Verfahren zur Herstellung von 17ss-oxy-17a-alkinyl-Steroiden Die Erfindung bezieht sich auf ein neues Ver fahren zur Herstellung von 17ss-Oxy-17a-alkinyl- Derivaten sowohl von gesättigten wie auch von un gesättigten Steroiden der Androstan- und 3 : 5-Cyclo- androstanreihe, welche vorzugsweise in den Ringen A und B durch mindestens eine der Gruppen OH, Alkyl-O- oder O substituiert sind.
Die erfindungsgemäss herstellbaren 17a-Alkinyl- Derivate sind auf Grund ihrer biologischen Eigen schaften oder auch als Zwischenprodukte für die Herstellung von Verbindungen mit wertvollen biolo- gischen Eigenschaften nützlich. Beispielsweise erhält man nach dem erfindungsgemässen Verfahren Ver bindungen, welche den progestational wirkenden Homologen von Äthisteron und 6a-Methyläthisteron entsprechen oder in diese umgewandelt werden können.
Die erfindungsgemäss herstellbaren neuen 17,B- Oxy-17a-alkinyl-Steroide der Androstan- und 3:5- Cycloandrostanreihe besitzen die folgende Restformel I bzw. II:

in der der Steroidkern auch ungesättigt sein kann und worin R eine Alkylgruppe mit höchstens 4 C-Atomen bedeutet.
Nach dem erfindungsgemässen Verfahren kön nen z. B. folgende neue Verbindungen hergestellt werden: 17,i-(Prop-l-inyl)-androst-5-en-3f,17ss-diol, 17 a-(But-l-inyl)-androst-5-en-3fl,17ss-diol, 17a-(Pent-l-inyl)-androst-5-en-3/3,17ss-diol, 17a-(Prop-l-inyl)-3 : 5-cycloandrostan-6/3,17ss- diol, 17a-(But-l-inyl)-3 : 5-cycloandrostan-6ss,17,B- diol, 17a-(Pent-l-inyl)-3 :
5-cycloandrostan-6ss,17ss- diol. Die obigen Verbindungen sind wertvolle Zwi schenprodukte für die Herstellung von 21-Methyl-, 21-Äthyl- und 21-Propyl-äthisteron, welche Verbin dungen bei oraler Verabreichung wertvolle progesta- tionale Eigenschaften zeigen.
6a-Methyl-17a-(prop- 1-inyl)-androst-5-en-3ss,17ss-diol ist ein wertvolles Zwischenprodukt für die Herstellung von 6a,21-Di- methyl - äthisteron, einem starken oral-wirksamen progestationalen Mittel.
Das erfindungsgemässe Verfahren zur Herstellung von 17f-Oxy-17a-alkinyl-Steroiden der Formel I bzw. 1I ist nun dadurch gekennzeichnet, dass man ein substituiertes 17-Keto-Steroid eines Androstans oder eines 3 : 5-Cycloandrostans der Restformel III bzw. IV

mit einem Alkinylmagnesiumhalogenid der Formel HalMgC-C-R, umsetzt, in welcher R eine Alkyl- gruppe mit höchstens 4 C-Atomen bedeutet.
Die erfindungsgemässe Umwandlung des substi tuierten 17-Keto-Steroids (III bzw. IV) in das ent sprechende 17a-Alkinyl-Steroid (I bzw. Il) erfolgt zweckmässig dadurch, dass man eine Lösung der Ver bindung der Formel III bzw. IV in einem Lösungs mittel, wie Tetrahydrofuran, zu einem überschuss des Alkinyl-magnesiumhalogenids, vorzugsweise des Bromids oder Jodids, in einem Lösungsmittel, wie z.
B. Tetrahydrofuran, zusetzt und das Gemisch auf Rückfluss erhitzt oder mehrere Stunden lang bei Zimmertemperatur stehenlässt. Hierauf kann der ge bildete Komplex zersetzt werden durch Zusatz von beispielsweise wässerigem Ammoniumchlorid, und das Steroid-Derivat kann in üblicher Weise, bei spielsweise durch Extrahieren, mit einem organischen Lösungsmittel, wie Äther, isoliert werden.
Das erfindungsgemässe Verfahren lässt sich auf 17-Keto-Steroide der Androstan- und 3:5-Cyclo- androstanreihe anwenden, welche verschiedene Sub- stituenten und ungesättigte Bindungen im Molekül aufweisen.
Oxy- und Acyloxygruppen werden im allgemei nen nicht an der Reaktion teilnehmen oder störend auf diese einwirken. Dasselbe gilt auch für isolierte oder konjugierte ungesättigte Bindungen, welche in den Ringen A, B und C vorliegen können, wenn sie nicht mit einer sauerstoffhaltigen Gruppe konjugiert sind. Auch Alkyl- und Alkoxygruppen in beliebiger Stellung (ausser an C17) treten nicht in die Reaktion ein.
Ketogruppen, insbesondere in den Stellungen 3, 6 und 12 (jedoch ausgenommen an C17), erfordern indessen im allgemeinen einen Schutz, damit deren Teilnahme an der Grignard-Reaktion vermieden werden kann. Ein solcher Schutz lässt sich leicht er reichen, beispielsweise durch Ketal- oder Enoläther- bildung, wobei die nachfolgende Regenerierung der Ketofunktion leicht möglich ist.
Insbesondere können als Ausgangsstoff folgende Verbindungen verwendet werden: 3ss-Oxy-(Acyloxy)-androst-5-en-17-on, 6,B-Oxy-3 : 5-cycloandrostan-17-on, die 3-Enoläther- und 3-Ketal-Derivate von An- drost-4-en-3,17-dion sowie von 11-oxydierten Androst-4-en-3,17-dionen. <I>Beispiel 1</I> 17a-(Prop-l-inyl)-aiidrost-5-en-3l,17f-diol In einem mit einem Tropftrichter,
Rührer und Rückflusskondensator versehenen Dreihalskolben wurde unter Stickstoff eine Lösung von Athyl- magnesiumbromid hergestellt aus 3,65 g Magnesium und<B>16,3</B> g Äthylbromid in 150 cm"- wasserfreiem Tetrahydrofuran. Die Lösung wurde unter Rück fluss 30 Minuten lang erhitzt und dann auf Zimmer temperatur abkühlen gelassen.
Hierauf ersetzte man den Rückflusskondensator durch einen Kondensator, welcher durch ein Gemisch aus Aceton und festem COz gekühlt wurde, und kühlte den Tropftrichter durch Zugabe von 20 cm3 Tetrahydrofuran von -60 C. In einem auf -60 C gekühlten Kolben wurden 8 g 1-Propin abgewogen, welche bei -60 C in 60 cm3 Tetrahydrofuran gelöst, in den Tropftrich ter übergeführt und rasch unter Rühren der Gri- gnard-Lösung zugesetzt wurden.
Es erfolgte Äthan- entwicklung, und das Rühren wurde 30 Minuten lang fortgesetzt, nach welcher Zeit die Äthanentwick- lung aufgehört hatte. Der Rückflusskondensator wurde dann wiederum an den Kolben angeschlossen, und es wurde unter Rühren tropfenweise eine Lösung von 4,3 g Dehydroisoandrosteron in 100 cm3 Tetra- hydrofuran zugesetzt. Es bildete sich ein gelatinöser Niederschlag, und das Gemisch wurde 2 Stunden lang auf Rückfluss erhitzt.
Der Komplex wurde mit 50g Ammoniumchlorid in 200 cm3 Wasser zersetzt, und das Produkt wurde mit Äther isoliert. Durch Entfernen des Lösungsmittels unter vermindertem Druck erhielt man einen freien Rückstand, welcher aus Aceton-Hexan in Form von farblosen Plättchen, Schmelzpunkt 178-180 C, kristallisierte. Durch Umkristallisieren aus wässerigem Methanol erhielt man das Monohydrat von 17a-(Prop-l-inyl)-androst- 5-en-3ss,17ss-diol in Form von Plättchen, Schmelz punkt 179-181 C, [a]D = -121 (c = 0,98; immer in Chloroform, wo nichts anderes angegeben wird).
<I>Beispiel 2</I> 17a-(But-l-inyl)-androst-5-en-3ss,17ss-diol In einem mit einem Tropftrichter, Rührer und Rückflusskondensator versehenen Dreihalskolben wurde unter Stickstoff eine Lösung von Äthyl- magnesiumbromid hergestellt aus 4,86 g Magnesium, 21,8 g Äthylbromid und 200 em3 wasserfreiem Tetrahydrofuran. Die Lösung wurde 30 Minuten lang unter Rühren erhitzt und dann auf Zimmer temperatur abkühlen gelassen.
Der Rückflusskonden- Bator wurde durch einen Kondensator ersetzt, wel cher mit einem Gemisch aus Aceton und festem C02 gekühlt wurde, und der Tropftrichter wurde durch Zugabe von 50 cm- Tetrahydrofuran gekühlt, wel ches auf -60 C abgekühlt war. In einem auf -60 C gekühlten Kolben wurden 12 g 1-Butin abgewogen, welche bei -60 C in 50 cm?, Tetrahydrofuran gelöst und der Grignard-Lösung rasch unter Rühren zu gesetzt wurden. Es entwickelte sich Athan, und das Rühren wurde 30 Minuten lang fortgesetzt.
Hierauf wurde der Rückflusskondensator wiederum mit dem Kolben verbunden, und es wurde eine Lösung von 5,8 g Dehydroisoandrosteron in 100 cm- Tetra- hydrofuran tropfenweise unter Rühren zugesetzt. Es bildete sich ein gelatinöser Niederschlag, und das Gemisch wurde unter Rückfluss 2 Stunden lang er hitzt.
Der Komplex wurde mit 50 g Ammonium chlorid in 200 cm-' Wasser zersetzt, und das Pro dukt wurde mit Äther isoliert. Durch Entfernen des Lösungsmittels unter vermindertem Druck erhielt man eine gummiartige Substanz, welche durch Zu gabe von Hexan zur Kristallisation gebracht werden konnte. Durch Umkristallisieren aus Aceton-Hexan erhielt man 17a-(But-l-inyl)-androst-5-en-3ss,17ss- diol in Form von Nadeln, Schmelzpunkt 78-81 C, [a] D -121 (c = 0,83).
<I>Beispiel 3</I> 17a-(Pent-l-inyl)-androst-5-en-3 ss,17ss-diol In Stickstoffatmosphäre wurde aus 4,86g Ma gnesium, 21,8g Äthylbromid und 200 cm3 wasser freiem Tetrahydrofuran eine Äthylmagnesiumbromid- Lösung hergestellt. Die Lösung wurde 30 Minuten lang unter Rückfluss erhitzt und dann auf Zimmer temperatur abkühlen gelassen. Der Lösung fügt man unter Rühren langsam 15 g 1-Pentin in 20 cm?, Tetrahydrofuran zu.
Es wurde Äthan entwickelt, und das Rühren wurde 30 Minuten lang fortgesetzt. Hierauf gab man unter Rühren tropfenweise eine Lösung von 5,8 g Dehydroisoandrosteron in 100 cm?, Tetrahydrofuran zu, wobei sich ein gelatinöser Nie derschlag bildete. Das Gemisch wurde 5 Stunden lang bei Zimmertemperatur gerührt und dann über Nacht bei Zimmertemperatur stehengelassen. Durch Zersetzung mit 50g Ammoniumchlorid in 200 cm3 Wasser und Isolieren des Produktes mit Äther erhielt man einen Gummi, welcher bei Zugabe von Hexan kristallisierte.
Durch Umkristallisieren aus Aceton- Hexan erhielt man 17a-(Pent-l-inyl)-androst-5-en- 3ss,17ss-diol in Form von Nadeln, Schmelzpunkt 67-69 C, [alD -120 (c = 0,97).
<I>Beispiel</I> 17a-(Hex-l-inyl)-androst-5-en-3ss,17ss-diol Unter Stickstoff wurde aus 4,86 g Magnesium, 21,8 g Äthylbromid und 200 cm?, wasserfreiem Te- trahydrofuran eine Äthyl-magnesiumbromid-Lösung hergestellt. Die Lösung wurde 30 Minuten lang unter Rückfluss erhitzt und dann auf Zimmertemperatur abkühlen gelassen. Zur Lösung wurden unter Rühren 18 g 1-Hekin in 20 cm3 Tetrahydrofuran langsam zugesetzt.
Es entwickelte sich Äthan, und das Rühren wurde 30 Minuten lang fortgesetzt. Hierauf wurde tropfenweise unter Rühren eine Lö sung von 5,8g Dehydroisoandrosteron in 100 cm3 Tetrahydrofuran zugegeben, wobei sich ein gelati nöser Niederschlag bildete. Das Gemisch wurde 2 Stunden lang auf Rückfluss erhitzt. Der Komplex wurde mit 50 g Ammoniumchlorid in 200 cm3 Wasser zersetzt, und das Produkt wurde mit Äther isoliert.
Durch Entfernung des Lösungsmittels unter vermindertem Druck erhielt man einen Gummi, wel cher aus Aceton-Hexan in Form von Nadeln, Schmelzpunkt 80-82 C, kristallisierte. Durch Um kristallisieren aus Aceton-Hexan erhielt man 17a- (Hex-l-inyl)-androst-5-en-3ss,17ss-diol in Form von Nadeln, Schmelzpunkt 80-82 C, die nach längerem Trocknen den Schmelzpunkt 70-72 C, [a] -114 D (c = 1,00), aufwiesen.
<I>Beispiel 5</I> 17a-(Prop-l-inyl)-3 : 5-cycloandrostan-6ss,17ss-diol In einem mit einem Tropftrichter, Rührer und Rückflusskondensator versehenen Dreihalskolben wurde aus 3,65 g Magnesium, 16,3g Äthylbromid und 150 cm3 wasserfreiem Tetrahydrofuran eine Äthyhnagnesiumbromid-Lösung hergestellt. Die Lö sung wurde unter Rückfluss 30 Minuten lang erhitzt und dann auf Zimmertemperatur abkühlen gelassen.
Der Rückflusskondensator wurde durch einen Kon densator ersetzt, welcher mit Aceton und festem CO., gekühlt wurde, und der Tropftrichter wurde gekühlt durch Zugabe von 50 cm3 Tetrahydrofuran von einer Temperatur von -60o- C. In einem auf -60 C gekühlten Kolben wurden 8 g 1-Propin abgewogen, welche dann bei -60 C in 50 cm3 Tetrahydrofuran gelöst, in den Tropftrichter übergeführt und der Gri- gnard-Lösung rasch unter Rühren zugesetzt wurden. Es entwickelte sich Äthan, und die Lösung wurde 30 Minuten lang gerührt.
Hierauf wurde der Rück flusskondensator wiederum an dem Kolben ange schlossen, und es wurde unter Rühren tropfenweise eine Lösung von 4,3 g 6ss-Oxy-3 : 5-cycloandrostan- 17-on in 100 cm3 Tetrahydrofuran zugesetzt. Die Lösung wurde dann 2 Stunden lang unter RückfIuss erhitzt. Der Komplex wurde mit 50 g Ammonium chlorid in 200 cm-' Wasser zersetzt, und das Produkt wurde mit Äther isoliert. Durch Entfernung des Lö sungsmittels unter vermindertem Druck erhielt man einen Gummi, welcher bei Zugabe von Hexan kri stallisierte.
Durch Umkristallisieren aus Aceton- Hexan erhielt man 17a-Prop-l-inyl-3 : 5-cycloandro- stan-6ss,17ss-diol in Form von Nadeln, Schmelzpunkt 199-201 C, [a] D<B>-12 </B> (c = 1,03).
<I>Beispiel 6</I> 17a-(But-l-inyl)-3 : -5-cycloandrostan-6ss,17ss-diol In einem mit einem Tropftrichter, Rührer und Rückflusskondensator versehenen Dreihalskolben wurde aus 3,65 g Magnesium, 16,3g Äthylbromid und 150 cm?, wasserfreiem Tetrahydrofuran eine Äthylmagnesiumbromid-Lösung hergestellt. Die Lö sung wurde 30 Minuten lang unter Rückfluss er hitzt und dann auf Zimmertemperatur abkühlen ge lassen.
Man ersetzte den Rückflusskondensator durch einen mit Aceton und festem C02 gekühlten Kon densator und kühlte den Tropftrichter durch Zugabe von 50 cm?, Tetrahydrofuran von -60 C. In einem auf -60 C gekühlten Kolben wog man 9,2 g 1-Butin, welches man bei -60 C in 50 ein?- Tetrahydrofuran löste, in Lösung in den Tropftrichter überführte und unter Rühren rasch der Grignard-Lösung zusetzte. Es entwickelte sich Athan, und das Rühren wurde 30 Minuten lang fortgesetzt.
Nun schloss man den Rückflusskühler wieder am Kolben an und setzte unter Rühren tropfenweise eine Lösung von 4,3 g 6ss-Oxy-3:5-cycloandrostan-17-on in 100 cm- Te- trahydrofuran zu. Die Lösung wurde 2 Stunden lang auf Rückfluss erhitzt. Durch Zersetzen des Kom plexes mit 50 g Ammoniumchlorid in 200 cm3 Wasser und Isolieren des Produktes mit Äther erhielt man einen Gummi, welcher aus Aceton-Hexan in Form von Prismen, Schmelzpunkt 166-169 C, kri stallisierte.
Durch Umkristallisieren aus wässerigem Methanol erhielt man 17a-(But-l-inyl)-3:5-cyclo- androstan - 6ss,17ss <I>-</I> diol in Form von Blättern, Schmelzpunkt 172-l74 C, [a]D -14,5 (c = 0,92). <I>Beispiel 7</I> 17a-(Pent-l-inyl)-3 : 5-cycloandrostan-6ss,17ss-diol Aus 3,65 g Magnesium, 16,3g Äthylbromid und 150 cm?, wasserfreiem Tetrahydrofuran wurde unter Stickstoff eine Lösung von Äthylmagnesiumbromid hergestellt.
Die Lösung wurde 3 Minuten lang unter Rückfluss erhitzt und dann auf Zimmertemperatur abkühlen gelassen. Unter Rühren setzte man langsam 13,6 g 1-Pentin in 50 cm3 Tetrahydrofuran zu. Es entwickelte sich Äthan, und das Rühren wurde 30 Minuten lang fortgesetzt.
Sodann gab man unter Rühren tropfenweise 4,3 g 6,B-Oxy-3 :5-cycloandro- stan-17-on in 100 cm3 Tetrahydrofuran zu und er hitzte die Lösung 2 Stunden lang unter Rückfluss. Der Komplex wurde mit 50g Ammoniumchlorid in 200 cm3 Wasser zersetzt, und das Produkt wurde mit Äther isoliert.
Durch Entfernen des Lösungs mittels unter vermindertem Druck erhielt man einen Gummi. Durch Perkolieren einer benzolischen Lö sung durch eine kurze Säule mit 10 g Tonerde und Entfernung des Lösungsmittels erhielt man einen Rückstand, welcher aus Benzol-Petroläther (Siede punkt 60-80 C) in Form von Nadelbüscheln kri stallisierte, Schmelzpunkt 140-144 C. Umkristalli sieren aus wässerigem Methanol ergab 17a-(Pent-1- inyl)-3:
5-cycloandrostan-6ss,17ss-diol in Form von Blättern, Schmelzpunkt 146-148 C, [a12' D -130 (c = 1,04).
<I>Beispiel 8</I> 6-Methyl-17a-(prop-l-inyl)-androst-5-en-3ss,17ss-diol Man ging gleich vor wie in Beispiel 1, verwen dete jedoch 6-Methyldehydroisoandrosteron-acetat anstelle von Dehydroisoandrosteron. Das Produkt wurde in üblicher Weise mit Äther isoliert und aus Aceton-Hexan gereinigt.
Das 6-Methyl-17a-(prop-l- inyl) - androst - 5 - en - 3ss,17ss-diol bildete Nadeln, Schmelzpunkt 115 C, [a] D -l05 (c = 0,5). <I>Beispiel 9</I> 17ss-Oxy-17a-(prop-l-inyl)-androst-4-en-3-on a)
Nach dem in Beispiel 1 beschriebenen Ver fahren wurde unter Verwendung von 8 g 1-Propin eine Lösung eines Grignard-Reagens hergestellt. Zu diesem Reagens gab man 4 g des von Androst-4-en- 3,17-dion abgeleiteten 3-Enoläthyläthers in 100 em3 Tetrahydrofuran und erhitzte das Gemisch 2 Stunden unter Rückfluss. Der Komplex wurde mit 50 g Ammoniumchlorid in 200 cm- Wasser zersetzt, und das Produkt wurde mit Äther isoliert.
Seine Lösung in 100 cm3 Äthanol wurde bei Zimmertemperatur 2 Stunden lang mit 5 cm-' konzentrierter Salzsäure behandelt, und das Produkt wurde mit Äther isoliert und aus Aceton-Hexan kristallisiert. Das 17f-Oxy- 17a-(prop-l-inyl)-androst-4-en-3-on bildete Nadeln, Schmelzpunkt 151-152 C, [a]D + 11 (c = 0,53 in Äthanol).
b) Nach dem in Beispiel 1 beschriebenen Ver fahren wurde unter Verwendung von 4 g 1- Propin eine Lösung eines Grignard-Reagens hergestellt. Zu diesem Reagens gab man 2 g des 3,3-Äthylendioxy- Derivates von Androst-4-en-3,17-dion in 50 cm3 Tetrahydrofuran und erhitzte das Gemisch 2 Stunden lang auf Rückfluss. Nach dem Kühlen behandelte man das Gemisch mit 25 g Ammoniumchlorid in 100 cm3 Wasser,
isolierte das Produkt mit Äther und behandelte 18 Stunden lang bei Zimmertempe ratur mit 25 cm3 9511ioiger Essigsäure. Durch Isolie ren des Produktes mit Äther unter anschliessender Reinigung aus Aceton-Hexan erhielt man 17fl-Oxy- 17a-(prop-l-inyl)-androst-4-en-3-on, Schmelzpunkt 151 C, welches mit einer Probe der gemäss a) her gestellten Substanz identisch war.
<I>Beispiel 10</I> 17ss-Oxy-6a-methyl-17a-(prop-l-inyl)-androst- 4-en-3-on Nach dem in Beispiel 1 beschriebenen Verfahren wurde unter Verwendung von 8 g 1-Propin eine Lösung eines Grignard-Reagens hergestellt.
Zu die sem Reagens gab man 4 g des 3,3-Äthylendioxy- Derivates von 6a-Methylandrost-4-en-3,17-dion in 100 ein- Tetrahydrofuran und erhitzte das Gemisch 3 Stunden unter Rückfluss. Nach Zersetzung des Komplexes mit wässerigem Ammoniumchlorid iso lierte man das Produkt mit Äther und behandelte 30 Minuten lang bei 100 C mit 50 cm-' 90oloiger Essigsäure. Dann goss man das Gemisch in Wasser, extrahierte mit Äther und kristallisierte das Produkt aus wässerigem Methanol.
Das 17ss-Oxy-6a-methyl- 17a-(prop-l-inyl)-androst-4-en-3-on bildete Plätt chen mit dem Schmelzpunkt 99-102 C, [a]20 +<B>10 </B> (c = 1,0 in Chloroform). <I>Beispiel 11</I> 17a-(But-1-inyl)-17ss-oxy-6a-methylandrost- 4-en-3-on Nach dem in Beispiel 2 beschriebenen Verfahren wurde unter Verwendung von 12 g 1-Butin eine Lösung eines Grignard-Reagens hergestellt.
Zu die sem Reagens gab man 6 g des 3,3-Äthylen-dioxy- Derivates von 6a-Methylandrost-4-en-3,17-dion in <B>100</B> cm? Tetrahydrofuran und erhitzte das Gemisch 2 Stunden unter Rückfluss. Man isolierte das Pro dukt wie im vorhergehenden Beispiel beschrieben, behandelte 30 Minuten lang bei 100 C mit 90 /oiger Essigsäure, isolierte das gebildete Material und rei nigte durch Umkristallisieren aus wässerigem Me thanol.
Das 17a-(But-l-inyl)-17ss-oxy-6a-methyl- androst-4-en-3-on schied sich in Form von Blättern ab, Schmelzpunkt 74-76 C, [a] D '-17 (c - 1,0 in Chloroform).
Process for the preparation of 17ss-oxy-17a-alkynyl steroids The invention relates to a new process for the preparation of 17ss-oxy-17a-alkynyl derivatives of both saturated and unsaturated steroids of androstane and 3: 5 -Cyclo androstane series, which are preferably substituted in rings A and B by at least one of the groups OH, alkyl-O- or O-.
