Procédé de préparation d'un nouvel antibiotique La présente invention a pour objet un procédé de préparation d'un nouvel antibiotique, par culture d'une souche de Streptomyces paucisporogenes dans un milieu aqueux contenant, à part des sels miné raux, une source d'azote et un hydrate de carbone, en aérobie jusqu'à ce qu'une activité substantielle se manifeste dans ledit milieu, puis extraction et puri fication de l'antibiotique.
Le Streptomyces paucisporogenes se trouve, en tre autres, dans la terre. On l'isole sous forme de culture pure conformément aux procédés courants. Il a été enregistré dans les bureaux de la titulaire sous le N T4915. Un échantillon de cette souche T 4915 a été déposé à la National Collection of Industrial Bacteria , The Commonwealth Mycolo- gical Institute, Ferry Lane, Kew (Surrey, Grande- Bretagne), sous le No NCIB 8790,
alors qu'un deuxième échantillon de la souche a été déposé à la American Type Culture Collection Washington D.C. (USA), sous le N ATCC 12596. Le Strepto myces paucisporogenes pousse difficilement sur les milieux de cultures solides habituellement utilisés. Il fournit des colonies composées de filaments dont la partie apicale, plus ou moins ramifiée, ne comporte qu'exceptionnellement des chaînes de conidies.
Sur milieux organiques gélosés complexes, le Streptomyces paucisporogenes est caractérisé mi- croscopiquement par un mycélium aérien blanc jau nâtre ; le feutrage du mycélium végétatif est épais, parfois craquelé. Il sécrète parfois une faible quan tité d'un pigment soluble brun jaune. Son dévelop pement est particulièrement bon sur un milieu com posé comme suit et ajusté à pH 7,5 : décoction dans un litre d'eau distillée de 200 g de graines de soja broyées, 20 g de glucose, 10 g de fécule de pomme de terre et 20 g de gélose.
Sur milieux chimiquement définis (Czapeck, Conn... etc.) la pousse est difficile : il y a toutefois solubilisation du malate de calcium.
Sur substrat naturel tel que la pomme de terre imprégnée de tampon à pH 7, le Streptomyces pau- cisporogenes se développe rapidement sous forme d'un enduit épais, cérébroïde, craquelé. Le mycé lium aérien est alors rare, blanc grisâtre à reflets jaunes.
Si la culture en surface du Streptomyces pau- cisporogenes est difficile, elle est par contre aisée en profondeur, plus spécialement dans les milieux à base de peptones et accompagnée d'une sécrétion plus marquée du pigment soluble. Le lait est rapi dement coagulé et peptonisé ; par contre, la sécré tion de diastases gélatinolytiques est très faible.
Le tableau suivant (I,) montre les différentes caractéristiques entre cet actinomycète nouveau et d'autres actinomycètes producteurs d'antibiotiques basiques.

Les bouillons de culture du Streptomyces paucisporogenes montrent une activité antibiotique qui augmen te avec le pH de la gélose sur laquelle on effectue le test de contrôle d'activité.
Ceci permet de classer le nouvel antibiotique dans le groupe des antibiotiques basiques : streptomycine, streptothricine, néomycine, fra- mycétine mais les indications fournies par le test de l'antibiose croisée font ressortir des différences essen tielles existant entre le streptomyces paucisporogenes et les micro-organismes producteurs des antibiotiques précités (Tableau II).

On peut, par exemple, opérer selon le principe de la culture dite en étages à partir d'une suspension aqueuse de la souche de Streptomyces paucisporogenes obte nue à la surface d'un milieu de culture solide gélosé. On inocule une série de fioles d'Erlenmeyer conte nant un milieu approprié, les abandonne à l'incuba tion sur agitateur mécanique à une température de préférence voisine de 30,, C, récolte aseptiquement le bouillon de culture, qui sert à ensemencer les fer menteurs industriels. La durée de la fermentation proprement dite est ordinairement comprise entre 48 et<B>150</B> heures, de préférence voisine de 70 heures.
L'aération des fermenteurs est réglée de façon que 0,5 à 2 volumes d'air par volume de milieu traver sent ledit milieu par minute ; l'agitation permet une répartition uniforme de l'air insufflé au sein de la masse liquide.
Le milieu nutritif est composé de sources d'azote telles que le corn steep, les farines, les résidus de distillerie, les levures ; de sources de carbone telles que les sucres, les dextrines, les amidons et de sels minéraux divers servant d'agent tampon et indispen sables à la croissance des cellules. On peut utiliser des huiles pour empêcher la formation de mousses.
Un milieu de culture convenant particulièrement bien à la production du nouvel antibiotique est, par exem- ple, composé comme suit: corn steep sec: 0,8 % ; farine de tourteaux de soja<B>:</B> 2,5 @0/0; dextrine : 1,3 0/0;
glucose: 1% ; carbonate de calcium: 0,2 % ; eau 94,2%. L'extraction de l'antibiotique s'effectue de pré férence sur résine échangeuse de cations.
Le filtrat de la culture est amené à pH voisin de 6 par addi tion d'acide et mis en contact avec une résine échan- geuse de cations telle que, par exemple, la résine marque Amberlite IRC 50 sous sa forme sodi que.
Après lavage à l'eau de la résine., on procède à son élution par une solution aqueuse basique, par exemple une solution saturée de triéthylamine. L'éluat est neutralisé, concentré sous vide et préci pité par adjonction d'un solvant approprié dans le quel le principe actif est insoluble, par exemple, le méthanol ou l'acétone.
La purification de l'antibiotique peut être réalisée par reprise dans un solvant approprié ou par l'inter médiaire de dérivés définis d'où l'on repasse aisé ment au produit initial. On peut, par exemple, trai ter le sulfate brut du nouvel antibiotique par l'al déhyde salicylique en présence de carbonate de so dium, isoler le dérivé salicylidène insoluble dans l'eau et le retransformer en sulfate purifié par une hydrolyse appropriée.
On peut également purifier le nouvel antibiotique par l'intermédiaire du sel cristal lisé qu'il forme avec l'acide p-(p'-hydroxyphénylazo)- benzène-sulfonique. L'antibiotique nouveau est actif <I>in vitro</I> sur de nombreux germes gram-positifs et gram-négatifs, peu actif ou inactif sur pseudomonas aeruginosa ATCC 10 145, inactif sur candida albi- cans et divers saccharomyces. Son action sur myco- bacterium tuberculosis est très nette.
Le tableau III ci-dessous montre à la fois le spectre antibactérien de l'antibiotique et les particularités qui le distin guent nettement des autres antibiotiques majeurs. Il a été établi en gélose par la méthode du gradient de Szibalski. Les valeurs rapportées sont exprimées en y/cm3 de gélose nutritive.

@ <SEP> W <SEP> rav@<B>Fi</B> L'activité sur Mycobactérium tuberculosis patho gène H 37 Rv montre les caractères particuliers du nouvel antibiotique ainsi que le grand avantage de son utilisation clinique. L'absence de résistance croi sée vis-à-vis de la framycétine et de la streptomycine est particulièrement importante.
En effet, une-pre- mière souche résistant à plus de 250 y/cm3 de sulfate de framycétine a été aisément inhibée par 50 y/cm3 de sulfate d'hydroxymycétine ; une seconde souche résistant à plus de 100 y/cm3 de sulfate de strepto mycine a été inhibée par 4 y/ce de sulfate d'hy- droxymycétine.
Le nouvel antibiotique montre une activité parti culièrement marquée<I>in vivo</I> vis-à-vis de la tuber culose. Contrairement à ce qui a été observé<I>in vitro</I> (voir tableau 111), l'hydroxymycine se trouve être pra tiquement d'une activité égale à celle de la strepto mycine. Il suffit en effet d'une dose de 1 mg/jour chez la souris pour obtenir une survie totale et l'ar rêt de l'évolution de toutes les lésions. Cette propriété remarquable fait donc de l'hydroxymycétine un anti biotique de choix dans le traitement de la tuberculose puisqu'elle permet son utilisation en remplacement de la streptomycine dans les cas, toujours plus nom breux, de streptomycine-résistance.
La toxité de l'an tibiotique nouveau peut être comparée à celle des autres antibiotiques. La toxicité aiguë, c'est-à-dire celle provoquée par une seule injection, a été dé terminée chez la souris par la dose léthale (DL 50), c'est-à-dire la dose provoquant la mort de 50 % des animaux auxquels on a injecté l'antibiotique.

L'antibiotique produit par le Streptomyces pau- cisporogenes est une substance basique contenant du carbone, de l'hydrogène, de l'oxygène et de l'azote et capable de fournir des sels avec les acides.
Les réactions caractéristiques suivantes ont été étudiées <I>Tableau V</I> Van Slyke + Ninhydrine + Fehling - Tollens - Benzidine - Phtalate d'aniline - Sakaguchi - Elson Morgan - Ces résultats ont permis les conclusions préli minaires suivantes quant à la constitution du nouvel antibiotique Présence de la totalité de l'azote sous forme de groupements aminés primaires,
absence de groupe ments réducteurs libres, absence de groupements gua- nidés. L'électrophorèse sur papier, la séparation à contre-courant dans le système eau-acétone-chloro- forme et la chromatographie sur papier Wahtmann No 1 dans les solvants butanol eau, acide acétique ;
butanol, eau, acide para-toluène-sulfonique ; butanol, pipéridine, acide para-toluène-sulfonique permettent d'affirmer la présence d'une seule substance active dans l'antibiotique obtenu par le procédé selon la présente invention.
Si l'on soumet le chlorhydrate de l'hydroxymycine à une méthanolyse chlorhydrique, on obtient par simple refroidissement du mélange réactionnel, la cristallisation d'un produit voisin de celui dénommé Fraction A, isolé par méthanolyse chlorhydrique de l'antibiotique framycétine et dé crit dans le Bulletin de la Société Chimique de France 1954, page 1453.
Il en diffère toutefois essentielle ment par ses valeurs analytiques qui permettent de lui attribuer la formule brute C1,.H.5N307. Son acti vité antibiotique représente environ 10 1% de celle de l'hydroxymycine. Si l'on benzoyle l'hydroxymycine dans les conditions habituelles de la réaction de Schotten-Baumann, à température ambiante, on aboutit aisément à un dérivé N-benzoylé cristallisé qui fond à 2320 C (sur bloc) [a]D = -i- 36o 2 (c=0,3 %, méthanol).
On aboutit à ce même dé rivé N-benzoylé par N,O-benzoylation totale de l'hy- droxymycine suivie d'une méthanolyse alcaline qui a pour effet de libérer les hydroxyles de la molécule. L'exemple suivant illustre l'invention.

Extraction de l'antibiotique brut</I> On centrifuge et filtre 1 litre de bouillon de cul ture obtenu selon 1 ou la, on ajuste le pH à 6 par addition de 1,1 cm3 d'acide sulfurique dilué (1 : 1) et met en contact avec 25 cm3 d' Amberlite IRC 50 préalablement traitée à la soude N et lavée à l'eau distillée jusqu'à neutralité.
La résine est lavée par 50 crus d'eau puis éluée par 3 volumes d'une solu tion aqueuse saturée de triéthylamine. L'éluat est neutralisé par l'acide sulfurique dilué (1 : 1) et con centré sous vide au 1/5000 de son volume initial. Le concentrai ainsi obtenu est acidifié à pH 3,5 par l'acide sulfurique 2 N et précipité dans 6 volumes de méthanol.
On essore, lave au méthanol, sèche sous vide sulfurique et obtient ainsi 700 mg de sulfate brut d'hydroxymycine ; titre = 400 u/mg, [ ]D = -f- 40 à 44p (c=1%, eau).
<I>C. Purification</I> <I>1. Purification du sulfate</I> On dissout 1 g de sulfate brut obtenu selon B dans 4 cm3 d'eau,- décolore par 0,6 g de charbon ac tif, on dilue à 7,5 cm3 et alcalinise par 0,75 g de bicarbonate de sodium, ajoute 0,7 cm3 d'aldéhyde salicylique et agite pendant deux heures à tempéra- turc ambiante. On filtre, lave à l'eau et sèche pour obtenir ainsi 1,25 g du dérivé salicylidène insoluble dans l'eau. Ce dernier est mis en suspension dans 5 em3 d'eau<B>;</B> on ajoute 3 cm3 d'acide chlorhydrique 2 N et chauffe cinq minutes à 600 C.
On refroidit, extrait au chloroforme et amène la phase aqueuse à pü 5 par addition de triéthylamine. On précipite dans 5 volumes de méthanol contenant 2 cm3 d'une solution méthanolique à 30% de sulfate de triéthyl- amine. On filtre, lave au méthanol et sèche sous. vide pour obtenir 0,6 g de sulfate purifié.
Ce produit se présente sous forme d'une poudre blanche amorphe, pratiquement insoluble dans les solvants organiques, [a]D = -I- 500 2 (c = 1%, eau), activité: 690 u/mg.
<I>la. Purification du sulfate</I> d'hydroxymycihe <I>par</I> <I>l'intermédiaire de sel de l'acide</I> p-(p'-hy- droxyphéhylazo)-benzèrae-sul <I>f</I> onique On dissout 3 g de p-(p =hydroxyphénylazo)-ben- zène-sulfonate de sodium dans 200 cm3 d'eau à 70p C, refroidit à 50p C et ajoute une solution de 1,2 g de sulfate d'hydroxymycine dans 20 cm3 d'eau.
On abandonne une nuit à la température ambiante, essore, reprend dans 10 crus de méthanol à 50p C, ajoute 15 cm3 d'eau -à 50,)C et laisse cristalliser deux heures à la température ambiante.
On essore, pour obtenir 1,9 g de p-(p'-hydroxyphénylazd)-benzène- sulfonate d'hydroxymycine. Ce produit se décompose vers 2000 C, [a]D = -f-370 5 (c = 0,5 %, mé- thanol).
Analyse <I>:</I> C 48,3 % H 5,0 % N 9,8'% S 8,0 0/0 1 g de ce sel est mis en solution dans 12 cm-' de méthanol.
On ajoute 4 cm3 d'acide chlorhydrique 2 N puis une solution de 4 cm3 de sulfate de tri- éthylamine dans 4 cm3 de méthanol<B>;</B> on essore, lave au méthanol et sèche sous vide pour obtenir 0,4 g de sulfate purifié d'hydroxymycine. L'amélioration d'activité du produit ainsi obtenu, par rapport au sulfate initial, se chiffre aux environs de 30 0/0.
Ainsi, au départ d'un sulfate d'hydroxymycine, préparé se lon B et titrant 400 u/mg, on obtient un sulfate ayant un titre de 600 u/mg. Une nouvelle purification fait atteindre la valeur de 750 u/mg, qui ne peut être dépassée, [a]D = -I- 50 à -I- 520.

Préparation du chlorhydrate</I> On dissout 1 g du sulfate purifié selon la dans 2 cm3 d'eau, ajoute une solution de chlorure de ba- ryum à 30 % contenant la quantité stoechiométrique de ce sel, filtre, concentre sous vide à 0,5 cm-3,
ajoute 5 cm3 de méthanol à 80 % et précipite dans 25 cm3 d'acétone. On filtre, lave à l'acétone et sèche sous vide pour obtenir 0,8 g de chlorhydrate d'hy- droxymycine, F (déc:) = 2000 C [a]D = -f- 520 (c = 10/0, eau).
Le produit se présente sous forme d'une poudre amorphe blanche, soluble dans l'eau et le méthanol aqueux, insoluble dans les solvants organiques.
<I>3. Préparation de la base</I> On dissout 1 g du sulfate purifié selon la dans 3 cm3 d'eau, ajoute, à la température de 300 C, la quantité stoechiométrique d'une solution à 10 % de baryte, refroidit à 20 C, filtre et concentre la so lution obtenue à 5 cm3. On ajoute 1 volume de mé thanol et précipite dans 15 volumes d'acétone.
On filtre, lave à l'acétone et sèche sous vide pour obte nir 0,6 g d'hydroxymycine, F (dée.) = 2000 C, pas de fusion franche instantanée jusqu'à 300 , [ ]D = + 63 0 2 (c = 1 0/0, eau), activité de l'ordre de 950 -<B>1000</B> u/mg. Process for the preparation of a novel antibiotic The present invention relates to a process for the preparation of a novel antibiotic, by culturing a strain of Streptomyces paucisporogenes in an aqueous medium containing, apart from mineral salts, a source of nitrogen and a carbohydrate, aerobically until substantial activity is manifested in said medium, followed by extraction and purification of the antibiotic.
Streptomyces paucisporogenes is found, among others, in soil. It is isolated as a pure culture according to standard procedures. It was registered at the licensee's offices under number T4915. A sample of this strain T 4915 has been deposited in the National Collection of Industrial Bacteria, The Commonwealth Mycological Institute, Ferry Lane, Kew (Surrey, Great Britain), under the number NCIB 8790,
whereas a second sample of the strain was deposited in the American Type Culture Collection Washington D.C. (USA), under the N ATCC 12596. Strepto myces paucisporogenes grows with difficulty on the solid culture media usually used. It provides colonies composed of filaments of which the apical part, more or less branched, only exceptionally contains chains of conidia.
On complex organic agar media, Streptomyces paucisporogenes is microscopically characterized by a yellowish-white aerial mycelium; the felting of the vegetative mycelium is thick, sometimes cracked. It sometimes secretes a small amount of a soluble yellow-brown pigment. Its development is particularly good on a medium composed as follows and adjusted to pH 7.5: decoction in a liter of distilled water of 200 g of crushed soybeans, 20 g of glucose, 10 g of apple starch. earth and 20 g of agar.
On chemically defined media (Czapeck, Conn ... etc.) growth is difficult: however, there is solubilization of calcium malate.
On a natural substrate such as potato impregnated with buffer at pH 7, Streptomyces pauisporogenes develops rapidly in the form of a thick, cerebroid, cracked coating. The aerial myceium is then rare, greyish white with yellow reflections.
If the surface culture of Streptomyces paucporogenes is difficult, it is on the other hand easy in depth, more especially in media based on peptones and accompanied by a more marked secretion of the soluble pigment. The milk is rapidly coagulated and peptonized; on the other hand, the secretion of gelatinolytic diastases is very low.

The new antibiotic is active <I> in vitro </I> on numerous gram-positive and gram-negative bacteria, little or inactive on pseudomonas aeruginosa ATCC 10,145, inactive on candida albicans and various saccharomyces. In fact, a first strain resistant to more than 250 µ / cm3 of framycetin sulfate was easily inhibited by 50 µ / cm3 of hydroxymycetin sulfate; a second strain resistant to greater than 100 µ / cc of streptomycin sulfate was inhibited by 4 µ / cc of hydroxymycetin sulfate.
The new antibiotic shows a particularly marked activity <I> in vivo </I> vis-à-vis tuber culosis.
