Im Hauptpatent wurde ein Verfahren be schrieben zur Herstellung eines ungesättigten polycyclischen Betons, dadurch gekennzeich net, dass man Androsten-5-diol-3 . 17-mono- acetat-17 in Gegenwart eines Leichtmetall- alkoholates oder eines Leichtmetallsalzalko- holates mit einem grossen -Überschuss einer Carbonylverbindung behandelt.
Man erhält dabei Testosteronacetat in sehr guter Aus beute.
Es wurde gefunden, dass man Testosteron erhalten kann, wenn man einen 17-Monoester des Androsten-5-diols-3.17 in Gegenwart eines Leichtmetallalkoholates oder eines Leichtmetallsalzalkoholates mit einem be trächtlichen Überschuss einer Carbonylverbin- dung behandelt und den so erhaltenen Testo- steronester, gegebenenfalls nach vorheriger Reinigung, verseift zu dem freien Testo- steron.
Das Testosteron ist eine bekannte Ver bindung; es soll als Heilmittel oder als Zwi- schenprodukt für die Herstellung von Heil- mitteln Verwendung finden.
<I>Beispiele:</I> 1. 0,600 g Androsten-5-diol-3. 17-acetat- 17 wird in 45 cm' Benzen und 8 cm' Aceton mit 2 g tertiärem Aluminiumbutylat 5 Stun den unter Rückfluss erwärmt. Darauf wird mit verdünnter Schwefelsäure ausgeschüttelt. Die Benzenschicht wird mit Natriumsulfat getrocknet und eingedampft. Der Rückstand wird umkristallisiert, z.
B. aus Petroläther, zwecks Reinigung des Testosteronacetats. Letzteres wird darauf in bekannter Weise verseift, z. B. mit Alkali, wobei reines Testo- steron vom Smp. 153 bis 155 erhalten wird. Gesamtausbeute etwa 470 mg =<B>78%</B> der theoretischen Ausbeute.
2. 0,5 g Androsten-5-diol-3.17-mono- benzoat-17 wird in 40 cm' Toluen und 7 em3 Cycloheganon mit 1 g tertiärem Äluminium- butylat 3 Stunden unter Rückfluss erhitzt. Darauf wird mit verdünnter Schwefelsäure ausgeschüttelt. Die Toluenschicht wird mit Wasser gewaschen, mit Natriumsulfat ge trocknet und eingedampft. Der Rückstand wird umkristallisiert, z.

B. Amylenhydrat), wie sekund"ire (z. B. Isopropylalkohol) kommen in Betracht. Als Carbonylverbindung können auch andere Ketone, sowie Aldehyde verwendet werden.
