-amino-4'-oxy- arsenobenzol- 3'-methylaminoformaldehydna- triumsulfoxylat.
Process for the preparation of 3-oxy-4- (bis-dioxypropyl) -amino-4'-oxyarsenobenzene-3'-methylaminoformaldehyde sodium sulfoxylate. In the patent of the Swiss patent no. <B> 183799 </B> a method for the presentation of aminoarsenobenzene monosulfoxylates which contain two oxyalkyl radicals on the same amino group is given.
It has now been found that therapeutically valuable compounds of the composition can also be used
EMI0001.0007
where R and R1 for nuclei of the benzene series, R_ for an alkyl radical which can contain at least one hydroxyl group, X and Y for alkyl radicals which contain at least one hydroxyl group, and Z for an alkali metal can be obtained, if you can get aminobenzolar acids ,
which may contain further substituents in the benzene nucleus, jointly converted into arsenobenzenes in a known manner and at any point in time during the process, simultaneously or successively, two oxyalkyl radicals in the amino group of one benzolaric acid and one oxalkyl or alkyl radical,
just as it introduces the formaldehyde sulfoxylate residue into the amino group of the other benzolar acid. The individual oxyalkyl radicals can be identical or different. The alkali metal can be sodium or another alkali metal such as potassium.
To introduce the oxyalkyl radicals, alkylene oxides, such as ethylene oxide, propylene oxide, isobutylene oxide and glycide, can be used, so that the radicals, for example, are attached to the nitrogen

to step. There can also be two different oxyalkyl radicals on the nitrogen.
The same radicals mentioned above can also be attached to the amino group which represents the -CHZ. SOlZ = rest carries, step. Furthermore, methyl, ethyl or propyl, for example, can be attached to this amino group.
The present patent relates to a process for the preparation of 3-oxy-4 (bis-dioxypropyl) -amino-4'-oxy-arsenobenzene-3 '-methylaminoformaldehyde sodium sulfoxylate, which is characterized in that 1 mol of 3 - Oxy - 4 - aminobenzene -1- arsic acid, 1 mole of 3-methylamino-4-oxybenzene-l-arsic acid,
2 moles of glycide, a reducing agent and sodium formaldehyde sulfoxylate act on each other.
If you first react 1 mole of the 3 - oxy - 4 - aminobenzene-1-amino acid with 2 moles of glyoid, then 1 mole of the 3 - oxy - 4- (bis-dioxypropyl) obtained
- aminobenzene-1-arsinic acid in the presence of 1 mole of 3-methylamino-4-oxybenzene-l-arsinic acid with a reducing agent and brings about the effect of the arsenobenzene formaldehyde sulphonylate, so the 3 - oxy - 4 - (bis - dioxypropyl)
-amino-4'-oxy-arsenobenzene-3'-methylaminoformaldehyde sodium sulfoxylate.
The same compound is obtained when 3-oxy-4-aminobenzene-1-arsinic acid and 3-methylamino-4-oxybenzene-1-arsinic acid are reduced together to form the corresponding arsenobenzene,
this is reacted with sodium formaldehyde sulfoxylate to form the corresponding monosulfoxylate and 2 mol of glycide are allowed to act on the aqueous solution of this monosulfoxylate under heat.
The compound, isolated in a suitable manner, forms a yellow powder which is easily soluble in water and insoluble in ether and ethyl alcohol. It is said to be used as a remedy.
<I> Example: </I> 13 g of 3 - oxy - 4 - (bis-dioxypropyl) - aminobenzene-1-arsic acid, which is obtained by reacting 3 - oxy - 4 - aminobenzene-1-arsic acid with 2 mol of glycide is prepared, with 8.44 g of 3 - methylamino - 4 - oxybenzene-1-arsinic acid, presented according to "Reports of the German Chemical Society", Volume 45 (1912), page 2131, and 10,
25 g of potassium iodide, mixed well, dissolved in 250 cm3 of 10% hydrochloric acid, sucked off with the addition of animal charcoal and 23 cm3 of 50% hypophosphorous acid added. The temperature rises to around <B> 30 '</B> C while the liquid is discolored.
When the reaction is complete, the yellow solution is stirred up with 250 cm 'of ice-cold concentrated hydrochloric acid, the inorganic salts which have separated out are filtered off with suction and the fil is precipitated in ethyl alcohol. Here, hydrochloric acid 3 - Ogy - 4 - (bis - dioxypropyl) - amino-3'-methyl-amino-4'-oxyarsenobenzene separates out as a yellow precipitate, which is filtered off with suction and washed with ether. The compound dissolves easily in water and contains 18.77% As.
If 13.5 g of the hydrochloric acid salt are dissolved in aqueous methyl alcohol and mixed with a solution of 20 g of sodium formaldehyde sulfoxylate in 40 cubic meters of water at about 27 ° C., a precipitate separates out, most of which dissolves when sodium carbonate is added . The residue is filtered off and the clear, yellow filtrate is precipitated in ethyl alcohol.
A yellow precipitate of 3-oxy-4 - (bis - dioxypropyl) - amino-4'-oxyarsenobenzene - 3 '-methylaminoformaldehyde sodium sulfoxylate is formed
EMI0002.0137
which is sucked off and washed with ether. The preparation is soluble in water and contains 19.3% As.
The same compound is obtained when a mixture of 23.3 g of 3-oxy-4-aminobenzene-1-arsinic acid and 24.7 g of 3-methyl-amino-4-oxybenzene-1-arsinic acid in 600 cm-10 Dissolve% i, ger hydrochloric acid, decolorize with animal charcoal and add <B> 8 </B> 0 cm '50% hypophosphorous acid,
and a solution of 27g of potassium iodide in 27cm 'of water at about. 45 C reduced to 3-oxy-4-amino-3'-methyl-amino-4'-oxyarsenobenzene hydrochloride and then converted into the corresponding monosulfogylate in a known manner by means of sodium formaldehydsulfoxylic acid.
If two moles of glycide are allowed to act on the aqueous solution of the sulfoxylate in the heat, a yellow precipitate of 3-oxy-4- (bi.s-dioxy-propyl) separates after cooling after the addition of ethyl alcohol and ether. -amino-4'-oxy-arsenabenzol-3'-methyl aminoformaldehyde sodium sulfoxylate, which shows the same properties and the same pharmacological effectiveness as the body obtained in the first way.
The two connections are therefore identical.
The 3-oxy-4-aminobenzene-l-arsinic acid used as the starting material can, according to the information provided by the German Patent Register No.
244166. It condenses with excess glycol in the heat to form 3 - oxy-4- (bis -dioxypropyl) -aminobenzene-l-arsic acid, a colorless powder that dissolves easily in water and methyl alcohol and contains 3.41% N.
