Zwecks Herstellung von unsymmetrisehen Verbindungen kann man auch von Verbin dungen ausgehen, deren C"H2"-Gruppen ver schieden sind bzw. auch je ein Mol zweier ver schiedener Alkylierungsmittel oder zweier ver schiedener sekundärer bzw. tertiärer Amine nacheinander zur Einwirkung bringen.
Bei der Herstellung der diquartären Ver bindungen wird zur Erzielung völliger Um setzung in den meisten Fällen zweckmässig er wärmt. Ebenso werden die Umsetzungen der a,co-Bis-(halogenalkanoyloxy)-alkane mit den sekundären Aminen vorteilhaft in der Wärme vorgenommen.
Die Umsetzungen können in An- oder Abwesenheit von Lösungsmitteln durchgeführt werden; als solche kommen zum Beispiel Aceton, Butanon, Essigester, Benzol, Toluol, Xylol, Äther, Dioxan, Methanol, abs. Äthanol oder auch ein Überschuss an Alkylie- rungsmittel in Frage.
Als Ausgangsprodukte kommen z. B. a,o)- Bis-ehloraeetoxy-, a,co-Bis-bromacetoxy, a,w- Bis-jodacetoxy-hexan, -heptan, -oktan, -nonaii, -dekan, -undekan, -dodekan, tridekan und -te- t.radekan in Betracht, sowie beliebig in beiden Acylgruppen monohalogeniertes a,
co-Di-pro- pionyloxy-, a,co-Di-butyryloxy-, a,co-Di-isobu- tyryloxy-, a,co-Di-valeryloxy-, a,co-Di-isovale- ryloxy-, a,orBis-(a'-methyl-butyryloxy)- und a,co-Bis-trimethylaeetoxy-hexan, -heptan, -ok- tan, -nonan, -dekan,
-dodekan und -tetradekan. Diese Verbindungen können zum Beispiel mit Dimethylamin, Methyl-äthylamin, Methyl-pro- pylamin, Methyl-isopropylamin, Methyl-allyl- a.min, Methyl-butylamin, Methyl-isobutylamin, Methyl-amylamin, Diäthylamin, Äthyl-propyl- amin, Äthyl-butylamin,
Äthyl-amylamin, Di- propylamin, Diisopropylamin, Diallylamin, Dibutylaminen und Diamylaminen bzw. Tri- methjamin, Methyl-diäthylamin oder Tri- äthylamin umgesetzt werden.
Als Alkyliei-Lingsmittel kommen insbeson dere Alkylhalogenide, wie Methyljodid, Mc- thylbromid, Äthyljodid, Äthylbromid, Äthyl- ehlorid, Propyljodid, Propylbromid, Isopro- pylbromid, Allyljodid, Butylbromid,
Isobutyl- bromid und Amy lbromid sowie Schwefelsäure ester, wie Dimethylsulfat und Diäthvlsulfat. in Frage. Allgemein reagieren die Jodverbin dungen am leichtesten, die Chlorverbindungen am schwersten, wobei man quartäre Ammo- niumjodide oder -bromide auch mittels Sil berchlorid in die quartären Ammoniumchlo- ride überführen kann. Nötigenfalls werden die Umsetzungen in Druckgefässen ausgeführt..
Cxegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung einer Diammo- niumverbindung, welches dadurch gekenn zeichnet ist, dass man auf ein Mol 1,10-Bis-(a- diäthylamino-acetoxy)-dekan zwei Mol Äthyl- jodid einwirken lässt.
Die erhaltene neue Verbindung, das 1,10- Bis- (a-triäthylammonium-acetoxy) -dekan-di- jodid, schmilzt bei 145-148 . Sie soll thera peutische Verwendung finden.
<I>Beispiel:</I> Zu einer Mischung aus 17,4 Teilen 1,10- Dekandiol, 100 Volumteilen abs. Benzol und 20 Volumteilen abs. Pyridin werden, zu einer Herstellung des Ausgangsstoffes für das be anspruchte Verfahren, unter Rühren 27 Teile frisch destilliertes Chloracetylchlorid zuge- tropft, wobei die Temperatur durch Kühlen finit Eis und Wasser unterhalb 20 gehalten wird.
Man rührt anschliessend etwa 14 Stun den bei Raumtemperatur, gibt dann Äther zu und wäscht die abgetrennte ätherische Lö sung mehrmals mit Wasser. Nach Trocknen mit Natriumsulfat wird der Äther abdestil- liert und der Rückstand im Hochvakuum frak tioniert. Das gesuchte 1,7.0-Di-(chloracetoxy)= dekan geht unter 0,05 mm Druck bei 160 bis 164 über, es kristallisiert beim Stehenlassen.
1,3 Teile dieses 1,10-Di-chloracetoxy-dekans werden mit 50 Volumteilen abs. Benzol und 7.6 Teilen Diäthylamin gemischt und im ver schlossenen Gefäss etwa 14 Stunden auf 100 erhitzt. Nach dem Erkalten verrührt man mit so viel 2n-Salzsäure, dass der wässerige Teil gerade schwach kongosauer reagiert, schüttelt diesen abgetrennten Teil Rdederholt mit Äther aus, versetzt die wässerige Lösung dann unter guter Eiskühlung vorsichtig mit überschüssi ger 30 0%o iger Natronlauge und nimmt die aus geschiedene Base unverzüglich in Äther auf.
Nach Trocknen mit Kaliumcarbonat destilliert man zunächst den Äther ab und anschliessend unter 12 mm Druck im siedenden Wasserbad alle weiteren flüchtigen Anteile. Die zurück bleibende rohe Base wird mit Aktivkohle be handelt und dann davon abfiltriert.
4,0 Teile dieses 1,10-Bis-(a-diäthylamino- acetoxy)-dekans werden als Beginn einer Aus führung des beanspruchten Verfahrens in 15 Teilen trockenem Aceton gelöst und mit 3 Vo- lumteilen Äthyljodid etwa 14 Stunden unter Rückfluss gekocht. Die Lösung wird nach Er kalten vorsichtig mit trockenem Äthylacetat versetzt, so dass sie bei Zimmertemperatur ge rade noch klar bleibt, und dann auf -10 gebracht, bis Kristallisation eintritt.
Das quar- täre Salz wird abgesaugt, mit einer Mischung von Aceton und abs. Äther gewaschen und im Vakuum bei 50-60 getrocknet. Das Dijodid des 1,10-Bis-(a-triäthyl-ammonium-acetoxy)- dekans ist etwas hygroskopisch, es schmilzt bei 145--1480.
