Description

  

  Verfahren zur     Gewinnung    und     Reindarstellung    von     spezifischem        Tuberkulose-          Euglobulin    aus Blutserum, sowie aus     Egsudaten    und     Tr        anssudaten    erkrankter  Individuen.

      Im Hauptpatent ist ein Verfahren     zut     Gewinnung     und.        Reindarstellung    von spezifi  schem     Tuberkulose-Pseudoglobulin    aus Blut  serum, sowie aus     Exsudafien    und Trans  sudaten erkrankter Individuen beschrieben  worden, bei welchem durch     elektroosmotisehe     Behandlung der genannten Ausgangsstoffe  zuerst das     Euglobulin    entfernt     wird.     Es ist dort ausführlich dargelegt, dass  das     Euglobulin    diejenige Serumeiweiss  fraktion ist, welche alle in dem Blut-.

    Serum     bezw.        Exsudaten    und     Transsudaten     ursprünglich vorhandenen Antigene ent  hält. Diese Antigene     sind    teilweise die  Krankheitserreger selbst     bezw.    ihre Abbau  produkte, teilweise die von diesen Krank  heitserregern im Organismus produzierten  Gifte,     sogenaunte        Toxine.    Es ist bekannt,  dass man mit diesen Stoffen entweder die  Infektion auf andere     Individuen    übertragen  oder mindestens Krankheitserscheinungen  derselben     Art    bei gesunden     Individuen    aus  lösen kann.

   Dies hängt davon ab, ob es sich    um die wirklichen noch lebenden Krankheits  erreger (Bakterien oder andere Virusarten)  oder aber um die     sogenannten    Toxine han  delt, welche im Blut, wie auch in den ge  samten Ergüssen der erkrankten Individuen  zu kreisen pflegen. Es ist auch bekannt, dass  die beschriebenen Stoffe sich in den patho  logischen. Ergüssen meistens in     viel    grösserer  Menge     bezw.    stärkerer Konzentration vor  finden als im Blutserum derselben Patien  ten.

   Es ist bekannt. dass diese Stoffe, be  sonders. bei chronisch verlaufenden Krank  heiten, bei wiederholter Einverleibung, nicht       nur    die Widerstandsfähigkeit des Erkrank  ten bedeutend zu erhöhen imstande sind,  sondern, wie die aktive Immunisierung. die  Herstellung grösserer Mengen von Anti  körpern gestattet, welche den     Krankheits-          prozess    günstig beeinflussen     bezw.    zur Aus  heilung bringen. Fernerhin ist es bekannt,  dass die wiederholte Einverleibung derarti  ger Antigene gesunde, der Infektion aus  gesetzte Personen gegenüber einer nachträg     #,         liehen Infektion in hohem Masse zu schützen       vermögen.     



  Die vorliegende Erfindung lehnt sich an  das im Hauptpatent beschriebene Verfahren  an und bezweckt die Gewinnung und     R,ein-          darstellung    von spezifischem     Tuberkulose-          Euglobulin    aus Blutserum, sowie aus     Eg-          sudaten    und     Transsudaten    erkrankter Indi  viduen.

   Sie kennzeichnet sich dadurch, dass  durch     ülektroosmotische    Behandlung der ge  nannten Ausgangsmaterialien bei dem     iso-          elektrischen        Punkt        Englobulin    mit den  darin enthaltenen     Antigenen    zur     Ausflockung     gebracht., dasselbe dann von der Flüssigkeit  getrennt und unter Benutzung entsprechen  der     Alkalimengen    in Lösung gebracht wird.

    <I>Beispiel</I>  In der Bauchhöhle von tuberkulösen     Pa-          tien        ten    befinden sieh häufig sehr grosse  Mengen sogenannter     Ascitesflüssigkeit.    Die  selbe wird gesammelt. und     zur    Entfernung  von festen Bestandteilen durch Papier     fil-          triert.    3 Liter dieser Flüssigkeit werden in  den     Mittelraum    eines     Dreizellenapparates     gebracht und der Einwirkung eines     elek-          trisehen    Stromes unterworfen.

   Die Span  nung     beträgt    anfänglich nur 20 bis 30 Volt,  während die     Stromstärke    zirka. 10 bis 12  Ampere beträgt. Innerhalb von etwa. 2 Stun  den sinkt die Stromstärke auf 0.05 bis  0,1 Ampere, während die Spannung auf 200  bis 220     Volt    ansteigt.

   In der Flüssigkeit  scheidet sieh nunmehr. bei Eintritt des     iso-          elekt-rischen        Punktes,    hier bei einer     Wasser-          atoffionenkonzentration    von etwa     6.-i-        Pi-I.       ein flockiger Niederschlag ab,     welcher    das       Euglobulin    mit den darin enthaltenen Anti  genen darstellt und mit Hilfe der Zentri  fuge gesammelt wird. Dieser Niederschlag  wird dann unter     V.,:rwendung    von schwach  alkalischer Kochsalzlösung zu einer trüben  Flüssigkeit gelöst.

   Die so erhaltene Lösung  enthält neben spezifischem     Tuberkulose-          a.ntigen    von hoher     Wirksamkeit:    häufig ge  ringe Mengen unversehrter Tuberkelbazillen.  welche     mittelst    Filtration durch Bakterien  kerzen aus der Flüssigkeit entfernt werden  können. Die klare, völlig bakterienfreie Lö  sung wird als Antigen zum Schutze gesunder  Individuen oder auch zur Therapie     kranker     Individuen verwendet.

   It is characterized in that by electroosmotic treatment of the named starting materials at the isoelectric point, englobulin with the antigens contained therein is flocculated, then separated from the liquid and brought into solution using the corresponding amounts of alkali.

    <I> Example </I> In the abdominal cavity of tuberculous patients there are often very large amounts of so-called ascites fluid.
