5 g Rohrzucker, 5 g Difco-Trypton, 1 g Natriumnitrat, 0,5 g sek.
Process for the preparation of oxypregnanes The present invention relates to a process for the preparation of saturated and unsaturated 14-oxy-pregnanes substituted in the 13-position by a free or functionally modified carbinol, formyl or carboxyl group. The new 14-oxy-pregnane and their derivatives, such as B. 14-oxy-aldosterone, show an increased effectiveness compared to the compounds not hydroxylated in the 14-position.
The new 14-oxy-pregnanes are obtained when the corresponding compounds, unsubstituted in the 14-position, are subjected to the action of cultures of fungi of the genera Mucor, Helicostylum, Pleospora or Curvularia or of corresponding enzymes.
In addition to those in the 3- and 20-positions, but especially in the 11-position, the starting materials preferably have a free or functionally modified oxy or oxo group. They can be saturated or contain double bonds, e.g.
B. 1- and / or 4-position, also in 5-, 6-, 7-, 8-, 9:11, 11:12 or 16-position, or additional substituents be seated, such as free or modified Oxy or oxo groups, and also epoxy groups or halogen atoms, for example in the 2-, 4-, 5-, 6-, 7-, 8-, 9-, 12-, 15-, 16-, 17- or 21-position , or methyl groups, e.g. B.
in 17a position. As far as the substituents are concerned, the starting materials can be of any steric configuration and can also be present as racemates. They can also belong to the so-called Nor and / or Homo series, in particular the 19-Nor and D-Homo series. Particularly important starting materials are z.
B. 18-Oxy- and 18-Oxo-progesterone, 17a, 18-Dioxy- and 17a-Oxy-18-progesterone, 18-Oxy- and 18-Oxo-cortexon, 17a, 18-Dioxy- and 17a- Oxy-18-oxo-cortexon, aldosterone, 18-oxy-corticosterone and -11-dehydro-corticosterone, 11-epi-18-oxy- and 11-epi-18-oxo-corticosterone, 17a-oxy-aldosterone,

B. with an aliphatic, aromatic or heterocyclic carboxylic acid, for example acetic acid, propionic acid, benzoic acid or furancarboxylic acid or an etherified oxy groups, z.
B. the tetrahydropyranyl-oxy, benzyloxy or triphenylmethoxy group, into consideration and as functionally modified oxo groups advantageously ketalized or
acetalized oxo groups, derived in particular from a dihydric alcohol, such as the ethylenedioxy group. The starting materials can also have a free or functionally modified carboxyl group, mainly in the 13-position, primarily an esterified or lactonized carboxyl group.
Good results are achieved above all with the following types of fungus and the corresponding enzymes: Pleospora gaeumanni, Mucor griseocyanus, Mucor parasiticus, Helicostylum piriforme and Curvularia pallescens. The media known per se are suitable for the cultivation of these fungi, e.g. B.
those with sugars, such as glucose or lactose, with peptones, corn steep liquor, soy products and the like, and with mineral salts, or synthetic nutrient solutions. One works in particular under aerobic conditions, for example in shaking telkultur, or submerged with stirring and air supply. The fungi mentioned differ from other microorganisms, e.g. B. the bacteria, through good growth under relatively simple cultivation conditions. As I said, the reaction can take place in the mentioned fungal cultures themselves or with the help of the enriched or separated enzymes, so z.
B. in a suspension of the separated mushroom mycelium, the homogenized mushroom mycelium or in filtrates or water-containing extracts th thereof.
The process products can be isolated by known methods. Your separation can z. B. by extracting the reaction mixture with an organic solvent, e.g. B. methylene chloride or ethyl acetate. Chromatography is particularly suitable for the further purification of the extract obtained in this way, e.g. on alumina or silica gel, application of distribution methods, z.
B. the countercurrent process, or the separation using Girard reagents, such as trimethylammonium or pyridinium acetic acid hydrazide. Following these cleaning methods or instead of them, recrystallization can finally be carried out from organic or aqueous organic solvents.
The 14-oxy-steroids obtainable according to the invention and their derivatives are distinguished by an increased effectiveness compared with the therapeutically active compounds which are not hydroxylated in the 14-position. As mentioned, the compounds have a free or functionally modified carbinol, formyl or carboxyl group in the 13-position and preferably a free or functionally modified oxy or oxo group in the 11-position;

If the process products do not have the configuration and the substituents of therapeutically ver usable steroids, they can be used as inter mediate products for their preparation, eg. B. serve the above compounds.
The reaction products obtainable according to the invention can be converted in a manner known per se into their functional derivatives, such as oxygen, sulfur or nitrogen derivatives, for example esters, ethers, enol esters, enol ethers, ketals, thioethers and thioketals, and also hydrazones, oximes and enamines, furthermore, oxy groups can be dehydrogenated to oxo groups.
In the esters and enol esters, the acid radicals can be those of any organic or inorganic acids, such as aliphatic, alicyclic, araliphatic, aromatic or heterocyclic carboxylic, thionic carboxylic, thiol carboxylic or sulphonic acids, preferably of formic acid, acetic acid, chloroacetic acids, trifluoroacetic acid, propionic acid , Butyric acids,

If during the hydrolysis, in particular that with alkaline agents, 9,11- or 12,11-halohydrins have been converted into the corresponding 9,11- or 11,12-oxo compounds, the latter can be converted into the latter by the action of hydrohalic acids, in particular Convert fluoric or hydrochloric acid back into the desired 9,11 or 12,11 halohydrins.
Example 1 For a shaking culture of Pleospora gaeumanni well developed at 28 and 4 days old in 500 cm3 70 percent. aqueous beer wort with 0.5 em3 sperm oil is given a solution of 125 mg aldosterone in 10 cm3 acetone under sterile conditions. The suspension is shaken for a further 4 days at the same temperature. The mycelium is then separated off and washed well with water and ethyl acetate. The united clear solutions are shaken out with ethyl acetate.
The ethyl acetate solutions are washed with water, dried and evaporated in vacuo. The residue is in 80 per cent. Dissolved methanol and extracted several times with petroleum ether. The ethanol solutions are then completely evaporated in a vacuum.
The paper chromatogram of the residue (propylene glycol-toluene) shows, in addition to a little aldosterone, the somewhat slower 14a-oxyaldosterone. The entire residue is separated by means of a preparative paper chromatogram (propylene glycol-toluene). The zones corresponding to 14a-oxy-aldosterone are cut out and treated with 50%. Methanol extracted several times.
The methanol is then removed in vacuo, the remaining aqueous solution is shaken out several times with ethyl acetate, the combined ethyl acetate solutions are washed with water, dried and evaporated in vacuo, pure 14a-oxyaldosterone being obtained as residue.
The incubation of aldosterone can also be carried out in 500 cm3 of a well-developed aqueous culture of Curvularia pallescens containing the following additives:
5 g cane sugar, 5 g Difco tryptone, 1 g sodium nitrate, 0.5 g sec. Potassium orthophosphate, 0.25 g magnesium sulfate, 0.25 g potassium chloride, 5 mg iron sulfate heptahydrate, 1.25 g calcium carbonate and 0.5 cm3 sperm oil. The work-up is carried out as indicated above.
The 14a-oxy-aldosterone obtained in this way can be esterified as follows: 0.2 cm3 of pyridine and 0.4 cm3 of acetic anhydride are poured over 100 mg. The solution is left to stand for 15 hours at 20 and then evaporated with the addition of water in a normal vacuum and finally in a high vacuum at 35.
The residue obtained is 14a-oxy-aldosterone-18,21-diacetate. Example 2 For a shaking culture of Pleospora gaeumanni well developed at 28 and 4 days old in 500 cm3 70 percent. watery wort with 0,
5 cm3 of sperm oil is given under sterile conditions a solution of 125 mg of 1-dehydro-aldosterone (obtainable from aldosterone by microbiological dehydration in 1-position using the fungus Colo nectria decora) in 10 cm3 of acetone. The suspension is shaken for a further 4 days at the same temperature.
The reaction mixture is then worked up as indicated in Example 1 and the 1-dehydro-14a-oxy-aldosterone formed is also isolated as indicated by means of a preparative paper chromatogram (propylene glycol-toluene), with. it runs a little slower than the 1-dehydro-aldosterone.
If the 1-dehydro-14aoxy-aldosterone obtained in this way is poured over with 0.2 cm3 of pyridine and 0.4 cm3 of acetic anhydride, the solution is left to stand for 15 hours at 20 and then evaporated in a normal vacuum and finally in a high vacuum at 35, this gives 1-dehydro-14a-oxy-aldosterone-18,21-diacetate is the residue.
Example 3 125 mg (18 -> lss) lactone of 44-3,20-dioxolss-oxy-pregnen-18-acid are obtained as in Examples 1 and 2 with a culture of Pleospora gaeumanni implemented. The reaction product is also worked up as there and the resulting (18 -> 11ss) lactone of 44-3,20-dioxo-llss, 14a-dioxy-pregnene-18-acid isolated as indicated.
