t l'invention est de fournir une substance qui a une très bonne activité tumorale et qui peut être utilisée pour lutter contre le cancer.
Jusqu'ici, le 5-fluorouracile et divers de ses dérivés ont été connus comme agents antitumoraux. Mais aucun d'entre eux n'a fait preuve de l'efficacité attendue et il

sion lorsqu'ils sont utilisés. En outre, certains des dérivés du 5-fluorouracile sont très toxiques et peuvent avoir des effets secondaires.
L'invention vise donc des composés qui n'ont pas les inconvénients présentés par les dérivés du 5-fluorouracile classiques, ont une toxicité moindre, peuvent être dissous dans une substance huileuse à l'état stable, en sorte qu'ils peuvent être absorbes facilement dans le corps, et sont des agents utiles pour lutter contre le cancer.
L'invention vise donc des dérivés de N-phtalidyl-Sfluorouracile.

On peut donc ainsi préparer des dérivés de Nphtalidyl-5-f luorouracile. Tous ces composés ont une activité antitumorale et sont utiles pour lutter contre le cancer.
On donne ci-dessous les résultats de l'étude pharmacologique de dérivés de N-phtalidyl-5-fluorouracile suivant l'invention.
Activité antitumorale
(1) On implante du P-388 (1 x 106 cellules) dans

semaines. On administre oralement la substance d'essai en suspension ou dissoute dans une solution aqueuse à 0,3 %
de carboxyméthylcellulose, une fois par jour pendant 9 jours, en commençant un jour après l'implantation, et on évalue l'activité antitumorale par le rapport de jours de survie des souris traitées à ceux des souris-témoins. Chaque groupe.comprend cinq souris.
(2) On implante par voie sous-cutanée du Meth-A
(5 x 106 cellules) et du MH-134 (1 x 106 cellules) dans la paroi de la région abdominale de souris Balb/c et C3H/He respectivement. On administre oralement deux fois par jour pendant 20 jours, en commençant un jour après l'implantation, la substance à essayer en suspension ou en solution dans
une solution aqueuse à 0,3 % de carboxyméthylcellulose, et on détermine le poids de la tumeur 21 jours après l'implantation. On évalue l'activité antitumorale par le rapport pondé-

On a obtenu des résultats de toxicité semblables pour les autres composés suivant l'invention. Concentration dans le sang.

la dose de 1 mmole/kg. On recueille leur sang après des durées définies. Après avoir séparé le sérum du sang, on le fractionne en la substance inaltérée et en le 5-fluorouracile qui est le métabolite. Puis on mesure les concentrations de 5-fluorouracile par la méthode de la coupelle en couche mince, en utilisant Staphylococcus aureus 209P pour une quantité de 1/2 de gélose sensible pour l'essai (Eiken). On détermine les composés VI et VIII inaltérés par chromatographie en phase liquide à grande vitesse. On effectue l'essai en utilisant 3 à 4 souris pour chaque groupe.

Après avoir achevé l'addition goutte à goutte, on agite le mélange pendant 15 minutes, tout en refroidissant par de la glace et encore pendant 3 heures à température ambiante. Puis on ajoute 100 ml d'eau au mélange et on l'extrait au chloroforme. On lave la couche chloroformique
à l'eau et on la sèche sur du sulfate de sodium anhydre, puis on élimine le solvant par distillation. On lave le résidu deux fois par 50 ml d'hexane normal et on y ajoute
20 ml de chloroforme. On recueille les cristaux qui se déposent par filtration pour obtenir 0,26 gramme (10 %) de

Après avoir achevé l'addition au goutte à goutte, on agite le mélange à température ambiante pendant 4 heures. Au mélange de réaction obtenu on ajoute 50 ml d'eau et on extrait le mélange obtenu par du chloroforme. On lave la couche chloroformique à l'eau et on la sèche sur du sulfate de sodium anhydre, puis on enlève le solvant par distillation. On lave le résidu deux fois par 25 ml d'hexane normal et le soumet à une chromatographie sur une colonne de gel de silice. On concrétise une substance huileuse incolore obtenue de l'éluat chloroformique par trituration avec une petite quantité d'acétate d'éthyle.
Le rendement est de 1,22 gramme (74 %). On recristallise dans l'acétate d'éthyle pour obtenir des granulés incolores ayant un point de fusion de 154 à 157[deg.]C.

Dans 5 ml de diméthylformamide, on dissout 4,68 grammes de N,-benzoyl-5-fluorouracile et on ajoute à la solution agitée, tout en refroidissant par de la glace, 0,80 gramme d'hydrure de sodium (à 60 % de teneur). Puis on ajoute goutte à goutte au mélange une solution de 4,67 grammes de 3-bromophtalide dans 10 ml de diméthylformamide. Après avoir achevé l'addition au goutte à goutte, on agite le mélange tout en refroidissant par de la glace, pendant
15 minutes, puis pendant 1 heure à température ambiante. On verse le mélange réactionnel obtenu dans 200 ml d'eau et on extrait le mélange par du chloroforme. On lave la couche organique à l'eau et on la sèche sur du sulfate de sodium anhydre. On enlève ensuite le solvant par distillation et on lave le résidu par de l'éther de pétrole.
On recristallise les cristaux obtenus dans de l'acétate d'éthyle pour obtenir 3,63 grammes (50 %) d'une poudre incolore ayant un point de fusion de 207 à 210[deg.]C. On soumet à nouveau la liqueur-mère à une chromatographie sur colonne de gel de silice pour obtenir 0,67 gramme (9 %) de cristaux. Ainsi on a obtenu au total 4,30 grammes (59 %) de N3-

The results of the pharmacological study of N-phthalidyl-5-fluorouracil derivatives according to the invention are given below.
Antitumor activity
