Verfahren zur Herstellung von Ringketonen
Gegenstand des Hauptpatentes Nr. 338189 ist ein Verfahren zur Herstellung von 2,6,6-Trimethyl-cyclo- hexandion-(1, 4).
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von Estern des 2,6,6-Tri- methyl-cyclohexanol-(4)-on-(1).
Das Verfahren gemäss der Erfindung ist dadurch gekennzeichnet, dass man auf 2,6,6-Trimethyl-cyclohexen-(l)-on-(4) eine organische Persäure einwirken lässt, das Reaktionsprodukt hydrolysiert, das entstandene 2,6,6-Trimethyl-cyclohexen-(2)-ol-(1)-on-(4) durch Einwirkung eines stark sauren Mittels zum 2,6,6-Trimethyl-cyclohexandion-(1,4) isomerisiert und die Ketogruppe in 4-Stellung zur Hydroxygruppe reduziert und verestert.
Das als Ausgangsmaterial verwendete 2,6,6-Tri- methyl-cyclohexen-(l)-on-(4) kann beispielsweise aus Isophoron hergestellt werden.
In der ersten Stufe des erfindungsgemässen Verfahrens wird das 2, 6,6-Trimethyl-cyclohexen-( l)-on- (4) mit einer organischen Persäure, z. B. Peressigsäure, Benzoepersäure, Phthalmonopersäure oder Perameisensäure, oxydiert. Das unbeständige primäre Oxydationsprodukt (wahrscheinlich eine Oxidoverbindung) wird ohne Isolierung zum 2,6,6-Trimethyl-cyclohexen (2)-ol-(1)-on-(4) hydrolysiert. In einer zweckmässigen Ausführungsform der ersten Verfahrensstufe wird das 2,6,6-Trimethyl-cyclohexen-(l)-on-(4) in Eisessiglösung mit Peressigsäure bei 0-35 C oxydiert, die Oxydationsmischung durch Zugabe von 30%iger Natronlauge schwach alkalisch gestellt und hierauf bei Zimmertemperatur 1-2 Stunden geschüttelt.
Das 2,6,6-Tri- methyl-cyclohexen-(2)-ol-(l)-on-(4) kann durch Extraktion mit Äther isoliert werden und bildet ein fast farbloses Ö1; Siedepunkt 110-112 C/0, 1 mm, und = 1,501, U.V.-Absorptionsmaximum bei 226 rqu (Ei = 1110 in Petrolätherlösung). Das Phenylsemicarbazon des Ketoalkohols schmilzt bei 189-190 C und besitzt im U.V.-Spektrum Absorptionsmaxima bei 240,5 m, und 285 m, (E: = 807 und 778 in Feinsprit).
In der zweiten Stufe des erfindungsgemässen Verfahrens wird das 2,6,6-Trimethyl-cyclohexen-(2)-ol (l)-on-q) zum 2,6,6-Trimethyl-cyclohexandion-(1,4) isomerisiert. Diese Isomerisierung erfolgt durch Einwirkung stark saurer Mittel. Hierzu eignen sich starke Säuren wie Schwefelsäure, p-Toluol-sulfonsäure, Phosphorsäure usw., stark sauer reagierende Salze, wie Zinkchlorid, Kaliumbisulfat usw., oder Ansolvosäuren (d. h. Säuren, die keine Protonen abgeben), wie z. B. BF3. Vorteilhaft erwärmt man das 2,6,6- Trimethyl-cyclohexen-(2)-ol-(1)-on-(4) mit einer geringen Menge (1-10% bezogen auf den Ketoalkohol) des stark sauren Mittels.
Eine zweckmässige Aus führungsform dieser Stufe besteht darin, dass man das 2,6,6-Trimethyl-cyclohexen-(2)-o1-(l)-on-(4) in einem inerten Lösungsmittel, beispielsweise Benzol oder Toluol, mit 2% p-Toluolsulfonsäure, bezogen auf das Gewicht des Ketoalkohols, kocht. Das gebildete 2,6,6-Trimethyl-cyclohexandion-(l 4) bildet farblose Kristalle von kampferähnlichem Geruch, Schmelzpunkt 63-65 C, welche im U.V.-Absorptionsspektrum zwischen 220 und 280 my kein Maximum aufweisen.
Das Phenylsemicarbazon des Dions schmilzt bei 218 bis 220 C und besitzt im Ü.V. -Absorptionsspektrum ein Maximum bei 250 mm (es = 1030 in Feinsprit).
Die Reduktion des 2,6, 6-Trimethyl-cyclohexan- dions-(1,4) lässt sich beispielsweise durch katalytische Partialhydrierung durchführen. Nach Aufnahme von einem Mol Wasserstoff ist praktisch nur die Ketogruppe in Stellung 4 reduziert. Als Katalysatoren eignen sich Platin, Raney-Nickel usw. Das entstandene 2,6, 6-Trimethyl-cyclohexanon-(l)-ol-(4) be steht aus einer öligen Mischung der beiden möglichen cistrans-Isomeren. Das eine Isomere kristallisiert beim Stehen des Öls aus und bildet farblose Kristalle; Schmelzpunkt 56"C. Das Produkt kann auf übliche Weise verestert werden. Beispielsweise wird daraus durch Einwirkung von Acetanhydrid und Pyridin das 4-Acetoxy-2, 6'6-trimethyl-cyclohexanon-(1) erhalten.
Die Verfahrensprodukte haben Riechstoffeigenschaften und können deshalb als Komponenten in Riechstoffgemischen und Parfüms verwendet werden.
Sie sind ausserdem wertvolle Zwischenprodukte für die Synthese von natürlichen Farbstoffen, wie Zeaxanthin, Kryptoxanthin usw.
Beispiele 2, 6, 6-Trimethyl-cyclohexen- (2)-ol- (1)-on- (4)
Zu 138 g 2,6'6-Trimethyl-cyclohexen-(l)-on-(4) in 50 cm3 Eisessig gibt man innert 2 Stunden bei 0-10 C unter Rühren 160 cm3 einer Lösung von Peressigsäure in Essigsäure (530mg Peressigsäure pro cm3) und lässt die Mischung über Nacht bei 20"C stehen.
Hierauf wird die Mischung unter Zugabe von Eis mit 30%iger Natronlauge schwach alkalisch (pIl etwa -8) gestellt und die Reaktionsmischung 1 Stunde bei 20"C geschüttelt. Nun extrahiert man die Reaktionsmischung zweimal mit je 800 cm3 Äther und wäscht die Ätherextrakte einmal mit 200 cm3 gesättigter Ammoniumchloridlösung. Der nach dem Trocknen der vereinigten Ätherextrakte über Natriumsulfat und Abdampfen des Äthers erhaltene Rückstand wird im Hochvakuum destilliert.
Nach einem zwischen 70 und 80"C übergehenden Vorlauf wird das 2,6,6-Trimethylcyclohexen-(2)-ol-(1)-on-(4) als fast farbloses Ö1 erhalten; Siedepunkt 110-112 C/0, 1 mm, und = 1,501, U.V.-Absorptionsmaximum 226 mm (es = 1110 in Petrolätherlösung) nach einigem Stehen. Phenylsemicarbazon: Schmelzpunkt 189-190"C, U.V.-Absorp- tionsmaxima bei 240,5 und 285 mm (El = 807 und 778 in Feinsprit).
2,6,6-Trimethyl-cyclohexandion- (1,4) a) 100 g 2,6,6-Trimethyl-cyclohexen-(2)-ol-(l)-on (4) werden in 200 cm3 Benzol unter Zugabe von 2 g p-Toluolsulfonsäure über Nacht unter Rückfluss gekocht. Nach dem Abkühlen der Reaktionsmischung gibt man ihr 2 g Natriumbicarbonat zu, mischt gut durch und filtriert durch eine Säule von 200 g neutralem Aluminiumoxyd. Man wäscht die Säule noch mit 800 cm3 Benzol nach, dampft das Filtrat im Vakuum ein und gibt dem noch warmen flüssigen Rückstand unter Umschütteln 500 cm3 Petroläther (Siedebereich 80-110 C) zu. Nach dem Erkalten wird das auskristallisierte 2,6, 6-Trimethyl-cyclohexan dion-(l,4) abfiltriert und bei Zimmertemperatur im Wasserstrahlvakuum getrocknet.
Farblose Nadeln, Schmelzpunkt 63-65 C. b) 10 g 2,6, 6-Trimethyl-cyclohexen-(2)-ol-(1)-on-(4) werden mit 1 g Kaliumbisulfat 20 Stunden bei 100"C erwärmt. Nach dem Erkalten wird das Reaktionsprodukt in Äther gelöst und die Ätherlösung mit Wasser und Natriumbicarbonatlösung gewaschen.
Der nach dem Trocknen der Ätherlösung über Natriumsulfat und Abdampfen des Äthers erhaltene Rückstand wird wie unter a) beschrieben kristallisiert. c) 10 g 2,6,6-Trimethyl-cyclohexen-(2)-ol-(2)-on-(4) werden mit 1 g konzentrierter Schwefelsäure 5 Stunden bei 100"C erwärmt. Die Reaktionsmischung wird wie unter b) beschrieben aufgearbeitet. d) 10 g 2,6,6-Trimethyl-cyclohexen-(2)-ol-(l)-on-(4) werden mit 1 g Zinkchlorid 4 Tage bei 100"C erwärmt. Die Reaktionsmischung wird wie unter b) beschrieben aufgearbeitet. e) 100 g 2,6,6-Trimethyl-cyclohexen-(2)-ol-(1)-on- (4) werden in 200 cm3 Toluol wie unter a) beschrieben behandelt.
Man erhält ebenfalls das in farblosen Nadeln kristallisierende 2,6,6-Trimethyl-cyclohexan- dion-(1,4).
2,6,6-Trimethyl-4-acetoxy-cyclohexanon- (1)
100 g 2,6,6-Trimethyl-cyclohexandion-(1,4) werden in 500 cm3 Methanol in Gegenwart von 0,5 g Platinoxyd bei Zimmertemperatur und Normaldruck hydriert. Nach Aufnahme von 1 Mol Wasserstoff pro Mol des Dions wird die Hydrierung abgebrochen, der Katalysator abfiltriert und die Lösung im Vakuum eingeengt. Beim Stehen des Rückstandes kristallisiert eines der beiden raumisomeren 2,6,6-Trimethyl-cyclohexanol-(4)-one-(l) aus und kann durch Abfiltrieren und Kristallisieren aus Äther und Petroläther in reiner Form gewonnen werden; Schmelzpunkt 56 C, Siedepunkt 600 C/0,02 mm.
Man lässt 200 g 2,6,6-Trimethyl-cyclohexanol-(4)- on-(l) zusammen mit 100 g Pyridin und 100 g Acetanhydrid 15 Stunden bei 20"C stehen. Hierauf wird die Reaktionsmischung auf Eis gegossen und das Produkt durch Extraktion mit Äther ausgezogen. Nach dem Trocknen der Ätherlösung über Natriumsulfat und Eindampfen erhält man das 2,6,6-Trimethyl-4-acetoxycyclohexanon-(l) als fast farbloses Ö1; Siedepunkt 8P86"C/0,12 mm, n24 = 1,4560.
