Abstract

La présente invention se rapporte à des biocapteurs et à des procédés de détection de séquences d'acides nucléiques sélectionnés. Dans une des réalisations de l'invention, ce biocapteur comporte une électrode et une membrane bicouche. Ladite membrane comporte une couche inférieure et une couche supérieure. La couche inférieure se trouve à proximité de l'électrode, à laquelle elle est couplée de telle manière à qu'il existe un espace entre la membrane et l'électrode, la conductance ou l'impédance de la membrane étant fonction de la présence ou de l'absence de la séquence de l'acide nucléique sélectionné. Ladite membrane comporte un réseau serré de molécules amphiphiles et une pluralité de canaux ioniques comportant des monomères recouvrant une première moitié de la membrane, dispersés au sein de la couche supérieure, et des monomères recouvrant une seconde moitié de la membrane, dispersés au sein de la couche inférieure. Les monomères recouvrant la première moitié de la membrane sont aptes à une diffusion latérale au sein de la couche supérieure et les monomères recouvrant la seconde moitié de la membrane sont bloqués vis à vis d'une diffusion latérale au sein de la couche inférieure. Un premier ligand, susceptible de réagir spécifiquement avec la séquence de l'acide nucléique sélectionné, est fixé à une extrémité d'une fraction des monomères recouvrant la première moitié de la membrane, à proximité de la surface de la membrane et un second ligand, susceptible de réagir spécifiquement avec l'acide nucléique sélectionné ou avec un marqueur fixé à cet acide, est fixé à une extrémité du reste des monomères recouvrant la première moitié de la membrane, à proximité de la surface de la membrane.
