Abstract

La présente invention concerne des gènes chimères. Ces gènes comprennent un premier promoteur reconnu par une polymérase d'ARN dépendante de l'ADN, différente d'une polymérase II d'ARN eucaryote. Une zone d'ADN codant un ARN chimère qui comprend un UTR 5' une séquence de codage hétérologue riche en AU, un UTR 3', et éventuellement une séquence de terminaison reconnue par cette polymérase d'ARN. Le premier promoteur et la zone d'ADN codant l'ARN chimère sont liés de manière opérationnelle de telle sorte que lors de la transcription par la polymérase d'ARN, une espèce d'ARN sans addition de chapeau est produite qui comprend une première séquence de stimulation de traduction dérivée de la zone 5' de l'ARN génomique ou sous-génomique d'un virus de plante à ARN à brins positifs; une séquence de codage d'ARN hétérologue codant un polypeptide ou une protéine interressante, de préférence, à partir d'un gène riche en AT et enfin une deuxième séquence de stimulation de la traduction dérivée de la zone 3' de l'ARN génomique ou sous-génomique d'un virus de plant à ARN à brins positifs. Cette espèce d'ARN sans addition de chapeau peut être traduite dans le cytoplasme d'une cellule de plante pour produire la protéine ou le polypeptide. L'invention concerne également des cellules de plantes et des plantes comprenant ces gènes chimères, intégrés dans leur ADN nucléaire, ces cellules produisant ainsi les polymérases d'ARN correspondant aux promoteurs et aux terminateurs utilisés. En outre, l'invention a pour objet un procédé pour produire une plante exprimant une protéine ou un polypeptide codé par un gène hétérologue qui comprend les étapes consistant à transformer le génome nucléaire d'une cellule de plante avec les gènes chimères susmentionnés, et regénérer une plante transformée à partir de la cellule transformée.
