Abstract
Des sondes d'ADN spécifiques fluorescentes et des paires d'amorces flanquantes sont conçues sur la base des informations de séquence trouvées dans la région conservée terminale 3' non codante de flavivirus, par ex. les nucléotides 10653-10678 du virus de la dengue. L'amplification en chaîne par polymérase (PCR) fluorogène employant ces amorces et sondes produit des résultats qui permettent l'identification spécifique de flavivirus. Les analyses peuvent être tant qualitatives que quantitatives. Des conditions optimales d'analyse avec bruit de fond nul qui permettent la détection de faibles niveaux de flavivirus dans des spécimens cliniques sont également décrites. Spécifiquement, des virus de dengue isolés dans différentes régions géographiques peuvent être universellement détectés et identifiés par l'analyse fondée sur la RT-PCR fluorogène décrite. Ladite analyse RT-PCR fluorogène a permis de détecter facilement une virémie dans des sérums prélevés chez des patients atteints de la dengue.
