendioxy-4-pregnen- l l ss-ol-3-on Man vereinigt 30 g Hydrocortison mit 1500 cm3 Chloroform und gibt zu dieser Lösung ein gekühltes Gemisch aus 600 cm3 konzentrierter Salzsäure und 600 cm3 Formalin. Das Reaktionsgemisch wird 1 Stunde lang bei Zimmertemperatur gerührt.
Man trennt die beiden Schichten, extrahiert die wässerige Schicht mit Chloroform und vereinigt die organischen Lösungen, welche mit Natriumbisulfatlösung ge waschen, getrocknet und im Vakuum konzentriert werden. Der gesamte Rückstand wird auf mit Säure gewaschener Tonerde chromatographiert. Durch Eluieren der Säule mit Petroläther-Äther 1 : 4 erhält man 4 g kristallinen 17-20,20-21-Bismethylendioxy- 4 - pregnen -11ss - o1 - 3 - on-11-methoxymethyläther, Schmelzpunkt 160-165 C.
Aus dem Äther-Eluat erhält man 10g 17-20,20-21-Bismethylendioxy-4- pregnen-1lss-ol-3-on, Schmelzpunkt 217-222 C. <I>Beispiel 12</I> 17-20s20-21 Bismethylendioxy-4-pregnen- 11 ss-ol-3-on Man löst 2 g Hydrocortison in 100 cm3 Chloro form, welches mit Schwefelsäure geschüttelt, über Calciumchlorid getrocknet und destilliert wurde.
Zu diesem Gemisch gibt man unter Kühlen 60 cm3 kon zentrierte Salzsäure und 60 cm!' methanolfreies 37%aiges wässeriges Formaldehyd. Das Reaktions- gemisch wird 8 Stunden lang bei Zimmertemperatur gerührt.
Unter Befolgung des im vorhergehenden Beispiel beschriebenen lsolierungsverfahrens erhält man nach dem Läutern des erhaltenen Öls mit Äthylacetat kristallines 17-20,20-21-Bismethylen- dioxy-4-pregnen-llss-ol-3-on, Schmelzpunkt 200 bis 220 C.
<I>Beispiel 13</I> 17-20,20-21-Bismethylendioxy-9, l l ss-oxido- 4-pregnen-3-on Man suspendiert 10 g 9,llss-Oxido-4-pregnen- 17a,21-diol-3,20-dion in 500 cm3 Methylenchlorid und setzt diesem Gemisch 20 cm-' Salzsäure und 20 cm;' Formalin zu. Dieses Reaktionsgemisch rührt man 90 Minuten lang bei Zimmertemperatur. Durch Isolieren des Reaktionsproduktes in der oben be schriebenen Weise erhält man 7 g 17-20,20-21-Bis- methylendioxy-9,llss-oxido-4-pregnen-3-on, Schmelz punkt 180-190 C.
<I>Beispiel 14</I> 17-20,20-21-Bismethylendioxy-l-pregnen- 3,11-dion Ein Gemisch aus 4 g 1-Pregnen-17a"21-diol- 3,11,20-trion, 200 cm3 Chloroform, 80 cm3 konzen trierter Salzsäure und 80 cm3 Formalin wird bei Zimmertemperatur 48 Stunden lang kräftig gerührt. Darnach trennt man die chloroformische Schicht ab und wäscht mit drei kleinen Portionen 511/o-iger Ka- liumcarbonatlösung und anschliessend mit Wasser bis zur Neutralität des Waschwassers.
Man trocknet die chloroformische Lösung über Magnesiumsulfat und entfernt das Lösungsmittel im Vakuum. Man er hält eine hellgelbe ölige Substanz. Durch Kristalli sieren dieses Materials aus Aceton und Umkristalli- sieren aus Methylenchlorid-Methanol erhält man reines 17-20,20-21 - Bismethylendioxy-1-pregnen- 3,11-dion, Schmelzpunkt 230-233 C.
<I>Beispiel 15</I> 17-20,20-21-Bisäthylidendioxy-4-pregnen- 3,11-dion Man suspendiert 500 mg Cortison in 25 cm3 Chloroform und setzt dieser Suspension ein Gemisch aus 10 cm- konzentrierter Salzsäure und 10 cm3 40 iaigem wässerigem Acetaldehyd zu. Dieses zwei- phasige Reaktionsgemisch wird 72 Stunden lang bei Zimmertemperatur gerührt. Hierauf wird die chloroformische Schicht abgetrennt und die anorga nische Schicht mit Chloroform extrahiert.
Die ver einigten chloroformischen Auszüge werden mit ge sättigter Natriumbicarbonatlösung gewaschen, ge trocknet und unter reduziertem Druck vom Lösungs mittel befreit. Man erhält einen amorphen Rückstand im Gewicht von 720 mg.
Man chromatographiert die ses Material an mit Säure gewaschener Tonerde und eluiert das 17-20,20-21-Bisäthylidendioxy-4- pregnen-3,11-dion mit Äther-Chloroform-Gemischen. <I>Beispiel 16</I> 17-20,20-21-Bisbutyraldioxy-4-pregnen- 3,
11-dion Zu einer Suspension von 500 mg Cortison in 25 ein- Chloroform gibt man ein Gemisch aus 10 cm-' 40 /aigem wässerigem Butyraldehyd und 10 cm3 konzentrierter Salzsäure. Das entstehende zweiphasige Reaktionsgemisch wird 68 Stunden lang bei Zimmertemperatur gerührt. Hiernach trennt man die beiden Phasen und extrahiert die wässerige Schicht mehrmals mit Chloroform.
Die chlorofor- mischen Auszüge werden mit der ursprünglichen organischen Schicht vereinigt und mit einer gesättig ten Natriumbicarbonatlösung gewaschen. Die chloro- formische Lösung wird getrocknet und unter redu ziertem Druck zu einem Öl konzentriert, welches 1,2 g wiegt. Dieses Öl wird in Benzol gelöst und auf einer Säule aus 30 g mit Aceton und Säure gewa schener Tonerde adsorbiert. Man eluiert mit Petrol- Äther (6:4 bis 2:8).
Diese Eluate ergeben beim Konzentrieren 17-20,20-21 - Bisbutyral - dioxy - 4 - pregnen-3,11-dion als amorphen Feststoff.
Unter Verwendung von Prednisonacetat und Ca- pronaldehyd anstelle von Cortison und Butyraldehyd erhält man nach dem oben beschriebenen Verfahren 17-20,20-21- Biscaproaldioxy-1,4-pregnadien-3,11- dion.
<I>Beispiel 17</I> 17-20,20-21-Bispropionaldioxy-4-pregnen- l lss-ol-3-on Man löst 800 mg Hydrocortisonacetat in 40 cm3 Methylenchlorid und vermischt mit 15 cm3 Propion- aldehyd und 15 cms konzentrierter Salzsäure. Das erhaltene Gemisch wird 10 Stunden lang bei 30 C gerührt.
Hierauf trennt man die Schichten und extra hiert die wässerige Schicht mit 10 cm?, Methylen- chlorid. Die Methylenchloridlösungen werden ver einigt, mit verdünntem Natriumbicarbonat gewa schen und über wasserfreiem Magnesiumsulfat ge trocknet. Das organische Lösungsmittel wird durch Konzentrieren im Vakuum entfernt.
Der so erhaltene Rückstand wird auf 30g mit Säure gewaschener Tonerde chromatographiert. Durch Eluieren mit Pe- troläther-Äther und Entfernen der Lösungsmittel aus den Eluatfraktionen erhält man 17-20,20-21-Bis- propionaldioxy-4-pregnen-1 lss-ol-3-on.
<I>Beispiel I S</I> 17-20,20-21-Bisvaleraldioxy-9a-fluor-1,4- pregnadien-11 ss-ol-3-on Unter gleichem Vorgehen wie in Beispiel 17, wobei man jedoch als Steroid 900 mg 9a-Fluor-1,4- pregnadien-11ss,17a,21-triol-3,20-dion und anstelle des Propionaldehyds 15 cm3 Valeraldehyd verwen det, erhält man 17-20,20-21-Bisvaleraldioxy-9a- fluor-1,4-pregnadien-l lss-ol-3-on. <I>Beispiel 19</I> 17-20,
20-21-Bisbutyraldioxy-4-pregnen- 11/3-ol-3-on Man löst 500 mg Hydrocortison in 25 cm3 Me- thylenchlorid und fügt zur entstehenden Lösung 10 cm3 40 /diges wässeriges Butyraldehyd und 10 cm? konzentrierte Salzsäure. Das Reaktions gemisch wird bei Zimmertemperatur 6 Stunden lang gerührt. Man trennt die beiden Lösungsmittelschich ten und extrahiert die wässerige Schicht mit frischem Methylenchlorid. Die Lösungen im organischen Lö sungsmittel
werden vereinigt, mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Man entfernt das Methvlenchlorid durch Konzentrieren im Va kuum undychromatographiert das zurückbleibende Öl auf mit Säure gewaschener Tonerde. Aus den Eluaten mit Petroläther-Äther 4 : 1 erhält man reines 17-20, 20-21- Bisbutyraldioxy - 4 - pregnen -11 /3-ol-3-on als klare, glasige Substanz.
Die Struktur des 17-20,20-21-Bisbutyraldioxy- 4-pregnen-I1/3-ol-3-ons lässt sich bestätigen durch Erhitzen eines Teils der oben erhaltenen glasigen Substanz mit 50 cm-- 50 /o:iger Essigsäure auf dem Dampfbad während 8 Stunden.
Man entfernt die Essigsäure durch Vakuumdestillation und reinigt den Rückstand durch Extrahieren mit Methylenchlorid, Entfernen des organischen Lösungsmittels und Acety- lieren durch 10minutiges Erhitz-,n mit Pyridin-Acet- anhydrid. Das acetylierte Material wird auf Tonerde chromatographiert, wobei man in den Äther-Chloro- form-Eluaten 1 : 4 reines Hydrocortisonacetat erhält.
<I>Beispiel 20</I> 17-20,20-21-Bismethylendioxy-allopregnan- 11/3-ol-3-on Zu einer Lösung von 7,1g Allopregnan-11f,17a, 21-triol-3,20-dion in 350 cm33 Chloroform gibt man 140 cm-' kalte konzentrierte Salzsäure sowie 140 cm kaltes neutrales Formalin. Das zweiphasige Gemisch wird 2 Stunden lang gerührt. Man trennt die Lö sungsschichten und extrahiert die wässerige Phase zweimal mit je 100 cm33 Chloroform.
Die chloro- formischen Lösungen werden vereinigt, nacheinander mit Wasser, 50/Q.iger Natriumbicarbonatlösung und wieder mit Wasser gewaschen und dann über wasser freiem Magnesiumsulfat getrocknet. Sodann entfernt man das Chloroform im Vakuum und chromatogra- phiert den Rückstand über 180 g Tonerde.
Durch Eluieren der Säule mit Äthyläther erhält man, nach Umkristallisieren aus Äther-Petroläther, 17-20, 20-21-Bismethylendioxy-allopregnen-11/3-ol-3-on-11- methoxymethyläther. Durch weiteres Eluieren mit Athyläther erhält man 17-20,20-21-Bismethylen- dioxy-allopregnan-11/3-ol-3-on, das nach Umkristalli- sieren aus Äther einen Schmelzpunkt von 220 bis 225 C zeigt.
In gleicher Weise wie oben beschrieben erhält man, ausgehend von 6,5 g 9a-Fluor-1,4-pregnadien- 11 i3,17a,21-triol-3,20-dion, 17-20,20-21-Bismethy- lendioxy-9 a-fluor-1,4-pregnadien-11/3-ol-3-on.

It has been found that treating a steroid of the pregnane series with a dioxyacetone grouping in the 17-position with formaldehyde or a lower alkyl aldehyde in the presence of a strong acid produces a steroid disubstituted by methylenedioxy groups or substituted methylenedioxy groups,
in which all three carbon atoms of the dioxyacetone part become members of at least one of the five-membered rings formed here.

It is advantageous if both the starting material and the reaction product are soluble in the organic solvent, but the 17-20,20-21-bismethylenedioxy steroid is also obtained in good yield and quality if the end product is soluble in the organic solvent and that 17a, 21-dioxy-20-keto steroid used as starting material is insoluble.
Water-immiscible organic solvents which are suitable for carrying out the process are chloroform, methylene chloride, ethylene dichloride, carbon tetrachloride, ethylene dibromide, methylene bromide, benzene, ethyl ether and the like, the chlorinated solvents being the most expedient.
The reaction according to the invention is usually carried out at temperatures from about -30 to about i-50 ° C. The simplest way to work is at temperatures from 0 to about 35 ° C. at which satisfactory yields are achieved with most steroids.
The reaction time required to achieve optimal yields of 17-20, 20-21-bisalkylidene-dioxy-steroids depends to a certain extent on the reaction temperature used. It has also been shown that the reaction time required for good yields is a function of the type of 17a, 21-dioxy-20-keto steroid used as the starting material.
For example, in a chloroform-formalin-hydrochloric acid reaction system, the optimal conversion of cortisone to 17-20,20-21-bismethylenedioxy - 4 - pregnen-3,11-dione takes about 40-50 hours, while the conversion of prednisolone in 17-20,20-21-bismethylenedioxy-1,4-pregnadien-11/3-ol-3-one in high yield only 15 to 30 minutes are necessary.
However, these differences in the reactivity of the steroids pose no difficulties in quickly determining the correct reaction time under any reaction conditions. The course of the conversion of the 17a, 21-dioxy-20-keto part of a steroid of the pregnane series into the 17-20, 20-21-bisalkylidenedioxy derivative can easily be followed by using dianisole-bisdiphenyl-tetrazolium-chloride reagent which is known as the BT reagent for performing the BT test.

Using the method discussed above and, if necessary, following the course of the reaction with the aid of the BT test, any 17a, 21-dioxy-20-keto steroid of the pregnane series can be converted into the corresponding 17-20,20-2l -bisalkylidenedioxy steroid convict.
Functional groups or substituents in other parts of the pregnane molecule do not interfere with the process and in most cases are not destroyed or changed under the reaction conditions either. Since the reaction takes place in the presence of a strong acid, substituents in other parts of the molecule which are highly acid-sensitive can react while it is being carried out;
where such side reactions can occur but are undesirable, when it comes to, preferably temporarily an acid-resistant derivative of such substituents is formed. In the case of 11-oxy steroids, a reaction of the 11-hydroxyl group can be avoided by carrying out the process in a solvent system which is free from alcohols, in particular from lower alkanols.
In an alcohol-free system, for example, hydrocortisone and prednisolone can easily be converted into 17-20, 20-21-bismethylenedioxy-4-pregnen-lli3-ol-3-one or 17-20, 20- by treatment with formaldehyde and strong acid. Transfer 21-bismethylenedioxy-1,4-pregnadien-1 lss-ol-3-one.
If alcohol is present during the reaction, the 17-20,20-21-bismethylenedioxy derivative is of course also obtained, but the 11-oxy group is at the same time etherified to the alkoxymethyl group. However, this is split again when the bismethylenedioxy function is removed.

The steroids can have substituents such as oxy, keto, halogen or lower alkyl groups in other positions of the core composed of fused rings, for example in positions 2, 3, 4, 6, 7, 9, 11, 12, 14 or 15.

Usually, however, the greatest value of the new compounds lies in the fact that they represent intermediates in which the important 17-dioxyacetone side chain is chemically inert and inaccessible, so that other parts of the steroid molecule are safely subjected to reactions and conversions which would destroy an unprotected dioxyacetone group. For example, Claisen condensations can be carried out in the a-position to a carbonyl group without an undesired reaction occurring in the side chain.
In this way, 2-methylcortisone or 2-methyl-hydrocortisone can be produced from cortisone or hydrocortisone by first producing the 17-20, 20-21-bisalkylidenedioxy derivative, treating this one after the other with methyl oxalate and a methylating agent, and finally treated the 2-methyl- 17-20,20-21-bismethylenedioxy steroid with sulfuric acid,

The new connec tions also enable the use of a convenient method for selectively reducing a carbonyl group in ring A of the steroid nucleus without reducing the 20-keto substituent at the same time.
Once the desired reactions have been carried out on the steroid with the protected dioxyacetone side chain, the side chain can be easily regenerated by treating the 17-20,20-21-bisalkylidene dioxy compound with acid.
<I> Example 1 </I> 17-20,20-21-bismethylenedioxy-4-pregnen-3,11-dione 500 mg of cortisone are suspended in 40 cm3 of chloroform and 10 cm3 of formalin (40% aqueous formaldehyde solution) and adds 10 cm3 of concentrated hydrochloric acid. This two-phase system is vigorously stirred at room temperature for 52 hours. The aqueous phase is then made alkaline with aqueous sodium hydroxide, separated and extracted with chloroform.

The same product is obtained when a solution of cortisone in methylene chloride is heated with formalin and perchloric acid under the conditions described.
<I> Example 3 </I> 17-20,20-21-bismethylenedioxy-4-pregnen-3,11-dione A mixture of 10 cm3 formalin and 10 cm3 is added to a solution of 500 mg of cortisone acetate and 20 cm3 of chloroform concentrated hydrochloric acid. <I> Example 7 </I> 17-20,20-21-bismethylenedioxy-4,6-pregnadiene-3,11-dione A solution of 1.106 g of 4,6-pregnadiene-3,11,20-trione-17a , 21-diacetate in 50 cubic meters of anhydrous methanol is stirred for half an hour under a nitrogen atmosphere.

20-21-Bisbutyraldioxy-4-pregnen-11/3-ol-3-one Dissolve 500 mg of hydrocortisone in 25 cm3 of methylene chloride and add 10 cm3 of 40 / d aqueous butyraldehyde and 10 cm? concentrated hydrochloric acid. The structure of 17-20,20-21-bisbutyraldioxy-4-pregnen-I1 / 3-ol-3-one can be confirmed by heating part of the glassy substance obtained above with 50 cm-50% acetic acid the steam bath for 8 hours.
The acetic acid is removed by vacuum distillation and the residue is purified by extracting with methylene chloride, removing the organic solvent and acetylating by heating with pyridine acetic anhydride for 10 minutes. The acetylated material is chromatographed on alumina, giving 1: 4 pure hydrocortisone acetate in the ether-chloroform eluates.
<I> Example 20 </I> 17-20,20-21-bismethylenedioxy-allopregnan-11/3-ol-3-one To a solution of 7.1 g of allopregnan-11f, 17a, 21-triol-3.20 -dione in 350 cm33 chloroform is added to 140 cm cold concentrated hydrochloric acid and 140 cm cold neutral formalin. The two-phase mixture is stirred for 2 hours. The solution layers are separated and the aqueous phase is extracted twice with 100 cm33 of chloroform each time.
The chloroformic solutions are combined, washed successively with water, 50% sodium bicarbonate solution and again with water and then dried over anhydrous magnesium sulphate. The chloroform is then removed in vacuo and the residue is chromatographed over 180 g of clay.
Eluting the column with ethyl ether gives, after recrystallization from ether-petroleum ether, 17-20, 20-21-bismethylenedioxy-allopregnen-11/3-ol-3-one-11-methoxymethyl ether. Further elution with ethyl ether gives 17-20,20-21-bismethylene dioxy-allopregnan-11/3-ol-3-one, which, after recrystallization from ether, has a melting point of 220 to 225 ° C.
