Verfahren zur Herstellung von Acetyldigitoxin-a. Es wurde gefunden, dass man zu einem bisher unbekannten Acetyldigitoxin (Acetyl- digitoxin-a) gelangt, wenn das bekannte Aeetyldigitoxin (Acetyldigitoxin-ss) unter Ver wendung eines organischen Lösungsmittels gelöst wird, die Lösung einen pH--#'vert im Intervall von 5-8 aufweist und erhitzt. wird.
Das als Ausgangsstoff dienende Acetyl- digItoxin-ss ist ein Herzglykosid, das sowohl aus Lanatosid A durch Abspaltung des Glu- eoserestes, als auch durch Extraktion aus den Blättern der Digitalis ferruginea erhältlich ist. Es setzt sich aus dem Aglykon Digitoxi- gen in und 3 Mol Digitoxose zusammen; an einem Digitoxoserest ist eine Acetylgruppe gebunden.
Das neue Produkt entsteht durch Umlagerung des Acetyldigitoxin-ss; das Ace- t.yIdigitoxin-a unterscheidet sich durch die Stellung der Acetylgruppe vom Aeetyldi4ri- toxin-ss.
Das bisher unbekannte Acetyldigitoxin-a kristallisiert aus wässrigem Methanol in Plätt- ehen vom Smp. 9217-22,10; [a]r) = +5 0,8 (in Pyridin).
Das Aeetyldigitoxin-a soll als 'Therapeuti- eum verwendet werden, da es eine aus gesprochen starke und gleichmässige Herz wirkung ohne hohe Toxizität aufweist. <I>Beispiel 1.</I> Eine Lösung von 5 g Acetyldigitoxin-ss in 500 cm3 Methanol wird 1 Stunde lang am Rückflusskühler gekocht. Hierauf wird zur Abscheidung von unverändertem Ausgangs- material mit 150 cm3 Wasser versetzt und über Nacht im Kühlschrank stehen gelassen.
Das auskristallisierte Acetyldigitoxin-ss wird abfiltriert. Das Filtrat wird durch Eindamp fen im Vakuum vom ll:ethylalkohol befreit; hierbei scheidet sich das Acetyldigitoxin-a aus und wird nach mehrstündigem Stehen lassen der wässrigen Suspension im Kühl schrank abfiltriert. Zur weiteren Reinigung wird es mit der 20fachen Menge Aceton ver rieben und das vom Aceton befreite Produkt durch Auflösen in möglichst wenig Chloro form und Zugabe des 2- bis 3-fachen Volumens Äther umkristallisiert.
Das so erhal tene Produkt zeigt folgende Eigenschaften: Smp. 217 - 221 ; [a] D = + 5 0,8 (in Pyridin).
<I>Beispiel 2.</I>
5 g Acetyldigitoxin-ss werden in einem Gemisch von 500 cm3 Äthanol und 250 em3 Wasser, evtl. durch Erwärmen, gelöst, die Lösung 1 Stunde lang am Rüekfluss gekocht und hierauf im Vakuum zur Trockne ver dampft. Der Rückstand wird in 50o cm3 Methanol gelöst, mit 150 cm3 Wasser versetzt, mit einer Spur Acetyldigitoxin-ss geimpft und etwa 20 Stunden bei 0 stehen gelassen, wobei sich unverändertes Ausgangsmaterial kristal lin ausscheidet.
Dieses wird abfiltriert und kann von neuem dem Umwandltulgsverfahren unterworfen werden. Das Filtrat wird im Vakuum vom Methylalkohol befreit, wobei das Acetyldigitoxin-a auskristallisiert. Es wird nach mehrstündigem iStehenlassen der wässri- gen Suspension bei 0 abfiltriert, getroelmet und aus möglichst wenig Chloroform und nnterZusatz des zwei-bisdreifachenVolumens Äther umkristallisiert.
<I>Beispiel 3.</I>
5 g Acetyldigitoxin-ss werden in einem Gemisch von 500 em3 Isopropylalkohol und 1.00 cms Wasser, evtl. durch Erwärmen gelöst; die Lösung wird 1 Stunde lang am Rückfluss gekocht und darauf im Vakuum zur Trockne verdampft. Den Rückstand löst man in. 500 cm3 llethy Ialkohol und versetzt mit. 150 em3 Wasser, worauf die Ausscheidung des nicht umgewandelten Acetyldigitoxins-ss ein setzt.
Nach etwa 20stündigem Stehen in der Kälte (0-4 ) wird filtriert und das Filtrat im Vakuum vom Methanol befreit. Das hier bei sieh ausscheidende Acetyldigitoxin-a wird nach mehrstündigem Stehen in der Kälte (0-4 ) abfiltriert und getrocknet. Es wird ein- bis zweimal aus möglichst wenig Chloro form unter Zusatz des zwei- bis dreifachen Volumens Äther umkristallisiert.
Beispiel <I>4.</I>
5 g Aeet.yldigitoxin-ss werden in einer Mischung von 500 cm3 Aceton und 50 cm3 einer 0,1-n.Essigsäure,wenn nötig unter<B>Er-</B> wärmen gelöst. Die Lösung, deren pH 53 be trägt, wird während einer Stunde am Rück fluss gekocht, hierauf mit. verdünntem Alkali neutralisiert und durch Einengen im Vakuum vom Aceton befreit. Nach längerem Stehen lassen in der Kälte (0-4 ) wird die Ausschei dung abfiltriert und getrocknet.
Dann wird sie in der 100fachen Menge Methanol auf gelöst und. die Lösung mit<B>30</B> /o- (Vol) Wasser versetzt, worauf sich das nicht umgewandelte A.cetyldigitoxin-ss ausscheidet. Nach längerem Stehen in der Kälte (0--4 ) wird filtriert und das Filtrat im Vakuum vom Methyl alkohol befreit.
Die sieh hierbei aiLsseheidende Substanz wird nach längerem Stehen in der Kälte abfiltriert und getrocknet. Dann wird sie nochmals in der 100fachen Menge Me- thano) gelöst und die Lösung wiederum mit 301/o- (Vol) Wasser versetzt, wobei sieh beim Stehen in der Kälte noch der Rest der nicht umgewandelten Substanz ausscheidet, der ab- genutseht wird.
Das Filtrat wird im Vakuum zur Trockne verdampft und der aus Acetvl- digitoxin-a bestehende Rückstand ein- bis zweimal aus möglichst wenig Chloroform unter Zugabe des zwei- bis dreifachen Volu- m ens Äther <B>,</B> umkristallisiert.

The hitherto unknown acetyldigitoxin-a crystallizes from aqueous methanol in platelets with a melting point of 9217-22.10; [a] r) = +5 0.8 (in pyridine).
The acetyldigitoxin-a should be used as a therapeutic agent because it has an extremely strong and even cardiac effect without high toxicity. <I> Example 1. </I> A solution of 5 g of acetyldigitoxin-SS in 500 cm3 of methanol is boiled for 1 hour on the reflux condenser. 150 cm3 of water are then added to separate the unchanged starting material and the mixture is left to stand in the refrigerator overnight.
The acetyldigitoxin-SS which has crystallized out is filtered off. The filtrate is freed from II: ethyl alcohol by evaporation in vacuo; the acetyldigitoxin-a is separated out and, after the aqueous suspension has been left to stand for several hours, it is filtered off in the refrigerator. For further purification it is rubbed with 20 times the amount of acetone and the product freed from acetone is recrystallized by dissolving it in as little chlorine as possible and adding 2 to 3 times the volume of ether.
The product obtained in this way shows the following properties: mp 217-221; [a] D = + 5 0.8 (in pyridine).
<I> Example 2. </I>
5 g Acetyldigitoxin-SS are dissolved in a mixture of 500 cm3 ethanol and 250 cm3 water, possibly by heating, the solution is refluxed for 1 hour and then evaporated to dryness in vacuo. The residue is dissolved in 50o cm3 of methanol, mixed with 150 cm3 of water, inoculated with a trace of acetyldigitoxin-ss and left to stand at 0 for about 20 hours, during which the unchanged starting material is precipitated as crystals.
This is filtered off and can be subjected to the conversion process again. The filtrate is freed from methyl alcohol in vacuo, the acetyl digitoxin-a crystallizing out. After the aqueous suspension has been left to stand at 0 for several hours, it is filtered off, dripped and recrystallized from as little chloroform as possible and with the addition of two to three times the volume of ether.
<I> Example 3. </I>
5 g of acetyldigitoxin-ss are dissolved in a mixture of 500 em3 isopropyl alcohol and 1.00 cms of water, possibly by heating; the solution is refluxed for 1 hour and then evaporated to dryness in vacuo. The residue is dissolved in 500 cm3 of methyl alcohol and mixed with. 150 em3 water, whereupon the excretion of the unconverted acetyldigitoxin-s begins.
After standing in the cold (0-4) for about 20 hours, the mixture is filtered and the filtrate is freed from methanol in vacuo. The acetyldigitoxin-a which is secreted here is filtered off and dried after standing in the cold (0-4) for several hours. It is recrystallized once or twice from as little chloro form as possible with the addition of two to three times the volume of ether.
Example <I> 4. </I>
5 g of Aeet.yldigitoxin-ss are dissolved in a mixture of 500 cm3 of acetone and 50 cm3 of 0.1N acetic acid, if necessary with <B> warming </B>. The solution, the pH of which is 53, is refluxed for one hour, then with it. diluted alkali neutralized and freed from acetone by concentration in vacuo. After standing for a long time in the cold (0-4), the precipitate is filtered off and dried.
Then it is dissolved in 100 times the amount of methanol and. <B> 30 </B> / o- (vol) water are added to the solution, whereupon the unconverted A.cetyldigitoxin-ss precipitates. After standing in the cold for a longer period (0-4), it is filtered and the filtrate is freed from methyl alcohol in vacuo.
After standing for a long time in the cold, the substance that forms in this process is filtered off and dried. Then it is dissolved again in 100 times the amount of methano) and 301 / o- (vol) water is again added to the solution, whereby when standing in the cold the rest of the unconverted substance is excreted, which is eaten away.
The filtrate is evaporated to dryness in vacuo and the residue consisting of acetylditoxin-a is recrystallized once or twice from as little chloroform as possible with the addition of two to three times the volume of ether.
