Verfahren zur hydrierenden Aufspaltung von vieinalen Epoxyden. Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von Oxy- verbindungen durch hydrierende Aufspaltung von vicinalen Epoxyden, das heisst Epoxyden mit der Gruppierung

Die reduktive Aufspaltung von vicinalen Epoxyden zur Herstellung von Oxyverbin- dungen, insbesondere zur Einführung einzel ner Oxy-Gruppen in das Gerüst der Steroide,
ist an sich bekannt. Nach den bekannten Arbeitsweisen verläuft die Reaktion oft mit niedriger Ausbeute und es entstehen schwer zu trennende Gemische.
Es wurde nun gefunden, dass die reduktive Aufspaltung vicinaler Epoxyde milder und einheitlicher verläuft, wenn man als Reduk tionsmittel reaktionsfähige Metall-Hydride, insbesondere Dimetall-Hydride, wie Alka.li- metall-Aluminium-Hydride, in erster Linie Lithium-Aluminium-Hydrid, ferner aber auch Natrium- oder Lithium-Bor-Hydrid, ver wendet.
Die als Ausgangsstoffe dienenden vicina- len Epoxyde können aliphatischer, alicyeli- seher, aromatisch-aliphatischer und hetero eyclischerNatur sein und können insbesondere der Steroidreihe angehören. Die Reduktion wird vorzugsweise in An wesenheit eines Lösungsmittels, worin das 1Zeta11-Hydrid löslich ist, durchgeführt.
So wird zum Beispiel die Reduktion mit einem illkali-Aluminium-Hydrid, wie Lithium- Aluminium-Hydrid, in Gegenwart. eines in- differenten,wasserfreien orga.nischenLösungs- mittels, in dem das Metall-Hydrid löslich ist, insbesondere in Gegenwart von Äther, Tetra- hydrofuran, Butyläther oder dergleichen, durchgeführt.
Wenn zum Beispiel mit Na- trium-Bor-Hydrid reduziert wird, so arbeitet man vorzugsweise in Gegenwart von Wasser. <I>Beispiel 1.</I>

7 Gew.-Teile 3ss'-Acetoxy-16,17a-oxido-20- keto-5-allo-pr egnan vom Smp. <B>1810</B> (dar gestellt zum Beispiel durch Umsetzen von Z1 16-3ss-Acetoxy-20-keto-5-allo-pregnen mit Fenzopersäure oder Wasserstoffperoxyd in Eisessig)
werden in 300 Vol.-Teilen abs. Äther gelöst und tropfenweise unter gutem Um rühren mi einer Lösung von 4 Gew.-Teilen Lithiiun-Aluminium-Hydrid in 600 Vol.- Teilen trockenem Äther gegeben. Nach Ab flauen der ziemlich heftigen Reaktion wird die Lösung noch während einer Stunde zum Sieden erhitzt, dann vorsichtig mit Wasser und anschliessend mit verdünnter Schwefel säure versetzt. Die ätherische Schicht wird mit Wasser neutral gewaschen, getrocknet und eingedampft.
Der krystallisierte Rück stand lässt sich durch Umlösen aus Methanol Wasser reinigen und schmilzt dann bei 223 bis 224 . Es liegt das 3ss,17a,20ss-Trioxy-5-allo- pregnan (Substanz J von Reiehstein 1938) vor.
Aus dem Gemisch der Mutterlaugen lassen sich durch Acetylieren und chromato- graphische Reinigung folgende 17a-Oxy- Steroide isolieren: 3ss,20ss-Diacetoxy-17a-oxy- 5-allo-pregnan vom Smp. 161 (Substanz J- Diacetat von Reiehstein) und 'ass,20a-Diacet- oxy-17a-oxy-5-allo-pre-rrarr vom Smp. 2430 (Substanz 0-Diaeetat\ von Reichstein 1938).
In gleicher Weise lässt sich das in Stellung 3 unveresterte 3ss-Oxy-16,17a-oxido-20-keto-5- allo pregnan vom Smp. 181-182 mit Litlrillm-Alllminlllm-Hydrld in Äther um setzen. Die Reaktionsprodukte sind bei glei- eher Aufarbeitung wie oben wieder die Sub stanzen J und 0 von Reiehstein.

Teilen absolutem Äther gelöst und tropfenweise unter gutem Umrühren zu einer Lösung von 5 Gew.-Teilen Lithium-Alumi- nium-Hydrid in 500 Vol.-Teilen trockenem Äther gegeben. Das Reaktionsgemisch wird 15 Minuten gut durchgerührt, dann tropfenweise mit 200 Vol.-Teilen Wasser und anschliessend mit 400 Vol-Teilen l0proz. Schwefelsäure versetzt. -lach Zugabe von mehr Äther wird die ätherische Schicht. abgetrennt, mit Wasser neutral gewaschen, getrocknet und einge dampft.
Der krystallisierte Rückstand ist nach zweimaligem Umlösen aus Äthanol rein und schmilzt bei 148-14911. Es liegt das 3ss,5- Dioxy-koprostan vor. Das 3-1VIonoacetyl- Derivat dieser Substanz schmilzt bei 80-810.

5 Gew.-Teile 3-Keto-4ss,5-oxido-koprostan vom Smp. 116-117 (hergestellt. durch Oxy dation von Cholestenon mit alkalischem Wasserstoffperoxyd) werden in 200 Vol: Teilen abs. Äther gelöst und tropfenweise unter gutem Umrühren einer Lösung von 5 Gew: Teilen Lithium-Aluminiiun-Hy drid in 500 Vol.-Teilen trockenem Äther zugefügt.
Die Aufarbeitung des Reaktionsgemisches er folgt wie in den vorhergehenden Beispielen .Das Rohprodukt wird zur Reinigung einer chromatographisehen Analyse unterworfen. Auf diese Weise gelingt es, 3,5-Gew.-Teile 3a,5-Dioxy-koprostan vom Smp. <B>192-1930</B> (vgl. Beispiel 4) und 1,3 Gew-Teile 3ss,5- Dioxy-koprostan vom Snip. 148-14911 (vgl. Beispiel 3) zu isolieren.
