L'objet de la présente invention est un nouveau procédé d'obtention de la N,N-diéthyl-5-méthyl[1,2,4]triazolo[1,5-a] pyrimidine-7-amine, caractérisé en ce qu'on réalise une diéthylation

avec de l'acétaldéhyde et avec une réduction catalytique, ou en utilisant un agent réducteur, tel qu'un hydrure alcalin.

dine-7-amine est un composé connu répondant à la formule structurale I :

Ce produit a déjà fait l'objet de brevets (brevet britannique n[deg.] 1.148.629, brevet d'Allemagne de l'Est n[deg.] 55.967) et de publications (Tenor e col., Pharmazie 26, 534-1971; Pfeifer et col., Pharmazie 26, 539-549, 1971; idem 27, 752-1972; Takahashi e col., Arzneim. Forsch. 32, 214-1982). Ce produit est utilisé en thérapeutique comme vasodilatateur coronarien sous le nom de de Trapidil.

l'on réduit en présence d'acétaldéhyde suivant une réaction d'alkylation réductrice, dont le schéma est le suivant :

Dans la réaction mentionnée ci-dessus, la réduction peut être menée de diverses manières, soit comme réduction catalytique classique, soit en utilisant, comme réducteurs, divers types d'hydrures alcalins, comme le borohydrure de sodium ou le cyanoborohydrure ou d'autres encore.
Le procédé faisant l'objet de la présente invention est beaucoup plus simple et est réalisable avec les appareils ordinaires d'un laboratoire de chimie organique, ou avec les installations habituelles de l'industrie chimique. Le procédé donne des rendements optima et un produit brut qui est déjà d'une pureté importante (égale ou supérieure à 95%).
EXEMPLE 1.

mole) et de l'éthanol à 95% (200 ml), on refroidit le mélange à 2-5[deg.], puis on introduit de l'acétaldéhyde (23,3 g;0,053 mole; 30 ml) et du carbone palladié à 10% (2,0 g). Le récipient est fermé et mis sous vide, puis chargé d'hydrogène; cette dernière opération est répétée deux fois, puis on procède à l'hydrogénation à 3,5 atmosphères, jusqu'à ce que la quantité d'hydrogène soit consommée (1 mole). Le temps nécessaire pour l'hydrogénation est d'environ 2-4 heures. Après avoir éliminé tout l'hydrogène du récipient par introduction d'azote, on filtre la solution éthanolique à la trompe pour chasser le catalyseur qu'on lave sur filtre avec de l'éthanol (environ
20 ml).
Le filtrat et le liquide de lavage réunis sont évaporés sous pression réduite, pour obtenir de la sorte, comme résidu, le produit brut d'une pureté d'environ 95%. Ce produit brut peut être cristallisé dans du n-heptane. On obtient le produit pur (38,6 g; 0,19 mole; rendement de 94,0%) qui a un point de fusion de 104-105[deg.]C qui ne diminue pas lors d'un mélange avec un échantillon authentique du produit. Le chlorhydrate obtenu par les méthodes habituelles a un point de fusion de 213[deg.]C.
EXEMPLE 2.
Dans un ballon (500 ml), on dissout de la 5-méthyl-

méthanol (200 ml). A la solution, on ajoute de l'aldéhyde acétique
(2,86 g; 0,065 mole; 3,6 ml) et une quantité catalytique d'acide p-toluènesulfonique (500 mg). Tout en agitant, on refroidit le mélange à 10-15[deg.]C, puis par petites portions, on ajoute du cyanoborohydrure de sodium en maintenant une agitation efficace. Après l'addition, on agite encore pendant environ 2 heures, puis on élimine le solvant sous pression réduite et on reprend le résidu avec du chloroforme, on alcalise avec un peu d'ammoniac gazeux, on filtre et on évapore le filtrat sous pression réduite. On obtient le produit brut qui cristallise dans du n-heptane, pour donner ainsi le produit analytiquement pur (5,6 g; 0,27 mole; rendement de 90,9%), qui est identique à celui obtenu dans l'Exemple 1 et à un échantillon authentique de ce produit.
EXEMPLE 3.
En opérant de façon analogue à l'Exemple 2 mais en utilisant, comme réducteur, le borohydrure de sodium, on obtient

In the reaction mentioned above, the reduction can be carried out in various ways, either as a conventional catalytic reduction, or by using, as reducing agents, various types of alkaline hydrides, such as sodium borohydride or cyanoborohydride or others. .
The process which is the subject of the present invention is much simpler and can be carried out with the ordinary apparatus of an organic chemistry laboratory, or with the usual installations of the chemical industry. The process gives optimum yields and a crude product which is already of high purity (equal to or greater than 95%).
EXAMPLE 1.

mole) and 95% ethanol (200 ml), the mixture is cooled to 2-5 [deg.], then acetaldehyde (23.3 g; 0.053 mole; 30 ml) and carbon are introduced. 10% palladium (2.0 g). The container is closed and put under vacuum, then charged with hydrogen; this last operation is repeated twice, then the hydrogenation is carried out at 3.5 atmospheres, until the quantity of hydrogen is consumed (1 mole). The time required for the hydrogenation is approximately 2-4 hours. After having removed all the hydrogen from the container by introducing nitrogen, the ethanolic solution is filtered by suction to remove the catalyst which is washed on a filter with ethanol (approximately
20 ml).
The combined filtrate and washing liquid are evaporated under reduced pressure, so as to obtain, as residue, the crude product of a purity of about 95%. This crude product can be crystallized from n-heptane. The pure product is obtained (38.6 g; 0.19 mole; yield of 94.0%) which has a melting point of 104-105 [deg.] C which does not decrease when mixed with a sample authentic product. The hydrochloride obtained by the usual methods has a melting point of 213 [deg.] C.
EXAMPLE 2.
In a flask (500 ml), 5-methyl-

methanol (200 ml). To the solution, acetic aldehyde is added
(2.86 g; 0.065 mole; 3.6 ml) and a catalytic amount of p-toluenesulfonic acid (500 mg). While stirring, the mixture is cooled to 10-15 [deg.] C, then in small portions, sodium cyanoborohydride is added while maintaining effective stirring. After the addition, the mixture is stirred again for approximately 2 hours, then the solvent is removed under reduced pressure and the residue is taken up with chloroform, it is basified with a little ammonia gas, it is filtered and the filtrate is evaporated under reduced pressure . The crude product is obtained which crystallizes from n-heptane, thereby giving the analytically pure product (5.6 g; 0.27 mole; yield of 90.9%), which is identical to that obtained in Example 1 and an authentic sample of this product.

By following a procedure analogous to Example 2 but using sodium borohydride as the reducing agent,
