Abstract

Compositions, kits et procédés pour générer des bibliothèques hautement diversifiées de protéines telles que les anticorps par le biais de la recombinaison homologue in vivo ainsi que criblage de ces bibliothèques à la recherche de cibles de protéines, peptides et acides nucléiques, et ce au moyen d'un procédé à deux hybrides dans la levure. Le procédé destiné au criblage d'une bibliothèque des protéines de fusion de test à la recherche d'une protéine ou peptide cible comprend ce qui suit: exprimer une bibliothèque de protéines de test dans les cellules de levure, la protéine de fusion de test comprenant une première sous-unité polypeptidique dont la séquence varie à l'intérieur de la bibliothèque indépendamment du premier polypeptide ainsi qu'un peptide de liaison qui relie la première et la deuxième sous-unités de polypeptides; exprimer une ou plusieurs protéines de fusion cibles dans les cellules de la levure exprimant les protéines de test, chacune des protéines de fusion cibles comprenant une protéine ou un peptide cible; et sélectionner celles des cellules de levure dans lesquelles un gène rapporteur est exprimé, l'expression du gène rapporteur étant activée par la liaison de la protéine de fusion de test à la protéine de fusion cible.
