Verfahren zur Herstellung von 7 DiketoScyclopentanopolyhydrophenanthrenen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung-von 7, 11-Diketo syclopentanopolyhydrophenanthrenen aus den entsprechenden z18 (9)-7-Keto-ll-aeyloxy-eyelo- pentanopolyhydrophenanthrenen. Die erfin dungsgemäss hergestellten Verbindungen sind wertvolle Zwisehenprodukte für die Herstel hng von Steroidhormonen, welehe ein sauerstofftragendes C-Atom in 11=Stellung besitzen, zum Beispiel von Adrenalhormonen, wie Corticosteron, Cortison und Verbindung F.
Die erfindungsgemäss erhältlichen 7, 11-Di keto-eyelopentanopolyhydrophenanthrene be- sitzen in den Ringen B und C die folgende Struktur :

Das erfindungsgemässe Verfahren ist dadurch gekennzeichnet, dass man einerseits die @8(9)-Doppelbindung eines @8(9)-7-Keto-ll-acyloxy-pyelopentanopolyhydrophenanthrens hy- driert und anderseits die Acyloxygruppe hy drolysiert, worauf man das gebildete 7-Keto11 - oxy - eyelopentanopolyhydrophenanthren dnich Behandeln mit einem Oxydationsm. it- tel in die 7, 11 - Diketo - Verbindung berführt.
In der Regel erfolgt die Hydrierung vor der Hydrolyse, man kann aber die letztere auch vorausnehmen.

Zu den als Ausgangsmaterial f r das er findungsgemässe Verfahren in Frage kommen den ds (9)-7-Keto-11-acyloxy-cyclopentanopoly- hydrophenanthrenen gehören solche, welche am C-Atom in 17-Stellung eine Sterinseitenkette aufweisen, wie etwa @8(9) ,22-3,11-Diacyl- oxy-7-keto-ergostadien, @8(9)-3,11-DiaeyIoxy-7- keto-cholesten und @8(9),22-3,11-Diacyloxy-7- keto-stigmastadien, solche, die eine Gallensäureseitenkette am C-17-Atom auf- weisen, wie @8(9) - 3,11 - Diacyloxy - 7 - keto- cholsäuren, solche mit einer abgebauten Gal lesÏuresertenkette, wie @8(9)-3,
11-Diaeyloxy- 7-keto-bisnoreholsäuren, solche mit einer Car})- oxylgruppe, wie z18 Diacyloxy-7-keto äthiocholsäure, solche mit einer Acylgruppe in 17-Stellung, wie A8 (9)-3, 11-Diacyloxy-7-keto- pregnen, solche mit einer Sapogeninseiten- kette, wie @(9)-7-Keto -11 - aeyioxy - dehydrotigogeninaeylat und dergleichen.
Die Hydrierung der @8(9)-7-Keto-11-acyl- oxy-cyclopentanopolyhydrophenanthrene wird gewöhnlieh durchgeführt, indem die Verbindungen mit Zinkstaub und Essigsäure oder auch mit Wasserstoff in Gegenwart eines Platinkatalysators zusammengebracht werden.
Zur Hydrolyse der gebildeten 7-Keto-11- acyloxy-cyclopentanopolyhydrophenanthrene zu den entspreehenden 7-Keto-11-oxy-eyelo- pentanopolyhydrophenanthrenen wird mit Vorteil die alkoholische Lösung eines Alkali- hvdroxydes verwendet, etwa eine Lösung von Natrium oder Kaliumhydroxyd in Methanol oder Äthanol.
Die 7-Keto-11-oxy-Verbindung wird dann zum Beispiel mit Chromsäure oxydiert, wobei das entsprechende 7, 11-Diketo-eyelopentano- polyhydrophenanthren entsteht. Diese Oxy dation kann unter Verwendung von Chromsäure als Oxydationsmittel etwa so ausgeführt werden, dass man die 7-Keto-11-oxy-Verbindung in einem. gegen Chromsaure praktisch inerten organischen Losungsmittel l¯st, eine Lösung von Chromsäure in wässeriger Schwe- felsäure zusetzt und die Mischung etwa bei Zimmertemperatur ungefähr eine Stunde reagieren lϯt.
Beispiel
0, 100 g ?8(9),22-3,11-Diacetoxy-7-keto-ergostadien wurden in 5 em3 Essigsäure gelost, 0, 1 g Zinkstaub zugefügt und die Mischung wahrend etwa dreier Stunden auf 100 C er hitzt. Dann wurde die Reaktionsmisehung filtriert, das Filtrat mit Wasser verdünnt und das sich ausseheidende, gummiartige Material mit Benzol extrahiert, wodurch 22-3, 11-Di- acetoxy-7-keto-ergosten in fester, amorpher Form erhalten wurde.
Das so hergestellte @22-3,11-Diacetoxy-7- keto-ergosten wurde in 5 cm3 Sprozentiger, methanolischer Natriumhydroxydl¯sung aufgelost und die Lösung etwa eine Stunde am R ckflu¯ gekoeht. Die Reaktionslösung wurde dann mit Wasser verdünnt und das sich aus seheidende, gummiartige Material durch Dekantation abgetrennt, gewaschen und getrock- net. Die amorphe, feste Masse war @22-3, 11 Dioxy-7-keto-ergosten.
Nun wurde dieses in 5 em3 Aceton gelöst und der Lösung eine L¯sung von 7 mg Chromsäure in 1 cm3'2n-Schwefelsäure zugefügt.
Die erhaltene Mischung blieb etwa 20 Minuten bei Zimmertemperatur stehen, dann wurde im Vakuum eingedampft und der Rüekstand mit Ather extrahiert. Der Ätherextrakt wurde an sauer gewaschenem Aluminiumoxyd chro- matographiert, wodurch praktiseh reines @22- 3-Oxy-7, 11-diketo-ergosten mit einem Schmelz- punkt von 199 bis 200 C erhalten wurde.

The resulting mixture was left to stand for about 20 minutes at room temperature, then it was evaporated in vacuo and the residue extracted with ether. The ether extract was chromatographed on acid-washed aluminum oxide, whereby practically pure @ 22-3-oxy-7,11-diketo-ergosten with a melting point of 199 to 200 ° C. was obtained.
