Abstract
método para análise de sequência em tempo real de fragmentos de dna, oligonucleotídeo adaptador e kit para sequenciamento paralelo em tempo real e uso destes. a invenção está relacionada a um método para análise de sequência em tempo real de fragmentos de dna, compreendendo i) fornecer pelo menos uma amostra de fragmentos de dna para análise de sequência, ii) conectar um tipo de primeiro e segundo oligonucleotídeos adaptadores às extremidades 5' e 3' de uma fita de dna dos fragmentos de dna da amostra, respectivamente, em que um primeiro oligonucleotídeo adaptador compreende de 5' a 3' a) uma primeira sequência de ligação de células de fluxo, b) um sítio iniciador de sequenciamento de leitura 1, c) opcionalmente uma sequência aleatória, e d) uma sequência de código de barras específica da amostra, e um segundo oligonucleotídeo adaptador compreende de 5' a 3' d) uma sequência complementar à sequência de código de barras específica da amostra do primeiro oligonucleotídeo adaptador, c) opcionalmente uma sequência complementar à sequência aleatória, b) um sítio iniciador de sequenciamento de leitura 2 que pode ser (parcialmente) complementar ao sítio iniciador de sequenciamento de leitura 1, e a) uma segunda sequência de ligação de células de fluxo, em que o primeiro e o segundo oligonucleotídeos adaptadores de um tipo têm sequências de código de barras complementares, e sequenciamento dos fragmentos de dna compreendendo os oligonucleotídeos adaptadores conectados em um processo de sequenciamento por síntese.
