dadurch gekennzeichnet., dass man 4'-Deme- thy 1-podophyllotoxinglueosid mit. Diazomethan umsetzt, und den gebildeten Methyläther iso liert.
Es ist bekannt, dass die Rhizome von Po- dophyllumarten Wirkstoffe enthalten, die dank ihrer antimitotischen, tumorschädigen- den Wirkung krebsartige Tumoren in thera peutisch günstigem Sinne beeinflussen kön nen.
In Helv. Chim. Acta 37, 1747 (1954) wurde ein Verfahren beschrieben zur Isolie- ning von Podophyllotoxinglucosid aus den Rhizomen von Podophyllum emodi Wall.
Diesem Glucosid kommt wegen seiner mitose- hemmenden und tumorzerstörenden Wirkung therapeutische Bedeutung zu, zumal da. es sich von dem vorher schon bekannten und untersuchten, in Wasser unlöslichen Podo- phyllotoxin durch seine Wasserlöslichkeit unterscheidet.
Es wurde nun gefunden, dass das ebenfalls in Podophyllum emodi vor kommende 4'-Demethyl podophyllotoxingluco- sid durch Methylierung in das biologisch aktivere Podop-hyllotoxinglucosid übergeführt werden kann. 4'-Demethyl-podophyllotoxin- glucosid kann aus den bei der Reinigung von Podophyllotoxinghicosid nach dem in der oben zitierten Literaturstelle beschriebenen Verfahren anfallenden Mutterlaugen gewon nen werden.
Die wässrigen Lösungen, wie sie nach dem genannten Verfahren nach Abtren nen der Gerbstoffe anfallen, enthalten neben vorwiegend Podophyllotoxinglucosid auch das Dem,ethyl-podophyllotoxinglucosid. Nach Ab trennen des ersteren durch Ausschütteln mit verschiedenen Lösungsmitteln bzw. Lösungs- mittelgemischen (z. B. Chloroform unter Zu satz von Alkoholen) kann die Demethylver- bindung der wäss,rigen Phase, z.
B. mit Bu- tanol entzogen werden. Das aus der Bu- tanolfraktion durch Eindampfen erhaltene Präparat enthält die Demethylverbindung bereits stark angereichert. Es kann nach be kannten Methoden, z. B. durch Chromato- graphie oder Verteilung zwischen Wasser und mit Wasser nicht mischbaren Lösungsmitteln gereinigt werden. Als besonders geeignet er wies sich eine Säulenanordnung mit Silica- gel als Trägermaterial.

Da das Aglykon des Podophyllotoxinglucosids unter dem Einfluss von Hydroxylionen eine Verän derung erleidet, die das Verschwinden der biologischen Aktivität des Glucosids zur Folge hat, muss die Methylierning unter Be dingungen ausgeführt. werden, die das Zu standekommen alkalischer Reaktion verhindern. Aus diesem Grund eignet sich Diazomethan be sonders als Methylierungsmittel. Als Lösungs mittel für das Glucosid kommen z. B. niedere Alkohole, Aceton oder Essigester in Betracht.
Das Dia.zomethan wird, vorzugsweise in äthe rischer Lösung, im Überschuss, bei Zimmer temperatur oder in der Kälte zugegeben. Aus der Reaktionslösung kann das gebildete Podo- phyllotoxinglucosid durch Abdampfen des Lösungsmittels gewonnen und anschliessend durch Umfällen aus Aceton-Äther gereinigt werden. Gegebenenfalls ist die Reinigung noch durch Verteilungschromatographie, z. B. an einer Silicagelsäule und Elution mit Essig ester, zu vervollständigen. Wird z.
B. für die Umsetzung mit Diazomethan nicht die reine 4'-Demethylverbindung, sondern eine an De- methylverbindung angereicherte Fraktion, wie sie z. B. beider Isolierung aus der Droge anfällt, verwendet, dann kann nach der Me- thylierung das Reaktionsprodukt nach dem in der oben zitierten Literaturstelle beschrie benen Verfahren gereinigt werden.
Das Podophyllotoxingliicosid wirkt stär ker mitosehemmend als die Demethylverbin- Jung. Die beiden Glucoside zeichnen sich vor den nichtglucosidischen Inhaltsstoffen der Droge durch die Löslichkeit in Wasser und insbesondere durch ihre stark herab gesetzte Toxizität und damit wesentlich bes sere Verträglichkeit aus.
Das nach dem beschriebenen Verfahren erhaltene Podophyllotoxinglueosid soll thera peutisch verwendet werden.
<I>Beispiel</I> Man destilliert in eine Lösung von 500 mg 4'-Demethyl-podophyllotoxinglucosid in 15 cm3 Methanol so viel einer ätherischen Dia.zomethanlösung, bis sich das Reaktions gemisch bleibend gelb gefärbt hat. Die Lö sung wird nach zweistündigem Stehen bei 0 im Vakuum eingedampft, und der Einda.mpf- rückstand aus Aceton-Äther umgefällt.
Das so gewonnene Podophyllotoxinglucosid besitzt die in Helv. Chim. Acta. 37, 1747 (1954) an gegebenen Eigenschaften: Smp. 152-154 C, [a,] D =-74,7 (in Methanol).

It is known that the rhizomes of pedophyllum species contain active ingredients which, thanks to their antimitotic, tumor-damaging effect, can influence cancerous tumors in a therapeutically beneficial sense.
In Helv. Chim. Acta 37, 1747 (1954) describes a method for isolating podophyllotoxin glucoside from the rhizomes of Podophyllum emodi Wall.
This glucoside is of therapeutic importance because of its mitosis-inhibiting and tumor-destroying effect, especially since. it differs from the previously known and investigated, water-insoluble podophyllotoxin by its water solubility.
It has now been found that the 4'-demethyl podophyllotoxing glucoside, which also occurs in Podophyllum emodi, can be converted into the biologically more active podophyllotoxin glucoside by methylation. 4'-Demethyl-podophyllotoxin glucoside can be obtained from the mother liquors obtained in the purification of podophyllotoxin hicoside according to the process described in the literature cited above.
The aqueous solutions, as obtained after separating the tannins according to the process mentioned, contain not only podophyllotoxin glucoside but also demethyl-podophyllotoxin glucoside. After separating the former by shaking out with various solvents or solvent mixtures (e.g. chloroform with the addition of alcohols), the demethyl compound of the aqueous phase, e.g.
B. can be removed with butanol. The preparation obtained from the butanol fraction by evaporation contains the demethyl compound already highly enriched. It can be according to known methods, for. B. can be purified by chromatography or partitioning between water and water-immiscible solvents. A column arrangement with silica gel as the carrier material proved to be particularly suitable.

The podophyllotoxingliicoside has a stronger mitosis-inhibiting effect than the demethylverbin-Jung. The two glucosides are distinguished from the non-glucosidic constituents of the drug by their solubility in water and, in particular, by their greatly reduced toxicity and thus much better tolerability.
The Podophyllotoxinglueosid obtained by the method described is to be used therapeutically.
<I> Example </I> Distillation is carried out in a solution of 500 mg of 4'-demethyl-podophyllotoxin glucoside in 15 cm3 of methanol so much of an ethereal Dia.zomethane solution until the reaction mixture has become permanently yellow. After standing for two hours at 0, the solution is evaporated in vacuo, and the concentration residue from acetone-ether is reprecipitated.
The podophyllotoxin glucoside obtained in this way has the properties described in Helv. Chim. Acta. 37, 1747 (1954) on the properties given: m.p. 152-154 C, [a,] D = -74.7 (in methanol).
