La presente demande a pour objet l'utilisation de composés antagonistes de molécules de la famille des chemokines ou des antagonistes des produits dérivés de chemokines ayant conservé toutes les propriétés biologiques des chemokines, lesdits composés étant caractérises en ce qu'ils permettent d'obtenir l'inhibition d'une infection virale ou de ses effets in vitro ou in vivo~ lesdits composés présentant à
titre préférentiel une activité anti-virale vis-à-vis d'infections dues à des rétrovirus provoquant une immunodéficience chez un hôte, et en particulier chez un patient humain, notamment des rétrovirus de type HIV ou VIH
(virus de l'immunodéficience humaine).
s Les chemokines sont des molécules de la famille des cytokines, qui présentent des proprietés d'activation, notamment de cellules de la famille des leucocytes faisant intervenir notamment des proprietés chemoattractives, des propriétés de mobilisation du calcium par augmentation du calcium intracellulaire et des propriétés de relargage d'enzymes (exocytose). Ces chemokines sont connues pour leur rôle eventuel en tant que médiateurs de l'inflammation.
Les chemokines ont été decrites par leur structure par leur affinité
pour un ou plusieurs recepteurs et par leurs proprietés biologiques dans une publication de Baggiolini M. et al (Advances in Immunology (1994), vol. 55, pages 97-179).
Une publication de Wells T.N.C. et al (Journal of Leukocyte Biology, vol. 59, January 1996, 53-60) décrit la structure tri-dimensionnelle de - plusieurs chemokines et leurs recepteurs spécifiques ou non.
De façon génerale, ces chemokines sont caractérisees par la présence dans leur structure primaire, de résidus cystéine conservés (1 à
CA 022~72 1998-11-19 4 résidus notamment) sur la base desquels plusieurs sous-familles ont été
distinguees selon la position des deux premieres cystéines. Ces familles comprennent celles des protéines CXC ou des protéines CC. La présence de ces résidus cystéine induit la formation de ponts disulfure.
s Parmi ces chemokines, différentes molécules ont fait l'objet d'études spécifiques. A cet égard, on citera la molécule RANTES décrite dans la publication de Schall T.J. et al. (The Journal of Immunology, August 1, 1988, vol. 141, 1018-1025, No. 3). La molécule RANTES (abréviation de Regulated upon Activation, Normal T-Express and presumably Secreted) a o été décrite dans cet article, notamment à l'aide de la séquence nucléotidique de son ADN complémentaire et à l'aide de la séquence d'acides aminés de la phase ouverte de lecture. Les proprietés biologiques de RANTES ont eté décrites notamment dans l'article de Baggiolini précité.
Ces récepteurs, spécifiques ou non, ont été identifiés dans la publication de Wells précitée ou dans les articles auxquels elle se réfère.
D'autres chemokines telles que les chemokines MIP (Macrophage Inflammatory Protein) observées, isolées et purifiées à partir de macrophages de souris (WOLPE S.D. et al, J. Ex. Med. (1988), 167-170) ont été décrites du point de vue de leurs propriétés biologiques dans I'article de Baggiolini et al. (1994).
Une autre catégorie de chemokines, appelées MCP (Monocyte Chemotactic Protein) a été identifiée à partir de basophiles. Dans les familles MIP et MCP, on connaît notamment les chemokines MIP1 a, MIP1,B, MCP1, MCP2 et MCP3.
Dans une publication de Cocchi F. et al (Science, 15 Déce~"bre 1995, vol. 270, 1811-1815), les auteurs ont émis l'hypothèse que des molécules telles que RANTES, MlP1a et MIP1~ produites notamment par des Iymphocytes T CD8~, pourraient constituer des facteurs suppresseurs de la réplication des isolats primaires du VIH. Ils ont ainsi émis l'hypothèse CA 022~72 1998-11-19 que les protéines RANTES, MlP1a et MIP1~ produites par des Iymphocytes T exprimant des récepteurs CD8 pourraient participer au contrôie de l'infection par un rétrovirus VIH en tant que facteur suppresseur de la replication. Des anticorps obtenus contre les 5 chemokines RANTES ou MlP1(x ou MIP113 ont selon Cocchi et al., la capacite de bloquer partiellement ou totalement, lorsqu'ils sont utilisés conjointement, I'activité anti-HlV des chemokines.
L'article de Cocchi et al. précité ne contient pas de donnees expérimentales sur le rôle et le stade de l'intervention des chemokines o dans le contrôle de l'infection par un rétrovirus VIH.
Les auteurs de la présente demande se sont intéressés a l'effet que peuvent exercer les chemokines sur la réplication du retrovirus VIH et après avoir vérifié l'effet anti-viral des molécules telles que RANTES, MlP1a et MIP1~ vis-à-vis de HIV, les inventeurs de la présente demande 15 se sont interessés au mécanisme biologique permettant l'interférence de chemokines avec la replication du rétrovirus VIH dans les Iymphocytes T.
De façon surprenante, les inventeurs ont caractérisé des composés, dépourvus des activites biologiques des chemokines et ayant perdu notamment l'activite chemo-attractive des chemokines vis-à-vis des 20 cellules de type leucocytes, en particulier des Iymphocytes ou des monocytes, composés susceptibles néanmoins de jouer un rôle dans le contrôle, voire l'inhibition, des effets d'une infection virale, en particulier d'une infection de cellules par un rétrovirus de type VIH.
Ainsi, les inventeurs ont mis en évidence que des antagonistes de 25 chemokines, par exemple des antagonistes de chemokines de la famille CXC ou de la famille CC, notamment de RANTES et/ou de MIP et/ou de MCP peuvent s'opposer à la réplication d'isolats primaires du rétrovirus VIH dans des Iymphocytes du sang périphérique (PBL) normaux.
L'invention a donc pour objet un composé antagoniste encore 30 appelé ~ antagoniste ~ d'une ou plusieurs molecules de la famille des CA 022~72 1998-11-19 chemokines pour l'utilisation pour le traitement ou la prévention d'une infection due à un virus et plus particulièrement à un rétrovirus du type VIH. Dans le cadre de cette définition, le composé antagoniste est utilisé
notamment pour la préparation d'un médicament pour la prévention ou le traitement de l'infection due à un retrovirus de type VIH.
Dans le cadre de la présente description, le terme ~ antagoniste ~
fait référence à un composé dont la presence chez un hôte et notamment chez un patient humain, est susceptible d'affecter l'activité d'une ou plusieurs chemokines tout en ayant la capacité d'empêcher ou d'inhiber o une infection virale in vivo ou in vitro; en particulier, un tel antagonistepeut bloquer ou déplacer la liaison d'une ou plusieurs chemokines à un ou plusieurs récepteurs de chemokines, sur des cellules, par exemple, des Iymphocytes, des monocytes ou des macrophages, et, donc le cas échéant de façon indirecte, cet antagoniste est susceptible d'affecter les propriétés s biologiques d'activation cellulaire de ces chemokines. Les propriétés recherchées ainsi définies de cet antagoniste peuvent être avantageusement vérifiées grâce à un test effectue in vitro sur des Iymphocytes PBL normaux stimules par un mitogene et infectés par des isolats primaires de rétrovirus VIH.
Selon la présente demande, on entend par l'expression ~ prévention ou traitement d'une infection due à un rétrovirus de type VIH ~ la capacité
du composé antagoniste précité d'intervenir soit sur la replication virale d'isolats primaires de VIH dans des cellules participant à l'immunité de l'hôte, et en particulier dans des Iymphocytes du sang périphérique, par exemple des Iymphocytes T-normaux. Cette expression englobe également la prévention et le traitement des infections telles que les infections dites opportunistes ou des tumeurs précédant et/ou accompagnant le développement de la maladie SIDA et de ce fait, définies comme maladies reliées a l'infection par un rétrovirus VIH.
CA 022~72 1998-11-19 Le test effectué in vitro sur des Iymphocytes PBL (Iymphocytes du sang périphérique) infectés a l'aide de souches primaires de VIH, est une indication pertinente des propriétés thérapeutiques ou prophylactiques recherchées des antagonistes décrits.
s Le terme ~ VIH ~ comprend les virus et rétrovirus humains ou animaux susceptibles d'induire une immunodeficience, tels que les rétrovirus connus sous les noms de VIH-1 et VIH-2 et notamment les différents isolats caractérisés jusqu'à ce jour, qu'il s'agisse d'isolats primaires ou d'isolats modifiés pour répondre à des contraintes de culture o in vitro. ou des clones HIV infectieux provenant d'isolats primaires.
Les composés antagonistes selon l'invention permettent, dans le cadre du traitement ou de la prévention d'une infection due à un rétrovirus de type VIH, de diminuer la réplication et/ou ia dissémination du rétrovirus dans l'organisme de l'hôte infecte et/ou de corriger le déficit immunitaire résultant de cette infection ou ses manifestations.
~ Selon un premier mode de réalisation de l'invention, I'antagoniste est un compose susceptible d'entrer en competition avec une ou plusieurs chemokines, pour la liaison à un ou plusieurs récepteur(s) cellulaire(s) de cette (ces) dernière(s).
Les molécules appartenant à la famille des chemokines sont connues pour avoir au moins pour certaines d'entre elles, la capacite de lier différents récepteurs cellulaires présents sur les cellules dérivées des lignées Iymphoïde ou myéloïde, et en particulier plusieurs récepteurs présents sur des cellules de type Iymphocytes, monocytes, basophiles, ou eosinophiles.
L'antagoniste utilisable dans le cadre de la présente demande, peut selon un premier mode particulier de réalisation de l'invention, être sélectionné pour sa capacité à interférer avec l'interaction normale entre les chemokines et leur(s) récepteur(s) et en particulier à modifier l'accès, par exemple la reconnaissance conformationnelle d'une ou plusieurs CA 022~72 1998-11-19 chemokines vis-à-vis de son(ses) récepteur(s) ou encore à bloquer la liaison d'une ou plusieurs chemokines avec un ou plusieurs recepteurs cellulaires naturels de ces chemokines.
Avantageusement, cet antagoniste se substitue à une ou plusieurs 5chemokine dans l'activité de liaison à un ou plusieurs recepteurs.
En dépit de l'existence d'une multi-spécificité de liaison des chemokines à des récepteurs celluiaires, I'invention a egalement pour objet selon un autre mode de realisation parti~culier, des composés antagonistes selectionnés pour leur affinité spécifique vis-à-vis d'un orécepteur déterminé ou d'un nombre déterminé de récepteurs de chemokines .
Selon une variante de réalisation de l'invention, I'antagoniste mis en oeuvre est choisi pour sa capacité à deplacer la liaison d'une ou plusieurs chemokines vis-à-vis d'un ou plusieurs recepteurs cellulaires de ces dernieres.
. Selon un mode de réalisation particulierement avantageux de l'invention, I'antagoniste a une activité de compétition avec une ou plusieurs chemokines choisies parmi RANTES, MIP1 a, MIP1~, MCP1, MCP3.
20Des parentés de structures ont été constatées entre les différentes chemokines précitees, en particulier entre les chemokines RANTES et MIP, tant au niveau de la séquence d'acides aminbs formant la structure primaire de ces chemokines que de leurs structures secondaires ou tri-dimensionnelles telles qu'établies par la technique de spectroscopie de 2sRésonnance Magnétique Nucléaire (Wells T.N.C. et al, Journal of Leukocyte ~iology, vol. 59, Janvier 1996, pages 53-60).
Les inventeurs ont constaté que des antagonistes de la molécule RANTES sont par exemple capables de lier les récepteurs de plusieurs chemokines en particulier ceux de RANTES, MCP3 et MCP1. De tels 30antagonistes présentent des propriétés tout-à-fait intéressantes, du fait de CA 022~72 1998-11-19 leur capacité à occuper les sites normalement occupés par RANTES, ladite occupation entralnant une activité anti-virale vérifiable notamment in vitro sur des Iymphocytes normaux infectés par des isolats primaires de ~ VIH.
Un composé antagoniste intéressant dans le cadre de la réalisation de l'invention est par exemple un compose dont la structure est dérivée de la structure d1une chemokine, par exemple d'une chemokine choisie parmi les molécules RANTES, MIP ou MCP définies précédemment.
On peut considerer qu'une structure est dérivée de celle d'une o chemokine lorsque le composé ayant cette structure est différent par exemple dans sa nature, sa composition, sa conformation, par rapport à la chemokine de référence tout en conservant les propriétés de ladite chemokine pour la réalisation de la fonction d'antagoniste définie ci-dessus, par exemple la capacité de se lier à un ou plusieurs récepteurs .
13 De façon particulièrement avantageuse, cet antagoniste est en outre caractérisé en ce qu'il est dépourvu des propriétés biologiques de la chemokine ou des chemokines dont il affecte l'activite.A cet égard, I'invention vise des antagonistes de chemokines dépourvus des propriétés de chemoattraction habituellement reconnues aux chemokines et 20 avantageusement à la molécule RANTES.
D'autres propriétés biologiques peuvent être atténuees voire supprimees dans lesdits antagonistes.
Par exemple, I'antagoniste peut être caractérise en ce qu'il est dépourvu de la capacité de transduction d'un signal dans les cellules 25 auxquelles il est lié par l'intermédiaire d'un ou plusieurs récepteurs déterminés. En d'autres termes, I'antagoniste est caractérisé en ce qu'il ne transduit aucune activité biologique à travers le ou les récepteurs auxquels il est lie. De façon essentielle, cet antagoniste conserve neanmoins l'activité antivirale reconnue a la molécule RANTES, à tout le moins a un CA 022~72 1998-11-19 niveau permettant de réduire la réplication ou la dissémination virale ou leur effet et de façon avantageuse, de l'inhiber.
Outre la diminution, voire la disparition des propriétés chemoattractives dévolues à la molecule native RANTES, un antagoniste selon l'invention peut egalement être caractérisé en ce qu'il est depourvu de la capacite naturelle de la molécule RANTES d'une part à induire la libération ou l'entrée du calcium dans les cellules auxquelles elle est liée par un récepteur et/ou d'autre part, à affecter l'exocytose par relargage d'enzymes à partir de ces cellules.
o Ainsi, I'invention met avantageusement à disposition des molécules ayant des effets anti-viraux d'une ou plusieurs chemokines qui sont néanmoins dépourvues des effets secondaires des chemokines actives.
Un antagoniste interessant selon l'invention peut en outre être caractérisé en ce qu'il s'agit d'un polypeptide dérive d'une chemokine, dont la structure comprend au moins un, voire deux ponts disulfure.
~ Ainsi, selon un mode de realisation préféré de l'invention pour l'utilisation dans le traitement d'une infection due à un rétrovirus ViH, on aura recours à un antagoniste de type polypeptidique, dérivé du polypeptide RANTES répondant à la sequence d'acides aminés représentée à la figure 1, par substitution, addition ou deletion d'au moins un résidu d'acides aminés, ledit antagoniste étant dépourvu des proprietés chemo-attractives des chemokines vis à vis des leucocytes, en particulier des Iymphocytes.
Le terme ~ polypeptide ~ fait référence à une structure d'acides aminés comprenant de 10 à 100, de préference de 10 à 70 acides aminés, laquelle structure peut être utilisee sous forme linéaire ou sous forme conformationnelle. A cet égard, I'invention concerne avantageusement des antagonistes ayant une structure correspondant à la structure conferée par la présence de plusieurs et notamment de deux ponts disulfure telle qu'on CA 022~72 1998-11-19 I'observe dans les cytokines, notamment dans les molécules RANTES, MIP et MCP.
Un polypeptide dérivé du polypeptide RANTES, utilisable dans le cadre de l'invention est par exemple obtenu par delétion de tout ou partie 5 de sa séquence N-terminale et en particulier par délétion de tout ou partie de la séquence comprise entre les residus d'acides aminés 1 et 15 ou entre les acides aminés 1 et 9 ou 1 et 8 (y compris les résidus extrêmes) de la séquence d'acides amines representée a la figure 1 C.
Selon un mode de realisation préféré de l'invention, I'antagoniste o utiiisable pour le traitement ou la prévention d'une infection due a un rétrovirus VIH est un polypeptide répondant à la séquence d'acides aminés suivante (appelée RANTES 9-68 ):

La suppression de ces résidus diminue les effets de signalisation à
travers les récepteurs de chemokines occupés, tout en gardant i'activite antivirale de cet antagoniste.
Les séquences d'acides aminés sont identifiees par référence au code à une lettre habituellement utilise.
Cet antagoniste peut être utilise sous la forme de sa structure primaire ou sous une forme tri-dimensionnelle par exemple sous une forme reproduisant les caractéristiques essentielles de la structure conformationnelle de la molécule RANTES a tout le moins de la partie de cette molécule impliquée dans la liaison à son ou ses récepteur(s) CA 022~72 1998-11-19 1() cellulaire(s). Cette structure a été rapportée dans l'article de Wells T.N.C.
précité et de Chung C.W. (Biochemistry 34, 9307-9314).
Alternativement ou au contraire selon une caractéristique supplémentaire, I'antagoniste utilisable dans le cadre de l'invention est un polypeptide dérivé du polypeptide RANTES lequel est representé par sa séquence d'acides amines à la figure 1 C, par délétion de tout ou partie de sa séquence C-terminale comprise entre les résidus d'acides aminés 50 et 68, de préférence 60 et 68 de la séquence représentée à la figure 1 C.
Un antagoniste dérivé du polypeptide RANTES par substitution ou o par mutation peut alternativement être caractérise en ce que environ 5% à
environ 10% des résidus d'acides aminés, à l'exception des résidus cystéine impliqués dans la formation de ponts disulfure, sont remplacés.
Les résidus d'acides aminés substitués peuvent l'être par des acides aminés neutres, ou constituant une substitution conservatrice ou 15 choisis le cas échéant en tenant compte de l'encombrement stérique.
A titre d'exemple des residus Iysine peuvent être remplaces par des résidus isoleucine, leucine ou valine.
A titre d'exemple, de tels antagonistes utilisables pour la réalisation de l'invention répondent à l'une des séquences d'acides aminés 20 suivantes:

Selon une variante de l'invention, I'antagoniste mis en oeuvre est caractérisé en ce qu'il s'agit d'une molécule RANTES dont l'extrémité N-terminale a éte allongée par addition d'au moins un résidu d'acide aminé
ou alternativement d'une molécule RANTES dont les extrémités N- et C-20 terminales ou dont les séquences internes à ces régions N- ou C-terminales ont été allongées par addition d'un ou plusieurs résidus d'acides aminés dans des conditions permettant d'obtenir l'effet antagoniste recherché.
L'invention a egalement pour objet une séquence de nucléotides 25 comprenant une sequence codant pour un polypeptide dérive du polypeptide F~ANTES repondant à la séquence d'acides aminés représentée à la figure 1, par delétion de tout ou partie de la sequence N-terminale de ce dernier.

Une sequence de nucleotides selon l'invention peut le cas echeant comprendre également des séquences exogènes, notamment des sequences de régulation et/ou d'activation capables de contrôler et/ou de réguler l'expression de la séquence codant pour un antagoniste selon 25 I'invention dans une cellule hôte, in vitro ou in vivo.
Elle peut également être recombinee avec une autre séquence codante susceptible d'exprimer dans un hôte cellulaire un polypeptide d'intérêt, notamment dans le but de traiter ou de prevenir une infection due à un retrovirus VIH ou avec une séquence codante pouvant remplir la 30 fonction de molécule porteuse. Par exemple, une séquence de nucléotides de l'invention peut être fusionnée avec tout ou partie d'une séquence codant pour la ,~-galactosidase. Dans ce cas, la séquence codante de la ~-galactosidase contient avant le premier codon correspondant au premier CA 022~72 1998-11-19 acide aminé proline de la molécule R9-68 ou R8-68, une séquence peptidique du type X-glycyl-L-propyl, reconnue spécifiquement par une collagénase, permettant ainsi la réalisation d'une coupure spécifique après le residu glycine du polypeptide formé, libérant l'antagoniste. Cette technique est décrite dans la demande de brevet européen 0115974.
Selon un autre mode de réalisation de l'invention, la séquence de nucléotides est caractérisée en ce qu'elle est inserée dans un vecteur d'expression ou adsorbée sur un support susceptible d'être internalisé
dans une cellule eucaryote ou encore dans une bactérie capable d'entrer o dans des cellules eucaryotes telle que Shigella selon le procédé decrit par Courvalin P. et al. (C.R. Acad. Sci. Paris, 1995, 318:1207-12). Ce vecteur est avantageusement choisi pour permettre l'expression in vitro dans un hôte cellulaire, d'un antagoniste tel que defini précedemment ou pour permettre l'expression in vivo chez un hôte auquel il est administre, notamment chez un patient infecté par un rétrovirus VIH, à des fins d'expression transitoire ou continue d'un antagoniste tel que défini dans les pages qui précedent.
Les séquences de nucléotides ainsi definies dans le cadre de l'invention sont par exemple utilisables dans le cadre d'un protocole de thérapie génique visant à prévenir ou à traiter une infection due a un rétrovirus VIH. A cet égard, on pourra avoir recours à des vecteurs utilisables dans le cadre de ces therapies, tels que ceux décrits dans les demandes de brevet internationales publiées sous les numéros WO
95/14785 et WO 94124298.
Ces séquences de nucléotides peuvent aussi être administrees en faisant appel aux techniques décrites dans le brevet FR 2711670 ou dans la demande de brevet internationale WO 90/11092, pour l'expression in vivo.
De façon genérale, et compte tenu des caractéristiques particulières de l'infection par un rétrovirus VIH, on peut avoir recours à des vecteurs CA 022~72 1998-11-19 permettant l'administration ou le relargage contrôlé voire retardé de ces séquences nucléotidiques ou de leur produit d'expression chez l'hôte traité, afin d'obtenir un effet antiviral continu ou itératif à tout le moins sur une période suffisante pour diminuer voire inhiber l'infection par le 5 rétrovirus ou ses effets. A cet égard, des techniques telles que celles faisant appel à l'utilisation d'un gel de phosphate de calcium peuvent être utilisées pour obtenir un relargage avec effet retard d'un produit actif. De telles techniques sont par exemple décrites dans le brevet français 2 543 439.
Des séquences reconnues par des protéases cellulaires ou extracellulaires peuvent être utilisees pour permettre la production de formes matures des molécules antagonistes de RANTES à partir de ces vecteurs. La forme mature de la proteine RANTES est en effet produite après clivage d'une séquence de 23 acides aminés situés en amont du s premier residu (serine) de la forme active de la protéine.
Les taux sériques à atteindre chez les patients traités seraient de l'ordre de 1 à 100 nM.
L'invention a également pour objet un polypeptide correspondant au produit d'expression dans une cellule hôte, par exemple une cellule 20 bactérienne ou une cellule eucaryote, notamment une cellule de levure, une cellule de mammifère, par exemple CHO, d'une séquence de nucléotides telle que définie précédemment.

Un autre polypeptide antagoniste utilisable dans le cadre de l'invention est par exemple un poiypeptide RANTES modifié par addition d'un ou plusieurs résidus d'acides aminés en amont de son extrémite N-terminale. Par exemple, I'addition d'une méthionine en amont du premier résidu (sérine) de la forme mature de RANTES presenterait l'avantage d'inhiber la capacité chemoattractive de la chemokine tout en conservant sa capacité d'inhibition de l'infection virale, grâce à sa capacité non modifiée à se lier spécifiquement a son recepteur.
La production de molécules modifiees de RANTES ou d'autres polypeptides antagonistes peut être réalisée par synthèse chimique ou par CA 022~72 1998-11-19 recombinaison et expression dans une cellule procaryote ou eucaryote par exemple dans la levure ou chez E coli.
Selon un autre mode de réalisation avantageux de l'invention, le polypeptide est un antagoniste d'une ou plusieurs chemokines choisies 5 parmi RANTES, MIP ou MCP, caractérisé en ce qu'il est dérivé du polypeptide RANTES répondant à la sequence d'acides aminés représentée à la figure 1, par substitution, addition ou déletion d'au moins un résidu d'acides aminés, ledit antagoniste étant dépourvu des propriétés chemoattractives des chemokines par exemple vis-à-vis des leucocytes, en o particulier des Iymphocytes, ledit polypeptide étant sous forme d'un polypeptide de fusion, capable de lier le(les) récepteurs cellulaires de RANTES et/ou MIP etlou MCP.
Les polypeptides de l'invention peuvent être prépares par génie génétique ou par synthèse chimique par exemple sur un appareil (Applied S Biosystems) ou par synthèse sur phase solide décrite par Merrifiel, par exemple selon la technique décrite dans la demande WO 95/09868.
L'invention vise également une composition caractérisée en ce qu'elle comprend pour l'administration séparee ou simultanée un composé
antagoniste d'une ou plusieurs molécules de la famille des chemokines 20 telles que définies précédemment, et un ou plusieurs agents actifs susceptibles d'être utilisés dans le traitement d'une infection par un rétrovirus VIH, par exemple un agent utilisable en chimiothérapie tel qu'un composé anti-transcriptase inverse ou anti-protéase virale ou un composé
tel qu'un antagoniste du récepteur CD4 présent sur des Iymphocytes de 2s patients infectés par un rétrovirus VIH ou encore un composé utilisable en immunothérapie, par exemple une cytokine et notamment l'interleukine 2.
Le cas échéant, ladite composition comprend, outre le composé
antagoniste d'une ou plusieurs chemokines et une ou plusieurs substances actives en chimiothérapie antivirale,ettou un agent de traitement des CA 022~72 1998-11-19 infections opportunistes se développant lors de l'infection par un rétrovirus VIH ou lors de l'évolution de l'infection in vivo.
Les produits définis dans l'invention, qu'il s'agisse des antagonistes ou des séquences de nucleotides codant pour de tels antagonistes, le cas 5 échéant en combinaison avec d'autres éléments, sont utilisables pour le traitement de patients à tous les stades de l'infection par un retrovirus de type VIH et notamment pour le traitement des enfants nes de meres séropositives ou des personnels soignants contamines accidentellement.
Les composés de l'invention peuvent être à cet egard administrés par o injection par voie intraveineuse par exemple par perfusion, par voie intramusculaire ou sous-cutanée. Dans le cas de l'utilisation en thérapie génique des composés décrits dans l'invention, on pourra avoir recours a des cellules du patient, par exemple des fibroblastes, myoblastes ou cellules hématopoïétiques traitées avec des vecteurs appropriés contenant 15 les sequences nucléotidiques de l'invention, ces cellules étant ensuite réimp~antées chez le patient pour l'expression des susdites séquences de nucléotides.
L'efficacité du traitement ainsi effectue peut être evaluée en contrôlant et en suivant la charge virale, plasmatique et cellulaire et en 20 examinant l'évolution du nombre de Iymphocytes porteurs de récepteurs CD4, ainsi que le statut immunologique et inflammatoire du patient.

o Evaluation de l'activite antivirale de l'antagoniste RANTES 9-68 Un antagoniste de la chemokine RANTES a été défini et obtenu sous la forme d'un polypeptide tronqué dont la séquence d'acides aminés correspond aux résidus d'acides aminés 9 à 68 de la sequence d'acides aminés de la molecule RANTES telle que définie à la figure 1.
L'antagoniste RANTES 9-68 a été produit en utilisant la technique décrite dans la publication de Gong JH et al (J B C vol 271 N18, issue of may 3, 1996 p 10521 -10527).
Il a eté utilise à des concentrations variables de 1000 nM à 10 nMet a été
ajouté à des cultures cellulaires après infection par différents isolats de VIH. A cet effetl un clone d'un isolat primaire obtenu a partir du fluide cérébrospinal d'un patient atteint de SIDA (désigné par VIH JR-CSF
Koganagi Y et al Science 236: 819, 1987 et Cann AJ et al, J Virol 64:
4735, 1990) a éte utilisé ainsi qu'un clone PYU2 correspondant a une souche primaire de VIH ayant un tropisme pour ies Iymphocytes T et les monocytes et isolé à partir du cerveau (Li et al, J. Virol., 1992, vol 66-6587). Pour obtenir le clone JR-CSF, I'ADN proviral a ete transfecté dans des cellules COS7 et le surnageant obtenu après trois jours de la culture transfectée contenant les particules infectieuses a été utilisé pour infecter des PBL.
CA 022~72 1998-11-19 La capacité antivirale de R9-68 mise en évidence dans des cellules infectées par HIV pYU2 et HIV JR-CSF était similaire à celle de MlP1a En effet, des niveaux de suppression de la réplication virale supèrieurs ou égaux à 95% des cultures contrôles infectées ont été obtenus à la concentration de 10nM, la plus faible concentration de MlP1a a induire un tel effet suppresseur sur la réplication de HIV-JR-CSF ou HlV-pYU2 o Les propriétés antivirales de l'antagoniste R9-68 ont éte testées en déterminant la capacite de cet antagoniste à bloquer la réplication du virus VIH.
La capacité d'une molécule à bloquer la réplication du virus VIH a pu être testee en utilisant la technique mettant en oeuvre des leucocytes isolés sur un gradient de Ficoll à partir de donneurs sains, lesquels leucocytes ont été infectés in vitro avec des isolats viraux primaires, incapables de se repliquer dans des lignées cellulaires transformées.
~ A cet égard, des cellules de six donneurs difféfents, le plus souvent des blastes induits par un traitement à la phytohémaglutinine (PHA) pendant 48 à 72 heures ont été infectés pendant 2 ou 3 heures avec les isolats de VIH. La quantité d'inoculum viral a été estimee par quantification de la protéine GAG P24 et s'est avéree être comprise entre 150 à 5000 picogrammes pour 10 000 000 de cellules. Après des lavages pour éliminer les particules virales residuelles, les cellules ont éte cultivées sur 2~ des microplaques de 96 puits (2 x 105/200 microlitres) en présence des molécules testées purifiées dans un milieu de culture enrichi en interleukine 2. Les surnageants de culture ont éte collectés périodiquement et les cultures cellulaires ont éte repiquées avec des milieux frais et les molécules testées. Dans certains cas, les blastes traites avec la PHA ont également été ajoutés pour apporter à la culture infectée CA 022~72 1998-ll-l9 des cellules fraîches servant de cible pour VIH. La réplication virale a été
mesurée en utilisant le kit ELISA P24 (Dupont de Nemours) (seuil de détection: 3 pg/ml).
Ainsi, pour un des donneurs les celluJes infectées par pYU2 et traitées avec R9-68 à la concentration de 1000nM ont produit 7 pg/ml de protéine p24 au jour 11 (76 et 24 pg/ml aux jours 6 et 8 respectivement).
La protéine p24 n'a pas été détectée dans les cultures de ce même donneur infectees par JR-CSF en présence de R9-68 au jour 11 (62 et 15 pg/ml ont été détectés aux jours 6 et 8 respectivement). Des cultures o contrôles infectées par pYU2 et JR-CSF mais non traitées par R9-68 ont montré une réplication virale intense qui a généré 7 ng/ml et 113 ng/ml de protéine P24 au jour 11 respectivement. Au contraire de R9-68, I'antagoniste MR-IL8 (1000 nM de la protéine IL8 était dépourvu d'effet antiviral.
Pour approfondir la connaissance des mécanismes d'interférence dans. Ia réplication du VIH, induits par R9-68, des expériences visant l'analyse de l'internalisation de particules infectieuses et la rétrotranscription de l'ARN viral génomique en ADN proviral ont été
realisées.
Après infection par la souche JR-CSF de cellules mononuclées du sang activées par la PHA, en présence ou en l'absence de 100nM de R9-68, des extractions d'ADN ont été pratiquées 3h post-infection, ou d'ADN
20h post-infection. Les cultures infectées en présence de R9-68 ont eté
maintenues en présence de la molécule iusqu'à extraction de l'ARN ou de I'ADN. Par des techniques d'amplification par PCR soit du RNA g~enomique rétrotranscrit in vitro, soit de l'ADN proviral, une légère diminution de l'accumulation de RNA génomique viral intracellulaire a éte mise en evidence dans les cellules traitées par R9-68 par rapport aux contrôles de cellules infectées mais non traitées. En revanche l'analyse de l'ADN
proviral a révélé une profonde diminution du signal spécifique dans les CA 022~72 1998-11-19 cellules infectees en présence de R9-68. La spécificité du signal obtenu de I'ADN proviral était démontrée par son abolition dans des cellules infectées et traitées par l'inhibiteur de la réverse transcriptase virale, 3'-azido-3'deoxythimidine (AZT). Des résultats comparables ont eté obtenus s avec RANTES dans des experiences réalisées en parallèie avec celles ci-dessus décrites.
Ces résultats indiquent que l'interférence de R9~68 et aussi celle de RANTES, dans la réplication de l'isolat primaire HIV-JR-CSF aurait lieu précocément mais de preférence après internalisation du virus.
o Des données similaires concernant l'activité antivirale des chemokines RANTES et MIP1 a ou de l'antagoniste R9-68 ont été
obtenues lorsque des blastes traités avec la PHA provenant d'un autre donneur ont été infectés avec l'isolat JR-CSF ayant subi deux semaines de passage dans des cultures de PBL traitées avec l'IL2.
Dans tous les cas la viabilité des cellules traitées avec R9-68 ou avec RANTES était comparable à celle des cultures controles infectées mais non traitees Les méthodes de mesure ont ét faite en comparaison des thymidines tritiées ou du contage direct du nombre de cellules à la fin des pèriodes de culture. 2 PCT / FR97 / 00900 USE OF ANTAGONIST MOLECULES OF CHEMOKINES

HIV TYPE RETROVIRUSES.

A publication by Wells TNC et al (Journal of Leukocyte Biology, flight. 59, January 1996, 53-60) describes the three-dimensional structure of - several chemokines and their specific or non-specific receptors.

EXAMPLES
o Evaluation of the antiviral activity of the antagonist RANTES 9-68 A RANTES chemokine antagonist has been defined and obtained in the form of a truncated polypeptide whose amino acid sequence corresponds to amino acid residues 9 to 68 of the acid sequence amines of the RANTES molecule as defined in Figure 1.
The antagonist RANTES 9-68 was produced using the technique described in the publication by Gong JH et al (JBC vol 271 N18, issue of may 3, 1996 p 10521 -10527).
It has been used at varying concentrations from 1000 nM to 10 nM and has been added to cell cultures after infection with different isolates of HIV. For this purpose a clone of a primary isolate obtained from the fluid cerebrospinal of an AIDS patient (designated by HIV JR-CSF
Koganagi Y et al Science 236: 819, 1987 and Cann AJ et al, J Virol 64:
4735, 1990) was used as well as a PYU2 clone corresponding to a primary strain of HIV with a tropism for T cells and monocytes and isolated from the brain (Li et al, J. Virol., 1992, vol 66-6587). To obtain the JR-CSF clone, the proviral DNA was transfected into COS7 cells and the supernatant obtained after three days of culture transfected containing infectious particles was used to infect PBL.

retrotranscribed in vitro, i.e. proviral DNA, a slight decrease in the accumulation of intracellular viral genomic RNA was put in evidence in cells treated with R9-68 compared to controls for infected but untreated cells. However, DNA analysis proviral revealed a profound decrease in the specific signal in CA 022 ~ 72 1998-11-19 infected cells in the presence of R9-68. The specificity of the signal obtained from Proviral DNA was demonstrated by its abolition in cells infected and treated with the reverse viral transcriptase inhibitor, 3'-azido-3'deoxythimidine (AZT). o Similar data regarding the antiviral activity of RANTES and MIP1 a chemokines or the antagonist R9-68 were obtained when PHA-treated blasts from another donor were infected with the JR-CSF isolate that had undergone two weeks of passage through PBL cultures treated with IL2.
In all cases, the viability of cells treated with R9-68 or with RANTES was comparable to that of infected control cultures but not treated The measurement methods were made in comparison with tritiated thymidines or direct counting of the number of cells at the end of cultivation periods.
