La présente invention est relative a l'application en cosmetique et en pharmacle de la 7-~ -(acetylthio 4',5'-dihydrospiro randrost 4-ène 17,2'-(3'~) furan] 3-one ou spiroxasone en vue d'induire et de stimuler la croissance des cheveux et diminuer leur chute.
La croissance des cheveux et leur renouvellement sont principalement deter~ines par l'activité des follicules pileux. Leur activite est cyclique et comporte essentiellement trols phases à
savoir la phase anagène, la phase catagène et la phase télogène.
A la phase anagène active ou phase de croissance, qui dure plusieurs années au cours de laquelle les cheveux s'allo~gent, succède une phase catagène très courte et transitoire et une phase de repos appelee phase telogene de quelques ~ois.
A la fin de la période de repos les cheveux tombent et un autre cycle recommence. La chevelure se renouvelle donc en permanence chez l~tre humain et sur les 150 Q00 cheveux que comporte une chevelure, à chaque instant i0% environ sont au repos et seront donc remplacés en quelques is.
Dans un nombre important de cas, la chute des cheveux survient sur des sujets prédisposés génetiquement et elle atteint notamment les hommes. Il s'agit plus particulièrement de l'alopécie androgénetique ou sndrogénique ou encore androgeno-génétique. The present invention relates to the application in cosmetics and in pharmacies of 7- ~ - (acetylthio 4 ', 5'-dihydrospiro randrost 4-ene 17.2 '- (3' ~) furan] 3-one or spiroxasone to induce and stimulate hair growth and decrease hair loss.
Hair growth and renewal is mainly deter ~ ines by the activity of the hair follicles. Their activity is cyclical and essentially comprises three phases at namely the anagen phase, the catagen phase and the telogen phase.
At the active anagen phase or growth phase, which lasts several years during which the hair turns nice, there follows a very short and transient catagen phase and a phase of rest called the telogene phase of a few months.
At the end of the rest period the hair falls out and another cycle starts again. The hair is therefore constantly renewed in the human being and on the 150 Q00 hairs that a hair comprises, around i0% at all times are at rest and will therefore be replaced in a few is.
In a significant number of cases, hair loss occurs on genetically predisposed subjects and it reaches in particular men. It is more particularly alopecia androgenic or sndrogenic or androgenogenetic.

des cheveux puis de leur quantité. Il y a un appauvrissement progressif de la chevelure par régression des cheveux dits "terminaux" au stade de duvets. Des zones sont touchées préférentiel-lement notamment les golfes temporaux ou frontaux chez l'homme, et chez les femmes, on constate une alopécie diffuse du vertex.
On recherche depuis de nombreuses années dans l'lndustrie cosmétique ou pharmaceutique des compositions permettant de supprimer ou de réduire l'effet de l'alopécie androgéno-génétique et notamment d'induire DU de stimuler la croissance des cheveux ou de diminuer leur chute. On a proposé a cet effet en particulier les dérivés de la pyrimidine tels que le minoxidil ou des rétinoides tel que l'acide tout-trans rétinoique.
La demanderesse a découvert maintenant que la spiroxasone lorsqu'elle est utilisée en application topique avait une activité
surprenante sur la repousse des cheveux et permettait d'induire et de stimuler la croi~sa~ce de ceux-ci et diminuer leur chute.
La spiroxasone (DCI) ou 7-~ (acétylthio) 4'~5'-dihydrospiro ~androst 4-ène 17,2' (3'~) furan~3-one est un composé connu en particulier pour ses propriétés thérapeutiques diurétiques.
Afin de determiner l'efficacité et la rapidité d'action des compositions conformes à l'invention on utilise le trichogramme ou de pré~érence le phototrichogramme qui permet de déterminer entre autres le pourcentage de cheveux en phase anagene par rapport aux cheveux en phase télogene. L'examen du phototrichogramme a permis de déterminer que la spiroxasone étalt partlculièrement efficace en ce qui concerne le traite~ent de l'alopécie androgénétique.
Un objet de l'invention est constitué par des compositions cosmétlques ou pharmaceutiques utilisées en appllcation toplque, en vue d'induire et de stimnler la croissance des cheveux et diminuer leur chute.

The Applicant has now discovered that Spiroxasone when used topically had activity surprising on hair regrowth and allowed to induce and stimulate the growth ~ sa ~ ce of these and decrease their fall.
Spiroxasone (INN) or 7- ~ (acetylthio) 4 '~ 5'-dihydrospiro ~ androst 4-ene 17.2 '(3' ~) furan ~ 3-one is a compound known as particularly for its therapeutic diuretic properties.
In order to determine the effectiveness and speed of action of compositions according to the invention, the trichogram or pre ~ erence the phototrichogram which allows to determine among other things the percentage of hair in the anagen phase compared to the hair in telogen phase. Examination of the phototrichogram made it possible to determine that spiroxasone is particularly effective in the treat ~ ent of androgenetic alopecia.
An object of the invention consists of compositions cosmetics or pharmaceuticals used in topical application, in view to induce and stimulate hair growth and decrease their fall.

Les compositions destinées à etre utilisees en application topique sur le culr chevelu, en vue d'indulre et de stimuler la 5croissance des cheveux et diminuer leur chute sont essentiellement caractérisées par le fait qu'elles contiennent la spiroxasone dans un milieu solvant physlologiquement acceptable à base d'alcool.
La spiroxasone ou encore 7- ~(acétylthio) 4',5'-dihydrospiro /androst 4-ène 17,2' (3'~) furan~ 3-one peut être utilisée telle 10quelle ou sous forme te ses sels ou esters acceptables en cosmétique et en pharmacie. On peut citer plus particulièrement l'acétate.
La spiroxasone est de préférence utilisee dans des proportions comprises entre 0,1 et 15% en poids par rapport au poids total de la composition et de preference entre 1 et 10~ en poids.
lSLe milieu solvant physiologiquement acceptable à base d'alcool utilisé conformement à l'invention est de préference un milieu alcoolique ou hydroalcoolique pouvant contenir un ou plusieurs autres solvants acceptables en cosmetique ou en pharmacie.
Le mllieu alcoolique est de préférence un milieu anhydre c'est-à-dire contenant moins de 1% d'eau et contenant un ou plusieurs alcools inférieurs en Cl-C4 tels que l'alcool ethylique, l'alcool isopropylique ou l'alcool tertiobutylique, un mélange d'alcool éthylique et d'alcool isopropylique étant particulierement préféré.
Les alcools utilisés dans les milieux hydroalcooliques sont choisis parmi les alcools inferieurs en Cl-C4 indiqués ci dessus.
Les autres solvants utilisables dans les compositions conformes à l'inve tion sont en particulier choisis parmi les alkylèneglycols tels que le propylèneglycol, ou dans lesquels le groupement alkyle ou alkylène comporte entre 1 et 4 atomes de carbone et plus particulièrement le noéthyléther d'éthylèneglycol, le monomethyl-éther de propylèneglycol et le monoéthyléther de diéthylèneglycol.
Ces d~fférents milieux peuvent être épaissis en utilisant des agents épaississants ou gelifiants blen connus dans l'état de la technique tels que plus particulièrement les hétéroblopolysaccharides 3 20 ~ 17169 .,, V

Spiroxasone or 7- ~ (acetylthio) 4 ', 5'-dihydrospiro / androst 4-ene 17.2 '(3' ~) furan ~ 3-one can be used as 10which or in form of its salts or esters acceptable in cosmetics and in pharmacies. Mention may more particularly be made of acetate.
Spiroxasone is preferably used in proportions between 0.1 and 15% by weight relative to the total weight of the composition and preferably between 1 and 10 ~ by weight.
lS Physiologically acceptable alcohol-based solvent medium used in accordance with the invention is preferably a medium alcoholic or hydroalcoholic which may contain one or more others solvents acceptable in cosmetics or pharmacy.
The alcoholic medium is preferably an anhydrous medium, that is i.e. containing less than 1% water and containing one or more lower Cl-C4 alcohols such as ethyl alcohol, alcohol isopropyl or tert-butyl alcohol, a mixture of alcohol ethyl and isopropyl alcohol being particularly preferred.
The alcohols used in hydroalcoholic environments are chosen among the lower Cl-C4 alcohols indicated above.
The other solvents which can be used in the conforming compositions at the invention are in particular chosen from alkylene glycols such as propylene glycol, or in which the alkyl group or alkylene has between 1 and 4 and more carbon atoms particularly ethylene glycol noethyl ether, monomethyl-propylene glycol ether and diethylene glycol monoethyl ether.

Les milieux particulièrement préférés sont constitués par les milieux alcooliques essentiellement anhydres, hydroalcooliques comportant de préférence moins de 50% d'eau.
Une forme de réalisation particuli~rement préférée de l'invention consiste à utiliser un milieu alcoolique ou hydroalcoolique contenant au moins un alcool et l'un des solvants non alcooliques indiqués ci-dessus dans des proportions comprises entre 85 et 99,9% en poids par rapport au poids total de la composition.
Il est ~galement possible d'effectuer le mélange de la spiroxasone avec le milieu solvant au moment de l'emploi.
Les compositions conformes à l'invention peuvent contenir d'autres agents permettant d'induire et de stimuler la croissance des cheveux et de diminuer leur chute comme par exemple le Minoxidil et/ou les rétino~des.
Les compositions conformes à l'invention peuvent également contenir d'autres ad~uvants habituellement utilisés dans le domaine cosmétique- ou pharmaceutique en vue de réaliser les compositions topiques telles que plus particulièrement des agents tensio-actifs, les agents conservateurs, des agents alcalinisants ou acidifiants. Le pH de ces co~positions peut varier entre 3 et 9 et de préférence entre 5 et 8.
Les fo~mes de composition particulièrement préférées sont des lotions, des gels, des émulsions, des crèmes éventuellement pressurisés en aérosols.

W O 90/09164 20~7~69 PCT/FR90/001~2 Ces compositions peuvent être appliquées en faisant suivre eventuellement leur application d'un rinçage avec ! de l'eau ou éventuellement sous forme de shampooing auquel cas elles contiennent obligatoirement un agent tensio-actif ayant des propriétés détergentes, bien connu dans l'état de la technique.
Lorsque la composition est préparée tout juste au moment de l'emploi, il est possible de la conditionner sous forme d'un dispositif à plus$eurs compartiments ou "kit" dont l'un des compartiments ou l'un des eléments contient la spiroxasone et l'autre le milieu solvant. Le compartiment ou élement contenant la spiroxasone est réalise de preference en un materiau photoprotecteur.
Le traitement pour lutter contre la chute tes cheveux et favoriser la repousse consiste principalement à appliquer sur les zones alopeciques du cuir chevelu et les cheveux de l'individu, une composition telle que definie ci-dessus par exemple après lavage du cuir chevelu et des cheveux à l'aide d'un shampooing 4U peu de temps après un shampooing. Le mode d'application prefere consiste à
appliquer 2 ml de la composition selon l'invention sur la zone alopecique sur une surface de 200 à 300 cm2 du cuir chevelu à une frequence ~ournalière. Le traitement pouvant être poursuivi quelques annees.
L'efficacite du traitement est contrôlee de preference une fois par is au moyen d'un phototrichogramme. On a constate que la composition conforme à l'invention permettait d'obtenir des resultats particulièrement surprenants par l'augmentation du rapport AtT
(anagène/télogène).
Un autre objet de l'invention est constitue par l'utilisation de la spiroxasone pour la préparation d'une composition thérapeutique destinée au traitement des alopécies en particulier androgenetiques.
Le procede de traitement conforme a l'invention presente les caracteristiques d'un traitement cosmetique dans la mesure où il permet de soigner les cheveux et le cuir chevelu dans le sens cosmetique du terme c'est-à-dire leur apporter les substances qui lui manquent et les e~bellir.

Le procede conforme à l'invention constitue egalement un traitement therapeutique dans la mesure où la substance actlve agit sur le mecanisme biologique du cycle pilaire et permet notamx nt un traitement thérapeutique de l'alopécie androgénétique.

On prépare une lotion permettant de favoriser la repousse des cheveux et d'enrayer leur chute, de composition suivante :
- Spiroxasone 1 g - ~élange pondéral alcool éthylique (l/3) - Alcool ~sopropylique (2/3) qsp 100 g On évalue l'activité de la composition pour stimuler la repousse des cheveux par rapport à un placebo, en appliquant 2 ml de cette composition a raison d'une application par Jour sur une zone alopecique de 100 a 200 cm du cuir chevelu. En quelques mois de traitement, la composition ameliore efficacement la repousse des cheveux.

When the composition is prepared just at the time of the job, it is possible to condition it in the form of a device with multiple compartments or "kit", one of which compartments where one of the elements contains spiroxasone and the other the solvent medium. The compartment or element containing the spiroxasone is preferably produced from a photoprotective material.
The treatment to fight against hair loss and promoting regrowth mainly involves applying to alopecic areas of the scalp and hair of the individual, a composition as defined above for example after washing the scalp and hair using a 4U shampoo shortly after shampooing. Claims

1. Composition used topically, intended to induce and stimulate hair growth and reduce hair loss, characterized by the fact that it contains. in a solvent medium physiologically acceptable alcohol-based, at least the 7-? -(acetylthio)4',5'-dihytrospiro [androst 4-ene 17,2'-(3'H)furan]
3-one or spiroxasone and its salts and esters physiologically acceptable.

2. Composition according to claim 1, characterized by the fact that spiroxasone is present in the composition in proportions between 0.1 and 15% by weight relative to the weight total of the composition preferably between 1 and 10%.

3. Composition according to claim 1 or 2, characterized by the fact that the physiologically acceptable solvent medium based on alcohol medium is an anhydrous or hydroalcoholic alcoholic medium.

4. Composition according to claim 3, characterized by the fact that the alcoholic medium consists of a mixture of alcohols lower in C1-C4.

5. Composition according to claim 4, characterized by the fact that this medium also contains alkylene glycols or alkyl-mono and dialkylene glycol ethers.

6. Composition according to claim 3, characterized by the fact that the hydroalcoholic medium contains less than 50% water, one or several lower C1-C4 alcohols and one or more alkylene-glycols.

11. Composition according to any one of claims 1 to 10 for its application in the therapeutic treatment of alopecia androgenetics.

12. Method of cosmetic treatment of scalp hair characterized in that at least one composition according to any one of claims 1 to 10 on alopecic areas of the scalp.

13. Use of spiroxasone for the preparation of a composition intended for the therapeutic treatment of alopecia androgenetics.

Multi-compartment device or "kit" characterized in that a first compartment contains spiroxasone and that a second compartment contains a solvent medium of the physiologically acceptable spiroxasone based on alcohol, the content of the second compartment being intended to be mixed with the contents of the first compartment at the time of use.
