Verfahren zur Herstellung von neuen Aryloxyessigsäureamiden
Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung von neuen Aryloxyessigsäureamiden mit wertvollen pharmakologischen Eigenschaften. Es wurde gefunden, dass substituierte Aryloxyessigsäureamide entsprechend der Formel

worin Rl einen Alkyl-, Alkenyl- oder Cycloalkylrest oder einen gegebenenfalls durch Halogenatome oder niedermolekulare Alkyl- oder Alkoxygruppen substituierten Phenyl- oder Benzylrest, R2 Wasserstoff oder einen niedermolekularen Alkyl- oder Alkenylrest, R3 und R4 niedermolekulare Alkylreste und X die direkte Bindung oder eine Athylen- oder Vinylengruppe, -CH2-CH2- bzw.
-CH CH-, bedeuten, wobei Alkylreste R, und R2 auch unter sich direkt oder über ein Sauerstoffatom verbunden sein können, pharmakologisch wertvolle Eigenschaften, insbesondere hypnotische, narkotische und anästhetische Wirksamkeit besitzen.
Das erfindungsgemässe Verfahren zur Herstellung der Verbindungen der Formel I ist dadurch gekennzeichnet, dass man eine Aryloxyessigsäure der Formel

oder ein reaktionsfähiges funktionelles Derivat einer solchen auf ein primäres oder sekundäres Amin der Formel einwirken lässt.

Dazu geeignete reaktionsfähige funktionelle Derivate von Aryloxyessigsäuren der Formel II sind beispielsweise deren Halogenide und gemischte Anhydride mit niedrigen aliphatischen Carbonsäuren, insbesondere Essigsäure, sowie deren Ester, insbesondere die niederen Alkylester und der Phenylester.
Die Umsetzungen werden zweckmässig in Lösungs- oder Verdünnungsmitteln durchgeführt. Als Lösungsmittel für die Umsetzungen mit Säurehalogeniden und -anhydriden kommen einerseits inerte organische Lösungsmittel, wie Äther oder Benzolkohlenwasserstoffe und anderseits Wasser, für die Umsetzungen mit Säureestern auch Alkohole in Betracht. Bei den erfindungsgemäss durchgeführten Reaktionen mit Säurehalogeniden oder gemischten Anhydriden in organischen Lösungsmitteln kann ein Überschuss an dem umzusetzenden Amin oder auch eine tertiäre organische Base, wie Pyridin oder Dimethylanilin, als säurebindendes Mittel zugegeben werden. In Wasser können zu diesem Zwecke überdies auch anorganische Stoffe, wie Natrium- oder Kaliumcarbonat, Verwendung finden.
Als Beispiele für erfindungsgemäss als Ausgangsstoffe in Betracht kommende Aryloxyessigsäuren der Formel II seien die
2-Methoxy-4-acetyl-phenoxyessigsäure,
2-Methoxy-4-propionyl-phenoxyessigsäure,
2-Methoxy-4-n-butyryl-phenoxyessigsäure,
2-Methoxy-4-isobutyryl-phenoxyessigsäure,
2-Methoxy-4-isovaleryl-phenoxyessigsäure,
2-Methoxy-4-(butan-3'-onyl)-phenoxyessigsäure, 2-Methoxy-4-( 1',2'-buten-3'-ony -phenoxy essigsäure, 2-Methoxy-4-(S 1', 2'-penten-3'-onyl)-phenoxy- essigsäure, 2-Äthoxy-4-acetyl-phenoxyessigsäure,
2-Äthoxy-4-propionyl-phenoxyessigsäure,
2-Äthoxy-4-n-butyryl-phenoxyessigsäure und 2-Äthoxy-4-isovaleryl-phenoxyessigsäure genannt.
Diese Säuren, vorzugsweise aber funktionelle Derivate derselben, können beispielsweise mit Methylamin, ethylamin, Allylamin, Benzylamin, Anilin, Dimethylamin, Diäthylamin, N-Methylbenzylamin, N-Äthyl-anilin, N-n-Butyl-anilin oder andern primären und sekundären Aminen der aliphatischen, aromatischen, aromatisch-aliphatischen und cycloaliphatischen Reihe, sowie mit Pyrrolidin, Piperidin und Morpholin umgesetzt werden.
Im nachfolgenden Beispiel bedeuten die Teile Gewichtsteile, welche sich zu Volumteilen wie g zu cm verhalten. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
10 Teile 2-Methoxy -4- n - butyryl-phenoxyessig- säure werden in 70 Teilen absolutem Benzol suspendiert und unter Rühren bei Zimmertemperatur mit einer Lösung von 11 Teilen Oxalylchlorid in 20 Teilen absol. Benzol tropfenweise versetzt. Man rührt 1 Stunde bei Zimmertemperatur und weitere 2 Stunden bei 40 , wobei unter Aufschäumen Abspaltung von Kohlendioxyd und Chlorwasserstoff erfolgt. Die erhaltene klare Lösung wird im Vakuum bei 35 bis 400 eingedampft, wobei als Rückstand ein tiefgelbes öl zurückbleibt, welches das rohe 2-Methoxy-4-nbutyryl-phenoxyessigsäurechlorid darstellt.

Suitable reactive functional derivatives of aryloxyacetic acids of the formula II are, for example, their halides and mixed anhydrides with lower aliphatic carboxylic acids, in particular acetic acid, and their esters, in particular the lower alkyl esters and the phenyl esters.
The reactions are expediently carried out in solvents or diluents. Suitable solvents for the reactions with acid halides and anhydrides are on the one hand inert organic solvents such as ethers or benzene hydrocarbons and on the other hand water, and for the reactions with acid esters also alcohols. In the reactions carried out according to the invention with acid halides or mixed anhydrides in organic solvents, an excess of the amine to be reacted or a tertiary organic base, such as pyridine or dimethylaniline, can be added as an acid-binding agent. Inorganic substances, such as sodium or potassium carbonate, can also be used in water for this purpose.

2-ethoxy-4-propionyl-phenoxyacetic acid,
2-ethoxy-4-n-butyryl-phenoxyacetic acid and 2-ethoxy-4-isovaleryl-phenoxyacetic acid.
These acids, but preferably functional derivatives thereof, can, for example, with methylamine, ethylamine, allylamine, benzylamine, aniline, dimethylamine, diethylamine, N-methylbenzylamine, N-ethyl-aniline, Nn-butyl-aniline or other primary and secondary amines of the aliphatic, aromatic, aromatic-aliphatic and cycloaliphatic series, as well as with pyrrolidine, piperidine and morpholine.
In the following example, the parts mean parts by weight, which are related to parts by volume as g to cm. The temperatures are given in degrees Celsius.

10 parts of 2-methoxy -4- n - butyryl-phenoxyacetic acid are suspended in 70 parts of absolute benzene and, with stirring, at room temperature with a solution of 11 parts of oxalyl chloride in 20 parts of absolute. Benzene was added dropwise. The mixture is stirred for 1 hour at room temperature and for a further 2 hours at 40, with splitting off of carbon dioxide and hydrogen chloride taking place with foaming. The clear solution obtained is evaporated in vacuo at 35 to 400, a deep yellow oil remaining as residue, which is the crude 2-methoxy-4-n-butyryl-phenoxyacetic acid chloride.
