Abstract
La présente invention concerne une méthode de traitement des cancers par régulation des niveaux cellulaires de la protéine p53. Elle réside plus particulièrement l'utilisation d'un composé capable de moduler l'activité de la calpaïne.
Claims (16)
1. Utilisation d'un composé capable de moduler l'activité de la calpaïne pour la préparation d'une composition pharmaceutique destinée à réguler les niveaux cellulaires de la protéine p53.
2. Utilisation selon la revendication 1 caractérisée en ce que le composé est une protéine ou un polypeptide inhibiteur de l'activité de la calpaïne, ou une séquence d'acides nucléiques codant pour un tel polypeptide ou protéine.
3. Utilisation selon la revendication 2 caractérisée en ce que le composé est une protéine ou un polypeptide inhibiteur spécifique de l'activité de la calpaïne sur la protéine p53 sauvage, ou une séquence d'acides nucléiques codant pour un tel polypeptide ou protéine.
4. Utilisation selon la revendication 2 ou 3 caractérisée en ce que l'acide nucléique fait partie d'un vecteur.
5. Utilisation selon la revendication 4 caractérisée en ce que l'acide nucléique fait partie d'un vecteur viral, choisi parmi les adénovirus, les rétrovirus et les virus adéno-associés.
6. Utilisation selon la revendication 4 caractérisée en ce que l'acide nucléique fait partie d'un vecteur liposomal, lipidique
7. Utilisation selon l'une des revendications précédentes caractérisée en ce que le composé est un acide nucléique codant pour tout ou partie de la calpastatine.
8. Utilisation selon la revendication 7 caractérisée en ce que l'acide nucléique comprend tout ou partie de la séquence SEQ ID n° 1 ou d'un dérivé de celle-ci.
9. Utilisation selon la revendication 8 caractérisée en ce que l'acide nucléique est choisi parmi les séquences SEQ ID n° 1 et 2.
10. Utilisation selon la revendication 8 caractérisée en ce que l'acide nucléique est choisi parmi les dérivés des séquences SEQ ID n° 1 ou 2 codant pour des inhibiteurs spécifiques de la dégradation de la protéine p53 sauvage.
11. Utilisation selon l'une des revendication 1 à 6 caractérisée en ce que le composé est un dérivé de la calpaïne capable de dégrader spécifiquement les protéines p53 mutées.
12. Vecteur viral comprenant une séquence d'acide nucléique codant pour une protéine ou un polypeptide inhibiteur de l'activité de la calpaïne et choisi parmi les adénovirus, les rétrovirus et les virus adéno-associés..
13. Vecteur selon la revendication 12 caractérisé en ce qu'il comprend une séquence codant pour tout ou partie de la calpastatine.
14. Vecteur selon la revendication 12 caractérisée en ce qu'il comprend une séquence codant pour un dérivé de la calpaïne capable de dégrader spécifiquement les protéines p53 mutées.
15. Composition pharmaceutique comprenant une séquence d'acides nucléiques codant pour tout ou partie de la calpastatine ou pour un dérivé de lacalpaïne capable de dégrader spécifiquement les protéines p53 mutées.
16. Composition selon la revendication 15 formulée en vue d'une administration intra-tumorale.
