Mundpflegemittel
Vorliegende Erfindung bezieht sich auf ein Mundpflegemittel, das ein wasserlösliches Chlorophyllderivat und eine mit dem Rest einer höheren aliphatischen Carbonsäure N-acylierte Aminoearbonsäure enthält.
Die Mundpflege und Mundhygiene umfasst eine grosse Zahl der versehiedensten zu berücksichtigenden Faktoren, worunter u. a. auch die Anwesenheit gewisser Mikroorga- nismen, die im Munde aus Kohlehydraten Säuren bilden, der Mund- und Atemgeruch, die Reinigung der Zähne und Zahnfleisch- gewebe ohne Reizung usw.
Das erfindungsgemässe Mundpflegemittel ist für die Pflege und Hygiene des Mundes äusserst wirksam und besitzt viele erwünschte Eigenschaften. So übt das Mundpflegemittel eine verlängerte Wirkung in bezug auf die Verhinderung der Säurebildung aus, die mit einer wesentlichen Herabsetzung oder Ausschaltung der unerwünschten Mund-und Atemgerüche einhergeht. Es übt ferner auf die Mundhöhle und das Zahnfleischgewebe ohne Reizung eine erfrischende Wirkung a. us.
Geeignete N-acylierte Aminocarbonsäuren sind solche, deren Acylrest z. B. 12-16 Kohlenstoffatome enthält. Optimale Ergeb nisse erhält man z. B. mit den Dodecanoyl-, Tetradecanoyl-, Hexadecanoylderivaten. Die Formel solcher Verbindungen ist :

in der R-CO ein gesättigter Carbonsäurerest mit 12-16 Kohlenstoffatomen, R'Wasser- stoff oder eine niedrige Alkylgruppe (vorzugsweise Methyl) und X Wasserstoff oder ein lösliche Salze bildendes Kation bedeuten.
Diese Verbindungen lassen sich herstellen durch Kondensation einer den höheren ali phatisehen Carbonsäurerest liefernden a. cy- lierenden Substanz mit einer geeigneten Aminocarbonxsäure, Der von der Aminocarbonsäure herrührende Teil dieser aktiven Komponenten ist im allgemeinen abgeleitet von niedrigen, gesättigten aliphatischen Aminocarbonsäuren, mit bis zu 6-Kohlenstoff- atomen, in. der Regel von Monocarbonsäure- derivaten. Die Verbindungen leiten sich vorzugsweise von den a. minosubstituierten Alkan- carbonsäuren, z. B. den mono-aminosubsti tmerten Alkanmonocarbonsäu. ren, insbeson- dere den a-aminosubstituierten Säuren ab.
Geeignete Aminoearbonsäuren können von Glycin (NH2-CH2-COOH), Sarcosin (Nmethyl-glycin), Alanin (2-Aminoä, thancarbon- säurem, 3-Aminoäthancarbonsäure und Valin abgeleitet werden. Vorzugsweise verwendet man Aminocarbonsäuren mit etwa 2-5 Kohlenstoffatomen und insbesondere die Sarcosin- derivate und Homologe mit bis zu 5 Kohlen- stoffatomen im Aminosäureteil des Moleküls, da diese ausgezeichnete Ergebnisse zeitigen.
Die N-acylierten Aminocarbonsäuren werden in der Regel in Form der. freien Sä. uren oder vorzugsweise deren wasserlöslichen Salze verwendet. Im allgemeinen verwendet man die Alkalimetall (z. B. Na-, K-Salze usw.), Ammonium-, Guanidin-, Amin-und Alkylola. minsalze (z. B. Mono-, Di- und Tri äthanola. minsalze). Weitere spezielle Bei- spiele sind Na-N-lauroyl-sarcosinat, Na-N Myristoyl-sarcosinat, Na-N-iPalmitoyl-sarcosi nat, N-Lauroyl-sarcosin, na-N-lauroyl-glycinat. Die Alkalimetallsalze scheinen ungefähr die gleiche Wirksamkeit, wie andere wasserlösliche Salze zu besitzen und liefern in der Regel optimale Ergebnisse.
Diese N-acylierten Aminocarbo. nsäuren sollten in reiner oder praktisch reiner Form verwendet werden. Sie sollten praktisch frei sein von Seifen oder ähnlichem Fettmaterial, das bei den üblichen technischen Arbeits- weisen entsteht und die speziellen Eigenschaften der N-acylierten Aminocarbonsäuren neutralisiert oder wesentlich herabsetzt. In der Praxis sollte der Anteil an Seife weniger als 15"/o der N-acylierten Aminocarbonsäure betragen.
Die wasserlöslichen Chlorophyllderivate können von natürlichem Chlorophyllmaterial oder von verschiedenen öllöslichen Materialen und Derivaten, wie Phäophytinen, Phäophor- binen und Chlorophyllinen abgeleitet sein.
Beispiele für geeignete wasserlösliche Chlorophyllderivate (Chlorophyllin), die erfin dungsgemäss verwendet werden können, sind

Magnesium-Chlorphyllin-Natrium,
Nickel-Chlorophyllin-Natrium, Eisen-Chlorophyllin-Natrium,
Kwpfer-Chlorophyllin-Ealium,
Magnesium-Chlorophyllin-Kalium.
Das wasserlösliche Chlorophyllderivat und die N-acylierte Aminocarbonsäure üben ihre günstige Wirkung je nach der speziellen Mi sehung und deren Bestandteile in verschiedenen Mengen aus. Zur Erzielung bester oder wesentlicher Effekte sind jedoch die gegen seitigen Mengenverhältnisse von Bedeutung.
Im allgemeinen wird die N-acylierte Amino ca. rbonsäure in Mengen bis zu 5 Gew. /o, vor zugsweise von 1-4 Gew. /o, und das Chloro phyllderiva t in Mengen bis zu 1%, vorzugs weise 0, 01-1' /o der Mischung verwendet. Es wurde gefunden, dass das Gewichtsverhältnis Chlorophyllderivat zu N-acylierter Amino carbonsäure 1 : 500 bis 1 : 2 und vorzugsweise 1 : 100 bis 1 : 4 sein sollte, um eine Zusam menwirkung und günstige gegenseitige Be einflussung der beiden Komponenten zu er reichen.
Es ist für die N-acylierten Amino carbonsäuren charakteristisch, dass sie in Ge genwart von Speichel Proteinstoffe adsor bieren, die wieder freigesetzt werden, was zum Teil die säurehemmende Wirkung im
Munde während längerer Zeit gewährleistet.
Das wasserlösliche Chlorophyllderivat kann von geeigneten Proteinstoffen und andern adsorbierenden Flächen adsorbiert werden.
Obschon man z. Zt. noch nicht weiss, in welchem Ausma. ss eine bevorzugte oder selek tive Adsorption zwischen den zwei Bestand teilen stattfindet, wurde beobachtet, dass übermässige Anteile an Chlorophyllderivaten, die wesentlich über den oben angegebenen maximalen Mengen liegen, eine deutliche
Abnahme der Aktivität und Wirksamkeit der N-acylierten Aminoca. rbonsäure zur
Folge haben, so dass die Hemmung dei
Säurebildung gehindert wird. Mundpflege mittel, deren Gehalt an den Hauptbesta. nd- teilen jedoch im Bereiche der angegebenen
Grenzen liegt, zeigen jedoch eine deutliche und wesentliche Herabsetzung der.
Säure- bildung sowie der Mund-und Atmungs- gerüche, die bei getrennter Anwendung der betreffenden Bestandteile nicht erreichbar ist.
Ausserdem unterstützen die Komponenten gegenseitig die Löslichkeit, wodurch eine bessere Wirkung und Eindringung im
Munde erzielt wird.
Diese Kombination kann in jeder für die
Verwendung im Munde geeigneten Zuberei- tungsformen Anwendung finden, z. B. als
Zahnpasta, Zahnereme, Zahnpulver, flüssige
Zahnpflegemittel, Mundwasser, Tabletten
Pastillen, Kaugummi usw. Diese Präparate können verschiedene Hilfsstoffe in geeigneten Mengen enthalten, sofern diese keinen wesentlichen sehädigenden Einfluss auf die verbesserten Eigenschaften der Mischung aus wasserlöslichen Chlorophyllderivat und Nacylierter Aminoca. rbonsäure ausüben.
Dieser Mischung kann man irgendein geeignetes, in Wasser praktisch unlösliches Poliermittel, wie Calziumkarbonat, Di-und Tricalziumphosphat, unlösliches Nairmm- metaphosphat, Aluminiumhydroxyd, Magne siumkarbonat und Bentonit zusetzen. Die Verwendung wasserunlöslicher Calzium- und Magnesiumsalze, insbesondere Calziumkarbonat und/oder-phosphat, wird bevorzugt. Im allgemeinen bilden diese Poliermittel ge wichtsmässig den Hauptanteil der festen Bestandteile und unterstützen die Reinigung des Mundes.
Zur Herstellung von Zahnpulvern genügt es in der Regel, die verschiedenen Bestandteile mechanisch miteinander zu vermischen.
Bei Zahncremerezepten ist der Gehalt an flüssigen und festen Bestandteilen so aufeinander abzustimmen, dass eine pastöse Masse entsteht, die sich aus Tuben ausdrücken lässt.
Die Flüssigkeit in den Zahnpasten wird im allgemeinen vorwiegend Wasser, Glycerin, Sorbit, Propylenglykol und dergleichen so- wie geeignete Mischungen dieser Stoffe enthalten. Es ist gewöhnlich vorteilhaft, eine Mischung zu verwenden, die sowohl Wasser als auch ein Feuchthaltungs-oder Bindemittel wie Glycerin oder Sorbit enthält. Die Ge samtmenge der Flüssigkeit kann etwa 20 bis 75 Gew. des Präparates betragen.
Es empfiehlt sich, den Mundpflegemitteln ein Stabilisiermittel zuzusetzen. Solche Mittel sind die Ca-und Mg-Ionen unterdrüekenden Stoffe, die bewirken, dass sowohl das wasserlösliche Chlorophyllderivat als auch die Nacylierte Aminocarbonsäure in ihrer aktiven Form erhalten bleiben, besonders wenn die Zahnpflegemittel Calzium-und Magnesium enthaltende Poliermittel enthalten. Diese Zu sä. tze bewirken eine erhöhte Stabilität der Zahncreme, so dass die aktiven Bestandteile ihre Wirksamkeit selbst nach langer Lagerung beibehalten.
Geeignete Stabilisiermittel sind Ca-und Mg-bindende Mittel, wie z. B. wasserlösliche anorganische Polyphosphatsalze. Beispiele hierfür sind die Alkalimetallsalze der Pyrophosphor-, Tripolyphosphor-oder Tetraphos- phorsäuren, insbesondere Tetranatriumpyrophosphat, Dinatriumpyrophosphat, Kalium tripolyphosphat, Hexanatriumtetraphosphat und Natiumhexametaphosphat, wobei Pyrophosphate bevorzugt werden. Man kann auch lösliches Natrium-und Kaliummetaphos- phat verwenden.
Andere geeignete Stabilisier- mittel sind chelatbildende Mittel (Komplex- bildner), wie ALkalimetall und vorzugsweise Na°Salze der Äthylendiamin-tetraessigsäure und Zitronensäure. Diese Stabilisiermittel können in jeder geeigneten Menge, die je nach der besonderen Mischung bis zu 5%, vorzugsweise 0,1-3 Gew.%, betragen kann, verwendet werden.
In die Präparate kann man insbesondere, wenn es sich um Zahnpasten handelt, noch andere Hilfsstoffe in geeigneten Mengen einarbeiten. Vorzugsweise verwendet man Geliermittel, wie natürlicher Traganth, Polyvinyl- pyrrolidon, Na-Carboxymethylcellulose und Stärke. Ferner kann man lösliches Saccharin, geschmacks-und geruchgebende Öle, Konser- vierungsmittel, Alkohole und dergleichen im gewünschten richtigen Mengenverhältnis zusetzen. Weitere geeignete Zusätze sind die verschiedenen Ammoniakderivate, wie Harn- stoff, Diammoniumphosphat und Mischungen solcher Stoffe, die im Munde Ammoniak freisetzen.
Typische Rezepte enthalten 3 /o Harnstoff mit 5 /o Diammoniumphosphat und 10-25% Harnstoff mit 5% Diammo niumphosphat.
Das erfindungsgemässe Mundpflegmittel ka. nn auch in Form eines Mundwassers und Spülmittels verwendet werden. Diese enthalten gewöhnlich eine wirksame Menge an wasserlöslichem Chlorophyllderivat und an N-acylierter Aminocarbonsäure in einem ge- eigneten flüssigen Trägermaterial, vorzugs- weise wässrig-alkoholischer Natur, gelöst oder dispergiert. Die Alkoholkonzentration kann 5-70%, vorzugsweise 5-405, betragen.
Das erfindungsgemässe Mundpflegemittel kann auch in Form eines flüssigen Zahnreini gungsmittels, das in der Regel Schleimstoffe und gewünschtenfalls kleine Mengen Polier- mittel lmd Glycerin enthält, verwendet werden.
Eine weitere Ausführungsform des Erfindungsgegenstandes ist Kaugummi, der eine kleinere aber wirksame Menge der aktiven Bestandteile in der Gummigrundsubstanz, wie Chicle oder andere natürliche oder synthetische Gummi oder gummiartigen Materialien einverleibt enthöält.
Das pg der fertigen Präparate ist variabel und die Produkte können in Lösung einen beliebigen, mit ihrer Stabilität verträglichen pH-Wert aufweisen. Es ist allgemein wünschenswert, dass das peu des Produktes bei den angegebenen Prüfungsbedingungen im Bereich von 5-10, vorzugsweise zwischen 5, 5 und 9, liegt, wobei die optimalen Effekte bei praktisch neutralem pg von etwa 6-8 der LÏsungen erzielt werden. Die pu-'verte beziehen sich entweder auf ein flüssiges Produkt oder bei Pasten und Pulvern und ähnlichen Produkten auf 20%ige wässrige Auf schlämmungen.
Nachstehend werden einige spezielle Beispiele von erfindungsgemässen Mund-und Zahnpflegemitteln gegeben.
