Verfahren zur Herstellung p-substituierter Benzolsulfonamide
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer psubstituierter Benzolsulfonamide der Formel I

Das erfindungsgemässe Verfahren zur Herstellung von Verbindungen der Formel list dadurch gekennzeichnet, dass man eine Sulfamylcarbonsäure der Formel II

oder ein Halogenid einer solchen mit einer Hydroxylverbindung, einer Mercaptoverbindung, mit Ammoniak oder einem primären oder sekundären Amin oder einem Hydrazin, das mindestens ein H-Atom aufweist, kondensiert. So kann man z. B. Sulfamyldiphenyläther-carbonsäuren der Formel II in an sich bekannter Weise in ihre Halogenide überführen und diese mit Hydroxylverbindungen, Mercaptoverbindungen, Ammoniak, Aminen oder Hydrazinen umsetzen.
Als Hydroxylverbindungen kommen z. B. aliphatische, cycloaliphatische, araliphatische und heterocyclische Alkohole wie Methanol, Äthanol, Propanol, iso-Propanol, Butanol, iso-Butanol, sek.-Butanol, Amylalkohol, iso-Amylalkohol, n-Hexanol, n-Octanol, 2 Äthylhexanol, Allylalkohol, Cyclohexanol, Methylcyclohexanole, Benzylalkohol, a- und ss-Phenyl-äthanol, Zimtalkohol, Furfurylalkohol, Tetrahydrofurfurylalkohol, a-Hydroxy-tetrahydropyran, Acetonglyzerin, 1-Alkyl-4-hydroxy-piperidine, fl-Pyridyl-carbinol;
sowie Verbindungen mit weiteren freien oder gebundenen funktionellen Gruppen wie Methoxyäthanol, Äthoxyäthanol, Äthylenglykol, Propylenglykol, Gly- zerm und deren Monoester, Glykolsäure und Milchsäure und deren Ester, Phenoxyäthanol, Salicylsäure und Mandelsäure und deren Ester, Dialkylaminoalkanole und Piperidinoalkanole in Frage; als Mercaptoverbindungen z. B.
Methylmercaptan, Äthyl- mercaptan und Allylmercaptan; und als Amine beispielsweise Mono- und Dialkylamine wie Methylamin, Äthylamin, Isopropylamin, n-Butylamin, Allylamin, Dimethylamin, Diäthylamin und Di-n-butylamin; im weiteren Monoäthanolamin und Diäthanolamin, ss,y-Dihydroxy-propylamin; überdies Cyclohexylamin, Diamine wie N,N,N'-Trimethyl-äthylendiamin, N,N,N'-Triäthyl-äthylendiamin; aromatische und araliphatische Amine wie Anilin, N-Alkylaniline, Toluidine, Anisidine, Chloraniline, Benzylamin, p-Phenyl- äthyl amin, N-Methyl-benzylamin;
heterocyclische Amine wie 2-Amino-4-methyl-thiazol, 2-Amino-pyridin, gesättigte N-haltige Heterocyclen wie Piperidin, Pyrrolidin, Morpholin, Thiomorpholin, l-Alkylpiperazine; Aminocarbonsäuren und deren Ester, wie Glykokoll, Alanin, Asparaginsäure, Glut aminsäure, Anthranilsäure, p-Aminobenzoesäure, Aminonicotinsäure und ihre Alkylester; Aminosulfonsäuren und deren Derivate wie ss-Amino-äthansulfonsäure, Sulfanilsäure und Sulfanilamid, ferner z. B. Aminoäther und Aminosulfone wie Athoxy- äthyl-amin, Phenoxyäthyl-amin und Methylsulfonyl äthyl-amin. Schliesslich kann man Carbonsäuren der Formel II oder ein Halogenid einer solchen auch mit Hydrazinen oder substituierten Hydrazinen wie z. B.
Methylhydrazin, Äthylhydrazin, iso-Propylhydrazin, N,N-Dimethylhydrazin, N,N,N'-Trimethyihydrazin, Acetylhydrazin, Semicarbazid, Carbäthoxyhydrazin, Acetonhydrazon, Cyclohexanonhydrazon und Benzaldehydhydrazon umsetzen.
Beispiel 1
29,4 Teile 4-Sulfamyl-diphenyläther-4'-carbon- säure (Smp. 2784) werden in 60 Volumteilen Thionylchlorid und 0,5 Teilen Pyridin 15 Stunden bei 40 bis 45O gehalten, wobei eine klare Lösung entsteht.
Diese wird unter kräftigem Rühren in Eis und Wasser getropft. Das sich zuerst schmierig abscheidende, dann erstarrende Säurechlorid wird filtriert, mit Eiswasser verrieben, wieder filtriert und unter Rühren in 100 Volumteile 25 0/oige wässrige Ammoniaklösung eingetragen. Nach zweistündigem Rühren bei 20-30" wird das überschüssige Ammoniak durch Einleiten von Wasserdampf vertrieben, der pulverige Niederschlag abfiltriert und zuerst aus Wasser, dann aus Alkohol umkristallisiert. Das so erhaltene 4-Sulf amyl-diphenyläther-4'-carbonsäureamid besitzt einen Schmelzpunkt von 218".
In gleicher Weise, jedoch unter Verwendung von Äthylamin bzw. Diäthylamin an Stelle von Ammoniak werden 4-Sulfamyldiphenyläther-4'-carbonsäure äthylamid (Schmelzpunkt 214 ) und 4-Sulfamyl diphenyläther-4'-carbonsäure-diäthylamid (Schmelzpunkt 201 ) hergestellt.
Beispiel 2
Das gemäss Beispiel 1 aus 29,4 Teilen Carbonsäure hergestellte 4-Sulfamyl-diphenyläther-4'-carbon- säurechlorid wird unter Rühren in eine eiskalte Lösung von 40 Teilen Diäthanolamin und 100 Teilen Wasser eingetragen. Hierauf wird das Gemisch auf 90O erhitzt und langsam erkalten gelassen. Das auskristallisierte 4-Sulfamyl-diphenyläther-4'-carbonsäure diäthanolamid wird zuerst aus Wasser, dann aus Nitromethan umkristallisiert und zeigt einen Schmelzpunkt von 159".
Beispiel 3
Das gemäss Beispiel 1 aus 29,4 Teilen Carbonsäure hergestellte 4-Sulfamyl-diphenyläther-4'-carbon- säurechlorid wird unter Rühren in eine mit 50 Teilen Eis versetzte Lösung von 45,5 Teilen Amino-essigsäureäthylester in 50 Teilen Wasser eingetragen.
Gleichzeitig wird die Mischung durch Zugabe von Natriumbicarbonat lackmusneutral gehalten. Nach 2stündigem Rühren bei 15 wird das abgeschiedene 4-Sulfamyl-diphenyläther-4'-carbonsäure-carbäthoxy- methylamid abfiltriert und mehrmals aus verdünntem Alkohol umkristallisiert. Schmelzpunkt 184 .
Beispiel 4
Das gemäss Beispiel 1 aus 29,4 Teilen Carbonsäure hergestellte 4-Sulfamyl-diphenyläther-4'-carbonsäurechlorid wird unter Rühren in eine Mischung von 40 Teilen Äthanolamin, 40 Teilen Dioxan und 100 Teilen Eis eingetragen. Das Carbonsäurechlorid geht in Lösung, und nach etwa 15 Minuten beginnt das 4-Sulfamyl-diphenyläther-4'-carbonsäureäthanolamid zu kristallisieren. Das Reaktionsgemisch wird nun auf 70O aufgeheizt, wieder erkalten gelassen und filtriert. Der Schmelzpunkt liegt bei 190-192", nach dem Umkristallisieren aus Wasser oder Alkohol bei 197".
4-Sulfamyl-diphenyläther-4'-carbonsäure - diäthyl- aminoäthylamid wird in gleicher Weise hergestellt, wobei Diäthylaminoäthylamin anstelle von Sithanol- amin verwendet wird. Der Schmelzpunkt des aus Benzol umkristallisierten Produktes liegt bei 169".
Beispiel 5
29,4 Teile 4-Sulfamyl-diphenyläther-4'-carbon- säure (Smp. 278O) werden mit 60 Volumteilen Thionylchlorid und 0,6 Volumteilen Pyridin 15 Stunden bei 40-45" gehalten. Das überschüssige Thionylchlorid wird hierauf im Vakuum abgedampft, der Rückstand mit Benzol versetzt und nochmals eingedampft. Das zurückbleibende kristallisierte 4-Sulf amyl-diphenyläther-4'-carbons äurechlorid wird in 70 Teilen Benzol angeschlämmt und rasch zu einem Gemisch von 150 Teilen Benzol und 70 Teilen Piperidin zugegeben. Die Mischung kocht auf, und das Piperidid scheidet sich in farblosen Kristallen ab.
Das Benzol wird mit Dampf abgeblasen und der Rückstand heiss filtriert. Das so erhaltene 4-Sulfamyldiphenyläther-4'-carbonsäure-piperidid schmilzt bei 214-217", nach dem Umkristallisieren aus Alkohol bei 2200.
Verwendet man statt Piperidin eine entsprechende Menge Morpholin, so erhält man in gleicher Weise das 4-Sulfamyl-diphenyläther -4' - carbonsäure - mor- pholid vom Schmelzpunkt 221 (aus Alkohol).
Beispiel 6
Das wie in Beispiel 5 aus 29,4 Teilen Carbonsäure hergestellte 4-Sulfamyl-diphenyläther.4'-carbon- säurechlorid wird in 160 Teilen Methanol gelöst und das überschüssige Methanol auf dem Wasserbad abdestilliert. Der-klebrige Rückstand wird beim Verreiben mit Wasser fest. Der noch unreine 4-Sulfamyl-diphenyläther-4'-carbonsäure-methylester vom Schmelzpunkt 97-108" wird durch Umkristallisieren aus Benzol gereinigt; Schmelzpunkt 137-138".
Verwendet man statt Methanol Isopropylalkohol, so erhält man bei sonst gleicher Arbeitsweise den 4 - Sulfamyl - diphenyläther - 4'- carbonsäure-isopropyl- ester vom Schmelzpunkt 1 34o (aus Benzol).
Beispiel 7
Das gemäss Beispiel 5 aus 29,4 Teilen Carbonsäure hergestellte 4-Sulfamyl-diphenyläther-4'-carbon- säurechlorid wird mit 50 Teilen Benzylalkohol und 50 Teilen Pyridin 1 Stunde auf 1000 erhitzt. Das Pyridin wird mit dem überschüssigen Benzylalkohol im Vakuum abgedampft und der ölige Rückstand durch Zugabe von Methanol und Wasser zur Kristallisation gebracht. Der so erhaltene 4-Sulfamyldiphenyläther-4'-carbonsäure-benzylester wird abfiltriert. Sein Schmelzpunkt liegt bei 125-127", der des aus Benzol umkristallisierten Produktes bei 128".
In einer zu den beschriebenen Beispielen analogen Weise können erhalten werden: 4 - Sulfamyl - diphenyläther - 2' - carbonsäureamid mit einem Schmelzpunkt von 2040 (Umkrist. aus Äthanol) aus der 4-Sulfamyl-diphenyläther-2' carbonsäure vom Schmelzpunkt 2340 ; 4- Sulfamyl - diphenylsulfid - 4'-carbonsäureamid vom
Schmelzpunkt 219-221" aus der 4-Sulfamyl diphenylsulfid-4'-carbonsäure vom Schmelzpunkt 240-242"; 4- Sulfamyl - diphenylsulfon - 4'-carbonsäureamid vom
Schmelzpunkt 2760 (umgelöst aus Aceton und
Wasser) aus 4-Sulfamyl-diphenylsulfon-4'-carbon säure vom Schmelzpunkt 3350 ;
sowie 4-Sulfamyl-diphenyläther-4'-carbonsäure- hydrazid vom Schmelzpunkt 2362380.

The 4-sulfamyl-diphenylether-4'-carboxylic acid chloride prepared according to Example 1 from 29.4 parts of carboxylic acid is introduced into an ice-cold solution of 40 parts of diethanolamine and 100 parts of water with stirring. Example 5
29.4 parts of 4-sulfamyl-diphenylether-4'-carboxylic acid (melting point 278O) are kept at 40-45 "for 15 hours with 60 parts by volume of thionyl chloride and 0.6 parts by volume of pyridine. The excess thionyl chloride is then evaporated off in vacuo, benzene is added to the residue and the residue is again evaporated. The remaining crystallized 4-sulfamyl-diphenylether-4'-carboxylic acid chloride is suspended in 70 parts of benzene and quickly added to a mixture of 150 parts of benzene and 70 parts of piperidine. and the piperidide separates out in colorless crystals.
The benzene is blown off with steam and the residue is filtered hot. The 4-sulfamyldiphenylether-4'-carboxylic acid piperidide thus obtained melts at 214-217 ", after recrystallization from alcohol at 2200.
If a corresponding amount of morpholine is used instead of piperidine, 4-sulfamyl-diphenyl ether -4'-carboxylic acid morpholide with a melting point of 221 is obtained in the same way (from alcohol).
Example 6
The 4-sulfamyl-diphenylether.4'-carbonic acid chloride prepared from 29.4 parts of carboxylic acid as in Example 5 is dissolved in 160 parts of methanol and the excess methanol is distilled off on a water bath. The sticky residue solidifies when rubbed with water. The still impure methyl 4-sulfamyl-diphenylether-4'-carboxylate with a melting point of 97-108 "is purified by recrystallization from benzene; melting point 137-138".
If isopropyl alcohol is used instead of methanol, the 4-sulfamyl-diphenyl ether-4'-carboxylic acid isopropyl ester with a melting point of 1,340 (from benzene) is obtained with otherwise the same procedure.
Example 7
The 4-sulfamyl-diphenyl ether-4'-carboxylic acid chloride prepared from 29.4 parts of carboxylic acid according to Example 5 is heated to 1000 for 1 hour with 50 parts of benzyl alcohol and 50 parts of pyridine. The pyridine is evaporated off in vacuo with the excess benzyl alcohol and the oily residue is crystallized by adding methanol and water. The 4-sulfamyldiphenyl ether-4'-carboxylic acid benzyl ester thus obtained is filtered off. Its melting point is 125-127 ", that of the product recrystallized from benzene is 128".
