Verfahren zur Herstellung eines Derivates des 19-Nortestosterons Die Erfindung bezieht sich auf die Herstellung des biologisch hoch aktiven ss-Phenylpropionsäureesters des 19-Nortestosterons.
In der schweiz. Patentschrift Nr. 321320 ist be schrieben, dass physiologisch aktive Verbindungen der Cyclopentanopolyhydrophenanthrenreihe mit einer freien Hydroxylgruppe, welche mit einer Fettsäure verestert werden können, befähigt sind, mit ss-Phenyl- propionsäure oder einem funktionellen Derivat der selben unter Bildung physiologisch aktiver Derivate zu reagieren, die eine längere Wirkungsdauer auf weisen als die bisher bekannten Derivate der ent sprechenden Verbindungen und überdies in den meisten Fällen auch eine grössere Aktivität als die bekannten Derivate besitzen.
Diese Ester können z. B. durch Kondensation des Steroids mit ss-Phenylpropionsäureanhydrid herge stellt werden, und zwar entweder ohne Verwendung eines Lösungsmittels, in welchem Fall die Reaktions zeit durch Erhitzen verkürzt werden kann, oder in einem Lösungsmittel, z. B. Pyridin, Benzol oder einem andern geeigneten Lösungsmittel oder Lösungsmittel gemisch.
Sie können auch mit Hilfe eines ss-Phenyl- propionsäurehalogenids, vorzugsweise des Chlorids, in einem Lösungsmittel wie Pyridin oder einem Benzol-Pyridin-Gemisch, das genug Pyridin enthalten muss, um das während der Reaktion abgespaltene HCl zu binden, hergestellt werden. Auch andere funktionelle Derivate der ss-Phenylpropionsäure kön nen angewendet werden.
Es ist bekannt, dass Testosteron, zusätzlich zu seiner adrogenen Wirksamkeit, die Eigenschaft be sitzt, den Aufbau des Gewebes anzuregen. Die kli nische Anwendung dieser Verbindung auf Grund ihres proteinerhaltenden Effektes, häufig anabohscher Effekt genannt, wird jedoch durch den starken an- drogenen Effekt des Testosterons begrenzt. Aus gedehnte Untersuchungen beschäftigen sich daher mit der Entwicklung einer Verbindung mit starkem anabolischem Effekt bei geringer oder fehlender androgener Wirkung.
Es wurde nun gefunden, dass 19-Nortestosteron in gleicher Weise wie bereits beschrieben mit ss-Phenyl- propionsäure verestert werden kann und dass der so erhaltene Ester eine sehr starke anabolische Wirk samkeit in Kombination mit einer nur sehr schwachen androgenen Wirksamkeit aufweist, so dass dieser Ester für die Anwendung als Heilmittel mit protein erhaltendem Charakter besonders gut geeignet ist. Dieser neue Ester besitzt einen Schmelzpunkt von 94-96 C und eine (a)D = + 58 (in Chloroform).
19-Nortestosteron selbst ist bekannt, siehe z. B. Journ. Amer. Chem. Soc. <I>75,</I> 5366 (1953), wo seine Herstellung aus Oestron beschrieben ist. Das ameri kanische Patent Nr. 2698855 beschreibt einen andern anabolisch wirksamen Ester des 19-Nortestosterons, nämlich das Benzoat.
19-Nortestosteron-ss-phenylpropionat besitzt hohe anabolische und niedere androgene Wirksamkeit, wie durch Bestimmung des Effektes auf das Gewicht des Levator-Animuskels und der Samenbläschen bei kastrierten männlichen Ratten festgestellt wurde.
(Hershberger et a1., Proc. Soc. exp. Biol. and Med. 83, 175, 1953.) Männliche Ratten mit 40-50 g Körper gewicht wurden kastriert und vom folgenden Tag an einer Reihe täglicher subkutaner Injektionen unter worfen. Nach einer Woche der Behandlung wurden die Tiere getötet und der Levator-Ani-Muskel (MLA) und Samenbläschen gewogen. Die Gewichte des MLA wurden pro 100 g des ursprünglichen Körpergewichts berechnet, diejenigen der Samenbläschen sind die tatsächlichen Gewichte.
In Tabelle 1 wird ein Vergleich zwischen der freien Verbindung, dem Benzoat und dem ss-Phenylpropionat gegeben. In allen Fällen wurden die gleichen Mengen, bezogen auf 19-Nortestosteron, verabreicht. Alle Prä parate wurden in Arachis-Öl gelöst.

In zwei weiteren Versuchen wurden die ss-Phenyl- propionate von Testosteron und 19-Nortestosteron miteinander verglichen, wobei die gleiche Versuchs technik angewendet wurde. Im letzteren Fall wird, wie ersichtlich, der maximale Effekt bei einer Dosierung von 50-100 Mikrogramm erzielt, und zwar in einer Höhe, die mit 200 Mikrogramm Testosteron-ss-phenylpropionat kaum erreicht wird. Sorgfältige Abschätzung führt zu etwa zweifacher anabolischer Wirksamkeit und etwa der halben androgenen Wirksamkeit gegenüber Testosteron-ss-phenylpropionat. Die folgenden Beispiele erläutern verschiedene Ausführungsformen des Verfahrens zur Herstellung des ss-Phenylpropionsäureesters.
<I>Beispiel 1</I> 12 g ss-Phenylpropionsäureanhydrid werden einer Lösung von 10 g 19-Nortestosteron in 30 ml Pyridin zugegeben. Nach Stehenlassen über Nacht bei Raum temperatur wird die Lösung durch Zusatz von Wasser zersetzt. Das 19-Nortestosteron-ss-phenylpropionat kri stallisiert aus Methanol, F. p. 95-96 C. <I>Beispiel 2</I> Eine Lösung von 10 g 19-Nortestosteron in 30 ml Benzol und 10 ml Pyridin wird bereitet und in Eis gekühlt. Unter Rühren werden 7,5 g ss-Phenylpro- pionylchlorid tropfenweise zugesetzt.
Hierauf wird die Reaktionsmischung über Nacht bei Raum temperatur stehengelassen. Danach wird sie nach einander mit Wasser, verdünnter Natriumcarbonat- lösung und verdünnter Salzsäure gewaschen. Nach dem Trocknen wird die Benzollösung eingedampft und der Rückstand aus Methanol umkristallisiert. Der Schmelzpunkt des gebildeten Esters beträgt 94,5 bis 95,5 C.
<I>Beispiel 3</I> 10 g 19-Nortestosteron und 12 g ss-Phenylpropion- säureanhydrid werden zusammen während 5 Stunden auf 100 C erhitzt. Die Schmelze wird mit Wasser ge mischt, die feste Substanz mit verdünnter Natrium- carbonatlösung gewaschen, hierauf getrocknet und aus Methanol umkristallisiert. Der Schmelzpunkt beträgt 94-95,5 C.
Process for the preparation of a derivative of 19-nortestosterone The invention relates to the preparation of the biologically highly active β-phenylpropionic acid ester of 19-nortestosterone.
In Switzerland. Patent specification No. 321320 describes be that physiologically active compounds of the cyclopentanopolyhydrophenanthrene series with a free hydroxyl group, which can be esterified with a fatty acid, are able to react with ß-phenylpropionic acid or a functional derivative of the same to form physiologically active derivatives, which have a longer duration of action than the previously known derivatives of the corresponding compounds and, moreover, in most cases also have a greater activity than the known derivatives.
These esters can e.g. B. by condensation of the steroid with ss-phenylpropionic anhydride Herge provides, either without using a solvent, in which case the reaction time can be shortened by heating, or in a solvent, eg. B. pyridine, benzene or other suitable solvent or solvent mixture.
They can also be prepared with the aid of an β-phenylpropionic acid halide, preferably the chloride, in a solvent such as pyridine or a benzene-pyridine mixture, which must contain enough pyridine to bind the HCl split off during the reaction. Other functional derivatives of β-phenylpropionic acid can also be used.
It is known that testosterone, in addition to its arogenic effectiveness, has the property of stimulating tissue build-up. The clinical application of this compound due to its protein-preserving effect, often called an anabolic effect, is limited by the strong anrogenic effect of testosterone. Extensive investigations therefore deal with the development of a compound with a strong anabolic effect with little or no androgenic effect.
It has now been found that 19-nortestosterone can be esterified with β-phenylpropionic acid in the same way as already described and that the ester obtained in this way has a very strong anabolic efficacy in combination with only a very weak androgenic activity, so that it Ester is particularly well suited for use as a medicinal product with a protein-retaining character. This new ester has a melting point of 94-96 C and (a) D = + 58 (in chloroform).
19-nortestosterone itself is known, see e.g. B. Journ. Amer. Chem. Soc. <I> 75, </I> 5366 (1953), where its manufacture from oestron is described. American Patent No. 2698855 describes another anabolic ester of 19-nortestosterone, namely benzoate.
19-nortestosterone-ss-phenylpropionate has high anabolic and low androgenic effectiveness, as was determined by determining the effect on the weight of the levator animuscle and the seminal vesicles in castrated male rats.
(Hershberger et al., Proc. Soc. Exp. Biol. And Med. 83, 175, 1953.) Male rats weighing 40-50 g were castrated and given a series of daily subcutaneous injections from the following day. After one week of treatment, the animals were sacrificed and the levator ani muscle (MLA) and seminal vesicles were weighed. The weights of the MLA were calculated per 100 g of original body weight, those of the seminal vesicles being the actual weights.
In Table 1 a comparison between the free compound, the benzoate and the ß-phenylpropionate is given. In all cases, the same amounts based on 19-nortestosterone were administered. All preparations were dissolved in arachis oil.

In two further experiments, the ss-phenylpropionates of testosterone and 19-nortestosterone were compared with one another, using the same experimental technique. The results are shown in Table 2.

In the latter case, as can be seen, the maximum effect is achieved at a dosage of 50-100 micrograms, and at a level that is hardly achieved with 200 micrograms of testosterone-ss-phenylpropionate. Careful estimation leads to about twice the anabolic effectiveness and about half the androgenic effectiveness compared to testosterone-ss-phenylpropionate. The following examples illustrate various embodiments of the process for preparing the β-phenylpropionic acid ester.
<I> Example 1 </I> 12 g of β-phenylpropionic anhydride are added to a solution of 10 g of 19-nortestosterone in 30 ml of pyridine. After standing overnight at room temperature, the solution is decomposed by adding water. The 19-nortestosterone-ss-phenylpropionate crystallized from methanol, F. p. 95-96 C. <I> Example 2 </I> A solution of 10 g of 19-nortestosterone in 30 ml of benzene and 10 ml of pyridine is prepared and cooled in ice. 7.5 g of β-phenylpropionyl chloride are added dropwise with stirring.
The reaction mixture is then left to stand at room temperature overnight. It is then washed successively with water, dilute sodium carbonate solution and dilute hydrochloric acid. After drying, the benzene solution is evaporated and the residue is recrystallized from methanol. The melting point of the ester formed is 94.5 to 95.5 C.
<I> Example 3 </I> 10 g of 19-nortestosterone and 12 g of ß-phenylpropionic anhydride are heated together to 100 ° C. for 5 hours. The melt is mixed with water, the solid substance is washed with dilute sodium carbonate solution, then dried and recrystallized from methanol. The melting point is 94-95.5 C.
