Verfahren zur Herstellung von neuen Azepinen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Azepine, die wertvolle Aus gangsstoffe für die Synthese von Arzneimitteln dar stellen.
Das erfindungsgemässe Verfahren zur Herstellung von 5-Dibenzo[b,f]azepinen (welche auch als Imino- stilbene bezeichnet werden können) der Formel

in ein leicht spaltbares 5-Acylderivat überführt, auf dieses eine Halogen abgebende Verbindung, insbeson dere N-Brom-succinimid, einwirken lässt und das ent standene,
gegebenenfalls substituierte 5-Acyl-10-halo- gen-10,11-dihydro-5-dibenzo[b,f]azepin mit einem halogenwasserstoffabspaltenden Mittel behandelt und gleichzeitig oder anschliessend hydrolysiert.
Geeignete Ausgangsstoffe der Formel II sind neben dem Iminodibenzyl beispielsweise das 3,7-Di- chlor-iminodibenzyl, 2,8-Dichlor-, 1,9-Dichlor, 3,7- Dimethyl-, 2,8-Dimethyl- und 1,9-Dimethyl-imino- dibenzyl, welche aus den entsprechenden Diamino- dibenzylen durch Erhitzen ihrer Monohydrochloride oder Phosphate, oder durch Erhitzen der Basen mit Polyphosphorsäure erhältlich sind.
Zur Überführung in leicht spaltbare 5-Acylderivate kann man die ge nannten Iminodibenzyle beispielsweise mit Acetyl- chlorid, Chlorameisensäure-methylester oder -äthyl- ester unter Erwärmen umsetzen. Als Halogen ab gebende Verbindung eignet sich N-Brom-succinimid vorzüglich, ferner kommen beispielsweise N-Chlor- succinimid, N-Brom-phthalimid und N-Brom-acet- amid in Betracht.
Die Abspaltung von Halogenwas serstoff und die Hydrolyse in einem Arbeitsgang kann zum Beispiel mittels Alkalilauge in der Wärme durch geführt werden. Bei der Behandlung der 5-Acyl-10- halogen-iminodibenzyle mit einer tertiären organi schen Base, wie zum Beispiel Kollidin, in der Wärme oder beim Einwirkenlassen einer Alkalilauge in der Kälte erhält man zunächst die 5-Acyl-iminostilbene, deren Hydrolyse ebenfalls mittels Alkalilauge in der Wärme durchgeführt werden kann.
Die erfindungsgemäss herstellbaren Iminostilbene lassen sich in ihrer Iminogruppe in mannigfacher Weise substituieren. Beispielsweise erhält man Ver bindungen mit wertvollen antiallergischen und psy- choplegischen Eigenschaften, wie das 5-(ss-Dimethyl- amino-äthyl)-, 5-(f-Dimethylamino-propyl)-, 5-(y-Di- methylamino-propyl)- und 5-(ss-Pyrrolidino-äthyl)- iminostilben,
wenn man die aus Iminostilben und Natriumamid oder Lithiumamid erhältliche Natrium verbindung bzw. Lithiumverbindung mit den entspre chenden Dimethylaminoalkylhalogeniden bzw. mit ,B-Pyrrolidinoäthylhalogeniden umsetzt.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile, diese verhalten sich zu Volumteilen wie Gramm zu Kubikzentimeter. Die Temperaturen sind in Centigraden angegeben. <I>Beispiel 1</I> a) 100 Teile Iminodibenzyl werden mit 300 Tei len Acetylchlorid so lange unter Rückfluss gekocht, bis alles gelöst ist. Dann destilliert man das über schüssige Acetylchlorid ab und giesst den Rückstand in Benzin, wobei das 5-Acetyliminodibenzyl auskri stallisiert. Es wird aus wenig Alkohol auskristallisiert; F. 98 .
b) 45 Teile 5-Acetyl-iminodibenzyl und 33,8 Teile Bromsuccinimid' werden in 400 Volumteilen Tetrachlorkohlenstoff bei starker Beleuchtung unter Rühren und Rückfluss gekocht. Sobald das anfäng lich auf dem Boden des Kolbens liegende Bromsuc- cinimid verschwunden ist und auf der Flüssigkeit das Succinimid sich abgeschieden hat, wird abgekühlt und vom abgeschiedenen Succinimid abgesaugt.
Die Te- trachlorkohlenstofflösung wird mit Wasser gewaschen und das Lösungsmittel im Vakuum bei einer Bad temperatur von 60 abdestilliert. Das so erhaltene rohe 10-Brom-5-acetyliminodibenzyl wird mit der vierfachen Menge Kollidin während 5 Stunden auf 140-150 erwärmt. Das ausgeschiedene Kollidin- hydrobromid wird nach dieser Zeit abgesaugt und das Kollidin im Vakuum völlig abgedampft.
Der Rückstand wird in viel Äther aufgenommen, gründ lich mit 2-n. Salzsäure und Wasser gewaschen, ge trocknet und auf ein kleines Volumen eingeengt, wo bei das 5-Acetyl-iminostilben auskristallisiert. Es wird abgesaugt, aus viel Äther umkristallisiert; F. 117 bis 119 .
Zum 5-Acetyl-iminostilben gelangt man auch, wenn man 30 Teile des oben genannten rohen Bro- mierungsproduktes in 120 Volumteilen abs. Alkohol löst und mit 12 Teilen 50%iger Kalilauge versetzt. Die Kaliumbromidabscheidung beginnt sofort.
Man lässt 2 Stunden stehen, versetzt dann mit etwa 170 Teilen Wasser und rührt bis zur Kristallisation. Man saugt das 5-Acetyl-iminostilben ab, wäscht zuerst mit Wasser neutral und schlussendlich mit eisgekühltem Äther.
c) 5 Teile 5-Acetyl-iminostilben werden mit einer Lösung von 3 Teilen Ätzkali in 2 Teilen Wasser und 15 Volumteilen 96 /oigem Alkohol 18 Stunden unter Rückfluss gekocht. Hierauf giesst man die Lösung in Wasser, saugt die orangefarbigen Kristalle ab und kristallisiert aus viel Alkohol um. Das Iminostilben schmilzt bei 206-208 .
Zur gleichen Verbindung gelangt man, wenn man 5 Teile rohes Bromierungsprodukt mit 6 Teilen Atz- kali, 4 Teilen Wasser und 30 Volumteilen 96%igem Alkohol 18 Stunden unter Rühren zum Rückfluss er hitzt. Hierauf giesst man die Lösung in Wasser und saugt das Iminostilben ab.
<I>Beispiel 2</I> a) 57 Teile Iminodibenzyl werden in 400 Volum- teilen abs. Benzol gelöst und bei 60-70 wird eine Suspension von 12 Teilen Natriumamid in Toluol zugetropft. Nach dem Zutropfen hält man noch 2 Stunden unter Einleiten von Stickstoff im Sieden. Dann kühlt man auf 20 ab und tropft langsam 33 Teile Chlorameisensäureäthylester zu. Die Tempera tur steigt auf 30-35 .
Anschliessend wird das Reak tionsgemisch 5 Stunden unter Rückfluss gekocht, hier auf abgekühlt und mit Wasser zersetzt. Die benzo- lische Phase wird abgehoben, mit Wasser gründlich gewaschen, getrocknet und eingedampft. Der Rück stand wird aus Äther-Petroläther umkristallisiert. Das 5-Carbäthoxy-iminodibenzyl schmilzt bei 95 .
b) 27 Teile dieses 5-Acylderivates werden in 200 Volumteilen Tetrachlorkohlenstoff mit 18 Teilen Bromsuccinimid 4 Stunden bei Belichtung unter Rückfluss gekocht. Nach dieser Zeit wird abgekühlt und das ausgeschiedene Succinimid abgesaugt. Die Tetrachlorkohlenstofflösung wird mit Wasser gewa schen und im Vakuum bei einer Badtemperatur von 60 völlig eingedampft. Der Rückstand wird zur Rei nigung in 400 Volumteilen Benzin aufgenommen, fil triert und eingedampft.
Es hinterbleiben 45 Teile eines Öles, das das rohe 10-Brom-5-carbäthoxy- iminodibenzyl darstellt.
c) 24 Teile der rohen Bromverbindung werden in 60 Volumteilen Alkohol gelöst und mit 10 Teilen 50%iger Kalilauge versetzt, wobei die Temperatur auf 40 steigt. Man lässt eine Stunde bei Zimmertem peratur stehen, kühlt dann auf -5 ab und saugt die ausgeschiedenen Kristalle ab. Sie werden zuerst mit wenig Alkohol und dann mit Wasser ionenfrei ge waschen und getrocknet. Der Schmelzpunkt des 5- Carbäthoxy-iminostilbens liegt bei 126-127 .
d) 5 Teile 5-Carbäthoxy-iminostilben werden mit einer Lösung von 3 Teilen Ätzkali, 1 Teil Wasser und 15 Volumteilen Alkohol 20 Stunden unter Rückfluss gekocht. Man kühlt auf Zimmertempera tur ab, saugt das in Form von orangefarbenen Blätt chen anfallende Iminostilben ab und wäscht es gründ lich mit Wasser.
Zum Iminostilben gelangt man auch, wenn man 5 Teile der rohen Bromverbindung mit 10 Teilen Ätzkali, 2 Teilen Wasser und 30 Volumteilen Alko hol 20 Stunden unter Rückfluss kocht.
<I>Beispiel 3</I> a) 26 Teile 3,7-Dichlor-iminodibenzyl werden in 300 Volumteilen Benzol zum Sieden erhitzt und in nerhalb von zwei Stunden 110 Teile Acetylchlorid zugetropft. Anschliessend wird die Reaktionslösung noch 4 Stunden unter Rückfluss gekocht. Dann wird das Lösungsmittel und überschüssiges Acetylchlorid im Vakuum abgedampft und der Rückstand aus Alkohol umkristallisiert. Das 5-Acetyl-3,7-dichlor- iminodibenzyl schmilzt bei 147-148 .
b) 53 Teile 5-Acetyl-3,7-dichlor-iminodibenzyl und 37 Teile Bromsuccinimid werden in 500 Volum- teilen Tetrachlorkohlenstoff unter Belichtung so lange zum Sieden erwärmt, bis das Succinimid oben schwimmt, was etwa 21/2 Stunden in Anspruch nimmt. Man kühlt ab, saugt ab und dampft den Tetrachlor- kohlenstoff im Vakuum vollständig ab. Der Rückstand wird in 150 Volumteilen Alkohol Cr löst und mit 25 Teilen 50 /oiger Kalilauge versetzt.
Nach halbstündigem Stehen wird mit 150 Volumteilen Wasser versetzt, die Kristalle des 5-Acetyl-3,7-dichlor- iminostilbens werden abgesaugt, mit wenig kaltem Alkohol gewaschen und getrocknet; F. 184-185 .
c) 42,2 Teile 5-Acetyl-3,7-dichlor-iminostilben und 26 Teile Kaliumhydroxyd werden in 130 Volum- teilen abs. Alkohol 4 Stunden unter Rückfluss ge kocht. Nach dieser Zeit kühlt man ab und saugt das rohe 3,7-Dichlor-iminostilben ab. Man wäscht es zu erst mit Alkohol, dann gründlich mit Wasser und kristallisiert es aus Äther um; F. 282-284 .

The carbon tetrachloride solution is washed with water and the solvent is distilled off in vacuo at a bath temperature of 60 °. 5-Acetyl-iminostilbene is also obtained if 30 parts of the abovementioned crude brominated product are mixed in 120 parts by volume of abs. Alcohol dissolves and mixed with 12 parts of 50% potassium hydroxide solution. The potassium bromide deposition begins immediately.
The mixture is left to stand for 2 hours, then about 170 parts of water are added and the mixture is stirred until crystallization. The 5-acetyl-iminostilbene is suctioned off, washed first with water until neutral and finally with ice-cold ether.
c) 5 parts of 5-acetyl-iminostilbene are refluxed for 18 hours with a solution of 3 parts of caustic potash in 2 parts of water and 15 parts by volume of 96% alcohol. The solution is then poured into water, the orange-colored crystals are suctioned off and recrystallized from a lot of alcohol. The iminostilbene melts at 206-208.
The same compound is obtained if 5 parts of crude bromination product with 6 parts of caustic potash, 4 parts of water and 30 parts by volume of 96% alcohol are refluxed for 18 hours with stirring. The solution is then poured into water and the iminostilbene is suctioned off.
<I> Example 2 </I> a) 57 parts of iminodibenzyl are abs in 400 parts by volume. Benzene dissolved and at 60-70 a suspension of 12 parts of sodium amide in toluene is added dropwise. After the dropwise addition, the mixture is kept at the boil for a further 2 hours while passing in nitrogen. It is then cooled to 20 and 33 parts of ethyl chloroformate are slowly added dropwise. The temperature rises to 30-35.
The reaction mixture is then refluxed for 5 hours, cooled down here and decomposed with water. The benzol phase is lifted off, washed thoroughly with water, dried and evaporated. The residue is recrystallized from ether-petroleum ether. The 5-carbethoxy-iminodibenzyl melts at 95.
b) 27 parts of this 5-acyl derivative are refluxed in 200 parts by volume of carbon tetrachloride with 18 parts of bromosuccinimide for 4 hours with light exposure. After this time it is cooled and the succinimide which has separated out is suctioned off. The carbon tetrachloride solution is washed with water and completely evaporated in vacuo at a bath temperature of 60. For cleaning, the residue is taken up in 400 parts by volume of gasoline, filtered and evaporated.
There remain 45 parts of an oil which is the crude 10-bromo-5-carbäthoxy- iminodibenzyl.
c) 24 parts of the crude bromine compound are dissolved in 60 parts by volume of alcohol and 10 parts of 50% strength potassium hydroxide solution are added, the temperature rising to 40%. It is left to stand for one hour at room temperature, then cooled to -5 and the crystals which have separated out are filtered off with suction. They are first washed free of ions with a little alcohol and then with water and dried. The melting point of 5-carbethoxy-iminostilbene is 126-127.
d) 5 parts of 5-carbethoxy-iminostilbene are refluxed for 20 hours with a solution of 3 parts of caustic potash, 1 part of water and 15 parts by volume of alcohol. It is cooled to room temperature, the iminostilbene, which accumulates in the form of orange-colored leaves, is suctioned off and it is washed thoroughly with water.
Iminostilbene is also obtained if 5 parts of the crude bromine compound are refluxed with 10 parts of caustic potash, 2 parts of water and 30 parts by volume of alcohol for 20 hours.
<I> Example 3 </I> a) 26 parts of 3,7-dichloro-iminodibenzyl are heated to boiling in 300 parts by volume of benzene and 110 parts of acetyl chloride are added dropwise within two hours. The reaction solution is then refluxed for a further 4 hours. The solvent and excess acetyl chloride are then evaporated off in vacuo and the residue is recrystallized from alcohol. The 5-acetyl-3,7-dichloro-iminodibenzyl melts at 147-148.
b) 53 parts of 5-acetyl-3,7-dichloro-iminodibenzyl and 37 parts of bromosuccinimide are heated to boiling in 500 parts by volume of carbon tetrachloride with exposure until the succinimide floats on top, which takes about 21/2 hours. c) 42.2 parts of 5-acetyl-3,7-dichloro-iminostilbene and 26 parts of potassium hydroxide are abs in 130 parts by volume. Alcohol boiled under reflux for 4 hours.
