So können sohc tertiäre Carbinole ; welche in der Stellung 1 oder 2 einen geeigneten ungesät tigten Substituenten besitzen, zum Ringschlu# gebracht werden, wobei der D-Ring eines Stcroidsystems gebildet wird, welches in der Stellung 17 einen funktionalen Substituenten besitzt.
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung neuer, in Stellung I durch ein tertiares Hydroxyl und einen organisehen Rest substituierten Poly- hydrophenanthrene, welches dadurch gekennzeichnet ist, dass Polyhydrophenanthrene, welche im C-Ring die folgende Struktur besitzen

worin R und Ri die gleiche Bedeutung haben wie oben lmd R2 das organische Radikal der Organometallverbindung darstellt. Als Aus gangsstoffe verwendet man vorzugsweise Poly- hydrophenanthrenderivate, welche auch in Stellung 4b eine Methylgruppe tragen. Die Ausgangsstoffe können im Kern gesättigt oder ungesättigt sein und zusätzliche Substituenten an den Ringen A und B des Phenanthrenkernes besitzen. So können die Ringe A und B durci Hydroxyl-, Acyoxy-, Alkyl-, Äther-, Keta, l- und a. ndere Cruppen sub, stitu, iert sein.
Wenn die Ausgangsverbindung reaktionsfähige Ketosubstituenten im Ring A oder B auf weist,reagieren diese Gruppen ebenfa lls unter Bildung eines ditertiären Alkohols. Doch können solche Ketogruppen blockiert werden, indem man sio in eine Enoläther-oder Ketalgruppe umwandelt, die sich durch Hydrolyse 35 40 45 50 55 60 wieder in die Ketogruppe zurüeliverwandeln lassen.
Das erfindungsgemä#e Verfahren ist besonders wertvoll für die Herstellung neuer Dodecahydrophenanthrene der folgenden Formel :

worin R, einen durch Hydrolyse in die Keto gruppe umwandelbaren Substit. uenten bedeu- tet. Diese Verbindungen haben sieh für die Synthese therapeutisch wichtiger Steroidverbindungen, wie Cortison und dergleichen, als wertvoll erwiesen.
Vor allem kommen als Ausgangsstoffe ungesättigte Organometallverbindungen in Betracht, z. B. Grignardverbindungen von sub stituierten oder unsubstituierten Alkenen oder Alkinen. Beispielsweise kann man 2,4b-Dimethyl-2-methallyl-7-äthylendioxy-1,2,3, 4, 4a, 4-1). 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1on-4-ol oder das entsprechende 1, 4-Dion mit einem Magnesiumhalogenid eines Alkens (unter anderem All und dergleichen) oder Alkins (unter anderemAcetylen,Äthoxyaeetylen und dergleichen) zur Reaktion bringen und so naeh der hydrolytisehen Zersetzung die entsprechenden 1-Hydroxy-l-alkenyl-oder 1-Hydroxy-1-alkinyl-derivate erhalten.
Zur Herstell'ung von a-Äthoxyäthinyl-2,4bdimethyl-2-methallyl-7-äthylendioxyd-1,2,3,4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1 ol-4-on beispielsweise wird vorzugsweise eine Losung von 2, 4b-Dimethyl-9-methallyl-7-ä. thylen- dioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydro phenanthren-1, 4-dion in Benzol zu einer benzo- lischen Lösung von Äthoxyacetylen-magnesiumbromid gebracht, welche etwa die 2-5fache theoretische Menge an organometalliseher Verbindung enthält. Nach etwa 2-4stündiger Reaktion bei Zimmertemperatur wird die entstandene Additionsverbindung in wässriger Lö sung zum gewünschten tertiären Alkohol hydrolysiert.
Der tertiäre Alkohol kann in iibl, icher Weise durch Extraktion der Hydro- lysenmiscbung und Eindampfeil des Extraktes isoliert werden.
In analoger Weise kann aueh bei der Verwendung anderer Organomagnesiumhalogenide gearbeitet werden. Statt Benzol kann jedes andere geeignete Lösungsmittel verwendet werden. Da die Grignardverbindungen gewöhn- lich in ätherischem Medium bereitet werden, ist es günstig, d'ass die Addit. ionsreaktion auch in Äthyläther gut verläuft. Auch andere Lo- sungsmittel wie Toluol, Xylol, isopropyläther oder KohIenwasserstoff-Äther-Misehungen geben befriedigende Resultate.
Die angewendete Menge der Organometallverbindung selieint nichet kritisch zu sein. Zum Erreichen optimaler Ergebnisse wird gewöhn- lich das 2-bis 5faehe der theoretischen Menge zugesetzt ; doch wird der Reaktionsablauf durch Anwendung odeur oder kleinerer Mengen der Organometallverbindung nicht ungünstig beeinflusst. Bei der Verwendung eines Polyhydrophenanthren-4-ol-1-ons als Aus gangsmaterial muss berücksichtigt werden, da# ein Molekül. der Grignardverbindung durch die Reaktion mit dem freien Hydroxyl verbraucht wird.
Bei der Hydrolyse des gebildeten Komplexes wird dann diese Hydroxvl- gruppe rückgebildet.
Die als Zwischenprodukte bei dieser Grignardreaktion gebildeten Komplexe werden durch Zusatz der Reaktionsmischung xu Wasser zu den 1-Hyd'roxy-l-alkenyl-oder 1-Hydroxy-1-alkinylderivaten zersetzt. Der gewunsehte tertiäre Alkohol kann aus der wäss- rigen Mischung leicht durch Extraktion mit einem nicht wassermisehbaren Losungsmittel isoliert werden.
Auf diese Weise wird aus 2, 4b-Dimethyl-2- methallyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b,5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-4-ol-1-on und Äthoxyacethylenmagnesiumbromid oder Aliyl- magnesiumbromid 1-Äthoxyäthinyl-2, 4b-dimethyl-2-methallyl-7-äthylndioxy-1,2,3, 4, 4a, 4b, 5, G, 7, 8, 10, 10a-docleeahydrophenanthren-: 1, 4- diol bzw. l. Allyl-2, 4b-dimethyl-2-methallyl-7- äthylendioxy-1, 2, 3, 4,4a, 4b, 5, 6, 7, 8, 10, 10a-do deeahydrophenanthren-1, 4-diol erhalten. In gleicher Weise ergeben die entspreehenden 1, 4.-Diolle mit diesen Magnesiumbromidverbindungen die 1-01-4-on-verbindungen.
Die 1, 4 Diol-vPrbindlungen können nachträglieh, z. B. mit Hilfe des Chromtrioxyd-Pyridin-Komplexes, in die entsprechenden 1-Ol-4-on-verbindungen umgewandel. werden.
Als ungesättigte Organometallverbindun gcn gen kann man ferner zum Beispiel auch Alkalimetall- oder Erdalkalimentall-acetylide verwenden. Die Acetylide werden gewöhnlich durch Reaktion des Alkalimetalles, etwa Lithium oder Kalium, mit Aeetylen unter Verwendung von flüssigem Ammoniak als Beaktionsmedium hergestellt. Es ist am bequemsten, die Addi- tionsreaktion in der so entstehenden Acetylid- lösung in Ammoniak auszuführen. Am besten wird das Ammoniak im Laufe der Reaktion entfernt und die Mischung allmählich auf Zimmertemperatur gebracht.
Nach vollständigem Ersatz des flüssigen Ammoniaks dllreh ein inertes organisches Losungsmittel kann das Beaktionsprodukt zersetzt und das Produkt auf übliehe Weise isoliert werden. Diess Äthinylierungsreaktion kann auch in einem tertiären Alkohol, wie tertiä. rem Butanol oder Isopropanol, durchgeführt, werden.
In diesem Fall wird das Keton zu der alkoholischen Lö sung des Alkalimetalles am besten in Gegen- wart eines bis zu 5-10fachen Überschusses über die tlieoretisch natige Menge Alkaliaeet. ylid zugesetzt, da beste Ausbeuten erhalten werden, wenn mit einem grossen Alkali aeetylidüberschuss unter optima. len Bedingun- gen gearbeitet wird.
Anf diese Weise erhält man zum Beispiel aus 2, 4b-Dimethyl-2-methallyl-7-äthylendioxy1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1,4-dion oder der entsprechenden 4-011-on-verbindung mit Lithium- oder Kaliumaeetylid die entsprechenden 1-Athinyl-l- hydroxy-verbindungen. Analoge Dodecahydro phenanthrenverbindungen, welche statt der Metallygruppen andere ungesättigte Kohlen wasserstoffradikale und/oderstattderÄthy- lendioxygruppe andere blockierende Gruppen enthalten, z. B, Enoläther oder andere Ketale können, durch Reaktion mit einem Meta. llaeetylid ebenfallsindieentsprechenden 1-Äthinyl-l-hydroxy-derivate übergeführt werden.
Weiter können die genannten tertiären Alkohole der Dodecahydrophenanthrenreihe auch durch Umsetzung der 1-Keto-dodeca hydrophenanthrene mit Estern von bromierten niederen Fettsäuren in Gegenwart von metal- lisehem Zink hergestellt werden. Diese Reak- tion wird üblicherweise in einem inerten Lö sungsmittel, etwa einem Äther oder einem aromatischen Kohlenwasserstoff ausgeführt, vorzugsweise bei Rüekflusstemperatur.
Die nach dem erfindungsgemä#en Verfahren erhältlichen tertiä. ren Alkohole der Dodecahydrophenanthrenreihe und darunter besonders diejenigen, welche in l-Stellung einen ungesättigten, aliphatischen Substituenten von weiger als 5 C-Atomen a. ufweisen, sind wert- volle Zwischenprodukte der Steroidsynthese, da die Seitenkette durch geeignete Behandlung zum Ringschlussund damit zur Bildung des D-Ringes des Sterinsystems gebracht werden kann.
Beispiel
1-Äthoxyäthinyl-2,4b-dimethyl-2-methallyl 7-athylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-do decahydrophena. nthren-1-ol-4-on.
Eine Losung von Äthylmagnesiumbromid (0, 1526 Mol) wurde in üblicher Weise a. us 3, 7 g Magnesium und. einem Überschuss von Äthylbromid in 100 cm3 trockenem Äther her gestellt. 11, 74 g durch trockenen Äther auf ein Gesamtvolumen von 40 cm3 verdünntes Äthoxy- acetylen (0, 165 Mol) wurde aNmählich der Grignardlosung zugesetzt und die Mischung gerührt, bis die Äthanentwicklung aufhörte.
Zum Losen des Äthoxyacetyleinmagnesium- bromides wurden 120 cm3 trockenes Benzol zugesetzt.
Nun wurde der gerührten Grignardlösung rasch eine Loaung von 27 g 2, 4b-Dimethyl-2methallyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1, 4-dion (Smp.
108-109 C) zugesetzt, die Mischung zwei Stunden bei Zimmertemperatur stehengelassen und dann durch Eingiessen in Eiswasser zersetzt. Um die sich bildende Emulsion Zll brechen, wurde eine ausreichende Menge gesättig- ter, wässriger Ammoniumchloridlösung zuge- setzt, hierauf die Benzol-Ather-Schicht abgetrennt und einmal mit Wasser gewaschen. Die wässrige Schicht wurde nochmals mit 500 cm3 einer Benzol-Äther-Mischung (1 : 1) extrahiert und der einmal mit Wasser gewaschene Auszug mit dem ersten Extrakt verinigt. Nach Trocknen über wasserfreiem Natriumsulfat und d Abfiltrieren des Salzes wurden die Lo sungsmittel im Vakuum abdestilliert.
Aus der Losung des zurückbleibenden Oh, in Äther wurden Kristalle des gewünschten Produktes mit dem Schmelzpunkt 133-134 C erhalten.
Bei Verwendung einer bei 138-139 C schmel'zenden Modifikation des obengenannten Ausgangsmaterials wurden durch die gleiche behandlung zwei versehiedene Stereoisomere des 1-Äthoxyäthinyl-2,4b-dimethyl-2-methallyl7-äthylendioxy-1,2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodeeahydrophenanthren-1-ol-4-ons mit den Schmelzpunkten 131-132 C bzw. 159-161 C erhalten.
Beispiel 2
1-Äthoxyäthinyl-2,4b-dimethyl-2-methallyl7-äthylendioxy-1,2,3,4,4a,4b,5, 6, 7, 8, 10, 10a-do deeahydrophenanthren-1, 4-diol'.
Zu einer aus 0, 27 g Magnesium (0,011 Grammatom) und einem Überschuss von Äthyl- bromid hergestellten Äthylmagnesiumbromidlösung in 25 cm3 Ä ther wurden 1, 2 g Äthoxyacetylen gegeben. Das gebildete feste Äthoxy acetylenmagnesiumbromid wurde durch Zusatz von 25 cm3 3 trockenem Benzol gelost.
Zu dieser Lösung wurde 1, 00 g (0, 00277 Mol) kri stallines 2, 4b-Dimethyl-2-methallyl-7-äthylen- dioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydro- phenanthren-4-ol-1-on zugesetzt, die Mischung zwanzig Stunden bei Zimmertemperatur reagie- ren gelassen, die homogene Reaktionsmischung in Eiswasser gegossen lmd die wässrige Mi- sehung mit Äther extrahiert.
Während der Extraktion wurde eine gerade zum Verhindern einer Emulsionsbildung ausreichende Menge Ammoniumchloridlosung zugegeben. Der ätherische Extrakt wurde über wasserfreiem Magensiumaulfat getrocknet und eingedampft, wobei kristalQines 1-Äthoxyäthinyl-2, 4b-di methyl-2-methallyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1@ 4diol erhalten wurde. Nach Umkristallisation aus einer Äther-Petroläther-Misehung schmolz das reine produkt bei 109-111 C.
Beispiel 3
1-Allyl-2,-dimethyl-2-methallyl-7-äthylen- dioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10,10a-dodecahydro phena. nthren1, 4-diol.
Zu 10 g 2, 4b-Dimethyl-2-methallyl-7- äthlendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-4-ol-1-on wurden unter kräftigem Rühren 150 cm3 einer etwa halbmolaren, ätherischen Losung von Allylmagne siumbromid gegeben. Die Mischung wurde eine Stunde bei Zimmertemperatur gerührt, dann wurden ihr unter Kühlung 5 cm3 Wasser zugetropft. Die ätherisehe Lösung wurde dekan tiert. und der Bückstand mit 150 cm3 Äther extrahiert. Die vereinigten ätherischen Lösungen wurden im Vakummzueinemgelben Harz eingeda. mpft, welehes in einer Benzol-Petrol- äther-Misehung gelöst und an Aluminiumoxyd adsorbiert, wurde.
Durch Eluierung mit einer Äther-Petroläther-Mischung wurde farbloses, amorphes 1-Allyl-2, 4-dimethyl-2-methallyl-7- äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1, 4-diol erhalten.
Durch saure Hydrolyse lässt sieh der Äthy- lendioxysubstituent dieser Verbindung abspalten, wobei 1-Allyl-2,4b-dimethyl-2-methallyl1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1,4-diol-7-on entsteht.
Beispiel 4
1-Äthinyl-2, 4b-dimethyl-2-methallyl-7- äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-do decahydrophenant. hren-1-ol-4-on.
Ein in der Flamme getrockneter, mit einem Kaliumhydroxydrohr versehener 100-cm3-Kolben wurde in einer Trockeneeis-Aceton-Kälte- Mischung abgekühlt und mit etwa 25 em3 flus- siem Ammoniak beschickt. Metallisches Lithium (0, 18 g 0, 0257 Grammatom) wurde im Ammoniak gelost und bis zum Verschwinden der blauen Fa. rbe A. cetylen durch die Lösung geschickt.
Zur entstandenen Suspension von weissem Lithiumacetylid wurde im La. ufe von 5 Minuten unter Rühren eine Lösung von 0, 97 g (0, 00271 Mol) 2, 4b-Dimethyl-2-methallyl 7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1, 4-dion in 25 cm3 einer 1 : 1 Benzol-Äther-Mischung tropfenweise zugesetzt. Der Kolben wurde dann aus dem Kältebad genommen und unter fortgesetztem Rühren und langsamem Einleiten von Acety- ! en allmählich auf Zimmertemperatur erwärmt.
Dazu waren etwa 45 Minuten notig, während weleher Zeit der grösste Teil des Ammoniakes abdampfte. Rühren und Einleiten von Acetylen wurden weitere 45 Minuten bei Zimmertemperatur fortgesetzt, dann langsam 20 cm3 Wasser zugegebenund die beidlen Schichten getrennt.
Die wä#rige Schicht wurde zweimal mit Chloroform ausgezagen und die vereinigten Chloroform-und Äther-Benzol-Losungen über Magnesiumsulfat getrocknet. Beim Eindamp- fen hinterblieb ein kristalliner Riickstand von 1-Äthinyl-2,4b-dimethyl-2-methallyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1-ol-4-on mit dem Schmelzpunkt 220-222 . Durch Umkristallisation aus Äthyl- acetat stieg der Schmelzpunkt auf 223, 5 bis 224, 5 C.
Beispiel 5
1-Äthinyl-2,4b-dimethyl-2-methallyl-7 äthylendioxy-l, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-do deeallydrophenanthren-1-ol-4-on.
Eine Losung von 1, 8 g (0, 005 Mol) 2, 4b Dimethyl-2-methallyl-7-äthylendioxy-1, 2, 3, 4, 4a, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1,4dion in 30 em3 Benzol wurde durch Abdestillieren von 20 cm3 Benzol getrocknet. Nach Abkühlung auf Zimmertemperatur wurden 20 em3 einer 4%igen Kaliumlosung in tertiärem Butanol (0, 020 Grammatom) zugesetzt. Nun wurde während zweier Stuc- den Aeetylen in Blasen durch die kirschrote Losung hindurchgeleitet. Hierauf wurden weitere 40 cm3 Benzol zugefügt und die Mi sehung in 35 cm3 Wasser gegossen.
Die Schichten wurden getrennt, die wässrige Schicht zweimal mit Benzol extrahiert, die vereinigten benzolssehen Losungen über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand bestand aus Kristallen und einem gelben 61.
Das durch Umkristallisation aus Äthylacetat gereinigte 1-Äthinyl-2, 4b-dimethyl-2-metha. llyl- 7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1-ol-4-on schmolz bei 218-222 C.
Beispiel 6
1-Äthinyl-2, 4b-dimethyl-2-met. hallyl-7-ät. hy- lendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodeca hydrophenanthrenl-ol-4-on.
Ein trockener, in Aceton-Trockeneis- Wältemischung gekühlter Kolben wurde mit 50 cm3 flüssigem Ammoinak und 0, 9 g (0, 0231 Mol) metallischem Kalium beschickt.
Trockenes Acetylen wurde in Blasen durch die Lösung geschickt, bis die blaue Färbung verschwunden und eine farblose Suspension von Kaliumacetylid entstanden war. Nun wurde aus einem Tropftrichter eine Lösung von 0, 9 g (0, 0025 Mol) 2, 4b-Dimethyl2-methallyl-7-äthylendioxy-1,2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10,10a-dodecahydrophenanthren-1,4-dion in 50 cm3 einer Benzol-Äther-Mischung (1 : 1) schnell zufliessen gelassen. Die rotliche Suspen- sionwurdeausdemKühlbadgenommenund unter fortgesetzter Acetylenznfuhr langsam auf etwa, 20 erwärmt, wobei das Ammoniak abdampfte.
Im Laufe der Erwärmung verän- derte sich die Farbe von rötlieh über purpur nach b. ra. un. Nach Erreichen von 20 wurde die Mischung langsam.. in 20 cm3 Wasser ge gossen, getrennt, und die wässrige Schicht noch zweimal mit Benzol extrahiert. Die vereinigten Extrakte wurden über Magnesiumsulfat t getrocknet und das Benzol im Vacuum abgedampft, wobei Kristalle und ein gelbes Öl zurüekblieben. Das 61 wurde durch Auswaschen mit kaltem Methanol praktisch voll- ständig entfernt.
Das zurückbleibende kristal- line 1-Äthinyl-2, 4b-dimethyl-2-methallyl-7 äthylen-dioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydriophenanthren-1-ol-4-on schmolz bei 212-220 C. Das durch Umkristallisation aus Athylaeetat gereinigte Produkt zeigte einen Schmelzpunkt von 220-222"C.
Beispiel 7
1-Carbomethoxymethyl-2, 4b-dimethyl-2 methallyl-7-äthylendioxy-1, 2, 3, 4, 4a, 4b, 5, 6, 7, 8, 10, 10a-dodecahydrophenanthren-1-ol-4-on.
Eine Losung von 2, 3 g 2,4b-Dimethyl-2methallyl-7-äthylendioxy-1, 2, 3, 4, 4a., 4b, 5, 6, 7, 8, I 0, 10a-dodecahyd'rophenanthren-1, 4-dion in 50 cm3 trockenem Ät. her wurde mit 2, 5 g Zink (nach Aktivierung mit verdünnter Salzsäure getrocknet) und 0, 75 cm3 Methylbromacetat gerührt. Nach kurzer Verzögerung setzte die Reaktion ein und es schied sich auf dem Zink ein hellgelber, öliger Niederschlag aus. Im La-Lite von 24 Stunden wurden 4 weitere Por- tionen Zink (2, 5 g) und Methylbromacetat (0, 5 cm3) zugegebe. Währendd der ganzen Zeit wurde das Reaktionsgemiseh auf Rück flusstemperatur gehalten.
