Abstract

MONOCOTILEDÔNEAS HETEROZIGOTAS PARA CENH3 E MÉTODOS DE USO DAS MESMAS PARA INDUÇÃO HAPLOIDE E EDIÇÃO SIMULTÂNEA DE GENOMA. Trata-se de plantas monocotiledôneas heterozigotas para histona 3 centromérica (CenH3) e, opcionalmente, que expressam construtos de edição de genes para uso na indução de haploides de uma planta alvo monocotiledônea e, opcionalmente, edição de genes de passagem. As plantas indutoras haploides monocotiledôneas são tipicamente compostas por células vegetais diploides com apenas um alelo que codifica uma proteína CENH3 funcional. As células vegetais diploides também podem incluir, por exemplo, um alelo CenH3 que codifica a proteína CENH3 não funcional. Em algumas modalidades, o alelo que codifica proteína CENH3 não funcional é uma mutação por mudança da matriz de leitura, alelo nulo de proteína, um alelo nulo de RNA ou uma combinação dos mesmos. Haploid monocot inducer plants are typically composed of diploid plant cells with only one allele encoding a functional CENH3 protein. Diploid plant cells may also include, for example, a CenH3 allele encoding the non-functional CENH3 protein. In some embodiments, the non-functional CENH3 protein-encoding allele is a frameshift mutation, protein null allele, an RNA null allele, or a combination thereof. The haploid monocot plant inducer may also include gene editing machinery, such as a site-directed nuclease and, optionally, a guide RNA stably expressed by cells of the monocot plant. Also provided are methods for inducing the formation of a target haploid monocot plant while optionally simultaneously modifying the genome of the target monocot plant.
