Des exemples de groupes acyle que R'peut représenter sont les suivants : formyle, acétyle, trifluoroacétyle, propionyle, butyryle, adamantyl-carbonyle, benzoyle, phénylacétyle et cinnamyle. Les groupes de type aliphatique et uréthane aromatique que R'peut représenter sont de préférence les groupes mentionnés comme groupe X préféré protégeant l'atome d'azote terminal de type aliphatique et uréthane aromatique.
Les sels des peptides de l'invention avec des acides ou des bases pharmaceutiquement acceptables font partie du cadre de l'invention. De tels sels acides d'addition peuvent dériver d'une variété d'acides organiques et inorganiques comme les acides sulfurique, phosphorique, chlorhydrique, bromhydrique, iodhydrique, nitrique, sulfamique, citrique, lactique, pyruvique, oxalique, maléique, succinique, tartrique, cinnamique, acétique, trifluoroacétique, benzoique, salicylique, gluconique, ascorbique et les acides apparentés. De tels sels d'addition basiques peuvent dériver d'une variété de bases organique et inorganique comme l'hydroxyde de sodium, l'hydroxyde de potassium, la diéthylamine, la triéthylamine et la dicyclohexylamine.
La synthèse des peptides de l'invention s'accomplit par des méthodes classiques en solution. La synthèse consiste essentiellement en des condensations successives appropriées d'amino-acides protégés ou de peptides. La condensation est effectuée de façon que les peptides résultant possèdent la séquence désirée des 4 ou 5 restes d'amino-acides. Les amino-acides et les peptides, qui sont condensés selon des méthodes connues en soi dans la chimie des polypeptides, ont ceux de leurs groupes amino et carboxylique qui ne sont pas impliqués dans la formation de la liaison peptidique bloqués par un groupe protecteur convenable. Les groupes protecteurs sont susceptibles d'être éliminés par acidolyse, saponification ou hydrogénolyse.

La condensation entre un groupe amino d'une molécule et un groupe carboxyled'une autre molécule pour former la liaison peptidique peut s'effectuer au moyen d'un dérivé acyle activé comme un anhydride mixte, un azide ou un ester activé, ou par condensation directe entre un groupe amino libre et un groupe carboxyle libre, en présence d'un agent de condensation comme le dicyclohexylcarbodiimide, seul ou avec un agent empêchant la racémisation, comme le N-hydroxysuccinimide ou le 1-hydroxybenzotriazole.
Les dérivés hydrazido ou hydrazido substitués conformes à l'invention sont préparés par condensation du peptide N-protégé ou de l'amino-acide avec une hydrazine convenablement substituée, comme le benzylcarbazate, le t-butylcarbazate, l'adamantylcarbazate, la phénylhydrazine ou l'adamantylhydrazine, ou par réaction du peptide N-protégé ou de l'hydrazide d'amino-acide avec un agent convenable d'alkylation comme un chlorure d'alkyle, ou avec un agent convenable d'acylation comme le benzylchloroformate, le t-butylfluoroformate, le di-t-butyl-dicarbonate ou l'adamantylfluoroformate. La condensation peut être effectuée dans un solvant comme le diméthylformamide, la pyridine, l'acétonitrile, le tétrahydrofuranne ou la N-méthyl-2- - pyrrolidone. La température de réaction peut être entre - 300C et la température ambiante.
La durée de la réaction est généralement de 1 à 120 heures. Le schéma de la synthèse, les groupes protecteurs et les agents de condensation sont choisis de manière à éviter le risque de racémisation.

Les réactions d'élimination des protections sont effectuées selon des méthodes connues en soi dans la chimie des polypeptides. Les peptides dans lesquels Y représente OR sont préparés, par exemple, à partir de l'amino-acide à C-terminal estérifié par un alcool approprié. Les peptides dans lesquels Y représente OH peuvent être préparés, par exemple, par hydrolyse de peptides dans lesquels Y représente OR. Les peptides dans lesquels Y représente NH2'NHR ou NR peuvent être préparés par ammoniolyse des esters correspondants ou à partir d'un amino acide à C-terminal amidaté à l'aide d'une amine appropriée.
Activité biologique
Les composés de l'invention possèdent une activité intéressante favorisant la croissance chez les animaux comme indiqué par un ensemble d'essais in vivo- - in vitro sur la synthèse des protéines des tissus du foie comme décrit par K. Kammerer et A. Dey-Hazra dans Veterinär-Medizinische Nachrichten, 99-112 (1980). Ils présentent aussi une activité endocrinologique intéressante comme la libération de la prolactine et de l'hormone lutéinisante.
Evaluation de l'activité favorisant la croissance
Les peptides de l'invention, par exemple H-Phe-Pro-Pro-Trp-Met-NH, ont été essayés sur des rats à des doses quotidiennes allant de 1 à 100 ng/kg administrées par voie sous-cutanée pendant une période de une à quatre semaines. Ils ont montré un accroissement de la synthèse des protéines du foie comme déterminé par la méthode de Kämmerer et Dey-Hazra et une augmentation du poids du corps à la fin des quatre semaines d'expérimentation. De plus, le rapport de conversion de l'alimentation était amélioré.
Pour l'usage vétérinaire, l'administration des composés de l'invention aux animaux produisant de la nourriture peut être effectuée dans une gamme de doses allant de 1 à 100 ng/kg, selon les techniques vétérinaires usuelles pour le traitement à l'aide d'agents anabolisants

ou favorisant la croissance, nommément par implantation sous-cutanée ou sous forme stabilisée convenable en mélange avec l'alimentation. En conséquence, l'invention couvre aussi toute composition pharmaceutique ou vétérinaire comprenant un composé de l'invention ou un sel acceptable en pharmacie ou en pratique vétérinaire de ce composé en mélange avec un diluant ou un véhicule acceptable en pharmacie ou en pratique vétérinaire ; de plus, ces préparations peuvent être du type à libération contrôlée ou retardée de l'ingrédient actif.

On donnera maintenant plusieurs exemples de préparations de composés de l'invention, sans intention limitative.
Les valeurs Rf ont été déterminées sur des plaques pré-enduites d'une couche d'une épaisseur de 0,25 mm de gel de silice 60 F254 (Merck) d'une longueur de 20 cm, à l'aide des systèmes suivants de développement : Système A : benzène/acétate d'éthyle/acide acétique/eau =
100/100/20/10 en volume (phase supérieure).
Système B : benzène/acétate d'éthyle/acide acétique/eau =
100/100/40/15 en volume (phase supérieure).
Système C : n-butanol/acide acétique/eau = 4/1/1 en volume.
Système D : chloroforme/méthanol/hydroxyde d'ammonium à 32% = 55/45/20 en volume.
"E. Merck"est une marque commerciale.
Les anaylses TLC n'ont pas été exécutées selon des conditions standardisées. Les valeurs Rf peuvent par conséquent changer, particulièrement à des températures différentes. Les points de fusion ont été déterminés dans des tubes capillaires ouverts à l'aide de l'appareil de Tottoli et n'ont pas été corrigés.
La plupart des dérivés se ramollissent et se décomposent avant la fusion. Les solvants pour la cristallisation, la précipitation ou le broyage sont indiqués entre crochets. L'électrophorèse sur papier à haute tension a été effectuée à l'aide d'un appareil Pherograph-Original- - Frankfurt Type 64 de Schleicher et SchUll sur papier No 2317 au pH 1,2 (acide formique : acide acétique : eau = 123 : 100 : 777) à 1600 V (40 V/cm) et au pH 5,8 (pyridine : : acide acétique : eau = 450 : 50 : 4500) à 1400 V (32,5 V/cm).
Les produits ont été caractérisés par leur mobilité au pH

La couche organique a été séchée sur du sulfate de sodium anhydre et le solvant a été éliminé sous vide.
On a obtenu 4,041 g (rendement 93%) du composé

Après élimination du solvant par évaporation sous vide, le produit brut a été transformé en acide libre correspondant de manière usuelle et purifié par chromatographie sur colonne, sur gel de silice (Merck) 0,040-0, 063 mm en éluant avec le mélange chloroforme : : méthanol = 9 : 1 en volume ; on a obtenu 3,252 g du composé V (rendement 90%) sous forme d'une mousse par évaporation à partir d'éther de pétrole : RfA 0, 67.
Opération 2. Boc-pro-Trp-Met-NH2 (VI)
En partant de 1,722 g (8 mmoles) de Boc-Pro-OH et de 2,967 g (8 mmoles) de HCl. H-Trp-Met-NH2 (II) et en opérant comme à l'opération 1 de l'exemple 1, on a obtenu 3,828 g (rendement 90%) du produit VI à partir d'alcool

Une solution de 2,340 g (5 mmoles de HC1. H- -Pro-Trp-Met-NH2 (VII) dans 20 ml de diméthylformamide a été refroidie à OOC et on a ajouté 0,56 ml de N-méthyl- - morpholine suivis de 1,812 g de Boc-Phe-Pro-OH (V), de 0,676 g (5 mmoles) de 1-hydroxy-benzotriazole et de 1,135 g (5,5 mmoles) de dicyclohexylcarbodiimide.
Le mélange en réaction a été agité à la température de la pièce pendant 3 heures puis soumis à filtration et à évaporation sous vide. Le produit brut a été purifié par chromatographie sur colonne, sur gel de silice (Merck) 0,040-0, 063 mm en

éluant avec le mélange acétate d'éthyle : méthanol : eau = 70 : 30 : 2 en volume. On a obtenu 1,940 g (rendement 50%) du composé (VIII) à partir du mélange acétate d'éthyle/ /éther diéthylique : RfA 0, 15 ; RfB 0, 60. 1 Opération 5. HCl. H-Phe-pro-pro-Trp-Met-NH2 (IX)
En partant de 1,552 g (2 mmoles) de Boc-Phe- -Pro-Pro-Trp-Met-NH2 (VIII) et en opérant comme à l'opération 2 de l'exemple 1, on a obtenu 1,282 g du produit brut.
Ce dernier a été purifié par chromatographie sur colonne, sur gel de silice (Merck) 0,040-0, 063 mm en éluant avec le mélange chloroforme : méthanol = 82 : 18 en volume. 0n a obtenu 0,705 g du composé IX (rendement général 50%) à

Example 4 Preparation of Pyr-Pro-Trp-Val-OH (XIII)
2.63 g (5 mmol) of Pyr-Pro-Trp-Val-OMe (XII), prepared according to Example 3, operation 3, were dissolved in 15 ml of methanol and saponified using 7 , 5 ml IN sodium hydroxide at room temperature. The reaction was complete in 4 hours. The solution was diluted with 40 ml of water and concentrated in vacuo to half the volume, diluted again with 40 ml of water, cooled to OOC, acidified with 5N hydrochloric acid at pH 2 and finally subjected to extraction using ethyl acetate. The organic layer was washed until neutral using saturated sodium chloride solutions, dried over anhydrous sodium sulfate and the solvent was removed in vacuo.
