in der Arl und Are Phenylgruppen bedeuten, von denen mindestens die eine an einer oder mehreren ortho-Stellen durch eine Alkylgruppe mit höchstens 6 Kohlenstoffatomen substituiert ist, wobei die Grup pen Arl und Ar. keine andere Substituenten tragen.
Diese Äther können in ihre Säureadditionssalze und quaternäre Ammoniumverbindungen übergeführt werden. Als Salze kommen z. B. das Hydrobromid, das Hydrochlorid, das Sulfat und das Methansulfonat in Betracht.
Es sind bereits mehrere Verbindungen ähnlicher Struktur bekannt. So werden in der amerikanischen Patentschrift Nr. 2 595 405 die unsubstituierte Ver bindung und in der amerikanischen Patentschrift Nr. 2 706 198 und in den britischen Patentschriften Nrn. 708911, 769282 und 773290 einige in einer Phenylgruppe substituierte Verbindungen beschrie ben. Diese Patentschriften erwähnen auch einige Herstellungsverfahren für diese Verbindungen. Die Substanzen obiger Formel besitzen ausgesprochen atropinartige Eigenschaften sowie eine starke Anti- histaminwirkung.
Es wurde nun gefunden, dass Verbindungen mit wesentlich stärkerer Wirkung erhalten werden kön nen, wenn wenigstens einer der Phenylkerne einen besagten Alkylsubstituenten an einer oder mehreren ortho-Stellen enthält.
Die USA-Patentschrift Nr. 2 706198 beschreibt auch Verbindungen der Formel

in der R und R1 unter anderem niedere Alkylgruppen sein können, aber die in dieser Patentschrift be schriebenen alkylsubstituierten Verbindungen enthal ten alle einen oder mehrere Substituenten an der para- oder meta-Stelle der Phenylkerne. Ausserdem ergibt sich aus der Patentschrift, dass hauptsächlich die para-halogen-substituierten Verbindungen für wichtig gehalten werden.
Es hat sich gezeigt, dass vor allem die mono ortho-methyl-substituierte Verbindung eine hervor ragende atropinartige Wirkung besitzt, welche die der entsprechenden unsubstituierten Verbindung so wie die der mono-meta- und mono-para-methyl- substituierten Verbindung viele Male übersteigt.
Ausserdem ist die Toxizität von derselben Grössen ordnung oder sogar geringer als die der isomeren Verbindungen oder der unsubstituierten Verbindung, so dass die therapeutische Breite der mono-ortho- Methylverbindung wesentlich grösser ist als die der entsprechenden Verbindungen, welche die Methyl- gruppe nicht oder an einer andern Stelle im Phenyl- kern besitzen.
In nachstehender Tabelle werden die pharma kologischen Versuchsergebnisse, die bei Vergleichs versuchen an dem isolierten Caviadarm mit den an verschiedenen Stellen durch eine mono-Methylgruppe substituierten Verbindungen erhalten wurden, er wähnt. Die Wirkung der gleichfalls in diese Unter suchung miteinbezogenen unsubstituierten Verbin dung wurde als Vergleichsbasis gewählt.

Wenn bei Mäusen 25 mgikg 1,4-Dipyrrolidino- butyn-2 injiziert wurden, betrug die wirksame Dosis bei mindestens 509/a der Mäuse (ED.o) bei sub kutaner Injektion für die 2-Methylverbindung 2,5 mg pro kg Maus, für die 2-tert.-Butylverbiiidung 21 mg pro kg Maus.
Auch in dieser Hinsicht zeigte es sich, dass die 2-Methyl-benzhydryl-tropinyl-äther wieder hervor ragende Eigenschaften besitzen.
Das erfindungsgemässe Verfahren zur Herstellung der genannten Tropinyläther ist dadurch gekenn zeichnet, dass man eine Verbindung der Formel

zu einer Verbindung der Formel I umsetzt, wobei A und B Reste bedeuten, die sich bei der Umsetzung mit Ausnahme eines in einem von ihnen enthaltenen Sauerstoffatomes abspalten.
Wenn eines der Symbole A und B ein Halogen atom und das andere eine OH-Gruppe darstellt, wird die Reaktion im allgemeinen unter Zusatz einer säurebindenden Substanz ausgeführt. Wenn A das Halogenatom ist, kann man auch mit einem über schuss des Aminoalkohols, ohne Zugabe einer an dern säurebindenden Substanz arbeiten. Man kann z. B. derart vorgehen, dass man 1 Äquivalent des substituierten Benzhydrylhalogenids mit 2 Äquiva lenten des Aminoalkohols erhitzt, wobei Zugabe eines Lösungsmittels nicht erforderlich ist.

<I>Beispiel 1</I> 21,6 g 2-Methyl-benzhydrylchlorid (0,1 Mol) werden mit 25,8 g Tropin (0,2 Mol) bei einer Tempe ratur von 100-150 C während 15-30 Minuten er hitzt. Es bilden sich zwei Schichten, von denen die untere Schicht bei geringer Kühlung fest wird. Nach Kühlung des Reaktionsgemisches werden 100 bis 150 cm3 Äther zugesetzt, in dem sich die flüssige obere Schicht löst. Nach Abtrennung der Äther schicht werden 150 cm3 Wasser und darauf 25 cm 40'o/aige Bromwasserstoffsäure zugesetzt.
Der Nieder schlag, bestehend aus 2-Methyl-benzhydryl-tropinyl- äther-hydrobromid, wird durch Filtration abgetrennt und mit Äther und anschliessend mit Wasser gewa schen und schliesslich getrocknet.
Die Ausbeute beträgt 14 g (45 %). Nach Reini- gung durch Kristallisation schmilzt die Verbindung bei 222-22411 C.
<I>Beispiel 2</I> Tropin wird mit Thionylchlorid in Chlortropan umgesetzt. 1 Mol Chlortropan wird mit 1 Mol 2-Me- thyl-benzhydrol in Gegenwart von Natriumamid als säurebindende Substanz während 15-30 Minuten auf 100-150 C erhitzt.
Die Reaktionsmischung wird in der im Beispiel 1 beschriebenen Weise aufgearbeitet. Da die Mög lichkeit, dass quaternäre Salze gebildet werden, hier viel geringer ist als bei der obenbeschriebenen Me thode, ist die Ausbeute erheblich grösser;
sie beträgt 60 %. Schmelzpunkt nach Kristallisation 221-223 C. <I>Beispiel 3</I> 1,0 Mol ortho - Methyl - benzhydrol wird mit 1,1 Mol Tropin versetzt. Die Mischung wird auf 50-60 C erhitzt, bis eine homogene Schmelze er halten worden ist.
Dieser Schmelze werden 1,15 Mol p-Toluolsulfosäure zugesetzt, wobei die Temperatur bis auf 90-100 C ansteigt. Anschliessend wird 4 bis 5 Stunden unter erniedrigtem Druck auf 130-150 C erhitzt. Nach Abkühlen wird die Reaktionsmasse in einem Gemisch aus verdünnter Lauge und Äther auf genommen. Die Ätherschicht wird abgetrennt und mit verdünnter wässriger HBr-Lösung behandelt. Das Hydrobromid des 2-Methylbenzhydryl-tropinyl-äthers scheidet sich als ein Öl aus, das bald fest wird.
Der feste Stoff wird isoliert und aus Aceton oder aus einem Aceton-Äther-Gemisch kristallisiert. Aus beute<B>750/a.</B> Schmelzpunkt 223-224 C.
<I>Beispiel 4</I> In der im Beispiel 3 beschriebenen Weise wird aus ortho-Äthyl-benzhydrol, Tropin und p-Toluol- sulfosäure das Hydrobromid von 2-Äthyl-benzhydryl- tropinyl-äther hergestellt. Schmelzpunkt nach Kristal lisation 180 C. Ausbeute 67"/o,.
<I>Beispiel 5</I> In der im Beispiel 3 beschriebenen Weise wird aus 2,2'-Dimethyl-benzhydrol der 2,2'-Dimethyl- benzhydryl-tropinyl-äther hergestellt. Schmelzpunkt des Hydrobromids 194 C. Ausbeute 70 1/o.
<I>Beispiel 6</I> 25,9 g 2-tert.-Butyl-benzhydryl-chlorid (0,1 Mol) werden mit 25,8g Tropin (0,2 Mol) während 15-30 Minuten auf 100-l50 C erhitzt. Das Reaktions gemisch wird nach Kühlung mit Äther extrahiert. In die ätherische Lösung wird nach Trocknen über wasserfreiem NazC03 trockener Chlorwasserstoff ein geleitet. Das Hydrochlorid des 2-tert.-Butyl-benz- hydryl-tropinyl-äthers wird abgetrennt und aus Aceton umkristallisiert. Ausbeute 4011/a. Schmelz punkt 229-230 C.

These ethers can be converted into their acid addition salts and quaternary ammonium compounds. As salts come z. B. the hydrobromide, the hydrochloride, the sulfate and the methanesulfonate into consideration.
Several compounds of similar structure are already known. For example, US Pat. No. 2,595,405 describes the unsubstituted compound and US Pat. No. 2,706,198 and British Patents Nos. 708911, 769282 and 773290 describe some compounds substituted in a phenyl group. These patents also mention some manufacturing methods for these compounds. The substances in the above formula have extremely atropine-like properties and a strong anti-histamine effect.
It has now been found that compounds with a significantly stronger effect can be obtained if at least one of the phenyl nuclei contains a said alkyl substituent at one or more ortho positions.
U.S. Patent No. 2,706,198 also describes compounds of the formula

in which R and R1 can be, inter alia, lower alkyl groups, but the alkyl-substituted compounds described in this patent all contain one or more substituents at the para or meta position of the phenyl nuclei. The patent also shows that it is mainly the para-halogen-substituted compounds that are considered important.
It has been shown that the mono-ortho-methyl-substituted compound in particular has an excellent atropine-like effect, which exceeds that of the corresponding unsubstituted compound as well as that of the mono-meta- and mono-para-methyl-substituted compound many times.
In addition, the toxicity is of the same order of magnitude or even lower than that of the isomeric compounds or the unsubstituted compound, so that the therapeutic range of the mono-ortho-methyl compound is significantly greater than that of the corresponding compounds which do not have the methyl group or which have one elsewhere in the phenyl nucleus.
In the table below, the pharmacological test results obtained in comparative tests on the isolated Caviadarm with the compounds substituted at various points by a mono-methyl group are mentioned. The effect of the unsubstituted compound also included in this study was chosen as the basis for comparison.
