<B>Verfahren zur Herstellung eines neuen</B> 11-Keto-steroids. Den Steroiden mit Sauerstoff in 11-Stel- lung kommt grosse Bedeutung zu. Ein wich tiger Vertreter dieser Klasse von Verbindun gen ist zum Beispiel das Cortison, das d4-3,11, 20-Triketo-17a,21-dioxy-pregnen. Die bisheri gen Verfahren zur synthetischen Gewinnung solcher Steroide gehen von der Desoxychol- säure bzw.
ihren Derivaten aus, also von Ver bindungen, die in 12-Stellung eine Hydroxyl- gruppe aufweisen. Es hat sich aber gezeigt, class die Verschiebung von Sauerstoff aus der 12- in die 11-Stellung sehr umständlich ist und mehrere Operationen erfordert.
Zudem ist die als Ausgangsstoff dienende DesoxYchol- säure mir in relativ beschränkter Menge er liält.lich, so dass es zum Beispiel praktisch un möglich ist, das für die Therapie benötigte Cortison auf diesem Wege in ausreichender Menge zu fabrizieren. Es ist daher ein Be dürfnis, neue Quellen für die Herstellung die ses Heilmittels zu erschliessen.
Die gut zugäng lichen Sterine, wie Ergosterin, Stigmasterin oder Sitosterin, insbesondere aber Cholesterin, bilden zwar schon seit vielen Jahren wichtige Ausgangsstoffe für die Produktion der Sexual hormone. Sie waren dagegen bisher für die Gewinnung von Verbindungen mit Sauerstoff in 11-Stellung des intakten Steroidgerüstes ohne Bedeutung.
Es wurde nun gefunden, dass man zu einem 11-Ketosteroid gelangen kann, wenn man das Ergosterin-D-acetat mit einer Persäure oxy diert, auf das entstandene 9,11-Epoxyd ein hydrolysierend und gleichzeitig isomerisierend wirkendes Mittel einwirken lässt, das gebildete d 8,9;
22,23-3fl-Acetoxy-7,11-dioxy-ergostadien mit einem Oxydationsmittel behandelt, das Oxy dationsprodukt partiell reduziert und im so gebildeten 4 22.23-3ss-Acetoxy-7,11- diketo - ergo- sten die Carbonylgruppe in 7-Stellung durch Reduktion in die Methylengruppe umwandelt..

Das Verfahrensprodukt, das 4 22,23_3ss-Aeet- oxy-11-keto-ergosten vom F. =<B>125-1260</B> ist neu; es soll als Zwischenprodukt zur Herstel lung von therapeutisch wertvollen Verbindun gen verwendet werden.
Als Persäure für die Oxydation des Ergo sterin-D-a.cetats werden insbesondere Phthal- monopersäure oder Perbenzoesäure verwendet. Die Oxydation wird am besten in Gegenwart von Verdünnungsmitteln, wie organischen Lö sungsmitteln, z. B. halogenierten Kohlenwas- serstoffen oder Äthern, durchgeführt.
Als hydrolysierende und gleichzeitig iso- merisierende Mittel eignen sich verdünnte an organische Säuren, z. B. Schwefelsäure, in An wesenheit von Lösungsmitteln, wie Alkoholen, Ketonen oder Äthern, z. B. Aeeton oder Di- oxan.
Bei der Oxydation der 48,9-7,11-Dioxy-Ver- bindung kann ein Gemisch des<B>A8,9-</B> und des 8,9-Oxido-7,11-diketons entstehen. Für die Oxydation eignen sich beispielsweise Verbin dungen des sechswertigen Chroms, wie Chrom säure oder tert. Butylchromat, oder Perman- ganate. Man arbeitet dabei in Anwesenheit von Verdünntungsmitteln, wie wasserfreien oder wasserhaltigen Lösungsmitteln, z.
B. nie deren aliphatischen Säuren, wie Eisessig, Al koholen, wie tert. Butylalkohol, oder Ketonen, wie Aceton.
Für die Reduktion der 8,9-Doppelbindung bzw. 8,9-Oxidogruppe verwendet man vorteil haft naseierenden Wasserstoff, wie er zum Beispiel aus der Reaktion von Alkalimetallen bzw. deren Amalgamen mit Alkoholen, Wasser oder feuchten Lösungsmitteln, oder von Metal len, bzw. deren Salzen, beispielsweise Zink, Stannochlorid oder Eisen mit organischen oder anorganischen Säuren erhalten wird.
<I>Beispiel:</I> 1 Gewichtsteil Ergosterin-D-acetat (d7.8; 9,11;22,23-3fl-Aeetoxy-ergostatrien) wird in 10 Volumteilen Äther gelöst und mit 8 Volum- teilen einer ätherischen Phthalmonopersäure- Lösung versetzt, die 1,1 Äquivalente aktiven Sauerstoff enthält.
Die Reaktionslösung wird längere Zeit bei 0 und anschliessend bei Zim- mertemperatur belassen, dann mit wässeriger Natriumbicarbonatlösumg und \'Nasser gewa schen, getroeknet und eingedampft. Durch Umkristallisieren aus Mischungen von Äther und Methanol oder Äther und Aceton lässt sich das 47,8;22,2a-3fl-Acetoxy-9,11-oxido-ergo- stadien vom unveränderten Ausgangsmaterial abtrennen. Das Oxyd schmilzt bei 204 ; [a] D = -39,5 (c= 0,783 in Chloroform).
Die Te- tranitrometha.nprobe dieses Produktes ist im Gegensatz zum Ausgangsmaterial rein gelb.
0,2 Gewichtsteile des Oxyds werden in 150 Volumteilen Dioxan gelöst., mit 27 Volumteilen 2n-Sehwefelsäure versetzt und etwa 3 Minu ten bei Raumtemperatur heftig geschüttelt. Die Reaktionslösung wird sofort mit Äther und lNTatriumbiearbonat-Lösung behandelt. Die Ätherlösung wird rasch mit. Wasser gewaschen, getrocknet und eingedampft.
Das kristalli sierte Rohprodukt liefert nach Umlösen aus Aceton 0,14 Gewichtsteile 48,9;22.23_3f-Acetoxy- 7,11-dioxy-ergostadien vom F. = 269-270 ; [a] D = -I- 82 (c = 0,331 in Chloroform).
0,2 Gewiehtsteile fein pulverisiertes 48,9: 22.23_3fl_ Acetoxy-7,11-dioxy-er,;ostadien werden unter Eiskühlung mit 18 Volumteilen Eis- essig-Chromsäure-Lösung versetzt, die 0,00113 Gewiehtsteile aktiven Sauerstoff pro Volum- teil und 0,5 Volumteile 2n-Sehwefelsäure ent hält. Die Mischung wird etwa 3 Minuten in tensiv gerührt, worauf die Substanz gelöst ist.
Ansehliessend lässt man die Lösung etwa 14 Stunden bei Raumtemperatur stehen und ver setzt dann mit Wasser und Methanol, Nach etwa 2 Stunden nimmt man (las Oxydations produkt in Äther auf und wäscht den Äther mit verdünnter, eisgekühlter Natronlauge und anschliessend mit Wasser. Man erhält auf diese Weise 0,19 Gewichtsteile eines neutralen Oxy dationsproduktes. Durch fraktionierte Kristal lisation aus Methanol erhält man d 22,23_3f Acetoxy-8,9-oxido-7,11-diketo-ergosten vom F. = 130-131 ; [a]D =-73 (c = 0,501 in Chloroform) und in kleinerer Menged 8,9;22,23- 3ss-Acetoxy-7,11-diketo-ergostadien vom F. _ 132-135 .
3 Gewichtsteile der oben erhaltenen Mi schung von ds,9;22,23-3ss-Acetoxy-7,11-diketo- ergostadien und 422.23-3ss-Acetoxy-8,9-oxido- 7,11-diketo-ergosten werden in 250 Volumtei- len Eisessig gelöst und in die kalte Lösung 2,5 Gewiehtsteile Zinkstaub eingetragen. Hier auf erhitzt man die Mischung innerhalb von etwa 30 Minuten zur Siedehitze, wobei nach dem Beginn des Siedens innerhalb von etwa 25 Minuten in je 5 Portionen je 1,15 Gewichts teil Zinkstaub hinzugefügt wird. Die erkal tete Lösung wird vom unverbrauchten Zink abfiltriert und in Äther aufgenommen.
Die ä.therisehe Lösung wäscht man mit Wasser neutral, trocknet und verdampft sie. Nach Umkristallisation aus Aceton erhält man in praktisch quantitativer Ausbeute das 422,23- 3l-Aeetoxy-7,11-diketo-ergosten vom F.=195,5 bis 196 ; [a]D = -27 (c = 0;85 in Chloro form).
Zur Eliminierung der 7-Keto-Gruppe wer den 1,5 Gewichtsteile d22,23_3ss_Acetoxy-7,11-di- keto-ergosten mit 100 Volumteilen Triäthylen- glykol versetzt. Zur gebildeten Suspension gibt man unter Erwärmen auf dem Wasser bad so viel Methanol, bis alles gelöst ist. Hier auf werden zur obigen Lösung 2 Gewiehtsteile 1-lydrazinhydrat gegeben und etwa 15 Minu ten auf dem Wasserbad erwärmt.
Dann wer den nochmals 1,6 Gewichtsteile Hydrazin- li@#drat sowie 10 Gewichtsteile Natrium- hvdroxvd und 20 Volumteile Wasser zur Lö sung gegeben und diese innerhalb etwa einer Stunde auf 180 erwärmt, wobei die leicht flüchtigen Anteile abdestillieren. Anschlie ssend erhitzt man noch 2 Stunden auf 180 .
Nach üblieher Aufarbeitung wird das Reak tionsprodukt mit 50 Volumteilen Acetanhy- drid und 50 Volumteilen Pyridin eine halbe Stunde auf dem Wasserbad erwärmt, dann im Vakuum zur Trockne verdampft und der Rückstand an Aluminiumoxyd chromatogra- phiert. Die Petroläther-Benzol-(9:1)- und (8 :
2)-Fraktionen liefern nach Umlösen aus einer Mischung von Methanol und Wasser das d 22,23_3ss_Acetoxy-11-keto-ergosten vom F.=125 bis 126 ; [a] D = + 12,5 (c = 1,576 in Chloro form).
