Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen der Formel

worin Rt Wasserstoff, eine Alkyl-, Cycloalkyl-, Aryloder Aralkylgruppe und R2 eine Cyan-, Carboxyl-, Carbamid-, N-Hydroxycarbamid- oder Carbalkoxygruppe darstellen, welches dadurch gekennzeichnet ist, dass man Verbindungen der Formel

mit flüssigem Ammoniak umsetzt. Das Reaktionsprodukt kann gegebenenfalls noch mit einer Säure in ein Salz überführt und verestert werden.
Zur Verhinderung der Polymerisation können dem Reaktionsgemisch vorzugsweise Polymerisationsinhibitoren zugegeben werden wie z. B. Hydrochinon, p-Phenylendiamin, Diphenylamin, Phenyl-fl-naphthyl amin. Wenn in der Formel II das Symbol R2 eine Carbomethoxygruppe darstellt, dann erhält man zur Hauptsache das Säureamid. Bei den höher homologen Estern, wie z. B. dem Äthyl-, Propyl- oder Butylester, bleibt die Estergruppe zum grössten Teil erhalten.
Als Ausgangsverbindungen der Formel II werden vorzugsweise solche verwendet, in denen R1 Wasserstoff, Methyl, Äthyl, Phenyl oder p-Nitrophenyl bedeutet.
Als leicht zugängliche Stoffe zur Herstellung der Ausgangsverbindungen der Formel II können a"S-Di- chlorverbindungen der Formel

verwendet werden. Bei der Durchführung des erfindungsgemässen Verfahrens kann man direkt von diesen Verbindungen ausgehen, da sie bei der Zugabe von Ammoniak vorerst in die entsprechenden Verbindungen der Formel II übergeführt werden, die dann bei Fortsetzung der Reaktion zu den Verbindungen der Formel I führen.
Die erfindungsgemäss erhältlichen Verbindungen sollen als Zwischenprodukte für die Herstellung von Arzneimitteln, insbesondere für die Herstellung von a-Amino-ss-hydroxycarbonsäuren, 4-Amino-isoxazolidinon und seinen 5-Substitutionsprodukten verwendet werden.
Beispiel 1
78 g a"B-Dichlor-propionsäuremethylester werden langsam in einen Liter flüssiges Ammoniak eingetropft. Das Reaktionsgemisch wird während 3 Stunden gerührt und darauf zur Trockne eingedampft.
Der auf 0o C gekühlte Rückstand wird mit 150 cm3 vorgekühlter 30% iger alkoholischer Salzsäure versetzt.
Nach 15stündigem Stehen wird das Reaktionsgemisch unter Rühren mit 250 cm3 absolutem Alkohol versetzt und anschliessend filtriert. Der Rückstand wird einige Male mit siedendem absolutem Alkohol ausgezogen. Aus den vereinigten Filtraten erhält man durch Eindampfen das a-Chlor-ss < amino-propion- säureamid-hydrochlorid, das aus Alkohol umkristallisiert wird. Schmelzpunkt 145-146" C.
32 g a - Chlor-fl- amino-propionsäureamid-hydro- chlorid werden während 4 Stunden in 300 cm3 absolutem Methanol unter Einleitung von Chlorwasserstoff am Rückfluss gekocht. Darauf engt man die Lösung auf 150 cm3 ein und filtriert das abgeschiedene Ammoniumchlorid ab. Das Filtrat wird zur Trockne eingedampft, in 35 cm3 absolutem Methanol aufgenommen, die Mischung filtriert und das Filtrat mit 300 cm3 absolutem Äther versetzt und auf -15"C abgekühlt. Man erhält dann das kristalline a-Chlor-B-amino-propions äuremethylester-hyd'rochlo- rid, welches nach nochmaliger Umkristallisation bei 101-103"C schmilzt.
Aus dem a,fl-Dichlor-propionsäureäthylester erhält man durch Umsetzung mit flüssigem Ammoniak gemäss den Angaben im ersten Absatz das Hydrochlorid des a-Chlor-18-amino-propions äureäthylesters vom Schmelzpunkt 104-106". Wenn man vom a,fl- Dichlor - propionsäure - isopropylester ausgeht, erhält man in analoger Weise das Hydrochlorid des a Chlor-B-amino- propions äureisopropylesters vom Schmelzpunkt 91-92".
Beispiel 2
Zu einem Liter flüssigem Ammoniak gibt man unter Rühren 41 g a-Chlor-acrylsäuremethylester und lässt über Nacht stehen. Darauf wird zur Trockne eingedampft. Unter guter Eiskühlung fügt man 90 cm3 30%ige alkoholische Salzsäure zu und rührt während etwa 30 Minuten weiter. Der gebildete kristalline Rückstand wird abfiltriert und aus Alkohol Äther umkristallisiert, wobei das rohe a-Chlor-, B- amino-propionsäureamid-hydrochlorid erhalten wird.
Wenn man a-Chlor-acrylsäureäthylester in analoger Weise mit flüssigem Ammoniak umsetzt, so erhält man a-chlor-, B-amino-propionsäureäthylester.
Aus a-chlor-acrylsäureisopropylester erhält man in analoger Weise den a-Chlor-g-amino-propionsäure- isopropylester.
