Abstract
"CEPAS ESTáVEIS DE ZYMIINAS MOBILIS FERMENTADORAS DE XILOSE E ARABINOSE, TRANSPOSON PARA INSERçãO ESTáVEL DE GENES ALHEIOS DENTRO DE UM GENOMA BACTERIANO, VETOR PLASMìDEO CONTENDO O REFERIDO TRANSPOSON, BACTéRIA TRANSFORMADA COM O REFERIDO PLASMìDEO, VETOR PZB1862-IDHL-ARA ATCC COM NúMERO DE ASCENçãO PTA-1798, ELEMENTO TRANSPONìVEL MINI-TN5-TAL/TKT-AXILAXILB ATCC COM NúMERO DE ASCENçãO PTA-1795, CEPA DE BACTéRIA G8 ATCC COM NúMERO DE ASCENCçãO PTA-1796, CEPA DE BACTéRIA C25 ATCC COM NúMERO DE ASCENçãO PTA-1799, CEPA DE BACTéRIA AX ATCC COM NúMERO DE ASCENçãO PTA-1797, MISTURA DE REAçãO COMPREENDENDO MEIO FERMENTáVEL". A presente invenção inclui de forma breve um transposon para a inserção estável de genes alheios dentro de um genoma bacteriano, compreendendo pelo menos um operon apresentando estruturas de gene codificando enzimas selecionadas do grupo composto por xilAxilB, araBAD e tal/tkt, e pelo menos um promotor para a expressão das estruturas de gene na bactéria, um par de seq³ências de inserção invertidas, com os operons contidos dentro das seq³ências de inserção, e um gene de transposase localizado no lado das seq³ências de inserção. Apresenta-se ainda um vetor lançador plamídeo para a transformação de genes alheios dentro de um genoma bacteriano, compreendendo pelo menos um operon apresentando estruturas de gene codificando enzimas selecionadas do grupo composto por xilAxilB, araBAD e tal/tkt, pelo menos um promotor para expressão das estruturas de gene na bactéria, e pelo menos dois fragmento de DNAs apresentando homologia com um gene no genoma da bactéria a ser trasformada. O transposon e o vetor lançador são úteis para a construção de cepas Zymomonas mobilis significativamente diferentes de acordo com a presente invenção que são úteis para a conversão dos açúcares de pentose derivados de celulose em combustíveis e produtos químicos utilizando tecnologia de fermentação tradicional porque são estáveis para expressão em um meio não seletivo."ZYMYMES MOBILIS FERMENTING ZERO AND ARABINOSIS STORAGE CODES, TRANSPOSON FOR STABLE INSERTION OF ALIEN GENES WITHIN A BACTERIAL GENOME, PLASMID VECTOR CONTAINING THE TRANSPOSMED-PERMED BETTER TRANSFORMED WITH THE TRANSFORMED-PERMITORED PLASTER PTA-1798, TRANSPONABLE ELEMENT MINI-TN5-TAL / TKT-AXILAXILB ATCC WITH ASCENTION NUMBER PTA-1795, G8 ATCC BACTERIA CUP WITH PTA-1796 ASCENTION NUMBER, C25 ATC-C99 BUTTER CUP WITH N99 AX ATCC BACTERIA CEPA WITH RANGE NUMBER PTA-1797, REACTION MIX UNDERSTANDING A FERTILIZABLE MEDIA ". The present invention briefly includes a transposon for the stable insertion of foreign genes within a bacterial genome, comprising at least one operon having gene structures encoding enzymes selected from the group consisting of xylAxilB, araBAD and tal / tkt, and at least one. promoter for the expression of gene structures in the bacterium, a pair of inverted insertion sequences, with operons contained within the insertion sequences, and a transposase gene located on the side of the insertion sequences. Also provided is a plasmid launcher vector for transforming foreign genes into a bacterial genome, comprising at least one operon having gene structures encoding enzymes selected from the group consisting of xylAxilB, araBAD and tal / tkt, at least one promoter for expression. of the gene structures in the bacterium, and at least two DNA fragments showing homology to a gene in the genome of the bacterium to be transformed. The transposon and the launcher vector are useful for constructing significantly different Zymomonas mobilis strains according to the present invention which are useful for converting cellulose derived pentose sugars into fuels and chemicals using traditional fermentation technology because they are stable for expression in a non-selective medium.
