durch Behandeln mit einer aromatischen Carbon- säure in die entsprechenden Deserpidsäure-lactone umlagert, diese mit Methanol in Gegenwart kataly- tisch-wirksamer Mengen von Alkali-methylat um setzt und in den entstandenen Hydroxyestern die Hydroxy baruppe mit einem reaktionsfähigen Derivat der 3,4,
5-Trimethoxy-benzoesäure verestert.
Reserpin, ein Alkaloid der Rauwolfia-Arten, hat in den letzten Jahren eine grosse therapeutische Be deutung erlangt, da es auf Grund seiner blutdruck senkenden und psychisch beruhigenden Eigenschaften eine erfolgreiche Behandlung der Hypertonie und von psychischen Störungen erlaubt. Immerhin weist dieses Alkaloid nicht selten Nebenwirkungen auf, die z. T. unangenehm sind und manchmal zum Unterbruch der Behandlung zwingen können.
Am häufigsten treten psychische Depression, Müdigkeit, Magenhyperacidität, Diarrhoe, Dyspnoe oder Asthma, Oedeme (antidiuretische Wirkung), manchmal pep- tische Geschwüre, Parkinsonismus, Grand mal-An- fälle auf [H. A. Schroeder u. H.
M. Perry, J. Amer. Med. Assoc. <I>159,</I> 839 (1955); Editorial, Brit. Med. <I>J.1955,</I> 11, 1378<B>1</B> Es war daher wünschenswert, über ähnlich wir kende Verbindungen verfügen zu können, welche bei Beibehaltung der therapeutisch wertvollen Eigen schaften die Nebenwirkungen des Reserpins nicht oder nur in vermindertem Ausmass zeigen würden.
Es hat sich nun gezeigt, dass die erfindungs gemäss erhältlichen, bisher unbekannten Verbin dungen wertvolle therapeutische Eigenschaften be sitzen. Sie haben insbesondere blutdrucksenkende und psychisch beruhigende Eigenschaften, wobei einzelne Verbindungen die Wirksamkeit des Re- serpins übertreffen. Ferner zeichnen sie sich durch eine besonders gute Verträglichkeit und eine lang dauernde Wirkung aus. Die neuen Verbindungen eignen sich u. a. zur Behandlung der Hypertonie und zur Beruhigung von psychischen Erregungs- oder Angstzuständen neurotischer oder psychotischer Her kunft.
Sie sollen in der Therapie verwendet werden.
Das Verfahren wird beispielsweise so ausgeführt, dass ein substituiertes Iso-deserpidsäure-lacton der Formel II (hergestellt nach Schweiz. Pat. Nr. 361811) in Xylol gelöst und mit Pivalinsäure in einer Stick stoffatmosphäre gekocht wird. Das entstandene sub stituierte Deserpidsäure-lacton wird mit Methanol in Gegenwart katalytisch wirksamer Mengen von Alkali methylat in den entsprechenden Hydroxyester über führt und dieser z.
B. in Pyridin mit 3,4,5-Tri- methoxy-benzoylchlorid verestert.
Die verfahrensgemäss hergestellten Verbindungen sind bei Normaltemperatur kristallin. In den nach folgenden Beispielen erfolgen alle Temperaturanga ben in Celsiusgraden. Die Schmelzpunkte sind kor rigiert.
<I>Beispiel 1</I> (-)-10,11-Dioxy-methylen-deserpidin a) Isomerisierung: 0,970 g (-)-10,11-Dioxy- methylen-isodeserpidsäure-lacton (Formel I: R2 und R3 = -O-CH#-O-, R1, R4 und RG = H, R5 = CH3 - hergestellt nach Schweiz. Patent Nr.<B>361811)</B> werden in 20 cm- einer Lösung von Xylol)Pivalinsäure 9: 1 16 Stunden in Stickstoff atmosphäre am Rückfluss gekocht.
Nach dieser Zeit wird vom gebildeten kristallisierten Niederschlag ab filtriert, das Filtrat im Vakuum zur Trockne ver dampft und der Rückstand zwischen verdünnter Ammoniaklösung und Methylenchlorid verteilt. Der Rückstand der Methylenchloridlösung liefert aus Me thanol eine weitere Substanzmenge in Form von Na deln, welche mit dem direkt abgetrennten Kristalli- sat vereinigt und aus Methanol umkristallisiert wird.
(-)- 10,11-Dioxy-methylen-deserpidsäure-lacton wird aus Methanol in Nadeln vom Smp. 310-313 und [a]D = + 50 (c = 0,3 in Pyridin) erhalten.
b) Hydroxy-ester: 0,2 g (-)-10,11-Dioxy-methy- len-deserpidsäure-lacton werden unter Wasseraus schluss 11/2 Stunden in einer verdünnten Natrium- methylatlösung (hergestellt durch Auflösen von 28 mg Natrium in 10 cm-- wasserfreiem Methanol) am Rückfluss gekocht. Nach dem Erkalten wird die klare Lösung mit verdünnter Salzsäure genau neutra lisiert und im Vakuum zur Trockne verdampft.
Den Rückstand verteilt man zwischen Methylenchlorid und verdünnter Ammoniaklösung. Aus dem amor phen Rückstand kristallisiert der Hydroxy-ester in Nadeln vom Smp. 222-224 . [a]20 = -118 (c = 0,3 in Pyridin).
c) (-)-10,11-Dioxy-methylen-deserpidin: 0,2 g (-)-10,11-Dioxy-methylen-deserpidsäure-lacton wer den wie unter b) beschrieben in den Hydroxy-ester übergeführt, dieser direkt in amorpher Form in 2 cm3 Pyridin gelöst, die Lösung mit 0,23 g 3,4,5- Trimethoxybenzoylchlorid versetzt und der Ansatz 2 Stunden bei Zimmertemperatur stehengelassen.
Hierauf versetzt man mit 0,5 cm- Wasser, lässt 1/, Stunde bei Zimmertemperatur stehen, spült mit Methylenchlorid in einen Scheidetrichter, schüt telt nacheinander mit verdünnter Salzsäure, verdünn tem Ammoniak und Wasser aus, trocknet über Na triumsulfat und verdampft das Lösungsmittel im Vakuum.
Aus dem Rückstand kristallisiert= -118 Dioxy-methylen-deserpidin aus Methanol in Nadeln vom Smp. 251-253 . [a]D = -164 (c = 0,2 in Pyridin). <I>Beispiel 2</I> (+)-10,11-Dioxy-methylen-deserpidin a) Isomerisierung: 10,0 g (+)-10,11-Dioxy-me- thylen-isodeserpidsäure-lacton werden wie im 1. Bei spiel unter a) beschrieben mit Pivalinsäure,fXylol be handelt.
(+) -10,11-Dioxy-methylen-deserpidsäure- lacton kristallisiert aus Methanol in Nadeln vom Smp. 308-310 , [a]20 = -52 (c = 0,3 in Pyri- din).
b) (+)-10,11-Dioxy-methylen-deserpidin: 0,35 g (+)-10,11-Dioxy-methylen-deserpidsäure-lacton wer den wie im 1. Beispiel unter c) beschrieben in den entsprechenden Methylester und dieser direkt in das 3,4,5-Trimethoxybenzoat übergeführt.
Die Verbin dung kristallisiert aus Methanol in Nadeln vom Smp. 252-254 . [a]20 = -r 158 (c @ 0,2 in Pyridin). <I>Beispiel 3</I> 11-Äthoxy-deserpidin a) Isomerisierung: 0,62 g 11-Athoxy-isodeserpid- säure-lacton (Formel I: R3 = OCH.,CH3, R1, R2, R4 und R6 = H, R5 = CH3) werden nach der im 1.
Beispiel unter a) gegebenen Vorschrift mit Pivalin- säure@Xylol behandelt. 11-Äthoxy-deserpidsäure- lacton kristallisiert aus Methanol in Nadeln vom Smp. 321-323 [a]20 = + 92 c = 0,2 in Pyridin).
b) 11-Äthoxy-deserpidin: 0,4 g 11-Äthoxy-de- serpidsäure-lacton werden wie im 1. Beispiel unter c) beschrieben in den Methylester und dieser direkt in das 3,4,5 - Trimethoxy - benzoat übergeführt.
11-Äthoxy-deserpidin kristallisiert aus Methanol in Nadeln vom Smp. 282-283 , [a]20 = -158 (c = 0,2 in Pyridin). <I>Beispiel 4</I> 10-Benzyloxy-deserpidin a) Isomerisierung: 2,92 g 10-Benzyloxy-iso- deserpidsäure-lacton (Formel 1: R" = OCH2 * COHS, R1, R3, R4 und R, = H, R5 =<B>CH.)</B> werden wie im 1.
Beispiel unter a) beschrieben mit Pivalinsäure- Xylol behandelt und aufgearbeitet. Die chromato- graphische Trennung ergibt 10-Benzyloxy-deserpid- säure-lacton, das aus Methanol in Nadeln vom Smp. 260-265 , [a]20 = + 50" (c = 0,2 in Pyridin) er halten wird. b) 11-Benzyloxy-deserpidin: 0,464 g 11-Benzyl- oxy-deserpidsäure-lacton werden wie im 1.
Beispiel unter c) beschrieben in den Methylester und dieser direkt in das 3,4,5-Trimethoxy-benzoat übergeführt. Nach Reinigung über das Oxalat kristallisiert das 11-Benzyloxy-deserpidin aus Aceton/Wasser in Na deln vom Smp. 144-145 , [a120 = -149 .
<I>Beispiel 5</I> 11-Benzyloxy-deserpidin a) Isomerisierung: 0,567 g 11-Benzyloxy-iso- deserpidsäure-lacton (Formel I: R3 = OCH2, C6H5, R1, R., R4 und R, = H, R5 =<B>CH.)</B> werden wie im 1.
Beispiel unter a) beschrieben mit Pivalinsäurel Xylol behandelt und aufgearbeitet. 11-Benzyloxy- deserpidsäure-lacton kristallisiert aus MethanoljÄther in Prismen vom Smp. 271-272 , [a]20 = + 69 (c = 0,2 in Pyridin).
b) 11-Benzyloxy-deserpidin: 0,464 g 11-Benzyl- oxy-deserpidsäure-lacton werden wie im 1. Beispiel unter c) beschrieben in den Methylester und dieser direkt in das 3,4,5-Trimethoxy-benzoat übergeführt. Nach Reinigung über das Oxalat kristallisiert das 11-Benzyloxy-deserpidin aus Aceton/Wasser in Na deln vom Smp. 144-145 , 1a]20 = -149 .
<I>Beispiel 6</I> 10-Methoxy-deserpidin a) Isomerisierung: 0,765 g 10 - Methoxy-iso- deserpidsäure-lacton (Formel I: R2 = OCHS, R1, R3, R4 und Rs = H, R5 =<B>CH.)</B> werden wie im 1. Beispiel unter a) beschrieben mit Pivalinsäure/Xylol behandelt und aufgearbeitet.
Die chromatographische Trennung ergibt 10-Methoxy-deserpidsäure-lacton, das aus Methanol in Nadeln vom Smp. 283-284 , [a]20 = + 60 (c = 0,2 in Pyridin) erhalten wird.
b) 10-Methoxy-deserpidin: 0,475 g 10 Methoxy- deserpidsäure-lacton werden nach dem im 1. Beispiel unter c) beschriebenen Verfahren in den Methylester und dieser direkt in das 3,4,5-Trimethoxy-benzoat übergeführt. Die Verbindung kristallisiert aus Me thanol in Nadeln vom Smp. 160-161 , [a]20 = -161 (c = 0,3 in Pyridin).
<I>Beispiel 7</I> 11-Propoxy-deserpidin a) Isomerisierung: 1,55 g 11 - Propoxy - iso- deserpidsäure-lacton der Formel 1I, in welcher R1, R2, R4 und R, = H, R3 = OC3H7 und R5 = CH3, werden in 15 cm3 einer Lösung von Xylol-Pivalin- säure (8 :
2) 15 Stunden in einer Stickstoffatmosphäre am Rückfluss erhitzt, dann wird die Lösung im Vakuum zur Trockne verdampft und der Rückstand zwischen Methylenchlorid und verd. Ammoniak lösung verteilt. Aus dem Rückstand der Methylen- chloridlösung kristallisiert das 11-Propoxy-deserpid- säure-lacton aus Methanol in Prismen vom Smp. 286 bis 290 . [a] D = + 70 (c = 0,2 in Pyridin).
<I>b)</I> 11-Propoxy-deserpidin: 0,78 g des unter<I>a)</I> beschriebenen 11-Propoxy-deserpidsäure-lactons wer den unter Wasserausschluss in einer verd. Natrium methylatlösung (hergestellt durch Auflösen von 125 mg Natrium in 40 cm3 Methanol) am Rück fluss erhitzt. Nach dem Erkalten wird die klare Lö sung mit Salzsäure genau neutralisiert und im Va kuum zur Trockne verdampft.
Man verteilt darauf den Rückstand zwischen Methylenchlorid und verd. Ammoniaklösung. Man löst den Rückstand der Me- thylenchloridlösung in 10 cm3 Pyridin, versetzt die Lösung mit 0,95 g 3,4,5-Trimethoxy-benzoylehlorid und lässt die Lösung 2 Stunden bei Zimmertempera- tur stehen.
Hierauf versetzt man mit 2 cm3 Wasser, lässt 1/2 Stunde bei Zimmertemperatur stehen, spült mit Methylenchlorid in einen Scheidetrichter, schüt telt nacheinander mit verd. Salzsäure, verd. Ammo niak und Wasser aus, trocknet über Natriumsulfat und verdampft das Lösungsmittel im Vakuum. Aus dem Rückstand kristallisiert 11-Propoxy-deserpidin aus Methanol in Prismen vom Smp. 215-217 . [a]2() = -160 (c = 0,2 in Pyridin).
<I>Beispiel 8</I> 11-Isopropoxy-deserpidin a) Isomerisierung: 1,1 g amorphes 11-Iso- propoxy-isodeserpidsäure-lacton der Formel 1I, in welcher R1, R2, R4 und Rs = H, R3 = OCH(CH3)2 und R5 =<B>CH.,</B> werden in 10 cm3 einer Lösung von Xylol-Pivalinsäure (8 :
2) 15 Stunden in einer Stick stoffatmosphäre zum Sieden erhitzt und wie im Bei- spiel 1 unter a) angegeben aufgearbeitet. 11-Iso- propoxy-deserpidsäure-lacton kristallisiert aus Me thanol in Nadeln vom Smp. 307-309 . [a]20 = + 68 (c = 0,2 in Pyridin).
b) 11-Isopropoxy-deserpidin: 0,63 g 11-Iso- propoxy-deserpidsäure-lacton werden wie in Bei spiel 1 unter b) angegeben in den entsprechenden Methylester und dieser direkt in das 3,4,5-Trimeth- oxy-benzoat übergeführt. Die Verbindung kristalli- siert aus Alkohol in Nadeln vom Smp. 255-258 .
[a]20 = -155 (c = 0,2 in Pyridin). <I>Beispiel 9</I> 9-Methoxy-deserpidin a) Isomerisierung: 0,87 g 9-Methoxy-isodeserpid- säure-lacton der Formel 1I, in welcher R1 = OCHS, R2, R3, R4 und R6 = H, R5 =<B>CH.,</B> werden wie in Beispiel 1 unter a)
angegeben mit einer Lösung von Xylol-Pivalinsäure (9 : 1) behandelt und aufgearbei tet. 9-Methoxy-deserpidsäure-lacton kristallisiert aus Methanol in Nadeln vom Smp. 292-295 [apD = 57 (c = 0,2 in Pyridin).
b) 9-Methoxy-deserpidin: 0,2 g 9-Methoxy- deserpidsäure-lacton werden wie in Beispiel 1 unter b) beschrieben in den Methlyester und dieser direkt in das 3,4,5-Trimethoxy-benzoat übergeführt.
Da die Verbindung als freie Base nicht klar kristallisiert werden konnte, wurde sie in das Oxalat übergeführt, welches aus verd. Alkohol in Nadeln vom Smp. 203 bis 205 (sint. ab 180 ) kristallisiert. [a]20 = -133 (c = 0,2 in Pyridin).
<I>Beispiel 10</I> 11-Butoxy-deserpidin a) Isomerisierung: 1,02 g 11-Butoxy-isodeserpid- säure-lacton der Formel 1I, in welcher R1, R2, R4 und R, = H, R3 = OC4H9, R5 = CH.., werden wie in Beispiel 1 unter a) angegeben mit l0o/oiger Piva- linsäure in Xylol behandelt und aufgearbeitet.
11 Butoxy-desärpidsäure-lacton kristallisiert aus Metha nol in Prismen vom Smp. 259-260 . [a120 = + 49 (c = 0,2 in Pyridin).
b) 11-Butoxy-deserpidin: 0,85 g 11- Butoxy- deserpidsäure-lacton werden wie in Beispiel 1 unter b) angegeben in den Methylester und dieser direkt in das 3,4,5-Trimethoxy-benzoat übergeführt. Die Verbindung kristallisiert aus Essigester-Hexan in Prismen vom Smp. 203-204 . [a] 20 = -162 (c = 0,2 in Pyridin).

Reserpine, an alkaloid of the Rauwolfia species, has achieved great therapeutic importance in recent years, as it allows a successful treatment of hypertension and mental disorders due to its blood pressure-lowering and psychologically calming properties. After all, this alkaloid often has side effects, such as Sometimes they are uncomfortable and can sometimes force the treatment to be interrupted.
Mental depression, fatigue, gastric hyperacidity, diarrhea, dyspnea or asthma, edema (antidiuretic effect), sometimes peptic ulcers, parkinsonism, grand mal seizures occur most frequently [H. A. Schroeder et al. H.

Med. <I> J.1955, </I> 11, 1378 <B> 1 </B> It was therefore desirable to be able to have similarly acting compounds which, while retaining the therapeutically valuable properties, reduce the side effects of reserpine would not show or show only to a lesser extent.
It has now been shown that the previously unknown compounds obtainable according to the invention have valuable therapeutic properties. In particular, they have antihypertensive and psychologically calming properties, with individual compounds exceeding the effectiveness of reserpin. In addition, they are particularly well tolerated and have a long-lasting effect. The new compounds are u. a. for the treatment of hypertension and for calming states of mental excitement or anxiety of neurotic or psychotic origin.
They are intended to be used in therapy.
The process is carried out, for example, in such a way that a substituted iso-deserpidic acid lactone of the formula II (manufactured in Switzerland. Pat. No. 361811) is dissolved in xylene and boiled with pivalic acid in a nitrogen atmosphere. The resulting sub-substituted deserpidic acid lactone is carried out with methanol in the presence of catalytically effective amounts of alkali methylate in the corresponding hydroxy ester and this z.
B. esterified in pyridine with 3,4,5-trimethoxy-benzoyl chloride.
The compounds produced according to the process are crystalline at normal temperature. In the following examples, all temperature information is given in degrees Celsius. The melting points are corrected.
<I> Example 1 </I> (-) - 10,11-Dioxy-methylene-deserpidine a) Isomerization: 0.970 g (-) - 10,11-Dioxymethylene-isodeserpidic acid-lactone (formula I: R2 and R3 = -O-CH # -O-, R1, R4 and RG = H, R5 = CH3 - made in Switzerland. Patent No. <B> 361811) </B> are pivalic acid 9 in a 20 cm solution of xylene) : 1 boiled under reflux in a nitrogen atmosphere for 16 hours.
After this time, the crystallized precipitate formed is filtered off, the filtrate is evaporated to dryness in vacuo and the residue is partitioned between dilute ammonia solution and methylene chloride. The residue of the methylene chloride solution yields a further amount of substance in the form of needles from methanol, which is combined with the directly separated crystals and recrystallized from methanol.
(-) - 10,11-Dioxymethylene-deserpidic acid-lactone is obtained from methanol in needles with a melting point of 310-313 and [a] D = + 50 (c = 0.3 in pyridine).
b) Hydroxy ester: 0.2 g of (-) - 10,11-dioxy-methylene-deserpidic acid-lactone are mixed with water for 11/2 hours in a dilute sodium methylate solution (prepared by dissolving 28 mg of sodium in 10 cm-- anhydrous methanol) is refluxed. After cooling, the clear solution is exactly neutralized with dilute hydrochloric acid and evaporated to dryness in a vacuum.
The residue is partitioned between methylene chloride and dilute ammonia solution. The hydroxy ester crystallizes from the amorphous residue in needles with a melting point of 222-224. [a] 20 = -118 (c = 0.3 in pyridine).
c) (-) - 10,11-Dioxy-methylene-deserpidine: 0.2 g (-) - 10,11-Dioxy-methylen-deserpidic acid-lactone who are converted into the hydroxy ester as described under b), this dissolved directly in amorphous form in 2 cm3 of pyridine, 0.23 g of 3,4,5-trimethoxybenzoyl chloride were added to the solution and the mixture was left to stand for 2 hours at room temperature.
Then it is mixed with 0.5 cm water, allowed to stand for 1 /, hour at room temperature, rinsed with methylene chloride in a separating funnel, shaken out successively with dilute hydrochloric acid, dilute ammonia and water, dried over sodium sulfate and the solvent evaporated in the Vacuum.
= -118 Dioxymethylene-deserpidine crystallizes from the residue from methanol in needles with a melting point of 251-253. The residue is then partitioned between methylene chloride and dilute ammonia solution. The residue of the methylene chloride solution is dissolved in 10 cm3 of pyridine, 0.95 g of 3,4,5-trimethoxy-benzoylechloride are added to the solution and the solution is left to stand for 2 hours at room temperature.
Then add 2 cm3 of water, leave to stand for 1/2 hour at room temperature, rinse with methylene chloride in a separating funnel, shake out one after the other with dilute hydrochloric acid, dilute ammonia and water, dry over sodium sulfate and evaporate the solvent in vacuo. 11-Propoxy-deserpidine crystallizes from the residue from methanol in prisms with a melting point of 215-217. Since the compound could not be clearly crystallized as a free base, it was converted into the oxalate, which crystallizes from dilute alcohol in needles with a melting point of 203 to 205 (sint. From 180). [a] 20 = -133 (c = 0.2 in pyridine).
