Abstract
Patente de Invenção:<B>"MéTODO DE APERFEIçOAMENTO DA PRODUçãO DE BIOMASSA OU DE UM PRODUTO DESEJADO A PARTIR DE UMA CéLULA"<D>. A produção de biomassa ou de um produto desejado a partir de uma célula pode ser aperfeiçoada por indução da conversão de ATP para ADP sem efeitos primários sobre outros metabólitos ou funções celulares, o que se consegue por expressão de uma atividade ATPase não-acoplada, na referida célula e incubando a célula com um substrato adequado para produzir a referida biomassa ou produto. Isto é convenientemente realizado expressando-se na referida célula a parte solúvel (F~1~) de H^ +^-ATPase (tipo F~ 0~F~ 1~) ligada à membrana ou de uma parte de F1 apresentando atividade ATPase. O organismo do qual a ATPase F~ 1~ ou partes deste é derivado, ou em que a ATPase F~ 1~ ou partes deste é expressada, pode ser selecionado dentre organismos procariontes e eucariontes. Em particular, o DNA codificando F~ 1~ ou uma parte desta pode ser derivado de bactérias e microorganismos eucarióticos, tais como leveduras, outros fungos e linhas de células ou organismos superiores e pode ser selecionado do grupo que consiste do gene que codifica a subunidade <225> de F~ 1~ ou de uma parte desta e de várias combinações do gene ou parte com os genes que codificam as outras subunidades de F~ 1~ ou partes deste. O método pode ser usado, inter alia, para otimizar a formação da biomassa ou de um produto desejado por uma célula, expressando diferentes níveis de atividade ATPase não-acoplada na célula, incubando a célula em um substrato adequado, medindo a velocidade de conversão do substrato em biomassa ou o produto desejado em cada nível de expressão ATPase, e escolhendo um nível de expressão ATPase ao qual a velocidade de conversão é otimizada.
