worin X und Y Wasserstoff, Halogen oder nieder molekulare Alkylreste, und R1 und R, niedermole kulare Alkylreste, welche auch unter sich direkt oder über ein Sauerstoffatom verbunden sein können, be deuten, sowie ihre Salze und quatemären Ammonium verbindungen sind bisher nicht bekanntgeworden.
Es wurde nun gefunden, dass diese Verbindungen pharmakologisch wertvolle Eigenschaften, insbeson- dere spasmolytische, antiallergische und hypnotische Wirksamkeit, besitzen. Sie können überdies zur Poten zierung der Wirkung von Arzneimitteln sowie zur Be handlung von Geisteskrankheiten Verwendung finden.
Das erfindungsgemässe Verfahren zur Herstellung von Verbindungen der Formel 1 ist nun dadurch ge kennzeichnet, dass man ein 5-(2',3'-Epoxy-propyl)- azepin der Formel

umsetzt. Die erhaltenen Reaktionsprodukte kann man gewünschtenfalls durch Behandlung mit anorgani schen oder organischen Säuren in ihre Salze,
oder durch Umsetzung mit reaktionsfähigen Estern von aliphatischen oder araliphatischen Alkoholen in qua- ternäre Ammoniumverbindungen überführen.
Die Umsetzungen der Epoxypropylverbindungen mit den sekundären Aminen können in An- oder Ab wesenheit von Lösungs- oder Verdünnungsmitteln durchgeführt werden. Man kann als Verdünnungsmit tel beispielsweise einen überschuss an dem umzuset zenden sekundären Amin verwenden und die Reak tion in diesem Falle z. B. bei der Siedetemperatur des Amins unter Normaldruck durchführen. Ferner kann man die Reaktion auch durch Erwärmen der Komponenten in einem inerten Lösungsmittel, wie z. B. Benzol, Toluol oder Xylol, ferner Äthanol oder Methanol, ausführen.
Insbesondere bei Verwendung leichtflüchtiger Amine, wie z. B. Dimethylamin, kommt auch die Umsetzung der Komponenten in einem geeigneten inerten Lösungsmittel im Auto klaven in Betracht.
Die Ausgangsstoffe der Formel II können aus dem 5-Dibenzo[b,f]azepin, welches im folgenden als Iminostilben bezeichnet wird, oder dessen C-Substi- tutionsprodukten durch Umsetzung ihrer Alkalimetall- v erbindungen, insbesondere ihrer Natriumverbindun- gen, mit Epichlorhydrin erhalten werden.
Das Imino- stilben und seine C-Substitutionsprodukte sind ihrer seits aus dem 10,11-Dihydro-5-dibenzo[b,f]azepin (Iminodibenzyl) bzw. dessen C-Substitutionsproduk- ten durch N-Acylierung, Bromierung in 10-Stellung mittels Bromsuccinimid und aufeinanderfolgende oder gleichzeitige Bromwasserstoffabspaltung und Hydro lyse, z.
B. mittels Alkalilaugen, erhältlich. Geeignete Ausgangsstoffe der Formel II sind z. B. das 5-(2',3'- Epoxy-propyl)-iminostilben, das 5-(2',3'-Epoxy-pro- pyl)-3,7-dichlor-iminostilben und das 5-(2',3'-Epoxy- propyl)-3,7-dimethyl-iminostilben. Diese Epoxypro- pyl-Verbindungen können beispielsweise mit Dime- thylamin, Methyl-äthylamin,
Diäthylamin, Di-n-pro- pylamin, Methyl-n-propylamin, Methyl-isopropylamin, Äthyl-n-propylamin, Di-n-butylamin, Methyl-n-butyl- amin, Di-iso-butylamin, Pyrrolidin, Piperidin oder Morpholin zur Reaktion gebracht werden.
Mit anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phos phorsäure, Methansulfonsäure, Äthandisulfonsäure, Essigsäure, Bernsteinsäure, Fumarsäure, Maleinsäure, Apfelsäure, Weinsäure, Citronensäure, Benzoesäure und Phthalsäure bilden die tertiären Basen, wie schon gesagt, Salze, welche zum Teil wasserlöslich sind.
Durch Umsetzung mit reaktionsfähigen Estern, insbesondere Halogeniden oder Sulfaten von alipha- tischen oder araliphatischen Alkoholen, z.
B. von Methyljodid, Dimethylsulfat, Äthylbromid, Äthyl- jodid, DiäthyIsulfat, n-Propylbromid, n-Butylbromid, Allylbromid, Allyljodid, Benzylchlorid, Benzylbromid oder p-Chlor-benzylchlorid kann man, wie oben schon erwähnt,
aus den tertiären Aminen der Formel I monoquaternäre Ammoniumverbindungen herstellen, wobei das aliphatisch gebundene Stickstoffatom reagiert.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile; diese verhalten sich zu Volumteilen wie g zu em3. Die Temperaturen sind in Celsiusgraden angegeben.
<I>Beispiel 1</I> 38,6 Teile Iminostilben werden in 400 Volum- teilen abs. Benzol unter Erwärmen gelöst und 20 Teile Epichlorhydrin zugegeben. Hierauf lässt man unter starkem Rühren bei 50-60 innerhalb einer halben Stunde eine Suspension von 8 Teilen Natriumamid in Toluol zutropfen. Anschliessend wird das Reak tionsgemisch 2 Stunden unter Rückfluss gekocht, hierauf abgekühlt und im Vakuum von den Lösungs mitteln befreit.
Der Rückstand wird mit Wasser ver setzt und mit Äther erschöpfend extrahiert. Die ätherische Lösung wird gründlich mit Wasser ge waschen, mit Natriumsulfat getrocknet und einge dampft. Bei der Destillation des Rückstandes im Hochvakuum aus einem Hickmann-Kolben geht das 5-(2',3'-Epoxy-propyl)-iminostilben unter 0,02 mm Druck bei l50-160 über.
24,9 Teile 5-(2',3'-Epoxy-propyl)-iminostilben werden mit 75 Teilen reinem Diäthylamin 16 Stunden unter Rückfluss gekocht. Anschliessend wird der LTber- schuss an Diäthylamin im Vakuum abdestilliert und der Rückstand in Äther aufgenommen.
Der ätheri schen Lösung werden die basischen Anteile mit 30 % iger Essigsäure entzogen, die essigsaure Lösung wird mittels konz. Natronlauge alkalisch gestellt und das ausgeschiedene<B>öl</B> in Petroläther aufgenommen.
Nach dem Trocknen engt man diese Lösung auf ein kleines Volumen ein, wobei das 5-(3'-Diäthylamino- 2'-hydroxy-propyl)-iminostilben vom Schmelzpunkt l05-106 auskristallisiert.
<I>Beispiel 2</I> 24,9 Teile des nach Beispiel 1 erhaltenen 5-(2',3'- Epoxy-propyl)-iminostilbens werden mit 50 Volum- teilen Piperidin 16 Stunden unter Rückfluss gekocht. Nach dem Abkochen wird das Reaktionsgemisch in Wasser gegossen und erschöpfend mit Äther extra hiert. Die Ätherlösung wird mit 2n Salzsäure ausge zogen, und die darin übergegangenen basischen An teile werden durch Alkalischstellen mittels konz. Na tronlauge wiederum ausgefällt und in Äther aufge nommen.
Die Ätherlösung wird über Natriumsulfat getrocknet und eingedampft. Bei der Destillation des Rückstandes im Hochvakuum erhält man das 5-(3'-Piperidino-2'-hydroxy-propyl)-iminostilben vom Kp.0.092 186-188 .
In analoger Weise erhält man unter Verwendung von 50 Teilen Pyrrolidin anstelle des Piperidins das 5-(3'-Pyrrolidino-2'-hydroxy-propyl)-iminostilben vom Kp.o.002 185<B>0</B>.
