Verfahren zur Herstellung von Tetracyclin Die Erfindung betrifft ein Verfahren zur Herstellung des Antibioticums Tetracyclin auf dem Weg der Fermentation, und zwar durch Züchtung neuer Mikroorganismen in Nährlösungen unter aeroben Bedingungen.
Tetra.eyclin ist ein Antibioticum von brei ter Wirksamkeit., dessen Struktur zuerst in einem Bericht im Journal of the American Chemieal Society, Band 74, Seite 4976 (1952) angegeben worden ist.
Die Herstellung dieses Antibiotieums durch katalytische Reduktion von Chlortetracyclin ist. gleichfalls schon be schrieben worden (Journal of the American Chemieal Society, Band 74, Seiten 4621-23 [1.9.531).
Obwohl. nun das Antibioticum Tetracyelin bereits auf chemischem. Weg aus Chlortetra cyelin, einem Fermentationsprodukt, gewon nen worden ist, liegen in der direkten Her stellung des Tetracyclins durch einen Fermen tationsprozess doch noch erhebliche Vorteile. Offensichtlich fällt dabei die ganze Anlage für die Hydrierung des Chlortetraeyelins fort.
Ferner ist der teure Hydrierungakatalysator nicht mehr notwendig und schliesslich sind auch die Gefahren. beseitigt, die stets mit einem Hydrierungsvorgang verbunden sind.
Es ist gefunden worden, dass Stämme von Mikroorganismen der Gattung Streptomyees, wenn sie unter aeroben Bedingungen gezüch tet werden, das Antäbiotieum Tetraeyclin er zeugen.
Damit diese neuen Stämme, die hin sichtlich ihrer Arten noch nicht identifiziert sind, jederzeit zu Vergleichszwecken zugäng- lich :sind, wurden Proben davon bei der ameri kanischen Sammlung von Kulturarten (Ame- rican Type Culture Collection, 2029 M Street, N. W., Washington, D. C.) hinterlegt und die ser Sammlung von Mikroorganismen unter der Bezeichnung ATCC No. 11652, 11653 und , 11654 einverleibt.
In der Sammlung der An- inelderin werden diese Organismen unter den Nummern J-9618, K-6030-1 und K-6030-4 ge führt.
Die charakteristischen Eigenschaften der , Kulturen dieser Mikroorganismen sind in den nachstehenden Tabellen festgehalten. Die Farbangaben, die mit einem R versehen sind, beziehen sich auf Ridkway, Color Standards and Nomenclature. .

sondern auch die Verwendung von :Mutatio- nen, welche aus diesen Organismen düreh ver- sehieclene Mittel. wie Rönt\renstrahlen, Ultra- violettst-rahlen, ?,2-Dichlor-N-metliyldiäthil- amin-hvdrochlorid lind dergleiehen hergestellt werden können.
Die das Teti-aci-clin erzeugenden Stämme von Streptomvees können auf Ausstrichen voll Emerson-Agar oder andern geeigneten Aledien aufbewahrt werden, oder Uran kann die Or-a- nis-men in der eingefrorenen Form troeknen, um ;
sie später wieder zum Leben zli erweeken. Kulturen auf Ausstrichen oder die kalt ge trockneten Präparate können rxi sterilen<B>Nähr-</B> medien.
in kleinen Kolben zugesetzt werden, urn kleine Mengen des Antibiotieums oder hin- i,eichende Men--en von dem Mikroorganismus für die Aussaat auf grossen Nährböden zxi erzeugen.
Im allgemeinen soll das Fermenta- tionsmedinin. eine- Isolilelivdratquelle u ud eilie Stickstoffquelle, vorzugsweise ill organiseller Form, enthalten.
Eine Anzahl versehiedener _1 Metallsalze hat ebenfalls einen günstimen Eixi- fluss, da diese Salze die Bildung des Anti- bioticunis fördern.
Oft liefert aber der Gre- brauch von Leitungs%vasser genügende Men gen dieser Metalle, tim ein gutes Wachstum sieherzustellen. Als Beispiel für Koblehydrat- quellen seien genannt:
Helasse, Glukose, Stärke, C,rlz zerin us-#v. Creeignete organische= Stichsto@f- quellen sind:
Sovabohnenmehl, WeizexiZluten, Erdnussmehl, Hydrokate voll Kasein und andern Proteinen usw. Gewisse Rohstoffe elit- halten Substanzen,
die das @@Tachstuni fördern und daher voll Wert für die Gewinnung ma_a- maler Ausbeuten an dem Antibiotieum sind.
Zti diesen Rohstoffen gehören Maismaische, Schlempe, Hefeeerakt usw. Salze, wie Na triumehlorid, Na.triuninitrat, Kaliumpliosphat und- hla!#nesiumsiilfat haben aueli einigen Wert. Obgleieh die Organismen nicht dazu neigen,
den BH-Wert des Mediums während ihres Waelisturng in nennenswerter Weise zu ändern, kann doeh die Verwendung von Puffersubstanzen wie ("aleiumkarbonat von Vorteil sein.
Welni während des Waehstuins der Organismen, die unter aeroben Bedingun- gen eingeta-uclit sind, Schaumbildung auftritt, isst ein Zusatz von Ölen, wie Specköl, Soy aö1 oder derglei,ehen, zuveel@nrässim.
Die Züchtung der ausgewählten Stämme von Streptompees unter aeroben Bedingungen in eingetanelxter Form ist für die -Erzeugung des Antibiotieums in grossem Massstab beson ders geeignet. Wenn eine Verinen- tation in grossem Massstab begonnen werden soll, ist es zweekinässig, dass vorbereitete, steri- lisierte Fermentationsmedium mit 1-10 /o V'olinnprozenten einer reinen Kultur des frisch @r-ezüeliteten, ausgewählten, Tetraeyelin erzeu genden Stammes von Streptomyees zu impfen.
Die antibiotische Aktivität de Produktes, das von den oben, beschriebenen Stämmen von Streptomyees erzeugt. wird, kann mit den be kannten Standardmethoden der Mikrobiologie für die Untersuchung der Antibiotiea geprüft werden, indem man verschiedene Mikroorga nismen benutzt.
Ebenso können verschiedene physikalische Methoden herangezogen werden, uni den Gehalt eines gegebenen Musters an Antibiotienin zu ermitteln, beispielsweise die T'Itra.violettabsorptio.n bei bestimmten Wellen längen oder die Infrarotabsorption. Das reine, kristalline, wasserfreie, amphotere T.etraeyelin wird dabei als Standard mit. einer W irksamkeit von 7.000 nieg;'mg benutzt.
Die Pa.pierchroma- to;raphie isst besonders gut geeignet, um den Tetraeyclingehalt sowohl eines rohen Mate rials als auch reinerer Produkte zii erkennen --ind festzustellen.
Wiewohl die rohe Fermentatio.nsbrühe, welche bei der Züchtung der neuen, ausge wählten, Tetracyelin erzeuPenden Stämme von Sti#eptomyces in einem Nährmedium unter aeroben Bedingungen im eingeta:uehten Zu stand entsteht, für sieh selbst Weit hat, bei- spielsweise als Zusatz zu Tierfutter oder als Mittel zur Förderung des Pflanzenwueh;
ses, ist es manchmal erwünscht, das Antibiotieum als trockenes Material entweder in roher oder ge- reiiii-ter Form mi gewinnen. Zu diesem Zweck kann man sich verschiedener Methoden bedie- lien. Beispielsweise kann die ganze Brühe ent weder vor oder nach der Filtration des Mycels im gefrorenen Zustand getrocknet werden, wobei man ein. Rohprodukt mit. einem erheb liehen Gehalt an Antibiotieum, erzielt.
Es isst vorteilhaft, diese Troeknungsoperation bei einem pH-Wert durchzuführen, der weder zii sauer noch zii basisch ist, da das Antibioticum sonst dur.eh. eine solche Behandlung in seiner Wirksamkeit herabgesetzt werden kann. Das Antibiot%cum \I'etraeyclin kann aus der Fermentationsbrühe auf verschiedene Weise abgeschieden werden.
Es ist zu empfehlen, vor Anwendung dieser Methoden das Myeel durch Filtration von der Brühe zu trennen. Die beste Ausbeute an Antibioticum wird in dem wäss- rigen Filtrat erhalten, wenn das Gemisch vor der Filtration mit einer starken Säure, wie Salzsäure, Schwefelsäure, Phosphorsäure oder dergleichen, auf einen pH-@Vert von etwa 2-3 eingestellt. wird. Obwohl eine mässige Erhöhung der Temperatur dazu helfen kann, :einige von den fein verteilten Verunreinigungen zu koagulieren und dadurch ein klareres Filtrat zu ergeben, wird doch eine höhere Temperatur besser vermieden.
Bei dem pH-Wert von 2-3 kann leicht eine teilweise Zerstörung des Anti- biotieums stattfinden, wenn die Lösung auf etwa. 40 C erhitzt wird oder wenn sie bei die sem pH längere Zeit stehenbleibt.
Zum Abfiltrieren des 1,lycel:s aus der Fer- menta.tionsbrühe können verschiedene Filter- substanzen benutzt werden. Besonders geeignet ist. Diatomeenerde. Das Filtrat kann dann ge trocknet oder weiteren Gewinnungsverfahren zum Zweck der Reinigung unterworfen wer den.
Eine Methode, die sieh für die Abschei- dung des Tetraeyelins aus solchen Fermenta- tionsbrühen als nützlich erwiesen hat, besteht in dem Zusatz von bestimmten mehrwertigen 1-letallionen, vorzugsweise in Form von wasser löslichen Salzen dieser Metalle.
Obgleich eine flewisse Menge solcher Metalle in der Fermen ta.tionsbrühe vorhanden sein kann, insbeson dere Calcitim, sofern Caleiumkarbonat als Puffersubstanz verwendet worden ist, genügt diese Menge oft nicht, um den Hauptteil des Antibiotieums niedermischlagen, wenn man die Brühe basisch macht. Die Ausfällung wird durchgeführt, indem man nach Hinzufügung der erforderlichen 1let.allsalze den pH-Wert auf schwach alkalische Reaktion einstellt. Im allgemeinen ist ein PH von etwa 7,5-9 am besten geeignet.
Es ist. zu empfehlen, mehr als nur eines der mehrwertigen, die Nieder schlagung bewirkenden Metalle zu benutzen da,die komplexen Produkte, welche sich in der Fermentationsbrühe mit einer Kombination dieser Metalle bilden, oft erheblich weniger löslich sind als diejenigen, die entstehen, wenn nur eines der mehrwertigen Metallionen zu gegen. ist.
Zu den Metallen, die sieh als beson ders nützlich für die Niederschlagung des Antibiotieums erwiesen haben, gehören Ba rium, Calcium, Magnesium, Strontium, Beryl lium, Quecksilber, Cadmium und andere Me tallionen, in erster Linie die der 2. Gruppe des periodischen Systems. Wie erwähnt, ist oft eine Kombination von 2 oder mehr dieser Me talle wirksamer als die Verwendung eines ein- ze-Inen 1#,fetalles.
Der komplexe Niederschlag aus dem Anti- biotieum und dien Metallionen, zusammen mit andern Bestandteilen der Fermentationsbrühe, kann von der zurückbleibenden Lösung durch Filtration, Zentrifugieren oder auf eine andere geeignete \V eise abgetrennt werden.
Man kann dann den Niederschlag mit einer kleinen Menge Wasser waschen, um die Reste der Fermentationsbrühe zu entfernen, die noch einen geringen Betrag an Antibioticum ent hält, und: das Rohprod:ükt kann darauf ge trocknet. und für bestimmte Zwecke verwen det werden, wenn die mehrwertigen Ionen ver hältnismässig nichttoxisch sind;
wie Calcium und Magnesium. Anderseits kann das feuchte oder trockene rohe Produkt in eine gereinigte Form des amphoteren Antibioticums über- ;eführt werden, indem man den Nied'ersehlag, mit einem Mittel behandelt; welches selektiv die Metallionen fällt, die man benutzt hat.
Wenn beispielsweise Calcium und Barium als Metalle verwendet wurden, kann man das Pro dukt mit verdünnter Schwefelsäure behandeln, um einen Niederschlag aus den Sulfaten von Calcitim und Barium und eine wässerige Lö sung des Antibioticums zu erzeugen:. Durch das A.bfiltrieren des festen Niederschlages wird: das Antibioticum abgetrennt.
Nach der Einstellung des pH-Wertes zur Vermeidung einer Zersetzung 'des Ant.ibioticums kann dit, konzentrierte Lösung, wie sie vorliegt, für ver schiedene Zwecke benutzt werden, oder man kann sie trocknen. 'Wenn das PH leicht alka lisch gemacht. wird, kann man aus der auf diese Weise erhaltenen konzentrierten wässe- riggen Lösung eine reinere .Form des ampho- teren Tetracyelins abscheiden.
Dieses Produkt ist im allgemeinen von genügender Reinheit, um es nach Verfahren, die in den oben erwähnten, früheren Veröff_ entliehungen be schrieben worden sind, zur Kristallisation zu bringen.
Eine andere Methode, die man zur Gewin nung des Tetracy clins auf der Fermüntations- brühe benutzen kann, besteht. darin, dass man das Antibioticum bei einem stark sauren oder schwach alkalischen PH mit bestimmten, mit Wasser nicht. misehba.ren, organischen Lö sungsmitteln extrahiert.
Im allgemeinen kann man diese Extraktion bei einem pH von etwa 1-2,5 oder von. 7,5-l.0 durchführen. Ver- sehiedene polare, mit Wasser nicht mischbare organische Lösungsmittel können zu diesem Zweck benutzt werden.
Zu ihnen gehören alkoholische Lösungsmittel, wie die Butanole, die Pentanole usw., sowie substituierte Alko hole, wie Benzylalkohol, Phenyläthylall@ohol, und bestimmte Substitutionsprodukte der CTly- kole, wie Phenyleellu lose, ferner mit. Wasser nicht mischbare Ketone, wie 3Tethylisobtityl- keton. Auch Ester kommen in Betracht, z. B.
Amylacetat, Propylaeetat, At.hylpropionatusw. Es hat sieh gezeigt, dass es zweckmässig ist, zti der filtrierten Ferment.ationsbrühe vor der Extraktion, insbesondere bei alkalischem pH, ein Fällungsmittel für mehrwertige Metall ionen zuzusetzen. Solche Mittel sind beispiels weise Polyphosphate, verschiedene Hydroxy- säuren, wie Cxhieonsäure, Zitronensäure, Wein säure usw.,
oder Aminosä.uren, wie Ätbylen- diamintet.raessigsäure. Diese Ma.ssnahmesichert, dass die mehrwertigen Metallionen in Form von Komplexen mit dem Tetracyelin und Be standteilen ,der Fermentationsbrühe in das Extraktionsmittel übergehen.
Die Extraktion kann nach. sta.tisehen Me thoden oder auf dem Weg, der kontinuierliehcn Gegenstrom-Extrahierung stattfinden. Eine besonders zweckmässige Methode der Extrak tion besteht.
in der Anwendung der im Handel erhältlichen und unter der Bezeichnung Pod- bielniak Extraktor bekannten Einrichtung. Im allgemeinen soll man mindestens ein Volti- irren des l.ösungsinittels auf jedes Zehntel- @-olumen der wässerigen Lösung des Antibioti- cums benutzen.
Erheblich höhere Mengen an Lösungsmittel können erforderlich werden., wenn das gewählte Lösungsmittel nicht die liöcliste Wirksamkeit aufweist., oder wenn die verwendete Apparatur nieht, rasch genug die gleichmässige.
Verteilung des Lösungsmittels liei-beizuführen vermag. Na,eh beendeter Extrak tion können die Lösizngsmittelphasen ver- vini;
t werden und das Antibiotieum kann daraus sowohl durch vollständige Verdamp- fung des Lösungsmittels als auch durch teil weise Entfernung desselben und Extraktion des Antibiot-icums mit einem kleinen Volumen verdünnter wässeriger Säure, vorzugsweise einer Mineralsäure, gewonnen werden. Das so erhaltene Konzentrat. kann für verschiedene Zwceke verwendet werden.
Das Produkt kann clurcli Einstellung des PH auf einen sehwa,eh -ilk < ilischen Wert-, z. B. etwa. 7,5, aus der kon zentrierten wässerigen Lösung, gefällt werden. Das auf diese Weise gewonnene Tetraey elin kann für gewisse therapeutische Zweeke Ver- cvendunr finden, oder es kann kristallisiert werden.
Eine weitere Methode zur Gewinnung des Tetracvclins aus der wie oben beschrieben lierge.stellten Fermentationsbrühe betrifft den Gebrauch gewisser organischer Basen als Aus- fä llungsmittel. Von besonderem Wert- sind dabei or;
anisehe Basen von hohem Molekular- gewicht, die in Wasser wenig löslich sind, wie lioehmolekulare, quaternäre Ammoniumsalze, z.
B. Cs-Cis Alkyitrimet.hylammonium- haloide oder Cs-Crs Alky1,dimethvlben7yl- ammoniumllaloide, auch lankettige Alkyl.- amine, sowohl primäre als auch sekundäre oder tertiäre, und eine Vielzahl anderer orga- niseher basiseher Substanzen.
Diese Fälhbngs- rnittel :scheinen Komplexe zu bilden, die nicht nur das Antibioticum, :sondern auch gewisse andere Komponenten enthalten, welche ge- wöhnlieh in den hergestellten Salzen vorhan den. sind. Solche Komponenten sind beispiels weise kleine Mengen mehrwertiger Metalle und organische Stoffe.
Nach dem Zusatz des basiseben o:rga.nischen Materials zu der filtrier- ten Tetraeyclin-Fermentation sbrühe soll der pH-Wert des Gemisches, vorzugsweise mit einem löslichen Alkali, wie Natriumhydr.oxyd, auf eine leicht basische Reaktion eingestellt werden, etwa auf 8-9,5. Der Niederschlag kann a,bfiltriert werden, zweckmässig nach kur zer Rührung.
Bin etwas reineres Produkt wird erhalten, wenn man die angesäuerte, filtrierte Tetracyelinbrühe mit einem der bei der zweiten Methode erwähnten Fällungsmittel für die Netallionen versetzt und erst dann den Zusatz der organis.ehen Base vornimmt..
Das Produkt, welches durch die vorstehend beschriebene Niedersehlaoung mit einer hoch- molekulaTen organischen Base gewonnen wor den ist, kann in eine Substanz übergeführt werden, die Salze des Tetraey clins enthält, oder man kann es auf amphoteres Tetraeyelin aufarbeiten.
Die einfachste Behandlung besteht darin, d'ass man den Niederschlag in einem polaren organischen Lösungsmittel von nied rigem Molekulargewieht auflöst. Der Zusatz einer starken Mineralsäure, wie konzentrierter Salzsäure, bewirkt die Bildung \von Tetra- ey elinhydrochlorid, das, sofern genügend kon zentrierte Lösungen verwendet wurden, kristal lisiert oder ausfällt. Die Komplexe von Tetra cyclin und der organischen Base haben auch einen guten Gebrauehswert,
z. B. als tech- nisehe Desinfektionsmittel.
Eine andere Methode zier Gewinnung des Antibioticunrs besteht darin, d-ass man die an gesäuerte Ferment.ationsbrühe nach dem Ab filtrieren des Myeels mit gewissen Arylazo- sulfonsäurefarbstoffen zusammenbringt, Da durch wird ein Farbstoffkomplex niederge schlagen., aus dem das Antibioticum dureh ähnliche Methoden abgeschieden werden kann, wie sie weiter oben beschrieben worden sind.
Beispielsweise. kan man das Gemisch in einem polaren organischen Lösungsmittel von nied rigem Molekulargewicht lösen. Die Einstellung des PH auf einen schwach alkalischen Wert bewirkt die Ausfällung von amphoterem Tetra- eyclin, sofern man hinreichend konzentrierte Lösungen benutzt hat..
Ausser mach. den im vorstehenden erörter ten Methoden der Ausfällung kann das Anti- bioteum. in teilweisse gereingter Forni aneli erhalten werden, indem man die Fermenta tion sbrühe mit Hilfe eines Adso #ptionsmittels, \wie Aktivkohle, aktive Erden oder ähnlichen Stoffen behandelt.
Die Adsorption kann bei einem px von etwa 3-8 ausgeführt werden, Lind man kann dass adsorbierte Antibiotieum zur Anreieherun, vom. Futtermitteln oder für andere Zlvecke verwenden.
Es ist. auch mög lich, das Tet.rae7Clin aus dem Adsorpt.ions- mittel durch Eluierung, beispielsweise mit einer verdünnten wässerigen Säure oder der verdünnten Lösung einer starken Säure in einem. polaren Lösungsmittel, zu gewinnen. Das Produkt kann in der erhaltenen Form benutzt, oder nach einem .der vorstehend be- sehriebenen Verfahren weitergereinigt werden.
Das Antibiotieum kann: auch aus der Fermen tationsbrühe an verschiedene Ionen.austau.sch- harze, insllesondere stark Saure, z. B. schwefel saure Harze, wie das Marl,:eliprodukt Dowex <B>1</B> <B>,</B> adsorbiert. werden.
Diese Adsorpt.ion wird am betten bei einem pR--V ert. von etwa 3-'- ausgeführt und dass Prodtakt kann so, wie es anfällt, Verwendung finden, oder man kann das Tetraey-elin, beispielsweise mit verdünntem Alkali, el.uieren.
ZV enn bei irgendeinem der vorstehend er ä.hnten Verfahren die Trennung des Tetra evelins von antibiotischeil Nebenprodukten der Fermeiitntion erforderlich werden sollte, so kann dies auf folgendem MVeg geschehen: E:
s wird znnäeht eine wässerige Lösung voll roheng Tetraei-elin nach einer der .oben erläu terten Methoden hergestellt. Diese Lösung wird angesäuert und mit einem mit. Wasser nicht mischbaren organischen Lesungsmittel extra hiert, insbesondere mit niedrigen Alkoholen, wie Butanol, Isobutanol. den Pentanolen, Hexa.nolen. Lind dergleichen.
Nach Beelidi- gun- der Extraktion werden die vereinigten Extrakte .auf praktisch neutrale Reaktion ein gestellt Lind die freie Tetraev elinbase kann dann durch Rühren der 3lisehung oder durch mehr- gtündiges Stehenlassen abgeschieden werden. The fermentation mixture from 10 such fermentations was combined and adjusted to a p11 of 2.5 with hydrochloric acid at room temperature.
Then the Alvcel was filtered off with the addition of diatomaceous earth as a filter aid. The filtrate was brought to pH 7.5 and extracted twice with 1C, volume butanol.
The butanol extracts were combined and concentrated to 1500 ml in vacuo. The butanol concentrate was then extracted several times with: approx. 200 ml of water which had been brought to a p11 of 1.9 with hydrochloric acid.
The combined aqueous extracts were adjusted to a pH of 7.5 with dilute sodium hydroxide solution and the crude, amphoteric Tetraeycl.in-containing product was filtered off with addition of diatomaceous earth. The material was further purified to obtain crystallized amphoteric tetracycline.
