Verfahren zur Darstellung -von 2-Keto-l-gulonsäure. Nach :dem Verfahren des Patentes Nr.171716 wird 2-Keto-l-o,ulonsäure dadurch erhalten, da.ss man auf 1-Sorbose eine Carbo- nylverbindung z.
B. Formaldehyd, Aceton, Methylä.thylketon oder Benzaldehyd, einwir ken, lässt, das so erhaltene Bismethylenäther- derivat in alkalischer Lösung oxydiert und die aus der so erhaltenen Lösung durch Zu satz einer starken Säure gewonnene Bisme- thylenäther-2-keto-1-gulorisäure zur Abspal tung der Carbonylverbindung mit Wasser bei saurer Reaktion erwärmt.
Es wurde nun gefunden. dass sich auch die Verbindungen von 1-Sorbose mit eycli- sehen Ketonen leicht in 2-geto-l-,,-ulonsäure iil-,erführen lassen. Die auf diese Weise ge wonnenen Zwischenprodukte kristallisieren :sehr gut, so da-ss sie leicht gereinigt werden können.
<I>Beispiel:</I> Teile gepulverte 1-Sorbose, 40 Teile Cyclohexanon und 2 Teile konzentrierte Schwefelsäure werden in einer Glasstöpsel- (lasche 20; Stunden auf :der Maschine ge schüttelt.
Die braune Lösung wird mit Äther verdünnt und mit Pottasehelösung ausge- achüttelt. Nach dem Trocknen wird die Lö sung durch Destillation von Ather und Cyclo- hexanon befreit und der Rückstand im Hoch vakuum destilliert. Man erhält ein fast farb loses, dickes<B>01</B> vom Siedepunkt 195-199 bei<B>0,2</B> mm, das glasig erstarrt und beim An reiben mit heissem Benzin fast augenblicklich partiell kristallisiert.
Die abgesaugten und mit Benzin nachgewaschenen Kristalle erwei sen sich als reine Monocycloheganon-Verbin- dung. Schmelzpunkt 161-162 . Die Di- cyclohexarionsorbose wird aus :der Benzinlö sung gewonnen.
Sie kristallisiert in feinen Nädelchen, welche bei 124 kor. schmelzen und in den meisten organischen Lösungs- mitteln ausser Benzin und Petroläther in der gälte leicht löslich sind. In der Wärme lö sen auch Benzin und Petroläther gut. In Wasser ist die Verbindung fast unlöslich. [a] D = - 23 (c = 2,07 in absolutem Methanol).
5 'feile Dicyclohexanon-l-sorbose werden mit einer Lösung von 0,51 Teilen Kalium hydroxyd in 15 Teilen Wasser aufge schwemmt, hierauf 30 Teile permauganat- festes Pyridin und eine Lösung von 0,95 Teilen Kaliumpermanganat in 25 Teilen Wasser zugegeben. Nun wird unter Erwär men auf<B>60-'</B> geschüttelt, bis die Farbe des.
Permanganates vollständig verschwunden ist, dann vom Braunstein abgesaugt und mit Wasser gewaschen. Das Filtrat wird mit Kohlensäure und Schwefelsäure versetzt, bis Phenolphtha.lein nicht mehr gerötet, Lack mus aber noch gebläut wird, und im Vakuum zur Trockne gebracht.
Der feste Rückstand wird mit Äther von unveränderten Resten der Dicyeloheganonsorbose befreit und hier auf durch mehrmaliges Auskochen mit ab solutem Alkohol das Kaliumsalz der Säure ausgezogen. Es kristallisiert beim Abkühlen .der filtrierten, alkoholieehen Lösung in Na deln, welche leicht löslich in Wasser, ziem lich schwer löslich in absolutem Alkohol und unlöslich in Äther und Benzol sind:
Durch Umkristallisieren aus absolutem Alkohol kann das Kaliumealz analysenrein erhalten werden. [a] = -15,6 (c = 0,8 in Wa.s- ser).
1,3 Teile des Kaliumsalzes werden in 4 Teilen Wasser gelöst, mit 2. Teilen Eis ver setzt und hierauf eine Lösung von 0,6 Teilen konzentrierter Salzsäure in 5 Teilen Wasser und 1 Teil Eis zugegeben. Die sofort aus fallende Säure wird abgesaugt, mit Eiswasser gewaschen und im ilochvall-uum getrocknet. Naeh dem Umkristallisieren aus Benzol- Benzin erhält man farblose Blättchen vom Schmelzpunkt 1d2-133 .
Die Dicyclo- heganon-?-keto-l-gulonsäure bildet farblose Blättchen vom Schmelzpunkt 132-133 , sie ist leicht löslieh in Äther, Benzol und ähn lichen Lösungsmitteln und schwer löslich in Benzin, Petroläther und dergleichen. In Wasser löst sie sich ziemlich schwer. [a] v = - 33,4 (c = 0,92 in Chloroform).
5 Teile Dicyclohexano.n-2-keto-l-gulon- säure werden mit 100 Teilen Wasser gekocht. Die Kristalle werden zunächst langsam klebrig, dann tritt allmählich Lösung ein. Hierauf wird im Vakuum zum Sirup ver dampft. Beim Impfen mit 2-Keto-1 b,-ulon- säure tritt sofort Xristallisatiou ein. Das erhaltene Produkt erweist sieh als identisch mit der 2-lieto-l-gulonsäure, welche nach (lern Verfahren des Patentes Nr. 171716 ge wonnen wird.
Process for the preparation of 2-keto-l-gulonic acid. According to: the process of patent no. 171716 2-keto-l-o, ulonic acid is obtained by adding a carbonyl compound to 1-sorbose, e.g.
B. formaldehyde, acetone, methyl ethyl ketone or benzaldehyde, einwir ken, the bismethylene ether derivative obtained in this way is oxidized in an alkaline solution and the bismethylene ether-2-keto-1 obtained from the solution obtained by adding a strong acid -Guloric acid heated with water to split off the carbonyl compound in an acidic reaction.
It has now been found. that the compounds of 1-sorbose with eycli ketones can easily be found in 2-geto-l - ,, - ulonic acid iil-. The intermediate products obtained in this way crystallize: very well, so that they can be easily cleaned.
<I> Example: </I> parts of powdered 1-sorbose, 40 parts of cyclohexanone and 2 parts of concentrated sulfuric acid are shaken in a glass stopper (20; hours on: the machine).
The brown solution is diluted with ether and shaken out with potase solution. After drying, the solution is freed from ether and cyclohexanone by distillation and the residue is distilled in a high vacuum. An almost colorless, thick <B> 01 </B> with a boiling point of 195-199 at <B> 0.2 </B> mm is obtained, which solidifies glassy and almost instantly partially crystallizes when rubbed with hot gasoline.
The crystals sucked off and washed with gasoline turn out to be pure monocycloheganone compounds. M.p. 161-162. The dicyclohexarionsorbose is obtained from: the gasoline solution.
It crystallizes in fine needles, which at 124 cor. melt and are easily soluble in most organic solvents except gasoline and petroleum ether in the cold. Gasoline and petroleum ether also dissolve well in the heat. The compound is almost insoluble in water. [a] D = - 23 (c = 2.07 in absolute methanol).
5 'files dicyclohexanone-l-sorbose are floated up with a solution of 0.51 part of potassium hydroxide in 15 parts of water, then 30 parts of permauganate-solid pyridine and a solution of 0.95 part of potassium permanganate in 25 parts of water are added. Now shake while heating to <B> 60- '</B> until the color of the.
Permanganates has completely disappeared, then sucked off the brownstone and washed with water. Carbon dioxide and sulfuric acid are added to the filtrate until phenolphthalene is no longer reddened, but varnish must still be blued, and then brought to dryness in a vacuum.
The solid residue is freed from unchanged residues of the dicyeloheganonsorbose using ether and the potassium salt of the acid is extracted here by boiling it out several times with absolute alcohol. When the filtered, alcoholic solution cools, it crystallizes in needles, which are easily soluble in water, rather sparingly soluble in absolute alcohol and insoluble in ether and benzene:
The potassium salt can be obtained in analytically pure form by recrystallization from absolute alcohol. [a] = -15.6 (c = 0.8 in water).
1.3 parts of the potassium salt are dissolved in 4 parts of water, 2 parts of ice are added and a solution of 0.6 part of concentrated hydrochloric acid in 5 parts of water and 1 part of ice is then added. The acid which immediately precipitates out is filtered off with suction, washed with ice water and dried in an interval of time. After recrystallization from benzene-gasoline, colorless flakes with a melting point of 1d2-133 are obtained.
Dicyclo- heganon -? - keto-l-gulonic acid forms colorless leaflets with a melting point of 132-133, it is easily soluble in ether, benzene and similar solvents and sparingly soluble in gasoline, petroleum ether and the like. It is very difficult to dissolve in water. [a] v = -33.4 (c = 0.92 in chloroform).
5 parts of dicyclohexano.n-2-keto-l-gulonic acid are boiled with 100 parts of water. The crystals slowly become sticky at first, then gradually dissolve. The syrup is then evaporated in a vacuum. When vaccinated with 2-keto-1b, -ulonic acid, crystallization occurs immediately. The product obtained proves to be identical to 2-lieto-l-gulonic acid, which is obtained according to the method of patent no. 171716.
