Verfahren zur Herstellung von Pyrazolonderivaten 5-Pyrazolone, die in 1-Stellung einen Ben- z-lrest und in 4-Stellung eine Dialkylamino- gruppe tragen, sind bislang nicht bekannt geworden.

wie katalytisch erregtem Wasserstoff, Amei sensäure, ZinkstaubAIineralsäure oder Zink- staub/Essigsäure, umsetzt. Als Ausgangsstoff wird das 1-Benzyl-2,3- dimethyl-pyrazolon-(5) verwendet.
Man arbeitet nach dem Verfahren gemäss der Erfindung in der Weise, dass man das 1 -Beilzyl-2,3-dimethyl-pyrazolon-(5) in be kannter Weise, z. B. unter Kühlung mit Na trittmnitrit in schwefelsaurer Lösung, in 4 Stellung nitrosiert. Die 4-Nitrosoverbindung lässt sich mit Natriumbisulfit zu Sulfamin- säure reduzieren und durch Verkochen mit einer Mineralsäure in das Amin überführen.
An Stelle des Natriumbisulfits können auch andere gebräuchliche Reduktionsmittel, wie Zinkstaub in mineral- oder essigsaurer Lösung, wobei man bei mittleren Temperatu ren, vorzugsweise bei 50 bis 60 C, arbeitet, oder Wasserstoff in Gegenwart von Kataly satoren der B. Gruppe des periodischen Sy stems, Verwendung finden.
Die 4-Aminoverbinclung lässt sich nach den verschiedensten Methoden alkylieren. Durch Kochen mit Formaldehyd und Ameisensäure erhält man aus ihr beispielsweise die Di- methylaminoverbindung. Die Ameisensäure kann hierbei durch andere Reduktionsmittel, z. B. Zinkstaub-Schwefelsäure, Zinkstaub-Es- sigsäure oder Wasserstoff in Gegenwart von Hydrierungskatalysatoren, wie Nickel, Palla dium, Platin, ersetzt werden. Ebenso kann inan an Stelle des Formaldehyds auch seine Polymeren, z.
B. Paraformaldehy d, einsetzen. Verwendet man an Stelle des Formaldehyds Acetaldehyd, so lässt sich die Aminogruppe auch durch Äthylreste substituieren.
Die Hydrierung mit Wasserstoff kann man so vornehmen, dass man die 4-Nitrosoverbin- dung in Methylalkohol löst und nach Zugabe des Katalysators mit Wasserstoff unter Druck, vorteilhaft zwischen 50 und 100 atm, und bei mässig erhöhten Temperaturen, vorzugsweise zwischen 25 und 30 C, bis zur Beendigung der Wasserstoffaufnahme behandelt.
Anschlie ssend hydriert man nach Zugabe von 30o/oiger wässriger Formaldehydlösung bei höheren Temperaturen, vorzugsweise bei 100 C, weiter bis zur Beendigung der Wasserstoffaufnahme. Man kann jedoch die Hydrierung auch so durchführen, dass man die Forznaldehydlösung sofort zusetzt, die Temperatur bis zur Beendi gung der Wasserstoffaufnahme zunächst bei etwa 25 bis 30 C hält und anschliessend bei etwa 1000 C zu Ende hydriert.
Für die Durchführung des Verfahrens hat sich herausgestellt, dass es häufig unnötig ist, die Aminov erbindung zu isolieren. Vielmehr kann man direkt von der 4-Nitroso-Verbin- dung ausgehen, die man in den meisten Fäl len ebenfalls nicht aus dem Reaktionsgemisch abzutrennen braucht.
Die nach dem angegebenen Verfahren her gestellten Produkte zeichnen sich besonders durch gute antipyretische und analgetische Wirkung aus. So zeigt beispielsweise das 1- Benzy 1- 2,3 - dimethyl-4-dimethylamino-pyra.zo- lon-(5) eine bedeutend stärkere analgetische Wirkung als das 1-Phenyl-2,3-dimethyl-4-di- methylamino-pyrazolon-(5) und ist leichter wasserlöslich.
<I>Beispiel 1</I> 202 Gewichtsteile 1-Benzyl-2,3-dimethyl- pyrazolon-(5) werden fein gepulvert und mit 500 Voltanteilen Wasser versetzt. Unter Küh lung durch Eis lässt man bei 0-50 C Innen temperatur gleichzeitig 550 Volumteile 2n- Schwefelsäure und 500 Volumteile 2ti-Na- triumnitrit-Lösung zufliessen.
Hierbei schei det sieh die grüne 4-Nitrosoverbindumg aus, die abgesaugt. und mit den üblichen Reduk tionsmitteln wie l@Tatriumliydrosulfit, Zink staub in mineralsaurer oder essigsaurer Lö sung, oder Wasserstoff in Gegenwart von Katalysatoren der Edelmetall- oder Nickel gruppe in die 4-Aminov erbindung übergeführt wird, welche nach Umkristallisation aus Ben zol oder Äthylaeetat bei 54 C schmilzt.
Sie ist leicht. löslich in _VVasser, mit Benzaldehyd erhält man aus ihr eine Benzyliden-Verbin- dung vom Schmelzpunkt 158 C.
22 Gewichtsteile des so erhaltenen 1-Ben- zyl-2,3-dimethyl-4-amino-pyrazolon-(5) wer den in<B>1.00</B> Volumteilen Wasser mit 25 CTe- wiclitsteilen 85o/oiger Ameisensäure und 25 Gewichtsteilen 30 o/aigem, wässrigem Formalde hyd 1.0 Stunden lang gekoelit. Nach dem Er kalten versetzt man mit überschüssiger kon zentrierter Natronlauge und schüttelt mehr mals mit Benzol aus.
Nach dem Trocknen und Abdestillieren des Benzols erhält man 22 Gewichtsteile 1-Benzyl-2,3-dimethyl-4-dime- thyjamino-pyrazolon-(5), das nach dem Um kristallisieren aus Cj'elohexan oder Äthylace- tat bei 75 C schmilzt. Statt durch Umkristal- lisieren kann man die erhaltene Verbindung auch durch Vakuumdestillation reinigen. Sie siedet bei 0,8 mm Druck bei 173-174 C.
Zu derselben Verbindung gelangt man, wenn man das 1-Benzyl-2,3-dimethyl-4-amino- pyrazolon-(5) in schwefelsaurer oder essig saurer Lösung bei 50-60 C mit Zinkstaub unter gleichzeitigem Zufügen von Formalde hyd oder Paraformaldehyd behandelt, oder wenn man es in Gegenwart von Formaldehyd oder Paraformaldehyd katalytisch mit Edel metallkatalysatoren bei geringem überdruck oder mit Nickelkatalysatoren unter erhöhtem Druck hydriert.
Beispiel N 202 Gewichtsteile 1-Benzyl-2,3-dimethl- py razolon-(5) werden, wie in Beispiel 1 be schrieben, nitrosiert. kocht 3 Stunden lang und führt danach die Methylierung durch Zugabe von Form aldehyd und Zinkstaub durch. Nach Neutrali sation mit konzentrierter Natronlauge und Ausrühren mit Benzol erhält man 200 Ge wichtsteile 1-Benzyl-2,3-dimethyl-4-dimethyl- amino-pyrazolon-(5), das durch Umkristalli- sieren oder Vakuumdestillation gemäss Bei spiel 1 gereinigt werden kann.
Beispiel <I>3</I> 202 Gewichtsteile 1-Benzyl-2,3-dimetKyl- pyrazolon-(5) werden, wie in Beispiel 1 be schrieben, nitrosiert, mit Bisulfit reduziert und anschliessend nach Zugabe von konzen trierter Schwefelsäure 3 Stunden lang ge kocht. Hierauf neutralisiert man mit Natron lauge, versetzt mit 270 Gewichtsteilen 85pro- zentiger Ameisensäure und<B>195</B> Gewichtstei len 4019/oigem wässrigem Formaldehyd und kocht 10 Stunden lang.
Nach Beendigung des Kocheis kühlt man ab, versetzt mit überschüs siger konzentrierter Natronlauge und arbeitet weiter wie in Beispiel 2 auf. Man erhält 200 Gewiehtsteile 1-Benzyl-2,3-dimethyl-4-dime- thylamino-pyrazolon- (5).
Beispiel <I>4</I> 23 Gewichtsteile 1-Benzyl-2,3-dimethyl-4-ni- troso-pyrazolon-(5) (Zersetzungspunkt 132 C), erhalten nach der in Beispiel 1 angegebenen Voi@sehrift, werden mit.
200 Volumteilen Me thanol, 25 Gewiehtsteilen wässrigem, 30o/aigem Formaldehyd und einem Gewichtsteil Palla- dium-Mohr mit Wasserstoff bei geringem t'berdruck bis zur Beendigung der Wasser stoffaufnahme hydriert. Nach Abtrennen vom Katalysator und Abdestillieren des Lösungs mittels erhält man in nahezu quantitativer Ausbeute das 1-Benzyl-2,3-dimethyl-4-dime- thyl-amino-pyrazolon-(5),
das durch Umkri- stallisieren weiter gereinigt werden kann. <I>Beispiel 5</I> 23 Gewichtsteile l.-Benzyl-2,3-dimethyl-4- iiitroso-pyrazolon-(5), erhalten nach der in Beispiel 1 angegebenen Vorschrift, werden mit 250 Volumteilen Methanol und 2,5 Ge wichtsteilen Raney-Nickel unter 50-100 atm Wasserstoffdruck bei 25-30 C bis zur Be endigung der Wasserstoffaufnahme behandelt.
Danach gibt. man 25 Gewichtsteile 30 o/oigen, wässrigen Formaldehyd hinzu und hydriert weiter bei 100 C bis zur Beendigung der Wasserstoffaufnahme. Nach Aufarbeitung ge mäss Beispiel 4 erhält man das 1-Benzyl-2,3- dimethyl-4-dimethylamino-pyrazolon-(5) in nahezu quantitativer Ausbeute. .
Man kann die Hydrierung auch so durch führen, dass man den Formaldehyd sofort zu setzt, zunächst bei Temperaturen von 25 bis 30 C bis zur Beendigung der Wasserstoffauf nahme arbeitet und danach bei 100 C zu Ende hydriert.
<I>Beispiel 6</I> 22 Gewichtsteile 1-Benzyl-2,3-dimethyl-4- amino-pyrazolon-(5), erhalten nach der in Beispiel 1 angegebenen Vorschrift, werden in 150 Volumteilen Wasser gelöst, mit 30 Ge wichtsteilen konzentrierter Schwefelsäure ver setzt und bei 50-60 C mit 50 Gewichtsteilen Zinkstaub unter Zutropfen von 15 Gewichts teilen Acetaldehyd behandelt.
Nach Aufarbei tung durch Neutralisieren und Ausschütteln mit Benzol erhält man das 1-Benzyl-2,3-di- methyl - 4 - diäthylamino - pyrazolon - (5), das durch Destillation im Vakuum und Umkristal- lisieren aus Äthylacetat gereinigt werden kann.
