Verfahren zur Herstellung von Primaquin. Die vorliegende Erfindung bezieht sieh auf die Herstellung des bereits bekannten Prima- quins und hat insbesondere zum Zweck, diesen Stoff unter Verwendung von leicht zugäng lichen Ausgangsmaterialien herzustellen.
Primaquin ist die Bezeichnung für den chemischen, Stoff, der als 6-Methoxy-8-(4'- amino-1'-methyl-bittylamino)-chinolin identifi ziert und im Journal! of the American Chemi- eal Soeiety Vol. 68, Seiten 15 bis 25 (1946) durch einen der Anmeld'er und seine Mitarbei ter beschrieben ist.
Dieser Stoff hat sehr wirk same therapeutische Eigenschaften zur Be kämpfung der Malaria, und insbesondere wurde noch gefunden, da.ss er ein kräftiges Mittel zur Behandlung gewisser Arten von Arthritis ist.
Es wurde nun ein verbessertes Verfahren zur Herstellung von Primaquin gefunden. Ge genstand der vorliegenden Erfindung ist also ein Verfahren zur Herstellung von Prima- quin und Zweck der Erfindung ist die Her stellung des Primaquins mit höheren Ausbeu- ten, und in grösserer Reinheit, als dies bisher möglich war.
Das erfindungsgemässe Verfahren zur Her stellung von Primaquin ist dadurch gekenn zeichnet, dass man 1,4-Dibrom-pentan mit einem All,alimetall-phthalimid in einem keine Hydroxylgruppen aufweisenden Lösungsmit tel erhitzt,
d@ass man das gebildete 4-Brom-1- phthalimido-pentan mit ss-1VTethoxy-8-amino- chinolin reagieren lässt. unter Bildung von 6- Methoxy-8-(1'-methyl - 4' - phthalimido - butyl- amino)-chinolin, und dass man dieses letztere Produkt mit Hydrazinhydrat behandelt, um durch Abspaltung von Phthalsäure die freie Base zu erhalten.
Bei der Durchführung der ersten Reak tionsstufe erhitzt man zweckmässig das 1,4 Dibrompentan mit. Kaliumphthahmid in Ace- tonlösung. Während dieser Reaktion reagiert das primär gebundene Bromatom am -Ende der Pentankette mit- dem Alkalimetall-Substituen- ten des Phthalimidkerns, so dass als Neben produkt Kaliumbromid entsteht.
Das Pentan- radikal vereinigt. sich dänn direkt, mit dem Kernstickstoffatom des Phthalimidradikals. Vorzugsweise verwendet man einen. stöchio- metrischen Überschuss von 1 bis 4=Dibrompen- tan, weil dann die Ausbeuten grösser sind. Im allgemeinen ist ein Überschuss von 5 0 bis 100% an Dibrompentan auf molarer Basis vorzuziehen.
Die Reaktionstemperatur wird ziveckmässigerweise zwischen 45 und 65 C gehalten. Die 'Temperatur des siedenden Ace tons unter atmosphärischem Druck liegt bei etwa 56 C, und da Aceton ein Lösungsmittel für beide Reaktionsteilnehmer ist, so wird es natürlich bevorzugt.
Die Reaktion verläuft selbständig, wenn die Reaktionsteilnehmer in Gegenwart. eines Überschusses von Aceton über einem Dampfbad erhitzt. werden, und. das Reaktionsgefäss wird vorzugsweise mit geeig neten Mitteln zum Kondensieren der ent wickelten Acetondämpfe und zum Zurückfüh- ren derselben in das Reaktionsgefäss versehen, Die Reaktion wird fortgesetzt, bis die 'Xli- sehung gegen Lakmus neutral ist, im allgemei nen während 24 bis 30 Stunden.
Vorzugsweise wird das Reaktionsgefäss mit geeigneten Mit teln zum Rühren oder zum Mischen der Re aktionsteilnehmer während des Reaktionsver- laufes versehen. Nach Beendigung der Reak tion kann man folgendermassen. vorgehen:
Die Lösung wird filtriert, um das gebildete Ka- liumbromid zu entfernen, und der Rückfluss- kondensator wird entfernt und durch einen gewöhnlichen Kondensator ersetzt; das Aceton wird zusammen mit, unverändertem 1,4-Di- brompentan abdestilliert und wiedergewonnen.
Das rolle Produkt wird im Vakuum destilliert und liefert eine Ausbeute an 4-Brom-l-phthal- imido-pentan, .die etwa 67 /o der theoretischen beträgt. Das Produkt ist ein Massgelbes Öl.
Die zweite Stufe der Synthese besteht darin, das gebildete 4-Brom-l-phthalimido- pentan (im folgenden als Verbindung I be zeichnet) mit 6-Methoxy - 8 - amino-chinolin, zweckmässig in einem Lösungsmittel, zur Re aktion zu bringen, wobei das freie Bromatom mit.
einem am Stickstoffatom des Aminosubsti- tuenten der Chinolinverbindung gebundenen Wasserstoffatom unter Bildung von 6- Methoxy-8-(1'-metlipl - 4' - phthalimido -butyl- amino)-chinolin reagiert. Diese Reaktion wird zweckmässig unter Erhitzen der Reaktions teilnehmer in einem geeigneten Lösungsmittel, wie zlun Beispiel Äthylalkohol, und unter Rühren durchgeführt.
Im allgemeinen ist die Reaktion in 60 bis 72 Stunden vollendet.; sie kann erleichtert werden, wenn man die Chino- linverbindung im Verhältnis von etwa 2 llol auf 1 Mol der Verbindung 1 anwendet. Nach Beendigung der Reaktion kann man folgen- z5 <B>Z,</B> vorgehen: Dein Gemisch wird ..eher zugesetzt; wobei ein fester Stoff ausgefällt wird.
Das Filtrat wird mit. wässerigem Ka- liumearbonat, dann mit Wasser gewaschen und zuletzt über Magnesiumsulfat getrocknet. Ein gelbes Produkt kristallisiert aus der Lö sung nach Zusatz von Alkohol und nach Ent- fernllng des Äthers; dieses Produkt wurde identifiziert, als 6-Methoxy - 8 - (Y- methyl-4'- phthaliniido-biit7lamino)-chinolin (im folgen- den als Verbindung 11 bezeichnet).
In der dritten Stufe wird die Plithalsäure- gruppe entfernt, wobei der Stickstoff an die Pentylgruppe gebunden bleibt und gleichzei tig an diesem Stickstoffatom zwei Wasser- stoffatome die Phthalsäuregruppe ersetzen und Phthalsäure abgespalten, wird.
Dies er folgt durch Behandlung der Verbindung 11 mit H.ydrazinhz-drat, zweekniässig in Alkohol bei Rückflusstemperatur. Nach einigen Stun den ist das alkoholisehe Lösungsmittel ab destilliert, und ein fester Körper beginnt sieh abzuscheiden, Dieser kann dann in wässerigem Kaliumhydroxyd gelöst und mit Äther extra hiert werden. Die Ätherlösun- wird dann zweckmässig mit Wasser -ewasehen und über Magnesiumsulfat getrocknet;
es ergibt. siel eine Ätherlösung der freien Base des Prima- quins. Aus dieser Lösung kann man Salze des Primaquins herstellen, welche direkt rein in Kristallform gewonnen werden können. Dazu wird die Lösung mittels einer Säure neutralisiert, zum Beispiel mit Salzsäure. Oxalsäure, vorzugsweise aber mit Phosphor säure.
Bei Zusatz von in absolutem Alkohol gelöster Oxalsäure bildet sieh ein gelber Nie derschlag von Prim.aquinmonooxalat. Der gelbe Niederschlag kann abfiltriert und aus 80pro- zentigem Alkohol oder aus Clvl@olmethyläthyl- äther umkristallisiert werden. Das Mono oxalat vom SN 13,272 schmilzt bei 182,7 bis 1ö5 C und das Diphosphat bei 197 bis 198 C.
Letzteres kann gewonnen werden durch Zusatz einer Lösung\ von 85prozentiger Phosphorsäure in 95prozentigem Äthylalko- hol unter Schütteln zu einer verdünnten iltherlösung der freien Base.
Der ausfallende gelbe Stoff geht durch weiteren Zusatz von Phosphorsäure in einen oran-efarbigen Stoff über, der nach Dekantieren des Lösungsmittels auf Zusatz von Alkohol hin kristallisiert lind nach LTmkristallisierung aus 90prozentigem Alkohol den angegebenen Schmelzpunkt auf weist, Die folgenden Beispiele zeigen, wie das erfindungsgemässe. Verfahren durchgeführt werden kann.
Ein Gemisch von 2 Mol 1,4-Dibrompentan und 1,5 Mol Kaliumphthalimid in 1,5 Litern Aceton wurde während 24 Stunden unter Rühren am Rückflusskühler erhitzt.. Nach Ab filtrieren des Kaliiunbromids, das mit. Aceton nachgewaschen wurde, wurde das Lösungs mittel und das überschüssige 1,4-ITibrom- pentan abdestilliert. Das rohe Produkt wurde bei etwa 167 C unter 0,25 mm Druck destil liert.
Zu Beginn der Destillation trat eine ge wisse Zersetzung auf, die bewirkte, dass Phthalimid, mit. dem Destillat sublimierte. Man erhielt mit einer Ausbeute von 67 % -die Verbindung I in Form eines Massgelben Öls folgender Zusammensetzung:

Wenn man an Stelle von Aceton höher siedende Lösungsmittel, zum Beispiel Dioxan oder Xylol, verwendet, so erfolgt als Neben reaktion eine weitgehende Entfernung des sekundären: Bromatoms in Form von Brom wasserstoff, unter Bildung von leicht poly merisierenden ungesättigten Verbindungen.
Eine Mischung von. 2 Mol 6-Methoxy-8- a.mino-chinolin und 1 Mol der gebildeten Verbindung I in einem Liter Äthylalkohol wurde unter Rühren während 72 Stunden am Rüekflusskühler erhitzt. Nach Abkühlung wurde Äther zugegeben und der feste Stoff wurde abfiltriert und. mit Äther gewaschen. Das Filtrat wurde mit wässerigem Kalium karbonat und dann mit Wasser gewaschen und über Magnesiumsulfat getrocknet.
Nach Abdestillieren des Äthers wurde Alkohol zu gesetzt, und die dunkelbraune Lösung wurde mit entfärbender Holzkohle gekocht, wodurch ein wesentlicher Teil der roten Farbe entfernt wurde. Wenn reine Ausgangsmaterialien ver wendet werden, so kristallisiert aus der Lö sung langsam ein gelbes Produkt aus. Die Ausbeute beträgt 40 bis 50 %.
Dieses Produkt. (Verbindung II) ergab folgende Analysenzahlen:

Gemisch von 0,06 Mol der Verbin dung I und 0,06 Mol 6-Methoxy-8-amino- ehinolin wurde zu 15 em3 Alkohol und zu 50 cm3 einer Lösung von 0,05 11_o1 Mono kaliumphosphat zugesetzt, welch letztere durch Zusatz von etwa 0,05 Mol Natriumhydroxyd auf den PH-Wert 8 gebracht wurde. Die Mi- sehung wurde gerührt und 79 Stunden lang auf 75 bis 80 erhitzt.
Die dunkelbraune ölige Schicht. wurde von cler wässerigen Lösung getrennt, mit. Kalium ka.rbonatlösung gewaschen, mit Äther ver- clünnt und über Magnesiumsulfat getrocknet. Das Lösungsmittel wurde abdestilliert und das zurückbleibende <B>Öl</B> wurde in Alkohol gelöst. Beim Abkühlen ergab sich eine rohe Aus- beute von 49%. eines festen Produktes.
Wenn bei dieser Reaktion die Verbindung I in rohem, nicht destilliertem Zustand verwen det wird, so bleibt das erhaltene Prodiukt (Verbindung II) -dunkel, sogar nach Behand- lung mit Holzkohle.
Anstatt eines kristalli sierten Produktes fällt ein. braunes öl aus, welches folgendermassen gereinigt werden kann: Das Öl wird mit wässerigem Brom, wasserstoff behandelt, um die Bromhydrate der umveränderten Chinolinverbindung und der Verbindung II niederzuschlagen.
Beide können durch Extraktion mit heissem Benzol getrennt werden, da das 6-Methoxy-8-amino- chinolin-bromhydrat in diesem Lösungsmittel nahezu unlöslich ist.. Nach Behandlung ,des Benzolextraktes mit überschüssiger Kalilauge wird das Lösungsmittel abdestilliert. Das er- lialtene blassgelbe Öl kann dann aus Alkohol auskristallisiert werden, wobei die Verbin- dttng II in reinem Zustand erhalten wird.
Die Verbindung II wurde durch zweistün dige Behandlung ihrer alkoholischen Lösung am Rückflusskühler mit der berechneten llense Hydrazinhydrat hydrolysiert. Der Alkohol wurde abdestilliert, bis ein fester Stoff sich abzuscheiden begann.
Dieser feste Stoff (Phthalsäure) wurde durch Zusatz von wässeriger Kalilauge gelöst. und die Lösung niit Äther extrahiert. Nach Waschen der Ätherlösung mit Wasser wurde sie über Ma- g-nesiumsulfat getrocknet. Sie enthielt. die freie Base des Primaquins.
Diese freie Base zersetzt. sich beim Ver such der Vakuumdestillation, gleichgültig, wie hoch das Vakuum ist. Es ist daher angebracht, für die Herstellung von Salzen direkt diese Lösung der Base zu verwenden. <I>Beispiel</I> II:
Ein Gemisch von 0,05 Mol des nach Bei spiel 1 aus 1,4-Dibrompentan und-.Kalium- phthalimid hergestellten 4 - Brom -1- phthal- imido - pentans und 0,1 Mol 6-llethoxy-8- amino-chinolin wurde unter Rühren eine halbe Stunde lang in einem Ölbad auf 125 C er hitzt. Die Erhitzung wurde dann eine Stunde lang bei 95 bis 105 C fortgesetzt.
Während dieses Erhitzungsprozesses bildete sich festes Material, nämlich 6-11ethoxy-8-amino-chinolin- br omhydrat.
- Dann wurden 25 em3 Alkohol hinzugefügt. Lind die Mischung 18 Stunden lang unter Rückfluss behandelt. Etwa 7 g 6-Methoxy-8- amino-chinolin-bromhydrat wurden abfiltriert. Die zurückbleibende braune Flüssigkeit wurde riüt Kaliumkarbonatlösung durchgerührt und mit Äther extrahiert:. Die Ätherschicht wurde über Magnesiumsulfat getrocknet und das Lösungsmittel entfernt.
Der ölige Rückstand wurde bei 103 bis 127 C unter 7 bis 10 Mikrons destilliert. Ein fester Stoff begann sich abzuscheiden und die Destillation wurde abgestellt. In das De stilliergefäss wurde Äther gegossen. Der feste Stoff löste sich nicht, dagegen löste sich das glasige Material am Boden dies Gefässes. Etwa. 3,3 g festes Material wurden abfiltriert. Nach Umkristallisierung aus Alkohol erhielt man oran-efarbene, nadelförmige Kristalle vom Schmelzpunkt 235 C. Dies kann ein dialkyliertes Produkt. sein.
Die Ätherlösung wurde verdampft und das erhaltene Öl aus Alkohol schwierig kri stallisiert.. Es ¯vurden etwa 4,8 g einer festen gelben Substanz erhalten, was einer Ausbeute von 24 % des erwarteten Phtlialimid-Z -isclien- produktes entspricht. Nach zweimaliger L'm- krist.allisierung aus Alkohol zeigte das feste Produkt. einen Schmelzpunkt von 89 bis 90,5 C.
Die Hydrolysierung des erhaltenen Produktes erfolgt wie in Beispiel I besehrie- ben.
<I>Beispiel</I> 11I: Ein Gemisch von 1,0 Mol Kaliumphthal- imid und 1;5 Mol 1,4-Dibrompentan in 1000 em3 Aceton wurde unter starkem Rühren 30 Stun den lang unter Rückfluss erhitzt, bis es gegen Lakmus neutral war. Nach Abkühlung wurde das Kaliumbromid abfiltriert und mit Aeet.on gewaschen.
Das Lösungsmittel und unverän dertes Dibrompentan wurden vom Filtrat un ter dem Druck der Wasserstrahlpumpe ab destilliert. Das erhaltene Produkt destillierte bei etwa 120 bis 130 C -unter 15 mm Druck.
Man er- hielt. eine Ausbeute von 67 % an reinem Pro- dukt. Ein Gemisch von 0,05 Mol des erhaltenen 4-Brom-l-phthalimidio-peiitans und<B>0,1</B> Mol 6 Methoxy-8-amino-chinolin in 25 em3 Alkohol wurde auf dem Dampfbad unter Rüekfluss 67 Stunden lang erhitzt.
Das kristallinische 6- 'L#lethoxy-8-amino-chinolin-bromhydrat, das als ?Nebenprodukt aus dem Reaktionsgemisch ab filtriert wurde, wog 9 g. Daraus konnte ge- schlossen werden, dass also etwa 71% der Verbindung I in der Reaktion verbraucht worden: waren.
Das Alkohol-Filtrat wurde mit einer wässe rigen Kaliumkarbonatlösung geschüttelt und mit Äther ausgezogen. Die vereinigten Äther schichten wurden mit Wasser gewaschen, über Magnesiumsulfat getrocknet und der Äther wurde abdestilliert.
Das ölige Material, das zurückblieb, wurde nicht destilliert, ' sondern aus Alkohol mit einiger Schwierigkeit auskristallisiert. Man erhielt. total 9,9 g, was einer Ausbeute an Verbindung II von. 51 1/o entspricht. Dieses Produkt wurde, wie in Beispiel I beschrieben, mit Hydrazinhy drat zur freien Primaquin- base hydrolysiert. -
Process for the preparation of primaquine. The present invention relates to the production of the already known primaquin and in particular has the purpose of producing this substance using easily accessible starting materials.
Primaquine is the name of the chemical substance that is identified as 6-methoxy-8- (4'-amino-1'-methyl-bittylamino) -quinoline and is used in the journal! of the American Chemical Soeiety Vol. 68, pages 15 to 25 (1946) by one of the applicants and his co-workers.
This substance has very effective therapeutic properties for combating malaria, and in particular it has been found to be a powerful agent for the treatment of certain types of arthritis.
An improved process for the production of primaquine has now been found. The subject of the present invention is therefore a process for the production of primaquin, and the purpose of the invention is the production of the primaquin with higher yields and in greater purity than was previously possible.
The inventive method for the manufacture of primaquine is characterized in that 1,4-dibromopentane is heated with an all, alimetallic phthalimide in a solvent which does not contain any hydroxyl groups,
that is, the 4-bromo-1-phthalimido-pentane formed is allowed to react with ss-1V-ethoxy-8-aminoquinoline. with formation of 6-methoxy-8- (1'-methyl-4'-phthalimido-butyl-amino) -quinoline, and that this latter product is treated with hydrazine hydrate in order to obtain the free base by splitting off phthalic acid.
When carrying out the first reaction stage, the 1,4 dibromopentane is expediently heated with. Potassium thahmid in acetone solution. During this reaction, the primarily bonded bromine atom at the end of the pentane chain reacts with the alkali metal substituents of the phthalimide nucleus, so that potassium bromide is formed as a by-product.
The pentane radical united. directly with the nuclear nitrogen atom of the phthalimide radical. Preferably one is used. stoichiometric excess of 1 to 4 = dibromopentane, because then the yields are greater. In general, an excess of 50 to 100 percent dibromopentane on a molar basis is preferred.
The reaction temperature is usually kept between 45 and 65.degree. The temperature of the boiling acetone at atmospheric pressure is about 56 C and, of course, since acetone is a solvent for both reactants, it is preferred.
The reaction proceeds automatically when the reactants are in the presence. an excess of acetone heated over a steam bath. be, and. the reaction vessel is preferably provided with suitable means for condensing the acetone vapors that have developed and for returning them to the reaction vessel. The reaction is continued until the lacquer is neutral, generally for 24 to 30 hours.
The reaction vessel is preferably provided with suitable means for stirring or for mixing the reaction participants during the course of the reaction. After the end of the reaction, you can do the following. Action:
The solution is filtered to remove the potassium bromide that has formed, and the reflux condenser is removed and replaced with an ordinary condenser; the acetone is distilled off together with unchanged 1,4-di-bromopentane and recovered.
The roll product is distilled in vacuo and gives a yield of 4-bromo-1-phthalimido-pentane, which is about 67 / o of the theoretical. The product is a mass yellow oil.
The second stage of the synthesis consists in reacting the 4-bromo-1-phthalimido-pentane formed (hereinafter referred to as compound I) with 6-methoxy-8-amino-quinoline, conveniently in a solvent, where the free bromine atom with.
a hydrogen atom bonded to the nitrogen atom of the amino substituent of the quinoline compound reacts to form 6-methoxy-8- (1'-metlipl - 4 '- phthalimido-butyl-amino) -quinoline. This reaction is expediently carried out with heating of the reaction participants in a suitable solvent, such as ethyl alcohol for example, and with stirring.
Generally the reaction is complete in 60 to 72 hours .; it can be facilitated if the quinoline compound is used in a ratio of about 2 mol to 1 mol of compound 1. After the reaction has ended, you can proceed as follows: Your mixture is ... more likely to be added; whereby a solid material is precipitated.
The filtrate is with. aqueous potassium carbonate, then washed with water and finally dried over magnesium sulfate. A yellow product crystallizes from the solution after the addition of alcohol and after removal of the ether; this product has been identified as 6-methoxy-8 - (Y-methyl-4'-phthaliniido-biit7lamino) -quinoline (hereinafter referred to as compound 11).
In the third stage, the phthalic acid group is removed, the nitrogen remaining bound to the pentyl group and, at the same time, two hydrogen atoms on this nitrogen atom replacing the phthalic acid group and phthalic acid being split off.
This he follows by treating compound 11 with H.ydrazinhz-drat, two-fold in alcohol at reflux temperature. After a few hours the alcoholic solvent is distilled off, and a solid body begins to separate. This can then be dissolved in aqueous potassium hydroxide and extracted with ether. The ether solution is then conveniently washed with water and dried over magnesium sulfate;
it results. An ethereal solution of the free base of the primaquine fell. This solution can be used to produce primaquine salts, which can be obtained directly in crystal form. To do this, the solution is neutralized using an acid, for example hydrochloric acid. Oxalic acid, but preferably with phosphoric acid.
When oxalic acid dissolved in absolute alcohol is added, a yellow precipitate of primary aquine monooxalate forms. The yellow precipitate can be filtered off and recrystallized from 80 percent alcohol or from Clvl @ olmethylethylether. The mono oxalate of SN 13.272 melts at 182.7 to 15 C and the diphosphate at 197 to 198 C.
The latter can be obtained by adding a solution of 85 percent phosphoric acid in 95 percent ethyl alcohol with shaking to a dilute ether solution of the free base.
The precipitated yellow substance changes to an orange-colored substance through further addition of phosphoric acid, which crystallizes after decanting the solvent on the addition of alcohol and has the specified melting point after crystallization from 90 percent alcohol. The following examples show how the invention. Procedure can be carried out.
A mixture of 2 moles of 1,4-dibromopentane and 1.5 moles of potassium phthalimide in 1.5 liters of acetone was heated for 24 hours with stirring on the reflux condenser. After the potassium bromide had been filtered off. Acetone was washed, the solvent and the excess 1,4-ITibrompentane was distilled off. The crude product was distilled at about 167 C under 0.25 mm pressure.
At the beginning of the distillation there was some decomposition which caused the phthalimide, with. sublimed to the distillate. Compound I was obtained with a yield of 67% in the form of a pale yellow oil of the following composition:
