umsetzt und die erhaltenen Reaktionsprodukte hydrolytisch zersetzt.
Die Araylaminoacetonitrile lassen sich, soweit sie nicht bekannt sind, nach bekannten Methoden (vgl.
Houben-Weyl, Methoden der organischen Chemie, 4. Aufl., 8. Band, S. 279 ff.) herstellen.
Die nach dem erfindungsgemässen Verfahren erhaltenen Verbindungen sind neue Substanzen und stellen wertvolle, sehr gut wirkende Analgetika dar.
Beispiel 1 1-(4-Methoxy-phenyl)-l-dimethylamino-
2-phenyl-athan

Aus 4, 6 g Magnesiumspänen in 40 cm3 wasserfreiem Äther wird durch Zutropfenlassen von 25, 2 g Benzylchlorid in 50 cm3 Äther unter Rühren in üblicher Weise die Grignardverbindung hergestellt.
Hierzu gibt man langsam unter Rühren 19 g 4 Methoxy-phenyl-dimethylamino-acetonitril (Kpgg = 108 bis 110 ) in 50 cm3 Äther gelöst, hinzu, erwärmt nach beendigter Zugabe noch zwei Stunden unter Rückfluss und zersetzt dann den Reaktionsansatz durch Zugabe von Eis und verdünnter Salzsäure bis zur sauren Reaktion. Nach dem Abtrennen der Ätherschicht, die verworfen wird, fügt man zur wässrigen Lösung etwas Ammonchlorid und dann Ammoniak bis zur alkalischen Reaktion zu. Das hierbei abgeschiedene Amin wird in Äther aufgenommen. Der nach dem Verjagen des Athers verbleibende Rück- stand wird im Vakuum destilliert.
Man erhält das tertiäre Amin vom Kp g =142 bis 143 in einer Ausbeute von 79 /o d. Th. ; wird Benzol als Lösungs- mittel verwendet, so erzielt man dasselbe Ergebnis.
Das durch Lösen des Amins in Äther und Ausfällen mit ätherischer Salzsäure erhältliche Hydrochlorid bildet farblose Kristalle vom F. = 187 .
Beispiel 2 1- (4-Methyl-phenyl)-1-dimethylamino-2-phenyl-dthan

Die Grignardverbindung aus 2, 3 g Magnesium und 12, 6 g Benzylchlorid in Äther wird in der oben angegebenen Weise mit 8, 7 g 4-Methyl-phenyl-di methylamino-acetonitril (Kp@, 7 = 104 ) umgesetzt und aufgearbeitet. Das tertiäre Amin vom Kapo, g =130 wird in einer Ausbeute von 74% d. Th. erhalten.
Wird die Umsetzung in Dibutyläther vorgenommen und der Reaktionsansatz auf 90 bis 100 erhitzt, so beträgt die Ausbeute 71 /o d. Th. Das farblose Hydrochlorid der Verbindung schmilzt bei 1890.

1-(4-Oxy-phenyl)-1-dimethylamino-2-phenyl-Ïthan

Wie in Beispiel 1 angegeben, setzt man das Grignardreagenz aus 2, 3 g Magnesium und 12, 6 g Benzylchlorid mit 5, 9 g 4-Oxy-phenyl-dimethylaminoacetonitril (F. = 75 ) um und arbeitet in der dort beschriebenen Weise auf. Das beim Verdampfen des Äthers verbleibende feste tertiäre Amin wird jedoch nicht destilliert, sondern aus Athanol umkristallisiert.
Es bildet farblose Kristalle vom F. = 136 bis 138 .
Ausbeute 59 /o d. Th. N ber. : 5, 81 O/o, gef. : 5, 83 /a.
Das Hydrochlorid dieser Verbindung schmilzt bei 173 .
Beispiel 4
1-(4-Isopropyl-phenyl)-1-dimethylamino 2-phenyl-äthan

Man setzt, wie in Beispiel 1 beschrieben, die Grignardverbindung aus 4, 6 g Magnesium und 25, 2 g Benzylchlorid mit 20, 2 g 4-Isopropyl-phenyldimethyl- amino-acetonitril (Kp0, 2 = 96¯) um. Das entstandene tertiäre Amin ist eine gelbliche Flüssigkeit vom Kpo o, = 93 bis 96¯. N ber. : 5, 24%, gef. : 5, 24 /o.

Beispiel 5 1- (4-Methoxy-phenyl)-l-dimethylamino-
2-(4-methyl-phenyl)-Ïthan

19 g 4-Methoxy-phenyl-dimethylamino-acetonitril lässt man in derselben Weise, wie in Beispiel 1 beschrieben, auf die Grignardverbindung aus 37 g p-Xylylbromid und 4, 6 g Magnesiumspänen einwirken. Man erhält das tertiäre Amin in einer Ausbeute von 67 /o d.
Th. als farblose Flüssigkeit vom Kp0.25 = 132 . Die Base erstarrt beim Stehen und schmilzt dann bei 45 . N ber. : 5, 2 %, gef. : 5, 35 %,

Man verfährt in derselben Weise wie in Beispiel 3 angegeben, unter Verwendung der Grignardverbindung aus 2, 3 g Magnesium und 18, 5 g p-Xylylbromid. Das aus Athanol umkristallisierte tertiäre Amin schmilzt bei 141 (Zers.). N ber. : 5, 49%, gef. : 5, 54 /o. Ausbeute 62 ouzo d. Th. Das Hydrochlorid dieser Verbindung schmilzt bei 188 .

17, 4 g 4-Methyl-phenyl-dimethylamino-acetonitril werden nach Beispiel 1 mit dem Grignardreagenz aus 4, 6 g Magnesium und 37 g p-Xylylbromid umgesetzt.
Das entstandene tertiäre Amin ist eine farblose Flüssigkeit vom Kp0.5=128¯. N ber. : 5, 54 I/o, gef. : 5, 52%. Ausbeute 67% d. Th. Das farblose, kristalline Hydrochlorid der Verbindung schmilzt bei 209 .
Beispiel 8 1-Phenyl-l-dimethylamino-2- nethyl-phenyl)-äthan

Nach Beispiel 1 wird die Grignardverbindung aus 4, 6 g Magnesium und 37 g p-Xylylbromid mit 16 g Phenyl-dimethylamino-acetonitril umgesetzt. In einer Ausbeute von 68 oxo d. Th. erhält man dabei das tertiäre Amin vom Kpo, l5 = 118 .
Beispiel 9 I-Phenyl-1-dimethylamino-
2- (4-methoxy-phenyl)-athan

Man stellt eine Grignardverbindung aus 19 g feinen Magnesiumspänen, 40 g p-Methoxy-benzylbromid und einigen Körnchen Jod in 100 cm3 wasserfreiem Ather her und lässt unter stetem Rühren 14 g Phenyl-dimethylamino-acetonitrll, in Äther gelöst, zutropfen. Nach beendeter Zugabe wird noch 21/2 Stunden unter Rückfluss erhitzt. Nach dem Erkalten zersetzt man das Reaktionsgemisch durch Zugabe von Eis und verdünnter Salzsäure, trennt die Atherschicht ab, welche verworfen wird, und macht die wässrige Lösung durch Zugabe von Ammoniak alkalisch.
Durch mehrmaliges Ausschütteln mit Ather wird das Amin aus der wässrigen Lösung extrahiert. Zur Reinigung des tertiären Amins wird die vereinigte Atherlösung mit etwa 5''Atiger Salzsäure mehrmals durchgeschüttelt. Die so erhaltenen wässrigsalzsauren Losun- gen werden vereinigt, durch Zugabe von Ammoniak alkalisch gemacht und mit Ather ausgeschüttelt. Die Atherlösung hinterlässt nach dem Verjagen des Lösungsmittels 8, 5 g Amin (Kpo 45 = 125 ). Der Schmelzpunkt des Hydrochlorids dieser Verbindung liegt bei 196 bis 197 .

The compounds obtained by the process according to the invention are new substances and are valuable, very effective analgesics.
Example 1 1- (4-methoxyphenyl) -l-dimethylamino-
2-phenyl-ethane

The Grignard compound is produced in the usual way from 4.6 g of magnesium shavings in 40 cm3 of anhydrous ether by adding 25.2 g of benzyl chloride in 50 cm3 of ether dropwise while stirring.
To this, 19 g of 4 methoxyphenyl-dimethylamino-acetonitrile (Kpgg = 108 to 110) dissolved in 50 cm3 of ether are slowly added, while the addition is complete, the mixture is heated under reflux for a further two hours and then the reaction mixture is decomposed by adding ice and dilute hydrochloric acid to acidic reaction. After separating off the ether layer, which is discarded, a little ammonium chloride and then ammonia are added to the aqueous solution until an alkaline reaction is obtained. The amine deposited in the process is taken up in ether. The residue remaining after the ether has been driven off is distilled in vacuo.
The tertiary amine is obtained with a boiling point of 142 to 143 in a yield of 79%. Th.; if benzene is used as the solvent, the same result is obtained.
The hydrochloride, which can be obtained by dissolving the amine in ether and precipitating it with ethereal hydrochloric acid, forms colorless crystals of F. = 187.
Example 2 1- (4-methyl-phenyl) -1-dimethylamino-2-phenyl-dthane

The Grignard compound of 2.3 g of magnesium and 12.6 g of benzyl chloride in ether is reacted and worked up in the manner indicated above with 8.7 g of 4-methylphenyldimethylaminoacetonitrile (bp @, 7 = 104). The tertiary amine from Kapo, g = 130, is used in a yield of 74% of theory. Th. Received.
If the reaction is carried out in dibutyl ether and the reaction mixture is heated to 90 to 100, the yield is 71 / o d. Th. The colorless hydrochloride of the compound melts in 1890.
