Toluol, Xylol usw. gelöst und unter Rühren und Zusatz eines ge eigneten Kondensationsmittels, z. B. Alkalihydroxyd, Alkaliamid, Alkalicarbonat oder Kupferpulver, mit 2-(N-Methyl-piperidyl-2')-1-chlor-äthan der Formel III bei Zimmertemperatur oder bei erhöhter Tempe ratur umgesetzt wird. Nach beendeter Umsetzung gewinnt man das Reaktionsprodukt durch entspre chende Aufarbeitung des Reaktionsgemisches und Destillation im Vakuum.
Die weitere Reinigung kann durch Überführung in ein geeignetes Salz und Rück gewinnung der freien Base erfolgen.
Im Hauptpatent wird die Herstellung von Di- phenylaminen beschrieben, die gegebenenfalls an einem oder beiden Phenyl-Resten einen einwertigen schwefelfreien Substituenten tragen. Diese Verbin dungen besitzen spasmolytische Wirkung; ferner die nen sie als Zwischenprodukte für die Herstellung von narkotisch, analgetisch und hypnotisch wirksamen Pharmaka.
Die erfindungsgemäss erhältlichen Verbindungen besitzen demgegenüber in m-Stellung eines Phenyl- restes einen schwefelhaltigen Substituenten in Form einer Alkylmercapto-Gruppe.
Sie sind bei Zimmertemperatur flüssig und bilden mit Säuren beständige, kristallisierte Salze. Sie be sitzen bemerkenswerte antimykotische Eigenschaften und sollen deshalb als Fungicide in der Therapie Ver wendung finden. Ferner dienen sie als Zwischenpro dukte für die Synthese von therapeutisch wirksamen Verbindungen. Sie liefern z. B. beim Behandeln mit elementarem Schwefel in Gegenwart oder bei Ab wesenheit eines Katalysators, wie Jod, oder mit Schwefeldihalogeniden Phenothiazin-Derivate mit wertvollen therapeutischen Eigenschaften, wie z. B.
Potenzierung des Effektes narkotisch, hypnotisch und analgetisch wirkender Pharmaka. Letztere Stoffe eignen sich deshalb zur Narkose-Vorbereitung, kön- nen aber ausserdem auch bei allergischen Erkrankun gen sowie als Spasmolytika oder als Neuroplegika in der Therapie verwendet werden.
<I>Beispiel 1</I> N-[m-Methylmercapto-phenyl]-N-[2-(N'-methyl- piperidyl-2')-äthyl-1 ]-anilin Man kondensiert m-Methylmercapto-anilin (Sdp. 163-165 116 mm Hg) mit dem Kaliumsalz der o-Chlor-benzoesäure und decarboxyliert die entstan dene N-(m-Methylmercapto-phenyl)-anthranilsäure (Smp. 139-l41 ) durch Erhitzen und anschliessende Destillation zum N-(m-Methylmercapto-phenyl)-anilin (Smp. 59-610)

15 g N-(m-Methylmercapto-phenyl)-anilin wer den in 75 cm3 abs. Xylol gelöst, mit 3,26 g fein pulverisiertem Natriumamid versetzt und während 2 Stunden bei einer Badtemperatur von 180 zum Sieden am Rückfluss erhitzt. Darauf lässt man, ohne das Erhitzen zu unterbrechen, innerhalb von 11/2 Std.
12,7 g 2-(N-Methyl-piperidyl-2')-1-chlor-äthan (Sdp. 840j10 mm Hg) gelöst in 12 cm3 abs. Xylol zutropfen und erhitzt, ohne das Rühren zu unterbrechen, das Gemisch während weiterer 3 Std. zum Sieden. Als dann wird abgekühlt und das überschüssige Natrium- amid durch Zusatz von 5 g Ammoniumchlorid zer setzt.
Die dreimal mit je 25 cm3 Wasser gewaschene Xylollösung wird einmal mit 30 cm3, dann dreimal mit je 10 cm3 3n Essigsäure ausgezogen. Man schüt telt die vereinigten Essigsäureauszüge mit 50 em3 Äther aus und macht sie mit 16 cm3 konz. Natron lauge phenolphthalein-alkalisch. Die Lösung der aus geschiedenen öligen Base in 100 em3 Benzol wird mit 30 cm3 Wasser ausgeschüttelt, über Pottasche ge trocknet, filtriert und unter vermindertem Druck ein gedampft.
Der Rückstand wird im Hochvakuum destilliert, wobei die unter 0,01 mm Hg bei 215 bis 218 übergehende Hauptfraktion aufgefangen wird. Das analysenreine N-[m-Methylmercapto-phenyl]-N- [2-(N'-methyl-piperidyl-2')-äthyl-1]-anilin hat den Siedepunkt 216 bei 0,01 mm Hg. <I>Beispiel 2</I> N-[m-Äthylmercapto-phercyl]-N-[2-(N'-metiayl- piperidyl-2')-äthyl-1 ]-anhin Man kondensiert m-Äthylmercapto-anilin (Sdp. 147-152 ,110 mm Hg)
mit dem Kaliumsalz der o-Chlor-benzoesäure und decarboxyliert die entstan dene N-(m-Äthylmercapto-phenyl)-anthranilsäure (Smp. 114-116 ) durch Erhitzen und anschliessende Destillation zum N-(m-Äthylmercapto-phenyl)-anilin (Sdp. 140 bei 0,007 mm Hg).
20,12 g N-(m-Äthylmercapto-phenyl)-anilin wer den in 90 cm3 abs. Xylol gelöst, mit 4,10 g fein pulverisiertem Natriumamid versetzt und während 2 Stunden bei einer Badtemperatur von 180 zum Sieden am Rückfluss erhitzt. Darauf lässt man ohne das Erhitzen zu unterbrechen innerhalb von<B>1</B>1/2 Std.
15,9 g 2-(N-Methyl-piperidyl-2')-1-chlor-äthan (Sdp. 84 bei 10 mm Hg) gelöst in 15 cm3 abs. Xylol zu tropfen und erhitzt ohne das Rühren zu unterbrechen das Gemisch während weiterer 3 Std. zum Sieden. Alsdann wird abgekühlt, und das überschüssige Natriumamid durch Zusatz von 5,0 g Arnmonium- chlorid zersetzt.
Die dreimal mit je 30 cm3 Wasser gewaschene Xylollösung wird einmal mit 40 cm3, dann dreimal mit je 15 cm-3 3n Essigsäure ausge zogen. Man schüttelt die vereinigten Essigsäureaus- züge mit 50 cm3 Äther aus und macht sie mit 25 em3 konz. Natronlauge phenolphthalein-alkalisch. Die Lö sung der ausgeschiedenen öligen Base in 110 cm-' Benzol wird mit 30 cm3 Wasser ausgeschüttelt, über Pottasche getrocknet, filtriert und unter verminder tem Druck eingedampft.
Der Rückstand wird im Hochvakuum destilliert, wobei die unter 0,005 mm Hg bei 205-207 übergehende Hauptfraktion aufge fangen wird. Das analysenreine N-[m-Äthylmercapto- phenyl] -N-[2-(N'-methyl-piperidyl-2')-äthyl-1 ]-anilin hat den Sdp. 206 bei 0,005 mm H1-1. <I>Beispiel 3</I> N-[m-lsopropylmereapto-phenyl]-N-[2-(N'-ntethyl- piperidyl-2')-äthyl-]]-anhin Man reduziert m-Isopropylmercapto-nitrobenzol (Sdp. 148-150 bei 11 mm Hg)
mit Stannochlorid oder Eisenspänen und Salzsäure zu m-Isopropyl- mercapto-anilin (Sdp. 142-144 bei 10 mm Hg), das mit o-chlorbenzoesaurem Kalium zur N-(m-Iso- propylmercapto-phenyl)-anthranilsäure (Smp. 114 bis 116 ) kondensiert wird.
Diese Verbindung wird durch Erhitzen und anschliessende Destillation zum N-(m- Isopropylmercapto-phenyl)-anilin (Sdp. 143 bei 0,005 mm Hg) decarboxyliert.
30,0 g N-(m-Isopropylmercapto-phenyl)-anilin werden in 135 cm-3 abs. Xylol gelöst, mit 5,77 g fein pulverisiertem Natriumamid versetzt und während 2 Std. bei einer Badtemperatur von 180 zum Sieden am Rückfluss erhitzt.
Darauf lässt man, ohne das Er hitzen zu unterbrechen, innerhalb von<B>11</B> .Std. 22,4 g 2-(N-Methyl-piperidyl-2')-1-chlor-äthan (Sdp. 84 bei 10 mm Hg) gelöst in 25 cm3 abs. Xylol zutropfen und erhitzt, ohne das Rühren zu unter brechen, das Gemisch während weiterer 3 Std. zum Sieden. Alsdann wird abgekühlt und das überschüs sige Natriumamid durch Zusatz von 10 g Ammo- niumchlorid zersetzt.
