COMPOSIÇÃO DERMOFITOCOSMÉTICA A BASE DO EXTRATO DAS FOLHAS DE Cochlospermum regium (Schrank) Pilger EM COMBINAÇÃO COM O EXTRATO DA ENTRECASCA DE Myracrodruon urundeuva All, a presente invenção compreende a obtenção de um dermofitocosmético a partir do extrato das folhas de Cochlospermum regium em combinação com o extrato da entrecasca de Myracrodruon urundeuva, em diferentes apresentações, tais como gel, creme, solução, pomada, cápsula, máscara, sabonete, aerossóis, pó, pasta e pellet, livre ou nano e microencapsulados em polímeros naturais ou não. The formulation based on C. regium with M. urundeuva is a dermophytocosmetic with antimicrobial action in addition to anti-inflammatory and healing action.
Description
[1] . A presente invenção descreve uma composição aditiva do extrato de Cochlospermum regium com o extrato de Myracrodruon urundeuva, bem como a obtenção de um dermofitocosmético que contêm os dois substratos, atóxico à célula humana, com ações anti-inflamatória e cicatrizante, e ações contra diversos microrganismos, em especial: Staphylococcus aureus, Escherichia coli, Streptococcus pyogenes, Enterococcus spp., e Candida spp., podendo ser usado na profilaxia ou no tratamento de infecções causadas por bactérias e fungos.[1] . The present invention describes an additive composition of Cochlospermum regium extract with Myracrodruon urundeuva extract, as well as obtaining a dermophytocosmetic containing both substrates, nontoxic to human cells, with anti-inflammatory and healing actions, and actions against various microorganisms , in particular: Staphylococcus aureus, Escherichia coli, Streptococcus pyogenes, Enterococcus spp., and Candida spp., and can be used in the prophylaxis or treatment of infections caused by bacteria and fungi.

[3] . A composição da invenção podem conter além da mistura aditiva mencionada, outros princípios ativos voltados a proporcionar efeitos específicos. Uma lista meramente explicativa e não limitativa de ativos inclui um ou mais dentre cicatrizantes, antibióticos, analgésicos, hemostáticos, antissépticos, antioxidantes, outros antifúngicos, anti-inflamatórios e antialérgicos.[3] . Dessa forma, uma ferida, independente da origem (queimaduras, lacerações na pele, úlceras...), apresenta uma abertura de fácil acesso ao nosso corpo, tornando propício o surgimento de infecções na pele, que atrasam o processo de cicatrização e ainda podem evoluir para infecções mais graves resultando em efeitos sistêmicos. Os patógenos mais comuns associados com infecções de pele incluem Staphylococcus aureus, Escherichia coli, Enterococcus spp., Streptococcus pyogenes e Candida spp.[4] . Os tratamentos de origem natural têm ganhado destaque nas formulações dermocosméticas, visto que o desenvolvimento de formulações naturais é uma alternativa aos produtos já utilizados no mercado, reduzindo os efeitos colaterais e o desenvolvimento de patógenos resistentes aos medicamentos.[5] . Dessa forma, os dermocosméticos requerem um cuidado especial na sua formulação, excluindo substâncias tóxicas e sintéticas nocivas à saúde.[17] . Além disso, tem-se observado uma preocupação mundial crescente em relação às políticas de sustentabilidade, conservação de recursos naturais, redução no impacto ambiental e geração de resíduos, e a busca por produtos mais seguros e biodegradáveis. Neste contexto, a incorporação de extratos vegetais em formulações dermocosméticas – dermofitocosméticos – com a finalidade de se obter produtos naturais menos agressivos à saúde e ao meio ambiente, vem atraindo a atenção do mercado cosmético e dos consumidores. Outro fator importante para o desenvolvimento de formulações naturais é o aumento de microrganismos resistentes às substâncias já utilizadas no mercado. Dessa forma, os dermofitocosméticos se apresentam promissores, visto que o Brasil possui uma vasta biodiversidade, proporcionando o desenvolvimento de tecnologias de produção econômica, ecológica e segura.[18] . Another important factor for the development of natural formulations is the increase in microorganisms resistant to substances already used in the market. In this way, dermophytocosmetics are promising, since Brazil has a vast biodiversity, providing the development of economic, ecological and safe production technologies.
[19] . Os extratos vegetais são conhecidos por apresentar ação terapêutica através da presença de compostos bioativos que são produzidos a partir do seu metabolismo secundário. Uma espécie vegetal que se destaca, é a Cochlospermum regium (Mart. ex Schrank) Pilg., planta integrante da biodiversidade brasileira, conhecida como algodãozinho-do-cerrado. Sua raíz é utilizada na medicina popular para o tratamento de infecções, abcessos, afecções da pele, feridas internas e externas, e também no tratamento de úlceras, cravos, espinhas e manchas da pele. Porém, a retirada de suas raízes leva a planta à morte, o que deixa a espécie mais suscetível a extinções. Dessa forma, nosso grupo tem investigado as folhas de C. regium, e recentemente mostrou que elas possuem atividade antimicrobiana e antibiofilme frente aos microrganismos Staphylococcus aureus, Escherichia coli, Candida tropicalis e Cryptococcus gattii.[19] . Plant extracts are known to have therapeutic action through the presence of bioactive compounds that are produced from their secondary metabolism. A plant species that stands out is the Cochlospermum regium (Mart. ex Schrank) Pilg., a plant that is part of Brazilian biodiversity, known as cottonzinho-do-cerrado. Its root is used in folk medicine to treat infections, abscesses, skin disorders, internal and external wounds, and also to treat ulcers, blackheads, pimples and skin spots. However, the removal of its roots leads the plant to death, which makes the species more susceptible to extinction. Thus, our group has investigated the leaves of C. regium, and recently showed that they have antimicrobial and antibiofilm activity against the microorganisms Staphylococcus aureus, Escherichia coli, Candida tropicalis and Cryptococcus gattii.
[20] . Outra espécie que se destaca, é a Myracrodruon urundeuva All., árvore encontrada no Brasil, conhecida como aroeira. Esta planta é utilizada na medicina popular para o tratamento de inflamação, dor, infecções, corrimento vaginal, entre outras indicações. Nosso grupo tem investigado a entrecasca de M.urundeuva, e recentemente mostrou que ela possui atividade antifúngica e antibiofilme contra C. albicans.[20] . Portanto a presente invenção refere-se a uma composição compreendendo o extrato aquoso das folhas de Cochlospermum regium e o extrato aquoso da entrecasca de Myracrodruon urudeuva e veículos/excipientes farmaceuticamente aceitáveis, na forma de gel, creme, solução, emulsão, tintura, pomada, aerossóis, máscara, sabonete, pó ou pasta, para uso no tratamento de feridas, infecções e afecções de pele.[21] . Após esse período o extrato é filtrado em membrana filtrante, sendo o extrato final liofilizado e acondicionado em frasco escuro e conservado em refrigerador com temperatura entre 10 ºC a 25 ºC.[22] . [25] . Foi avaliada a combinação do extrato de C. regium com M. urundeuva frente a Staphyloccus aureus resistente à meticilina. O ensaio foi realizado por meio da técnica de microdiluição em caldo. As concentrações utilizadas variaram de 3,9 a 500 µg/mL para C. regium, e 7,8 a 1.000 µg/mL para M. urudeuva. A combinação, C. regium + M. urundeuva, formou uma matriz de combinações de diferentes concentrações em microplacas de 96 poços de fundo chato. As microplacas foram incubadas por 24 h a 37 °C. 3,9 a 500 µg/mL para C. regium, e 7,8 a 1.000 µg/mL para M. urudeuva.[25] . The concentrations used ranged from 3.9 to 500 µg/mL for C. regium, and from 7.8 to 1,000 µg/mL for M. urudeuva. The combination, C. regium + M. urundeuva, formed a matrix of combinations of different concentrations in flat-bottomed 96-well microplates. The microplates were incubated for 24 h at 37 °C. 3.9 to 500 µg/mL for C. regium, and 7.8 to 1,000 µg/mL for M. urudeuva.

A atividade antibacteriana da combinação do extrato e do antifúngico foi baseada no índice de concentração inibitória fracionária (ICIF) que é definido como: ICIF = (CIM de ECR em combinação/CIM de ECR sozinho) + (CIM de EMU em combinação/CIM de EMU sozinho). Os critérios para a interpretação foram: sinérgico (ICIF ≤ 0,5), efeito aditivo (0,5 < ICIF < 1), indiferente (1 ≤ ICIF < 4) ou antagonista (ICIF ≥ 4).[26] .
