Verfahren zur Herstellung von Vitaminprodukten. Gegenstand der Erfindung ist ein Ver fahren zur Gewinnung wertvoller Vitamin produkte durch Kultivieren ausgewählter Stämme von Pilzen (Fungi), wobei in den Nährböden Substanzen mit Vitamin-1312-Wir- kung erhalten werden, die das Wachstum vor. Lactobaeillus laetis Dorner fördern und als Nahrungsergänzungsstoffe wichtig sind.
Das erfindun- cemässe Verfahren zur Hentellunt, von Produkten mit Vitamin-1312- Wirkung ist* dadurch gekennzeichnet, dass man einen Mikroorganismus kultiviert, wel cher aus denjenigen Stämmen von Pilzen aus gewählt wurde, die bei der Züchtung in einem flüssigen Nährmedium Gärbrühen lie fern, welche mindestens 20 LLD-Einheiten je Milligramm der in ihnen gelösten Feststoffe aufweisen, wobei das Kultivieren des Mikro organismus in dein flüssigen Nährmedium so lange Zeit erfolgt,
bis die Gärbrühe min destens den genannten Vitamingehalt aufweist und die wirksamen wasserlösliehen Stoffe so dann von der Brühe abgetrennt werden<B>',</B> und zwar in der Weise, dass praktisch eine Vitamin- zersetzuno- vermieden ist.
Die durch das erfindungsgemässe Ver fahren erhaltenen Vitaminsubstanzen können für verschiedene Zwecke verwendet werden, z. B. zur Anreieherung von Tiernahrung mit einem mangelhaften Gehalt am animalisehen Proteinfaktor.
Als Test der erfindungsgemäss gewonnenen Vitaminprodukte dient gewöhnlich ihre waehs- tumsfördernde Wirkung in bezug auf den Mikroorganismus Laetobaeill.us laetis Dorner (LLD) unter der von Shorb (,f. Biol. Chem. <B>1.69,</B> 455-6) beschriebenen Versuehsbedin- gungen. Die Wirksamkeit der Vitamin produkte, entsprechender Gärungsbrühen oder angereicherten Konzentraten,
kann durch eine inikrobiologisehe -#Vaehstumsbestimmungsme- thode unter Verwendung von LLD als Test- or,-anismus ermittelt werden, wobei als will kürlicher Standard für die Wirksamkeit von <B>1000</B> Einh.,'mg diejenige Menge eines gerei- ni-ten Leberextraktes dient, welche zur Er zielung des halben maximalen Wachstums be nötigt wird. Reines kristallines Vitamin BI2 hat etwa<B>11</B> Millionen LLD-Einli./mg.
Es ist schon lange bekannt, dass manche Tiernahrungsmittel und insbesondere die in der Hühnerzueht verwendete Nahrung, welche in der Hauptsache aus Pflanzenmaterial, z. B. Getreidekörner, Sojabohnenmehl, Alla und dergleichen, besteht, wohl in bezug auf den Gehalt an Aminosäuren-, Mineralstoffe und den Vitaminen bekannter chemischer Struk tur vollwertig sind, aber einen Mangel an einem oder mehreren Faktoren aufweisen, die für die maximale Waehstumsgesehwindigkeit der Tiere unerlässlich sind.
Ein solcher Fak tor wird gewöhnlich in den Geweben des Huhns aufgespeichert und durch das Ei auf das Küeken übertragen. Daher'sollten Hennen mit Rationen gefüttert werden, welche diesen Faktor enthalten, um genügende Brutfähig- keit der Eier und die Lebensfähigkeit der ausgebrüteten Kücken zu sichern. Die Kücken müssen die betreffende Substanz entweder direkt mit der Nahrung oder indirekt durch das Ei von der Henne erhalten, damit sie sich optimal entwickeln und übermässige Sterblich keit vermieden wird.
Es ist demnach ersieht- lieh, dass der genannte Faktor für die Wirt- sehaftlichkeit der Hühnerzucht grosse Bedeu tung besitzt.
Gute Nährmittel für Hühner enthalten den animalischen Protein-Faktor (im nach folgenden mit APF bezeichnet) bisher in der Form von animalischen Produkten, wie Leber- inehl, Fischmehl, Fischextrakte und derglei- ehen. Diese Zusätze sind in den benötigten Mengen schwer zu beschaffen, und ihr hoher Preis verteuert die IEIühnerzueht erheblich. Die genannten Produkte haben zudem den Nachteil, dass sie verhältnismässig geringe und wechselnde M.,engen des benötigten APP enthalten.
Eine ergiebigere und billigere Quelle dieses wichtigen Bestandteils der Nahrung ist daher ein wertvoller Fortschritt. Eingehende Versuche haben gezeigt, dass die durch Kultivieren von gewissen Pilz stämmen in wässerigen Nährmedien erhal tene LLD-wirksamen Gärungsrückstände eine reiche Quelle eines waehstumsfördernden Mit tels darstellen, welches praktiscli dem so- genannten animalisehen Protein-Faktor in der Förderung des Wackstums von Küeken gleichkommt.
Es wurde ferner gefunden, dass aus den genannten Gärangsrilekständen ein reines Vitamin B12 isoliert werden kann und bei einer an APF unzureichenden Ration der Zusatz von nur 0,0000031/o Vitamin BI.2 eine maximale Iffachstumsgeschwindigkeit bei Kücken ergibt. Man braucht zu diesem Zweck das Vitamin B12 nicht in reiner Form aus der Brühe zu gewinnen, sondern es können auch Konzentrate mit einem entsprechenden Gehalt an LLD-wirksamen Substanzen als Futterzusatz verwendet werden.
Beim erfin dungsgemässen Verfahren werden Pilzstämme kultiviert, welche Gärflüssigkeiten liefern, die wenigstens 20 LLD-Einheiten pro mg der ent- haltenen Feststoffe zeigen. Es hat sieh ge zeigt, dass die wasserlösliehen festen Bestand teile soleher Brühen eine Wirksamkeit in der Höhe von etwa 140 LLI)-Eiiih./mg. aufweisen.
Es können leicht Konzentrate hergestellt wer den, welehe 2000 LLD-Eiiih.,'mg oder mehr enthalten, währenddeni die bisherigen reich sten APF-haltigen Produkte, wie Fisehmehl und Fisehextrakte, nur etwa<B>1-3</B> LLD- Einh.Img zeigen.
Eine an APP mangelhafte Grundration für Hühner kann, wie bereits erwähnt, da durch in bezug auf APF vollwertig gemacht werden, dass ihr 0,0000031/o Vitaniiii- B12 zu gesetzt wird, an Stelle von<B>1</B> bis 51/o Fiseh- mehl oder Fischextrakt. Genannter Vitamin- B12-Zusatz entsprieht ungefähr<B>3130 000</B> LLD- Einheiten <B>je kg</B> Futter.
Ein Konzentrat von 2000 LLD-Einh./mg liefert einen Gehalt von <B>330 000</B> LLD-Einheiten <B>je kg</B> Futter bei einem Zusatz von<B>165</B> mg des Konzentrats zu<B>1 kg</B> Futter<B>(0,0165</B> 1/o).
Es ist bekannt, dass versehiedene wasser lösliche Gärprodukte als Nahrungszusatz ver wendet werden können, hauptsächlich auf Grund ihres Gestalts an Riboflavin und an dern Vitaminen bekannter Struktur, wie Biotin, Pantothensäure usw. Solche Zusätze enthalten nur geringe Mengen LLD-wirk- samer Substanzen.
Die lösliehen Bestandteile derBlitylgärung, welche erhaltenwerden dureh Eindampfung der naeh dem Abdestillieren des B-Litylalkohols als Rüekstand gewonnent,ii Flüssigkeit, haben nur etwa 4 LLD-Einh./m.- und die löslichen Bestandteile der Getreide- schlempe weniger als<B>1</B> l,
LD-Einh./mg. Bei einem solchen Gehalt kann man praktisch nicht an eine Gewinnung von Konzentraten denken, die für das Anreichern von an APF unzureichendem Futter dienen sollen.
Anders dagegen, wenn das Ausgangs material wenigstens<B>20</B> LLD-Einh./m- darin enthaltener Feststoffe aufweist. Zar Erzeu gung derartiger Gärbrühen geeignete Mikro organismen sind die FLiiigi, welehe im Buche von Smith und Raistrick An introduetion to industrial niveology (London, Edward Arnold & - Co.,<B>1938)</B> auf Seite 2 angegeben sind, das heisst Myxomyeeten,
Schizomyeeten und Eumyeeten. Die Sehizomyeeten sind be sonders geeignet, insbesondere gewisse Stämme der Gattung Streptomyees griseus, welche auch für die Herstellung der Antibiotiea Streptomyein und Grisein verwendet werden können.
Auch andere Gattun,gen, wie Strept. albidolla-,vms, Strept. eolombiensis nov. sp., Strept. roseoehromogenus und Strept. anti- biotieus, weisen Stämme auf, welche hohe Ausbeuten an LLD-wirksamen Substanzen liefern.
Andere verwendbare Schizom.veeten sind Glieder des Stammes Clostridium. Ver suche zeigten, dass ausgewählte Stämme von Clostridium tetanomorplium, Clostridium eo- ehlearium, Clostridium flabel.Iiierum und Clo- stridium butyrieum ebenfalls für die Durch führung des erfindungsgemässen Verfahrens gut geeignet sind.
Andere Fungi, welche sich für die technische Herstellung von Gärbrühen mit mindestens 20 LLD-Einh./mg der darin enthaltenen Peststoffe als brauchbar erwiesen haben, sind Torula, Eremotheeium ashybii und Escheriehia coli.
Das erfindungsgemässe Verfahren kann teilweise mit der Herstellung der Antibiotiea Streptomyein und Grisein einhergehen. Bei den Streptomycin- und Griseingärverfahren werden Maisehen niedriger Konzentration vergoren, wobei für eine verhältnismässig ge ringe Menge des Antibiotikums grosse Mengen Gärbrühe entstehen. Bei einer grossen Strepto- myeinanlage können täglich nach Abtrennung des Antibiotiums viele tausend Kubikmeter Gärflüssigkeit anfallen.
Bisher war nicht be kannt, dass die genannten Rückstände noch ein nützliches Produkt enthalten. Tatsächlich ist der Gehalt an andern Vitaminen als Vit amin Bl,2 zu gering, um die Gewinnung jener andern Vitamine wirtschaftlich zu machen. Bei der technischen Herstellung von Streptomycin bildete der Anfall von unverwertbarem Mate rial in grosser Menge bisher ein Problem.
Inzwischen hat sich gezeigt, dass die Gär- flüssigkeit nach der Abtrennung des Antibio tikums Streptomyein mittels lonenaustauseli- Harzen bedeutend mehr als 20 LLD-Einheiten <B>je</B> Milligramm darin enthaltener Feststoffe aufweisen, und aus den genannten Rüel#.stän- den Konzentrate von hoher LLD-Wirksamkeit erfindun'gsgemäss erhalten werden.
Für die Durchführung des erfindungs gemässen Verfahrens kann als Ausgangsmate rial jedes wässerige Nährmedium verwendet werden, welches üblieherweise zum Kultivie ren von Pilzen dient. Die üblichen Nähr flüssigkeiten enthalten eine Kohlenstoffquelle, eine Stiekstoffquelle, anorganische Salze und gegebenenfalls auch Waehstumsfaktoren. Der Kohlenstoff kann von einem Kohlehydrat ge liefert werden, z. B. von Dextrose, Maltose, Xylose, Invertzueker, Getreideauszüge und dergleichen.
Der Stickstoff kann von einem Ammoniumsalz, von Aminosäuren oder von Proteinen stammen, z. B. Sojabohnen, Hafer, Hefeextrakten, Produkten des Abbaus von Casein mit Tryptin, Fleisehextrakt, Blutmehl, proteinreiehem Fleisch und Knoehenmaterial, dem fein gemahlenen getrockneten Rüekstand von animalisehen Geweben, die frei von Haaren, Klauen, Horn, Blut, Darm- und Mageninhalt sind und mehr als 4,
4% Phosphor enthalten, ferner von Fisehmehl und Fisehextrakten, löslichen Bestandteilen der Schlempe und dergleichen. Falls ge wünscht, können die Pilze unter Verwendung von Proteinen oder Aminosäuren lmltiviert werden, das heisst ohne Kohlenhydrate in Nährmedium, in welchem Falle die Proteine bzw. Aminosäuren sowohl als Kohlenstoff- wie auch als Stickstoffquelle dienen.
Das Nährmedium wird zweckmässig steri lisiert Lind dann mit einem Stamm des ge wählten Pilzes geimpft. Das Gemisch wird der Inkubation unterworfen, bis die Gärflüssig- keit einen Gehalt von wenigstens 20 LLD je mg Feststoffe in der Brühe aufweist.
Die Impfung wird gewöhnlich unter der Ober fläche der Flüssigkeit ausgeführt und bei der für den verwendeten Pilz geeigneten Tempe ratur, Der Gehalt der Gärbrühe an LLD- Einheiten wird durch die wachstumsfördernde Wirkung auf Lactobaeillus laetis Dorner be stimmt, wie bereits beschrieben.
Am Ende der Glärperiode kann die Flüs sigkeit eingedampft werden, wobei der erhal tene Rückstand ohne weitere Behandlung als Zusatz züi Futtermitteln verwendet' werden kann, um sie mit APP anzureichern. Es wird gewöhnlich vorgezogen, die Gärflüssigkeit züi filtrieren, das Filtrat mit einem Adsorptions- mittel, wie Fullererde oder Aktivkohle, zu be handeln, das Adsorbat zu trocknen und dann als Futterzusatz zu verwenden.
Falls ein Futterzusatz frei von Adsorp- tionsmitteln verlangt wird, so wird das Ad- sorbat z. B. mit geeigneten Lösungsmitteln, wie wüsserige oder verdünnte alkoholisehe Lösungen von Pyridin oder a-Picolin, elLiiert und das gesammelte EILiat eingedampft. Diese Aufbereitung bewirkt eine partielle Reini gung der LLD-wirksamen Bestandteile Lind damit ein hoehwertiges Produkt.
Wird eine weitere Reinigung gewünseht, so kann das feste Konzentrat z. B. mit einem niedrigen aliphatischen Alkohol, wie Methyl alkohol, extrahiert und der alkoholisehe Extrakt durch eine mit Aktivtonerde gefüllte Kolonne geleitet werden, wobei die LLD-akti- ven Substanzen an Tonerde adsorbiert werden. Die Kolonne wird dann mit frischem Methanol eluiert, und diejenigen Fraktionen des Eluats, Yvelche ILD-Aktivität zeigen, werden ver einigt und eingeengt.
Die eingeengte alkoho- lisehe Lösung wird dann mit einem Lösungs mittel versetzt, in welchem die aktiven Sub stanzen unlöslich sind und das mit der alko- holisehen Lösung misehbar ist, z. B. mit Aceton. Der Niederschlag wird abfilltriert, aus wüsserigem Aeeton umkristallisiert und ge trocknet. Als Produkt wird ein kristallines Vitamin B12 erhalten.
<I>Beispiel<B>1:</B></I> Ein Gärbehälter von<B>15,2</B> m3 wurde mit <B>23 kg</B> konzentriertem Fleisehextrakt (etwa <B>70</B> D/o feste Bestandteile), 143<B>kg</B> eiD es Tryptin- Abbauproduktes von Casein,<B>72,5 kg</B> Natrium- ehlorid, <B>720 g</B> Ferrosullat-heptahydrat, 144g Kobaltonit.rat-hexahydrat, 20 Liter So.ja- bohnenöl lind etwa<B>13,2</B> m3 Wasser gefüllt.
Die Mischung wurde<B>30</B> Min. bei 120u<B>C</B> steri lisiert, dann auf etwa<B>9-80 C</B> abt-ekühlt und mit 1130 Liter der Kultur eines CT 'risein erzeu- genden Stammes von Streptoin#-ees griseus (bezeichnet mit<B>25</B> CT) geimpft.
Das geimpfte Nährmedium wurde bei einer Temperatur von etwa 2811 <B>C</B> während etwa 48 Stunden der<B>Gä-</B> rung überlassen, wobei kontinuierlich mit etwa 140 m3 Luft, pro Stunde durehlüftet wurde. Am Ende der Gärperiode wurde die Brühe durch Zusatz von Phosphorsi-Lire auf den pH Wert<B>3</B> eingestellt und filtriert. Der Filter kuchen wurde mit Wasser ausgewaseben. Die Waschflüssigkeit wurde mit dem Filtrat vei - einigt und durch Zusatz von Natronlauge leicht alkaliseh gemacht.
und noehmals fil triert. Die erhaltene Gärflüssi-keit zei-te den pff #Vert <B>7,8,</B> enthielt 1,71/o feste Bestandteile und zeigte 2400 LLD-Einh./einu. <B><I>A.</I></B> Ti-ock(%ii,1)
i-äpai-(it aws Strept. griseits <I>Gärflüssigkeit.</I> <B>379</B> Liter der erhaltenen filtrierten Gär flüssigkeit wurden bei verringertem Druck und einer Temperatur unter 400 C auf un.ge- fähr <B>22,7</B> Liter eingedampft. Dieses Konzen- Z, trat, mit, einer Wirksamkeit, von etwa 40000 LLD-Einheiten/em3, wurde darell Zerstüti- bung getrocknet.
Das Troehenpräparat ent- hielt etwa 3% Feuchtigkeit und zeigte eine Wirksamkeit von etwa 140 LLD-Einll./1-no, 1. <I>B.</I> Koltleiiadsoi,bat <I>aus</I> Strept. griseas Gärflüssigkeit.
<B>13,2</B> m3 der gleichen Gärflüssigkeit, wie unter<B>A</B> verwendet, wurden mit<B>72,5 kg</B> Aktiv kohle während<B>30</B> Minuten verrührt. Das Ad- sorbat wurde durch Filtrieren abgetrennt, in, <B>378</B> Liter Wasser stispendiert und nochmals abfiltriert. Es wurde ein feuchtes Kohle- adsorbat mit einem Wassergehalt von etwa <B>50</B> 1/o erhalten.
<B>195 -,-</B> des leuchten Kohleadsorbats wurden im Vakuum bei -1511 <B>C</B> etwa 20 Stunden ge trocknet, wobei<B>99 g</B> trockenes Kohleadsorbat erhalten wurden. Dieses Produkt zeigte eine Wirksamkeit von 470 LLD-Einh./--ig, bereeh- net aus der ursprünglichen Aktivität der Gär- brühe und der restlichen Aktivität der bei der Kohleadsorption verbleibenden Flüssigkeit.
<B><I>C.</I></B><I> Aus dein</I> Ehtat <I>des</I> Kohleadsorbats geivonne2ies <I>Konzentrat.</I>
Der Rest des gewaschenen leuchten Kohle- adsorbats, entsprechend 72,4<B>kg.</B> Aktivkohle, wurde durch Verrühren mit<B>632</B> Liter 25%i.-em Pyridin eluiert. Der eluierte Filter- kuchen wurde auf der Filterpresse zuerst mit 150 Liter 25%igem Pyridin während 20 Mi- nuten ausgezogen
und dann noch mit 1.50 Liter 251/oigem Pyridin naehgewasehen.
Die Eluate und Waschflüssigkeiten wur den vereinigt und<B>2/3</B> der Gesamtmenge bei vermindertem Druck auf 24 Liter ein gedampft, wobei die Dampftemperatur unter <B>350 C</B> betrug.
Das konzentrierte Eluat zeigte eine Wirk samkeit von etwa<B>250 000</B> LLD-Einh.jem3. Eine Teilmenge von<B>150</B> cm- 3 wurde im ge frorenen Zustand getrocknet und lieferte 14,1<B>g</B> eines Produktes mit einer Aktivität von 2800 LLD-Einh./mg. <B><I>D.</I></B><I> Kristallines</I> Vt(tMn B12- Ein festes Konzentrat, hergestellt aus <B>3,2</B> ms Gärbrühe durch Behandlung mit Aktivkohle und Aufarbeitung des feuchter- Kohleadsorbats, wie beschrieben unter C,
Sojabohnenniehl 70,01/o Dextrose, techn. <B>7,9</B> Cellulose <B>5,0</B> Weizenkeimöl 4,5 Caleiumglukonat <B>2,5</B> Glyeln 2,0 DL-31ethionin <B>0,9</B> CaC03 1,5 K2HPO4 <B>1,612</B> KH2P04 1.,0 NaC1 0,838 Mg S04. <B>7</B> 1120 0,51 CaHP04. 2 1120<B>0,
375</B> Ferrieitrat 0,138 Mn SO4. 4 H20 <B>0,025</B> Ki 0,004 wurde mit Methanol extrahiert und der alkoholisehe Extrakt durch eine Kolonne mit aktiver Tonerde geleitet, zwecks Adsorption des Vitamin B12. Die Kolonne wurde mit frischem Methanol eluiert und die Frak tionen mit starker LLD-Aktivität vereinhyt und eingeengt.
Z Lider konzentrierten alkoholi- sehen Lösung -wurde Aeeton zugesetzt und das ausgefällte rohe Vitamin B12 abfiltriert. Das Produkt wurde durch Ausfällen aus einer Äthanol-Lösung durch Zusatz von Aeeton mit nachfolgendem Umkristallisieren aus wässe rigem Aeeton gereinigt. Ausbeute:<B>106</B> mg kristallines Vitamin B12.
Der Wert der oben erwähnten Konzentrate und des kristallisierten Vitamin B1,2, als Er satz von Nahrungszusätzen mit dem animali- sehen Protein-Faktor, wird durch Fütterungs versuche illustriert, bei welchen das Wachs tum von Kücken unter einer APF-Mangeldiät verglichen wird mit dem Wachstum unter derselben Diät mit Zusatz der genannten Konzentrate oder kristallinem Vitamin BI2. Kücken-Fütteritnjsversuche. Die verwendeten Kücken wurden aus Eiern ausgebrütet, die von Hennen gelegt waren,
welche auf einer APF-Mangeldiät ge halten wurden.

<I>Beispiel 2:</I> In ein Gärgefäss von<B>15,2</B> n13 wurden ge- geben: 45<B>kg</B> konzentrierter FleisehextraK 143<B>kg</B> eines Tryptin-Abbauproduktes von Casein,<B>72,5 kg</B> Natriumehlorid, 20 Liter eines Sehaumverhütungsmittels und<B>13</B> 200 Liter Wasser.
Das Nährmedium wurde sterilisiert, geimpft, vergoren, angesäuert, abfiltriert und neutralisiert, wie beschrieben in Beispiel<B>1.</B> Die neutrale filtrierte Gärflüssigkeit wurde <B>30</B> Minuten mit 43,5<B>kg</B> Aktivkohle verrührt, um die LLD-aktiven Bestandteile zu adsorbie- ren. Das Kohleadsorbat wurde von der Flüs sigkeit abgetrennt und durch Suspension in etwa<B>380</B> Liter Wasser gewaschen und wie derum filtriert.
Das gewaschene Adsorbat wurde<B>30</B> Minuten mit<B>359</B> Liter 251/oigera a-Picolin verrührt. und das Eluat durch Fil trieren gewonnen. Der FilterkLichen wurde auf der Filterpresse mit 113 Liter 9-5% a-Pi- eolin <B>30</B> Minuten aus-ewasehen. Das Elnat wurde mit der Wasehflüssigkeit
vereinigt und dann unter vermindertem Druck auf ein Volumen von<B>22,7</B> Liter eingedampft, wobei die Dampftemperatur unter<B>250 C</B> gehalten wurde. Die Lösum, wurde mit<B>227</B> Liter Me thanol versetzt und ausgefällter inerter Stoif abfiltriert. Aus dem Filtrat wurden die LLD aktiven Bestandteile durch Zusatz der Me- thanollösung zu<B>681</B> Liter Äther ausgefällt. Der Niederschlag wurde abfiltriert, mit Äther gewaschen und im Vakuum bei<B>300 C</B> ge trocknet.
Ausbeute:<B>860 g</B> eines Trockenprä parats mit einer inikrobiologischen Wirksam keit von<B>1500</B> LLD-Einh.img. Der Wert dieses Materials als APF-Quelle im Vergleich mit rohem Leberextrakt und mit kristallinem Vitamin Blf! wurde biologisch durch Fütterungsversuche an Kücken be stimmt. Kücken-Pütterungsversuche. Die Kücken wurden aus Eiern ausgebrütet, n welche von Hennen gelegt waren, die mit einer APP-Mangeldiät gefüttert worden waren.
Ein Tag alte Küeken wurden auf eine ()'rrunddiät'gesetzt, welche mit derjenigen im Beispiel<B>1</B> identisch war, mit der Ausnahme ', dass der Gehalt an Sojabohnenmehl 40,01/0 und an Dextrose 37,91/o betrug.
Nachdem<B>5</B> Tage mit obiger Grunddiät ge füttert w-Lirde, wurden hinsiehtlieh des Kör- pergewiehts ausgeglichene Gruppen von je<B>7</B> Klieken, ausgewählt.
Zwei dieser Gruppen wurden weiterhin ausschliesslich mit der ge nannten Grunddiät gefüttert, die andern Gruppen mit derselben Grunddiät, ergänzt wie folgt:

Ferner zeigen die Versuche, dass die erreichte Waelistumszunahme unce- fähr dieselbe ist, wie bei Zusatz von 1% rohem Leberextrakt zur Nahrung, das heisst einer der besten bisher bekannten Quellen des APF. <I>Beispiel</I> 3.- Ein -Nährmedium,
folgender Zusammen- Setzung - Sojabohnenmehl 3% Dextrose 211/o Lösl. Anteil von Sehlempe 0,75% Kochsalz 0,25% Kobaltnitrat <B>1</B> Teil per Million als Co-,
+ Wasser ad<B>1000/0</B> wurde sterilisiert und mit<B>10</B> Vol.1/o einer vegetativen Kultur eines Streptomyein erzeu- el genden Stammes von Strept. grisens versetzt. Die geimpfte Brühe wurde bei 2711 C gehalten und die Flüssigkeit unter Durchlüftung ge rührt.
Die Gärbrühe wurde mit Phosphor säure auf den pl, Wert<B>3</B> eingestellt und so dann durch Kieselgur filtriert. Die filtrierte Brühe zeigte eine Streptomycin-Aktivität von <B>810</B> g/em3 und eine LLD-Aktivität von<B>2700</B> LLD-Einh./em3 oder 45 LLD-Einh./mg fester Stoffe in der Brühe.
<B>227</B> Liter dieser Brühe wurden durch eine Kolonne geleitet, welche<B>600 g</B> Karbonsäure- Ionenaustauschharz (IRC <B>50</B> der Resinolls Produets and Chemical Co.) enthielt. Das Streptomvein wurde an genanntes Harz ad- sorbiert. Die abfallenden Brühen wurden ver einigt und weiter verarbeitet.
<B><I>A.</I></B> Kohleadsorbat <I>der</I> von Streptomycin <I>befreiten</I> Gärbrühe. <B>75</B> Liter der von Streptomycin befreiten Brühe wurden mit<B>1600 g</B> Aktivkohle behan delt, wobei die Kohle 751/o der ursprünglich in der Brühe vorhandenen LLD-aktiven Sub stanzen adsorbierte. Das Kohleadsorbat wurde gewaschen und getrocknet.
Das Trockenpro dukt enthielt etwa<B>80000</B> LLD-Einh./mg. Es kann in gleicher Weise verwendet werden wie das Kohleadsorbat gemäss Beispiel<B>1.</B> <I>B.</I> Fullererde-Adsorbat <I>der von</I> Streptomycz##v <I>befreiten Brühe.</I>
40 Liter von Streptomycin befreiter Gär brühe mit<B>500</B> LLD-Einh./ems wurden mit Phosphorsäure auf den pH-Wert<B>2,5</B> einge stellt. Die angesäuerte Brühe wurde mit<B>600,g</B> Fullererde behandelt.
Das Adsorbat enthielt 90% der ursprünglich in der Brühe vorhan- denen LLD-Aktivität und lieferte durch Aus waschen und Trocknen ein festes Endprodukt mit einer Aktivität von<B>30 000</B> LLD-Einh./g. Dieses Produkt kann in der gleichen Weise verwendet werden wie das Kohleadsorbat gemäss Beispiel<B>1</B> und 2.
<B><I>C.</I></B> DurchZerstäube-ngetrockneteKonzentrate. <B>80</B> Liter der gleichen von Streptoniyein befreiten Brühe, welche bei<B>A</B> und B als Aus gangsmaterial dienten, wurden auf ein Volu men von 12 Liter bei vermindertem Druck eingedampft.
Von der konzentrierten Lös-u-my wurden 5440 cm3 durch Zerstäuben -letroeknet" wobei <B>335 g</B> eines TroekenprodLiktes erhalten wur den mit einer mikrobiolocischen Aktivität von <B>C</B> 1.40 LLD-Einh./rrig. Dieses Produkt kann in der gleichen Weise verwendet werden, wie die getrocknete Strept.-griseus-Brühe gemäss Bei spiel 1.
<I>Beispiel 4:</I> Es wurde ein Nährmechuin hergestellt, welches aus 1()/o eines Tryptin Abbaupro duktes von Casein, 0,31/o konzentriertem Fleischextrakt und <B>10</B> Teilen je Million Ko- baltonitrathexahydrat bestand, lind der pH- Wert auf<B>7</B> eingestellt.
Sieben Portionen dieses Mediums von<B>je 500</B> ems wurden in 2 Liter Erlenme- yer abgefüllt und während <B>30</B> Min. bei 11511 <B>C</B> sterilisiert. Dann wurde jede Flasche mit etwa<B>10</B> ein einer vegetativen Kultur von Strept. roseoehroiiio,--en-Lis geimpit ,Lind<B>7</B> Tage auf einer rotierenden SeliütL'el- maschine entwickelt.
Der Inhalt der Flaschen wurde dann vereinigt und abfiltriert.
Ein Liter der filtrierten Brühe (etwa <B>1500</B> LLD-Einh./em3) wurde bei verminder tem Druck bei einer Temperatur unter<B>350 C</B> eingedampft und aus dem gefrorenen Zustand getrocknet. Ausbeute<B>6,9 g</B> eines Produktes mit einer Aktivität von etwa<B>130</B> LLD-Einli./mg.
Ein anderer Teil der filtrierten Brühe <B>(2300</B> ems) wurde bei einem pH <B>= 7</B> während <B>30</B> Minuten mit<B>23</B> Aktivkohle verrührt. Das erhaltene Adsorbat wurde zweimal durch Verrühren während<B>30</B> Minuten mit einer Mischung von<B>125</B> em3 Äthanol, <B>12,5</B> em3 Pyridin und<B>112,
5</B> em33 Wasser eluiert. Die vereinigten Eluate wurden unter ver- mindertern Druck bei einer Temperaiur unter 3511 <B>C</B> eingedampft und aus dem gefro renen Zustand getrocknet.
Ausbeute: 2,3<B>g</B> eines Produktes mit einer Aktivität von etwa 440 LLD-Einh./mg. Der getrocknete (jä.rrüel,:staiid und das aus dem Kohleneluat gewonnene Troekenpräparat können in derselben Weise verwendet werden, wie die Produkte gemäss Beispiel<B>1.</B> <I>Beispiel<B>5:</B></I> Es wurde eine Strept.-albidoilavis-Ku.It-uir hergestellt und analog wie in Beispiel 4 für Strept. roseoehromogenus beschrieben ent wickelt.
Die Gärbrühe wurde mit Phosphor säure auf PH<B>3</B> eingestellt und der filtrierten <B>Z</B> angesäuerten Brühe _Natronlauge bis zur Neu- trallsation zugesetzt.
<B>1570</B> CM3 neutraler filtrierter Brühe mit einer Aktivität von<B>77</B> LLD-Einli./iiig der festen Bestandteile der Briiiie wurden<B>30</B> Min. mit<B>15,7 g</B> Aktivkohle verrührt. Das Adsorbat wurde abfiltriert und im Vakuum bei 450<B>C</B> während 20 Stunden getrocknet. Ausbeute: <B>50,7 g</B> (einschliesslich Filtermaterial) ge trocknetes Adsorbat mit einer Aktivität von etwa<B>30</B> LI-iD-Eiiih./nig, berechnet aus der Wirksamkeit der ursprünglichen neutralen gefilterten Brühe und derjenigen der von der Kohle ablaufenden Brühe.
Dieses Kohleadsorbat kann in der gleichen Weise verwendet werden wie die Produkte gemäss Beispiel<B>1.</B>
<I>Beispiel<B>6:</B></I> Das Nährmedium hatte die folgende Zu sammensetzung: Pfeiffers Trocken hefe<B>2 0/0</B> Kobaltnitrat <B>1-0</B> TeileproMillionalsCo- Dest. Wasser ad.<B>100</B> 1/o 40 em3 dieses A.,
fediums wurden in einer '#50-eM3-Plasehe sterilisiert und mit<B>5</B> Vol.II/o einer vegetativeii 48-Stunden-Kultur eines Grisein produzierenden Stammes von Strept. griseus (bezeichnet mit<B>25</B> CT) geimpft. Die geimpfte Brühe wurde<B>72</B> Stunden bei<B>280 C</B> entwickelt, wobei die Flasche ständig auf einer rotierenden Schüttelniasehine gesehüt- telt wurde.
Nach Vollendung der Gärperiode zeigte die Brühe 4800 LLD-Einh./cm3 oder etwa 220 LLD-Einh./mg fester Bestandteile der Brühe. Diese Brühe kann als Ausgangs material dienen zur Herstellung von festen Konzentraten mit APF--'#%Tirksainlzeit oder von reinem Vitamin B12, wie in Beispiel<B>1</B> be schrieben. <I>Beispiel<B>7:</B></I> Es wurden<B>8</B> Nährmedien hergestellt, deren Zusammensetzung in bezug auf anorganische Salze identisch war, die jedoch verschieden artige stiekstoffhaltige Zusätze aufwiesen.

Die geimpften Brühen -wurden 4 Tage bei 280<B>C</B> entwickelt, wobei die Flaschen ständig geschüttelt wurden.
Nach Beendigung der Gärperiode wurden die vergorenen Brühen geprüft. Die vom stickstoffhaltigen Zusatz zum Medium abhän gigen Resultate sind nachfolgend zusammen gestellt:

Wasser ad.<B>1000/0</B> Das Medium wurde in Portionen von je <B>100</B> em3 in fünf 125-eM3-Erlenmeyer-Kolben gegeben und sodann sterilisiert. Jede Flasche wurde mit einer verschiedenen Spezies von Clostridium geimpft, wie angegeben in der folgenden Tabelle.
Die geimpften Brühen wurden, unter Durchlüftung während einer Gärperiode von mehreren Tagen bei einer Temperatur von <B>350 C</B> entwickelt, und zwar ohne Umrühren der Brühen.

A concentrate of 2000 LLD units / mg provides a content of <B> 330 000 </B> LLD units <B> per kg </B> feed with an addition of <B> 165 </B> mg des Concentrate of <B> 1 kg </B> feed <B> (0.0165 </B> 1 / o).
It is known that various water-soluble fermentation products can be used as food additives, mainly because of their riboflavin and vitamins of known structure, such as biotin, pantothenic acid, etc. Such additives contain only small amounts of LLD-active substances.
The soluble components of the blityl fermentation, which are obtained by evaporation of the liquid obtained after the distillation of the B-lityl alcohol, have only about 4 LLD units / m. And the soluble components of the grain vinasse less than <B> 1 </B> l,
LD unit / mg. With such a content, it is practically impossible to think of obtaining concentrates which are intended to be used for the enrichment of feed which is insufficient in APF.
On the other hand, it is different if the starting material has at least 20 LLD units / m of solids contained therein. For the production of such fermentation broths suitable microorganisms are the FLiiigi, which are given on page 2 in the book by Smith and Raistrick An introduetion to industrial niveology (London, Edward Arnold & Co., 1938) is called Myxomyeeten,
Schizomyeetes and Eumyeetes. The Sehizomyeeten are particularly suitable, in particular certain strains of the genus Streptomyees griseus, which can also be used for the production of the antibiotics Streptomyein and Grisein.
Other genera too, such as Strept. albidolla-, vms, strept. eolombiensis nov. sp., Strept. roseoehromogenus and Strept. antibiotieus, have strains which deliver high yields of LLD-effective substances.
Other schizomata vets that can be used are members of the Clostridium tribe. Tests have shown that selected strains of Clostridium tetanomorplium, Clostridium eo- ehlearium, Clostridium flabel.Iiierum and Clostridium butyrieum are also well suited for carrying out the method according to the invention.
Other fungi which have proven to be useful for the technical production of fermentation broths with at least 20 LLD units / mg of the pesticides contained therein are Torula, Eremotheeium ashybii and Escheriehia coli.
The method according to the invention can in part be associated with the production of the antibiotics a streptomyein and grisein. In the streptomycin and gris fermentation processes, maize mares are fermented at a low concentration, with large amounts of fermentation broth being produced for a relatively small amount of the antibiotic. In a large streptomyel plant, many thousands of cubic meters of fermentation liquid can be produced every day after the antibiotic has been separated.
It was not previously known that the residues mentioned still contained a useful product. In fact, the content of other vitamins than vitamin B1, 2 is too low to make the production of those other vitamins economical. In the industrial production of streptomycin, the accumulation of unusable material in large quantities has so far been a problem.
In the meantime it has been shown that the fermentation liquid, after the antibiotic streptomyein has been separated off by means of ion exchange resins, has significantly more than 20 LLD units per milligram of solids contained therein, and from the above mentioned Rüel # .stän - the concentrates of high LLD effectiveness are obtained according to the invention.
Any aqueous nutrient medium which is usually used for cultivating fungi can be used as starting material for carrying out the method according to the invention. The usual nutrient fluids contain a carbon source, a nitrogen source, inorganic salts and possibly growth factors. The carbon can be supplied by a carbohydrate, e.g. B. of dextrose, maltose, xylose, invert sugar, cereal extracts and the like.
The nitrogen can come from an ammonium salt, from amino acids or from proteins, e.g. B. soybeans, oats, yeast extracts, products of the breakdown of casein with tryptin, meat extract, blood meal, protein-free meat and bone material, the finely ground, dried residue of animal tissues that are free from hair, claws, horn, blood, intestinal and stomach contents and more than 4,
Contains 4% phosphorus, as well as fish meal and fish extracts, soluble components of the vinasse and the like. If desired, the fungi can be activated using proteins or amino acids, that is to say without carbohydrates in the nutrient medium, in which case the proteins or amino acids serve as both a carbon and nitrogen source.
The nutrient medium is appropriately sterilized and then inoculated with a strain of the selected fungus. The mixture is subjected to incubation until the fermentation liquid has a content of at least 20 LLD per mg of solids in the broth.
The inoculation is usually carried out under the surface of the liquid and at the temperature suitable for the fungus used. The LLD unit content of the fermentation broth is determined by the growth-promoting effect on Lactobaeillus laetis Dorner, as already described.
At the end of the fermentation period, the liquid can be evaporated, and the residue obtained can be used as an additive for animal feed without further treatment in order to enrich it with APP. It is usually preferred to filter the fermentation liquor, treat the filtrate with an adsorbent such as fuller's earth or activated charcoal, dry the adsorbate and then use it as a feed additive.
If a feed additive free of adsorbents is required, the adsorbate is z. B. with suitable solvents, such as aqueous or dilute alcoholic solutions of pyridine or a-picoline, elLiiert and evaporated the collected EILiat. This treatment causes a partial cleaning of the LLD-active components and thus a high-quality product.
If further cleaning is desired, the solid concentrate can be added e.g. B. with a lower aliphatic alcohol, such as methyl alcohol, extracted and the alcoholic extract passed through a column filled with active alumina, the LLD-active substances being adsorbed on alumina.
