Procédé de préparation d'une nouvelle oxazoline thérapeutiquement active. La présente invention a pour objet un pro cédé de préparation d'une forme t.hérapeu- tiquement active d'une nouvelle oxazoline, utilisable comme antibiotique ou comme inter médiaire dans la production d'agents théra peutiques.

Le terme structural se réfère à la relation spatiale des groupes respective ment fixés sur les deux atomes de carbone asymétriques.
Par analogie avec la nomenclature adop tée par Rebstock et al (J. A. C. S. Vol. 71 pp. 245S-2473) en connexion avec l'amino- diol diastéréoisomère, le 2-diehloracétamido-1- p-nitrophényl-propane-l,3-diol (également connu sous le nom de ehloramphénicol), les formes isomères de structure sont nommées dans le présent mémoire érythro et thréo respectivement.
Le présent brevet se rapporte à la prépa ration des formes thérapeutiquem.ent actives de l'isomère thréo du composé selon la for mule ci-dessus.
La forme thréo peut exister comme racé- mate d'isomères optiquement actifs ou comme isomère séparé, c'est-à-dire comme DL-thréo, L-thréo et. D-thréo.
Il est à signaler que la représentation eonfigurationnelle des isomères connus D et L n'a aucune relation avec le sens réel de rota tion, mais se réfère à la configuration con cernant l'atome de carbone alpha.
Les isomères DL- et D-thréo seuls ont un intérêt thérapeutique direct du fait. qu'ils possèdent une activité de même nature que celle des formes DL- et D-thréo du 2-dichlor- acétamido-1-p-nit.rophényl-propane-1,3-diol et. sont ainsi utilisables dans les mêmes domaines thérapeutiques. La. présente invention se rap porte donc uniquement à la préparation des isomères DL-thréo et D-thréo de ladite oxazo- line. Leur degré de tolérance est comparable à celui de la substance connue.
En outre, ils présentent. également sur eux l'avantage qu'ils sont pratiquement sans saveur alors que ce ehloramphénicol et. le mélange racémique le contenant sont fortement amers. En outre, le composé préparé selon le présent procédé, c'est-à-dire les formes Dl.- et D-thréo de l'oxazoline, est. important. comme intermédiaire pour la. préparation du chloramphénicol et du mélange racémique duquel le chloramphénicol est le constituant D.
Le procédé selon l'invention pour la pré paration d'une forme thérapeutiquement active de la thréo-\?-dichlorométhyl-5-p-nitrophényl- 4-hydroxyméthyl-42-oxazoline est caractérisé en ce qu'on fait réagir le DL- ou le D-thréo 2- amino-1-paranitroph6nyl-propane-1,3-diol de formule

où R représente un alcoyle, X un atome d'oxygène ou de soufre, ou avec. un sel d'addi tion d'acide d'un tel éther. L'éther utilisé est.
donc un imino-éther ou un imino-thio-éther. Le mélange réactionnel résultant contient l'oxazoline selon la formule indiquée, qui peut être séparée, par exemple par cristallisation fractionnée ou par chromatographie. Si on désire l'isomère D-thréo selon ladite formule à l'état pur, il est préférable d'em ployer comme matière première l'amino-diol D-thréo à l'état pur, car l'oxazoline est assez instable et par suite difficile à isoler par les méthodes classiques.
L'invention est. illustrée par les exemples suivants Exemple .1: 1,65 g de D(-)-thréo-2-amino-1-p-nitro- phényl-propane-1,3-diol, préparés par la méthode décrite par Rebstock et al (J. A. C. S.
Vol. 71 pp. 2458-2473) sont dissous à chaud dans 10 cms de py ridine sèche et traités avec 1,8 g de chlorhydrate de dichloracétiminoéthyl- éther. Le mélange est abandonné au repos à la température ordinaire pendant quelques heures et le chlorhydrate de pyridine est séparé par filtration.
Par évaporation du sol vant et cristallisation du résidu dans le méthanol et recristallisation également dans le méthanol, on obtient, comme produit secon daire de la réaction, la. D-thréo-2-dichloro-- méthyl - 4 - p - nitr ophény lhy droxy méthyl - J'_ - oxazoline, fondant à 143-141-".

3,18; N, 9,17; Cl, 23,2%. Les eaux mères de cette cristallisation sont. évaporées à sec et le résidu soumis à la cristallisation fractionnée dans le méthanol, l'acétate d'éthyle et enfin le benzène. On obtient ainsi le produit cherché qui est la DL-thréo-2-dichlorométh@#1-5-p-nitro- phényl-4-hydroxyméthZ,1-.d2-oxazoline, fondant à 128-129 .
Trouvé: C, 43,3; H, 3,3; N, 9,1; Calculé pour: C11H1o04N.Cl.: C, 43,25; H, 3,18; N, 9,17D/o. <I>Exemple 3:</I> 1,1 g de DL-thréo-2-amino-1-p-nitrophényl- propane-1,3-diol dans 6,6 em3 de pyridine anhydre sont traités avec 1,3 g de chlor- hydrate de dichloraeétimino-6thyl-thio-éther. La solution résultante est abandonnée une nuit au repos à température ordinaire puis filtrée. Le solide est lavé avec un peu de pyri- dine anhydre.
Le filtrat réuni aux lavages est évaporé sous vide et donne une gomme r ougeâtre <B>à</B> qu'on dissout dans 10 CM3 de méthanol chaud. Le solide cristallisé qui se sépare est recristallisé dans le méthanol. C'est la DL-thréo-2-dichlorométhyl-4-p-nitrophényl- hydroxyméthyl-d2-oxazoline, fondant à 162 à 163 , qui s'est formée comme produit secon daire dans la réaction.
Les eaux mères sont évaporées sous vide et donnent une gomme qui se solidifie par trituration avec un peu d'alcool méthylique. Le solide ainsi obtenu, recristallisé dans l'acétate d'éthyle, donne le produit cherché, la DL-thréo-2-dichlorométhyl-5-p-nitrophényl- 4-hy droxyméthyl-d2-oxazoline, fondant à 128 à 130 .
Process for preparing a novel therapeutically active oxazoline. The present invention relates to a process for preparing a therapeutically active form of a novel oxazoline, which can be used as an antibiotic or as an intermediate in the production of therapeutic agents.

A compound of the above formula contains two asymmetric carbon atoms and can exist in isomeric forms both structurally and optically. The term structural refers to the spatial relationship of the groups attached to the two asymmetric carbon atoms, respectively.
By analogy with the nomenclature adopted by Rebstock et al (JACS Vol. 71 pp. 245S-2473) in connection with the diastereoisomeric amino-diol, 2-diehloracetamido-1- p-nitrophenyl-propane-l, 3-diol (also known as ehloramphenicol), the structural isomeric forms are referred to herein as erythro and threo respectively.
The present patent relates to the preparation of therapeutically active forms of the threo isomer of the compound according to the above formula.
The threo form can exist as a racemate of optically active isomers or as a separate isomer, i.e. as DL-threo, L-threo and. D-threo.
It should be noted that the eonfigurational representation of the known D and L isomers has no relation with the real direction of rotation, but refers to the configuration concerning the alpha carbon atom.
The DL- and D-threo isomers alone are of direct therapeutic interest because of this. that they have an activity of the same nature as that of the DL- and D-threo forms of 2-dichloracetamido-1-p-nit.rophenyl-propane-1,3-diol and. can thus be used in the same therapeutic fields. The present invention therefore relates only to the preparation of the DL-threo and D-threo isomers of said oxazoline. Their degree of tolerance is comparable to that of the known substance.
In addition, they present. also over them the advantage that they are practically tasteless while this ehloramphenicol and. the racemic mixture containing it are strongly bitter. Further, the compound prepared according to the present process, i.e. the D1.- and D-threo forms of oxazoline, is. important. as an intermediary for the. preparation of chloramphenicol and the racemic mixture of which chloramphenicol is the constituent D.
The process according to the invention for the preparation of a therapeutically active form of threo - \? - dichloromethyl-5-p-nitrophenyl-4-hydroxymethyl-42-oxazoline is characterized in that the DL- is reacted. or D-threo 2-amino-1-paranitroph6nyl-propane-1,3-diol of formula

therefore an imino-ether or an imino-thio-ether. The resulting reaction mixture contains the oxazoline according to the given formula, which can be separated, for example by fractional crystallization or by chromatography. If the D-threo isomer according to said formula is desired in the pure state, it is preferable to employ as the raw material the amino-diol D-threo in the pure state, since the oxazoline is quite unstable and therefore difficult to isolate by conventional methods.

The resulting solution is left to stand overnight at room temperature and then filtered. The solid is washed with a little anhydrous pyridine.
The filtrate combined with the washings is evaporated in vacuo and gives a red gum <B> to </B> which is dissolved in 10 cm3 of hot methanol. The crystalline solid which separates is recrystallized from methanol. DL-threo-2-dichloromethyl-4-p-nitrophenyl-hydroxymethyl-d2-oxazoline, melting point 162 to 163, formed as a side product in the reaction.
The mother liquors are evaporated off under vacuum and give a gum which solidifies by trituration with a little methyl alcohol.
