La présente invention concerne un procédé permettant de mesurer l'activité de la cathepsine B en présence d'autres enzymes protéolytiques, telles que la trypsine. La présente invention concerne, en outre, de nouveaux substrats qui sont hautement spécifiques de la cathepsine B. Le procédé selon l'invention est d'un intérêt particulier pour la détection de niveaux anormaux de l'activité de la cathepsine
B dans les fluides corporels, tels que le sang humain et
dans les tissus corporels.
Les protéinases sont des enzymes qui digèrent des protéines et des chaînes polypeptidiques. Ces enzymes sont hautement sélectives pour la coupure de sites des substrats polypeptidiques. Cette activité enzymatique est précisément régulée dans un système biologique normal. Une défaillance dans cette régulation peut avoir des conséquences biologiques sérieuses. De nombreux cas de maladies ont été attribués à de telles défaillances ou dérèglements dans l'activité des protéinases (ainsi que décrit par A.J. Barrett dans Proteinases in Mammalian Cells and Tissues, North-Holland, New York, édit. (1979) ; par A.J. Barrett et J.K. Mc Donald dans Mammalian Proteases : A Glossary and Bibliography (vol 1 (1980), Academic Press, New-York)).
La cathepsine B est une protéinase (thiol) cystéine lysosomique normale. Il a été récemment montré (A.J. Barrett et J.K. Mc Donald dans l'ouvrage précité ; M. Sandler dans Enzyme Inhibitors as drugs, édit. (1980), University Park Press, Baltimore)) qu'une défaillance apparente dans
la régulation de l'activité de la cathepsine B peut conduire
à divers cas de maladies. Plusieurs maladies en relation
avec une baisse de collagène et une dégradation de la glycoprotéine structurelle sont liées à une activité accrue de
la cathepsine B. En particulier, l'existence d'une corréla-tion entre l'activité de la cathepsine B et la maladie métastatique a été suggérée par plusieurs recherches, dont une élévation de l'activité de la cathepsine B, ou de la cathepsine analogue à la cathepsine B, dans le sang des patients atteints d'une maladie métastatique [ainsi que décrit par R.J. Pietras et al. Obstet. Gynecol. 52, 321-327

Aussi, un procédé permettant de déterminer de façon précise et sélective l'activité de la cathepsine B dans les fluides corporels de mammifères faciliterait le diagnostic, tel que par détection ou surveillance.
Les dosages de diverses activités d'enzymes sont des procédés de routine dans un laboratoire clinique. De tels dosages comportent, par exemple, la mise en contact de l'échantillon contenant l'enzyme avec un substrat de synthèse (défini comme la substance ou le composé sur lequel agit l'enzyme) qui est coupé sélectivement par cet enzyme en produits qui changent de couleurs :

Ainsi, des substrats pour la cathepsine B seraient des peptides ou des composés analogues aux peptides qui, par coupure, permettent l'analyse des produits. Des méthodes de dosage qui ont été utilisées pour la cathepsine B [ainsi que décrit par A.J. Barrett et par R.J. Pietras dans les ouvrages précités ; et par A.J. Barrett dans Biochem. J. 187,
909-912 (1980)] ont fait appel à des substrats peptidiques contenant un ou plusieurs amino-acides arginyle ou lysyle fixés sur un groupe indicateur pouvant être coupé par une enzyme (par ex. Bz - Arg - NA, CBZ - Lys - PNP, CBZ - Arg Arg - MNA, CBZ - Ala - Arg - Arg - AFC, CBZ - Phe - Arg AMC, voir la signification des abréviations ci-dessous).
Bien que la coupure sélective des substrats de synthèse par la cathepsine B n'est pas encore bien comprise, ces substrats couramment utilisés peuvent, par la nature de leurs groupes chargés positivement, être également coupés par les protéases analogues à la trypsine (sérine). Ainsi, les dosages
de la cathepsine B utilisant un des ces substrats de lysyle ou d'arginyle dans les fluides corporels de mammifères sont rendus compliqués par la présence de protéases analogues à
la trypsine en quantités importantes. Ainsi, on ne sait pas comment obtenir une sélectivité élevée pour la détection de l'activité de la cathepsine B en présence de trypsine ou d'enzymes analogues à la trypsine.
Aussi, l'un des buts de la présente invention est de proposer des substrats hautement spécifiques pour la cathepsine B. Un autre but de l'invention est de doser sélectivement l'activité de la cathepsine B en présence d'autres enzymes protéolytiques, par exemple de trypsine. L'invention a encore pour but de déterminer l'activité de
la cathepsine B dans les fluides corporels et les tissus de mammifères. D'autres buts de l'invention apparaîtront à la lecture de la description de l'invention.
La présente invention fournit un procédé permettant de tester l'activité de la cathepsine B dans les fluides corporels ou les tissus de mammifères en mesurant
la vitesse avec laquelle est libéré un groupe indicateur (X) par coupure d'un substrat peptidique de synthèse hautement sélectif pour la cathepsine B. Des substrats contenant des groupes indicateurs appropriés pour la détection de la cathepsine B ont pour formule générale :

mule générale, CBZ est utilisé pour Z et AMC est utilisé pour X. L'anhydride mixte se réfère au type de méthode de couplage. Les réactions 1 à 4 sont décrites en détail cidessous.
(1) Réaction CBZ - Cl : Dans quelques cas, les dérivés CBZ-R étaient disponibles dans le commerce, dans les autres cas, ils ont été préparés par la méthode suivante : l'amino-acide R (25 mmoles) et 50 mmoles de bicarbonate de sodium ont été dissous dans 50 ml d'eau dans un ballon de 100 ml. Du carbo-benzyloxychlorure (25,5 mmoles) a été ajouté sous agitation à la température ambiante en 5 parties aliquotes pendant une heure. La solution a été agitée ensuite pendant encore deux heures, elle a été extraite par l'éther deux fois, puis versée goutte à goutte dans 100 ml de 1 M HC1. Le produit
qui précipite tout d'abord est semi-solide, puis il se solidifie en le laissant reposer. Le produit brut a été lavé avec de l'eau, séché et utilisé sans purification ultérieure. Le rendement moyen était de 55 %.
(2) Couplage avec AMC : On a additionné dans un ballon de
50 ml, 11,4 mmoles d'amino-acide R, bloqué, 15 ml de THF anhydre et 11,54 mmoles de TEA. Après dissolution du composé, la solution a été refroidie sur de la glace et 11,4 mmoles
de chloroformate d'isobutyle ont été ajoutés. La solution
a été agitée pendant 10 minutes sur de la glace et ensuite une solution refroidie de 11,4 mmoles d'AMC dans 21 ml de
DMF a été ajoutée. On a continué l'agitation sur de la glace pendant une heure, puis à la température ambiante pendant
une nuit. Le mélange réactionnel a été filtré et le THF a
été éliminé du filtrat dans un évaporateur rotatif. Le résidu a été placé dans un entonnoir à décantation de 125 ml
avec 20 ml de CH2C12 et 30 ml d'une solution aqueuse de HC1
à 10 % , puis secoué vigoureusement. Après extraction de la phase organique avec une seconde partie aliquote d'acide aqueux, le produit a précipité dans la phase organique. La phase aqueuse a été décantée et le précipité a été recueilli par filtration et lavé avec de l'éther. Le produit a été séché et utilisé sans purification ultérieure. Le rendement moyen était de 41 %.
(3) Déblocage : Le groupe de blocage N-carbobenzyloxy a été enlevé de Z-R.-AMC par traitement par le bromure d'hydrogène/ solution d'acide acétique glacial (HBr-HOAc) ou par hydrogénation catalytique. Dans les deux exemples, on a utilisé
-t-BOC au lieu de CBZ et le déblocage a été réalisé par traitement par l'acide formique.
HBr/HOAc : Dans un ballon de 500 ml, on a ajouté 3,8 mmoles

15 minutes. Le mélange réactionnel a été dilué avec 500 ml d'éther et le précipité résultant a été recueilli par filtration sous azote. Le produit a été remis en suspension trois fois dans 100 ml d'éther et refiltré. Le produit a été séché et utilisé sans purification ultérieure. Le rendement moyen était de 98 %.
Hydrogénation catalytique : Dans un ballon sous pression de
500 ml, on a ajouté 8,5 mmoles de Z-R1-AMC, 4 g de perles
de palladium-polyéthylèneimine (Pd-PEI) et 200 ml de méthanol. Le ballon a été mis sous pression avec 1,4 kg/cm<2> d'hydrogène et secoué pendant six heures, puis a été laissé reposer pendant 18 heures de plus. Après que les perles de catalyseur se soient déposées, l'alcool a été décanté et les perles ont été lavées deux fois avec 50 ml de méthanol. Afin de faciliter la purification, le sel de chlorhydrate a été préparé en ajoutant 12,5 ml de HC1 aqueux 1 N. Le solvant
a été éliminé sous vide et le résidu a été redissous et réévaporé deux fois avec 50 ml d'alcool dénaturé. Le résidu a été trituré avec de l'alcool dénaturé et laissé reposer

Acide formique : Dans un ballon de 500 ml, on a ajouté

a été utilisé sans autre purification.
(4) Couplage des peptides : Dans un ballon de 50 ml ont été

Après dissolution des composés, les deux solutions ont été refroidies sur de la glace, et 3,2 mmoles de chloroformate d'isobutyle ont été ajoutés à la solution de CBZ-R2. refroidie a été ajoutée au mélange réactionnel CBZ-R2. On a poursuivi l'agitation pendant une heure sur de la glace, puis pendant la nuit à la température ambiante. Le mélange réactionnel a alors été ajouté goutte à goutte sous agitation dans 950 ml d'une solution aqueuse de bicarbonate de sodium à 5 %. Le précipité a été recueilli par filtration et lavé trois fois avec de l'eau. Le rendement brut moyen était de 90 %.
Purification : Les produits finaux ont été purifiés par la recristallisation qui suit et dans quelques cas par des recristallisations additionnelles et HPLC. Le produit

acétique glacial sous agitation et en chauffant doucement. On a ajouté 54 ml d'acétonitrile et cette solution a été chauffée sous reflux. La solution très chaude a été filtrée et 80 ml d'eau ont été ajoutés au filtrat par petites aliquotes en chauffant et sous agitation. Un précipité blanc qui s'est formé lors du refroidissement, a été filtré et lavé à l'eau. Le produit recristallisé a été séché sous vide et le rendement de cristallisation moyen était de 80 %.
Le rendement global et les modifications de synthèse sont donnés pour chaque exemple, ci-dessous.
Dosage :
l'activité de la cathepsine B est détectée en contrôlant la vitesse de libération du groupe indicateur X. Le groupe indicateur peut être un fragment de molécule dont la fluorescence ou l'absorbance change lorsqu'il est coupé. Tout indicateur connu, tel que AMC, HMC, AFC, AQ ou PNA peut être utilisé. En variante, on peut également utiliser un indicateur marqué radioactivement.
Les procédés de dosage de l'enzyme, la cathepsine B, suivent les procédés standards et les principes généraux utilisés pour les enzymes (ainsi que décrit par A.J. Barrett dans l'ouvrage précité ; par W.P. Jencks (1969) dans Catalysis in Chemistry and Enzymology, Mc Graw Hill, New York). Une condition cinétique à l'état d'équilibre est réalisée
en utilisant des rapports de concentration enzyme/substrat appropriés (la concentration du substrat et sa constante Michaelis dépassent largement la concentration de l'enzyme) de manière à obtenir une valeur de vitesse précise dans la plus petite échelle de temps du laboratoire.
Le dosage de l'activité de la cathepsine B est réalisé en ajoutant une faible quantité d'un échantillon de

aqueuse ayant un domaine de pH pour lequel la cathepsine B est active, couramment pH 5-6,5, de préférence pH 5,5 contenant EDTA et un activateur thiol (anti-oxydant biologique
ou autre agent réducteur convenable) tel que cystéine ou dithiothréitol. Après une courte période d'activation par exemple de 1 à 2 minutes, le substrat dissous dans un solvant compatible, tel que méthanol, éthanol, acétonitrile ou DMF,

Les résultats de l'interaction enzymatique avec les exemples de substrats 1 à 17 et les produits classiques du commerce CBZPhe-Arg-AFC (Exemple 18), CBZ-Ala-Arg-Arg-AFC (Exemple 19) et Bz-Val-Lys-Lys-Arg-AFC (exemple 20) sont donnés dans le tableau 1.

Conformément à un autre mode de réalisation de l'invention, l'activité de la cathepsine B peut être mesurée qualitativement ou semi-quantitativement par mise en contact d'un échantillon préparé d'un fluide provenant d'un mammifère avec un substrat approprié selon l'invention. PROCESS FOR DETERMINING CATHEPSIN B IN THE PRESENCE OF OTHER PROTEOLYTIC ENZYMES AND COMPOUNDS USEFUL THEREOF.
The present invention relates to a method for measuring the activity of cathepsin B in the presence of other proteolytic enzymes, such as trypsin. The present invention further relates to novel substrates which are highly specific for cathepsin B. The method according to the invention is of particular interest for the detection of abnormal levels of cathepsin activity
B in bodily fluids, such as human blood and
in body tissue.
Proteinases are enzymes that digest proteins and polypeptide chains. These enzymes are highly selective for cleavage of sites on polypeptide substrates. This enzymatic activity is precisely regulated in a normal biological system. A failure in this regulation can have serious biological consequences. Many cases of disease have been attributed to such failures or disturbances in the activity of proteinases (as described by AJ Barrett in Proteinases in Mammalian Cells and Tissues, North-Holland, New York, ed. (1979); by AJ Barrett and JK Mc Donald in Mammalian Proteases: A Glossary and Bibliography (vol 1 (1980), Academic Press, New-York)).
Cathepsin B is a normal lysosomal cysteine proteinase (thiol). It has recently been shown (A.J. Barrett and J.K. Mc Donald in the aforementioned book; M. Sandler in Enzyme Inhibitors as drugs, ed. (1980), University Park Press, Baltimore)) that an apparent failure in
regulation of cathepsin B activity can lead to
to various cases of disease. Several related diseases
with a fall in collagen and a breakdown of the structural glycoprotein are linked to an increased activity of
cathepsin B. In particular, the existence of a correlation between the activity of cathepsin B and metastatic disease has been suggested by several studies, including an increase in the activity of cathepsin B, or of the cathepsin analogous to cathepsin B, in the blood of patients with metastatic disease [as described by RJ Pietras et al. Obstet. Gynecol. 52, 321-327

Also, a method for precisely and selectively determining the activity of cathepsin B in mammalian body fluids would facilitate diagnosis, such as by detection or monitoring.
Assays of various enzyme activities are routine procedures in a clinical laboratory. Such assays include, for example, contacting the sample containing the enzyme with a synthetic substrate (defined as the substance or compound on which the enzyme acts) which is selectively cut by this enzyme into products which change colors:

Thus, substrates for cathepsin B would be peptides or compounds analogous to the peptides which, by cleavage, allow the analysis of the products. Assay methods which have been used for cathepsin B [as described by A.J. Barrett and by R.J. Pietras in the aforementioned works; and by A.J. Barrett in Biochem. J. 187,
909-912 (1980)] used peptide substrates containing one or more arginyl or lysyl amino acids attached to an indicator group which can be cut by an enzyme (eg Bz - Arg - NA, CBZ - Lys - PNP , CBZ - Arg Arg - MNA, CBZ - Ala - Arg - Arg - AFC, CBZ - Phe - Arg AMC, see the meaning of the abbreviations below).
Although the selective cleavage of synthetic substrates by cathepsin B is not yet well understood, these commonly used substrates can, by the nature of their positively charged groups, also be cleaved by proteases analogous to trypsin (serine). So the dosages
of cathepsin B using one of these lysyl or arginyl substrates in mammalian body fluids are complicated by the presence of proteases analogous to
trypsin in significant quantities. Thus, it is not known how to obtain a high selectivity for the detection of the activity of cathepsin B in the presence of trypsin or of enzymes analogous to trypsin.
Also, one of the aims of the present invention is to provide highly specific substrates for cathepsin B. Another aim of the invention is to selectively measure the activity of cathepsin B in the presence of other proteolytic enzymes, by example of trypsin. Another object of the invention is to determine the activity of
cathepsin B in body fluids and mammalian tissues. Other objects of the invention will appear on reading the description of the invention.
The present invention provides a method for testing the activity of cathepsin B in body fluids or mammalian tissue by measuring
the speed with which an indicator group (X) is released by cutting a synthetic peptide substrate highly selective for cathepsin B. Substrates containing indicator groups suitable for the detection of cathepsin B have the general formula:

General mule, CBZ is used for Z and AMC is used for X. Mixed anhydride refers to the type of coupling method. Reactions 1 to 4 are described in detail below.
(1) CBZ-Cl reaction: In some cases, the CBZ-R derivatives were commercially available, in the other cases, they were prepared by the following method: the amino acid R (25 mmol) and 50 mmol sodium bicarbonate was dissolved in 50 ml of water in a 100 ml flask. Carbo-benzyloxychloride (25.5 mmol) was added with stirring at room temperature in 5 aliquots for one hour. The solution was then stirred for another two hours, it was extracted with ether twice, then poured dropwise into 100 ml of 1 M HCl. The product
which first precipitates is semi-solid, then it solidifies leaving it to stand. The crude product was washed with water, dried and used without further purification. The average yield was 55%.
(2) Coupling with AMC: We added in a flask of
50 ml, 11.4 mmol of amino acid R, blocked, 15 ml of anhydrous THF and 11.54 mmol of TEA. After dissolution of the compound, the solution was cooled on ice and 11.4 mmol
isobutyl chloroformate was added. The solution
was stirred for 10 minutes on ice and then a cooled solution of 11.4 mmol of AMC in 21 ml of
DMF has been added. Stirring was continued on ice for one hour, then at room temperature for
a night. The reaction mixture was filtered and THF was
was removed from the filtrate in a rotary evaporator. The residue was placed in a 125 ml separatory funnel
with 20 ml of CH2C12 and 30 ml of an aqueous solution of HC1
at 10%, then shaken vigorously. After extraction of the organic phase with a second aliquot of aqueous acid, the product precipitated in the organic phase. The aqueous phase was decanted and the precipitate was collected by filtration and washed with ether. The product was dried and used without further purification. The average yield was 41%.
(3) Unblocking: The N-carbobenzyloxy blocking group was removed from Z-R.-AMC by treatment with hydrogen bromide / glacial acetic acid solution (HBr-HOAc) or by catalytic hydrogenation. In the two examples, we used
-t-BOC instead of CBZ and the release was carried out by treatment with formic acid.
HBr / HOAc: In a 500 ml flask, 3.8 mmol was added

of the sample, the solution was stirred for a further
15 minutes. The reaction mixture was diluted with 500 ml of ether and the resulting precipitate was collected by filtration under nitrogen. The product was resuspended three times in 100 ml of ether and re-filtered. The product was dried and used without further purification. The average yield was 98%.
Catalytic hydrogenation: In a pressure balloon
500 ml, 8.5 mmol of Z-R1-AMC, 4 g of beads were added
of palladium-polyethyleneimine (Pd-PEI) and 200 ml of methanol. The activity of cathepsin B is detected by monitoring the rate of release of the indicator group X. The indicator group can be a fragment of a molecule whose fluorescence or absorbance changes when it is cut. Any known indicator, such as AMC, HMC, AFC, AQ or PNA can be used. Alternatively, a radioactively marked indicator can also be used.
The methods for assaying the enzyme, cathepsin B, follow the standard methods and general principles used for enzymes (as described by AJ Barrett in the aforementioned work; by WP Jencks (1969) in Catalysis in Chemistry and Enzymology , Mc Graw Hill, New York). A kinetic condition at steady state is achieved
using appropriate enzyme / substrate concentration ratios (the concentration of the substrate and its Michaelis constant greatly exceeds the concentration of the enzyme) so as to obtain a precise speed value in the smallest time scale of the laboratory.
The assay of the activity of cathepsin B is carried out by adding a small amount of a sample of

aqueous having a pH range for which cathepsin B is active, commonly pH 5-6.5, preferably pH 5.5 containing EDTA and a thiol activator (biological antioxidant

DOSAGE EXAMPLES
Purified cathepsin B was prepared from pig liver and neoplastic human liver using standard salting-out methods. Dosing has been carried out

fresh buffer at 37 [deg.] C containing the thiol activator in a 5 x 5 mm fluorimeter cuvette and incubated at 37 [deg.] C for 2 minutes, 20 μl of the substrate in DMF at a concentration ranging from 0 0.05 mM to 2 mM were then added with stirring. The increase in fluorescence was monitored over time at 37 [deg.] C for several minutes using a Perkin Elmer LS-5 spectrofluorimeter. For the AMC, the excitation wavelength was 350 nm, the emission wavelength was 460 nm and for the AFC the excitation wavelength was 385 nm and the emission wavelength of 490 nm. The fluorimeter was adjusted as follows: slit width 5 and 5 mm, response of filter 1, and the scale factor was set to 0-1.0.
The results of the enzymatic interaction with the examples of substrates 1 to 17 and the conventional commercial products CBZPhe-Arg-AFC (Example 18), CBZ-Ala-Arg-Arg-AFC (Example 19) and Bz-Val-Lys -Lys-Arg-AFC (Example 20) are given in Table 1.

The peptide substrates of the invention avoid or reduce the problems of interference due to enzymes analogous to trypsin, and have a sensitivity towards cathepsin B of 60 to 350 times larger than the peptides of Examples 18 to 20.
In accordance with another embodiment of the invention, the activity of cathepsin B can be measured qualitatively or semi-quantitatively by bringing a prepared sample of a fluid from a mammal into contact with a suitable substrate according to the invention. Typical clinical laboratory practices for obtaining such analyzes may include, for example, cytological analysis in which cells, tissues and organ samples are contacted with the substrate and other reagents for the purpose of selective staining. Other characteristic clinical analyzes include the use of a test strip which contains the substrate incorporated in a suitable, relatively inert support, which may be a cellulosic or nonwoven material, or a combination thereof.
When the sample is brought into contact with a test strip, the sample is absorbed by the test strip, causing a color change allowing a relatively rapid analysis.
