Abstract
Selon l'invention, pour mettre en évidence les interactions entre des récepteurs GPC (récepteurs comportant sept domaines transmembranaires qui sont en interaction avec des protéines régulatrices liant le nucléotide guanylique) ou avec des systèmes de transmission de signaux contrôlés par le récepteur GPC et des substances test (ligands, modulateurs) et pour rechercher des substances qui peuvent induire une interaction avec ce type de récepteurs ou de systèmes de transmission de signaux, il est prévu d'utiliser des lignées cellulaires transformées, par exemple de Ustilago maydis. Lesdites lignées cellulaires transformées présentent une voie de transduction de signaux contrôlée par le récepteur GPC, à rétroaction positive et se trouvent transformées, de manière à exprimer un récepteur GPC hétérologue. Les lignées cellulaires contiennent en outre un gène rapporteur dont l'expression peut être détectée selon des techniques de mesure. Ledit gène rapporteur est piloté par un promoteur inductible par stimulation d'un récepteur GPC. Le promoteur inductible par stimulation de récepteur est endogène et le gène rapporteur est endogène ou hétérologue.
