L'invention concerne des dérivés d'ergoline, ainsi qu'un procédé pour leur préparation.
L'invention fournit des 7-cyano-ergolines répondant à la formule générale I :

L'invention fournit également un procédé pour la préparation de dérivés d'ergoline répondant à la formule générale l définie ci-dessus, ce procédé consistant à faire réagir une ergoline de formule générale (II) :

R < nure de sodium, le cyanure de triméthyl-silyle et le cyanure de tétrabutyl-ammonium. De préférence, on effectue la réaction dans un solvant tel que le chloroforme, le dichlorométhane, le diméthylformamide ou le diglyme.
En règle générale, la réaction a lieu à une température de -70oC à 0 C pendant une heure.
Les sels d'ergoline-immonium de formule II peuvent être obtenus par l'un ou l'autre des procédés illustrés par le schéma suivant :

On fait réagir des dérivés connus d'ergoline de formule III dans laquelle R, et R ont les signi-
R1fications indiquées ci-dessus, avec du peroxyde d'hydrogène aqueux pour obtenir les N6-oxydes correspondants qui, par traitement modéré avec une solution de chlorure de mésyle dans du dichlorométhane, donnent les sels d'im- monium II (méthode A, voir P. Potier et al.,"J. Am.
Chem. Soc.", 90, 5622, 1968).

En variante (méthode B), les ergolinelactames connus de formule IV dans laquelle R, et R2 ont les significations indiquées ci-dessus, sont réduits partiellement en utilisant un agent approprié tel que l'hydrure de diisobutyl-aluminium dans du tétra-
R1hydrofuranne à une température de -1000C à -700C. Les énamines cycliques initialement formées (voir R. Lukes et J. Kovar, "Collection Czech. Chem. Commun.", 19,

Les dérivés d'ergoline suivant l'invention sont utiles comme produits intermédiaires pour la pré-

Les exemples suivants illustrent l'invention.
Exemple 1 7-cyan-6, 8p-diméthylergoline Méthode A :
Tout en agitant pendant 6 heures, on a chauf-

fé à reflux une suspension de 1 g de 6, ergoline dans 30 ml de méthanol et 4, ml de peroxyde d'hydrogène aqueux à 36%. A la solution refroidie, on a ajouté lentement 100 mg de charbon palladié à 10% en poids afin de détruire le peroxyde en excès et l'on a poursuivi l'agitation pendant une heure.
Après filtra-
8ss-diméthyl-tion et évaporation du méthanol, on a dissous le résidu dans du dichlorométhane, on l'a séché sur du sulfate de sodium anhydre et on l'a évaporé jusqu'à siccité pour obtenir 1 g du N-oxyde de 6, 8ss-diméthyl-ergoline. On a dissous ce dernier dans 40 ml de dichlorométhane et on a effectué un traitement à -70oC avec 0, 91 ml de chlorure de mésyle puis, après 3 heures, avec 4,19 g de cyanure de tétrabutyl-ammonium. On a agité le mélange pendant une heure, puis on l'a traité avec une solution aqueuse saturée de carbonate de potassium.

On a lavé la couche organique avec du chlorure de sodium aqueux, on l'a séchée et on l'a évaporée jusqu'à siccité. On a soumis le résidu à une chromatographie dans une colonne de gel de silice en utilisant un mé- lange d'acétate d'éthyle et de cyclohexane (1 : 1 en volume) comme éluant pour obtenir 0, 3 g du composé sous rubrique après cristallisation dans l'éther diéthylique.

ergolin-7-one dans 25 ml de tétrahydrofuranne, à une température d'environ -700C, on a ajouté 3, 93 ml d'une solution d'hydrure de diisobutyl-aluminium 1M dans du toluène. On a poursuivi l'agitation à -70 C pendant 30 minutes, puis on a ajouté 2 ml d'une solution d'hydroxyde de sodium à 15%. On a évaporé le mélange aqueux obtenu jusqu'à siccité. On a filtré le résidu en suspension dans l'eau et l'acétate d'éthyle et on a soumis la couche organique (séchée sur du sulfate de sodium anhydre) à une distillation sous pression réduite jusqu'à siccité.
Au produit brut dissous dans 20 ml de dichlorométhane, à une température de -10 C, on a ajouté une quantité stoechiométrique d'acide trifluoracétique et 0, 8 g de cyanure de tétrabutyl-ammonium.

On a agité le mélange pendant 30 minutes, puis on l'a traité comme décrit dans le procédé A pour obtenir 0, 17 g du composé sous rubrique.
Exemple 2 7-cyano-68a-diméthylergoline
En travaillant comme décrit à l'exemple 1 (méthode A), mais en utilisant la 6, 8a-diméthylergoline

The invention relates to ergoline derivatives and a process for their preparation.
The invention provides 7-cyano-ergolines corresponding to the general formula I:

wherein R represents a hydrogen atom or a methyl group and (a) R represents a hydrogen atom or a methyl group and R represents a hydrogen atom or (b) R. and R together represent a methylene group.
The invention also provides a process for the preparation of ergoline derivatives corresponding to the general formula I defined above, this process consisting in reacting an ergoline of general formula (II):

The following examples illustrate the invention.
Example 1 7-cyan-6, 8p-dimethylergoline Method A:
While stirring for 6 hours, it was heated

at reflux a suspension of 1 g of 6, ergoline in 30 ml of methanol and 4, ml of 36% aqueous hydrogen peroxide. To the cooled solution was added slowly 100 mg of 10% by weight palladium on carbon to destroy the excess peroxide and stirring was continued for one hour.
After filtering
8ss-dimethyl-tion and evaporation of methanol, the residue was dissolved in dichloromethane, dried over anhydrous sodium sulfate and evaporated to dryness to obtain 1 g of N-oxide of 6 , 8ss-dimethyl-ergoline. The latter was dissolved in 40 ml of dichloromethane and a treatment was carried out at -70oC with 0.91 ml of mesyl chloride and then, after 3 hours, with 4.19 g of tetrabutylammonium cyanide. The mixture was stirred for one hour, then treated with a saturated aqueous solution of potassium carbonate.

The organic layer was washed with aqueous sodium chloride, dried and evaporated to dryness. The residue was subjected to chromatography in a column of silica gel using a mixture of ethyl acetate and cyclohexane (1: 1 by volume) as eluent to give 0.3 g of the title compound after crystallization from diethyl ether.

ergolin-7-one in 25 ml of tetrahydrofuran, at a temperature of about -700C, 3.93 ml of a solution of 1M diisobutyl aluminum hydride in toluene was added. Stirring was continued at -70 ° C for 30 minutes, then 2 ml of 15% sodium hydroxide solution was added. The aqueous mixture obtained was evaporated to dryness. The residue suspended in water and ethyl acetate was filtered and the organic layer (dried over anhydrous sodium sulfate) was subjected to distillation under reduced pressure to dryness.
To the crude product dissolved in 20 ml of dichloromethane, at a temperature of -10 ° C., a stoichiometric amount of trifluoroacetic acid and 0.8 g of tetrabutylammonium cyanide were added.
